    Electrostatic correlations on the ionic selectivity of cylindrical membrane nanopores

    We characterize the role of electrostatic fluctuations on the charge selectivity of cylindrical nanopores confining electrolyte mixtures. To this end, we develop an extended one-loop theory that can account for correlation effects induced by the surface charge, nanoconfinement of the electrolyte, and interfacial polarization charges associated with the low permittivity membrane. We validate the quantitative accuracy of the theory by comparisons with previously obtained Monte-Carlo simulation data from the literature, and scrutinize in detail the underlying forces driving the ionic selectivity of the nanopore. In the biologically relevant case of electrolytes with divalent cations such as CaCl2 in negatively charged nanopores, electrostatic correlations associated with the dense counterion layer in the channel result in an increase of the pore coion density with the surface charge. This peculiarity analogous to the charge inversion phenomenon remains intact for dielectrically inhomogeneous pores, which indicates that the effect should be observable in nanofiltration membranes or DNA-blocked nanopores characterized by a low membrane permittivity. Our results show that a quantitatively accurate consideration of correlation effects is necessary to determine the ionic selectivity of nanopores in the presence of electrolytes with multivalent counterions.Peer reviewe

Nous examinons dans cet article les éléments empiriques et théoriques qui expliquent la hausse des prix récente et sa résistance aux retournements conjoncturels. En combinant la notion de bulle économique, les arguments de l’économie spatiale et une analyse d’économie politique, nous suggérons que la valorisation importante de l’immobilier en France est le résultat d’une logique rationnelle et conforte les intérêts des acteurs locaux. Dès lors, la forte valorisation peut être considérée comme une « bulle robuste », à même de résister à des chocs importants. Cette bulle organise un transfert intergénérationnel et peut avoir des effets positifs. Elle peut également renforcer la ségrégation spatiale, alimenter les inégalités territoriales et empêcher d’exploiter les économies d’agglomération possibles.     Innovative methods in European road freight transport statistics: A pilot study

    By using innovative methods, such as the automated transfer of corporate electronic data to National Statistical Institutions, official transport data can be significantly improved in terms of reliability, costs and the burden on respondents. In this paper, we show that the automated compilation of statistical reports is possible and feasible. Based on previous findings, a new method and tool were developed in cooperation with two business partners from the logistics sector in Austria. The results show that the prototype could successfully be implemented at the partner companies. Improved data quality can lead to more reliable analyses in various fields. Compared to actual volumes of investments into transport, the costs of transport statistics are limited. By using the new and innovative data collection techniques, these costs can even be reduced in the long run; at the same time, the risk of bad investments and wrong decisions caused by analyses relying on poor data quality can be reduced. This results in a substantial value for business, research, the economy and the society

    Hydrogen Phases on the Surface of a Strongly Magnetized Neutron Star

    The outermost layers of some neutron stars are likely to be dominated by hydrogen, as a result of fast gravitational settling of heavier elements. These layers directly mediate thermal radiation from the stars, and determine the characteristics of X-ray/EUV spectra. For a neutron star with surface temperature T\lo 10^6 K and magnetic field B\go 10^{12} G, various forms of hydrogen can be present in the envelope, including atom, poly-molecules, and condensed metal. We study the physical properties of different hydrogen phases on the surface of a strongly magnetized neutron star for a wide range of field strength BB and surface temperature TT. Depending on the values of BB and TT, the outer envelope can be either in a nondegenerate gaseous phase or in a degenerate metallic phase. For T\go 10^5 K and moderately strong magnetic field, B\lo 10^{13} G, the envelope is nondegenerate and the surface material gradually transforms into a degenerate Coulomb plasma as density increases. For higher field strength, B>>1013B>> 10^{13} G, there exists a first-order phase transition from the nondegenerate gaseous phase to the condensed metallic phase. The column density of saturated vapor above the metallic hydrogen decreases rapidly as the magnetic field increases or/and temperature decreases. Thus the thermal radiation can directly emerge from the degenerate metallic hydrogen surface. The characteristics of surface X-ray/EUV emission for different phases are discussed.     Expectations and emotions concerning infant transitions to ECEC : international dialogues with parents and teachers

    Although a large body of literature recognises the impact of parent-teacher relationships on infant everyday experiences, less is known about the emotional experience and associated expectations of the adults themselves during earliest transitions. In the context of a multi-site international investigation across five countries–Brazil, Finland, Scotland, New Zealand, and the United States–the present paper examines teacher and parent interviews to reveal expectations prior to and after the transition to ECEC, highlighting the associated emotions that arise during this process. Irrespective of whether expectations are met, parents universally express insecurities and fears in relation to the transition. Parents are aware of the impact that the various aspects of the ECEC setting have on their child, and acknowledge difficulty in relinquishing control of the care of their child. Despite these concerns, parents consistently articulate their strong trust in the institution and the professional expertise of the staff. Correspondingly, teachers are keenly aware of the importance of their role in supporting families, and hold certain expectations for how the transition experience will play out accordingly. Regardless of country of context, the study shows that clear communication surrounding both centre and parental expectations establishes high levels of trust and ameliorates anxiety.     Rapid detection of 2-hydroxyglutarate in frozen sections of IDH mutant tumors by MALDI-TOF mass spectrometry

    All isocitrate dehydrogenase (IDH) mutant solid neoplasms exhibit highly elevated levels of D-2-hydroxyglutarate (D-2HG). Detection of 2HG in tumor tissues currently is performed by gas or liquid chromatography-mass spectrometry (GC- or LC-MS) or biochemical detection. While these methods are highly accurate, a considerable amount of time for tissue preparation and a relatively high amount of tissue is required for testing. We here present a rapid approach to detect 2HG in brain tumor tissue based on matrix-assisted laser desorption ionization - time of flight mass spectrometry (MALDI-TOF). We analyzed 26 brain tumor samples with known IDH1 or IDH2 mutation and compared readouts to those from 28 brain tumor samples of wildtype IDH status. IDH mutant samples exhibited a clear positive signal for 2HG which was not observed in any of the IDH wildtype tumors. Our analytical pipeline allowed for 2HG detection in less than 5 min. Data were validated by determining 2HG levels in all tissues with a biochemical assay. In conclusion, we developed a protocol for rapid detection of 2HG levels and illustrate the possibility to use MALDI-TOF for the detection of metabolites on frozen tissue sections in a diagnostic setting

    Biological and technical variables affecting immunoassay recovery of cytokines from human serum and simulated vaginal fluid: A multicenter study

    The increase of proinflammatory cytokines in vaginal secretions may serve as a surrogate marker of unwanted inflammatory reaction to microbicide products topically applied for the prevention of sexually transmitted diseases, including HIV-1. Interleukin (IL)-1β and IL-6 have been proposed as indicators of inflammation and increased risk of HIV-1 transmission; however, the lack of information regarding detection platforms optimal for vaginal fluids and interlaboratory variation limit their use for microbicide evaluation and other clinical applications. This study examines fluid matrix variants relevant to vaginal sampling techniques and proposes a model for interlaboratory comparisons across current cytokine detection technologies. IL-1β and IL-6 standards were measured by 12 laboratories in four countries, using 14 immunoassays and four detection platforms based on absorbance, chemiluminescence, electrochemiluminescence, and fluorescence. International reference preparations of cytokines with defined biological activity were spiked into (1) a defined medium simulating the composition of human vaginal fluid at pH 4.5 and 7.2, (2) physiologic salt solutions (phosphate-buffered saline and saline) commonly used for vaginal lavage sampling in clinical studies of cytokines, and (3) human blood serum. Assays were assessed for reproducibility, linearity, accuracy, and significantly detectable fold difference in cytokine level. Factors with significant impact on cytokine recovery were determined by Kruskal−Wallis analysis of variance with Dunn’s multiple comparison test and multiple regression models. All assays showed acceptable intra-assay reproducibility; however, most were associated with significant interlaboratory variation. The smallest reliably detectable cytokine differences (P < 0.05) derived from pooled interlaboratory data varied from 1.5- to 26-fold depending on assay, cytokine, and matrix type. IL-6 but not IL-1β determinations were lower in both saline and phosphate-buffered saline as compared to vaginal fluid matrix, with no significant effect of pH. The (electro)chemiluminescence-based assays were most discriminative and consistently detected <2-fold differences within each matrix type. The Luminex-based assays were less discriminative with lower reproducibility between laboratories. These results suggest the need for uniform vaginal sampling techniques and a better understanding of immunoassay platform differences and cross-validation before the biological significance of cytokine variations can be validated in clinical trials. This investigation provides the first standardized analytic approach for assessing differences in mucosal cytokine levels and may improve strategies for monitoring immune responses at the vaginal mucosal interface

    TREX exposes the RNA-binding domain of Nxf1 to enable mRNA export

    The metazoan TREX complex is recruited to mRNA during nuclear RNA processing and functions in exporting mRNA to the cytoplasm. Nxf1 is an mRNA export receptor, which binds processed mRNA and transports it through the nuclear pore complex. At present, the relationship between TREX and Nxf1 is not understood. Here we show that Nxf1 uses an intramolecular interaction to inhibit its own RNA-binding activity. When the TREX subunits Aly and Thoc5 make contact with Nxf1, Nxf1 is driven into an open conformation, exposing its RNA-binding domain, allowing RNA binding. Moreover, the combined knockdown of Aly and Thoc5 markedly reduces the amount of Nxf1 bound to mRNA in vivo and also causes a severe mRNA export block. Together, our data indicate that TREX provides a license for mRNA export by driving Nxf1 into a conformation capable of binding mRNA
