    Efikasi Kombinasi Herbisida Penoxsulam Dan Butachlor Terhadap Gulma Pada Budidaya Padi Sawah (Oryza Sativa L.) Tanam Pindah

    Padi merupakan komoditas pangan utama di Indonesia. Penurunan produksi pangan khususnya padi akibat gulma berkisar antara 60 hingga 87%. Gulma adalah tumbuhan pengganggu yang merugikan kepentingan manusia sehingga manusia berusaha untuk mengendalikannya. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi herbisida penoxsulam+butachlor terhadap gulma, pertumbuhan dan produksi tanaman padi sawah. Penelitian ini dilaksanakan di Trimurjo, Lampung Tengah dan di Laboratorium Ilmu Gulma Fakultas Pertanian Universitas Lampung. Penelitian disusun dalam Rancangan Acak Kelompok (RAK) dengan 4 ulangan. Perlakuan terdiri atas dosis kombinasi herbisida penoxsulam+butachlor (7,5+300), (10,0+400), (12,5+500), (15,0+600) g/ha, penyiangan manual dan kontrol. Pengujian homogenitas dilakukan dengan uji Bartlett dan additifitas data dengan uji Tukey. Jika asumsi terpenuhi data akan dianalisis dengan sidik ragam. Apabila perlakuan menunjukkan perbedaan nyata maka dilakukan uji beda nilai tengah antar perlakuan dengan uji Beda Nyata Terkecil (BNT) pada taraf kepercayaan 5 %. Hasil penelitian menunjukkan bahwa kombinasi herbisida penoxsulam+butachlor pada dosis 7,5+300 sampai dengan 15,0+600 g/ha mampu mengendalikan pertumbuhan gulma total, gulma dominan Ludwigia hyssopifolia, Monochoria vaginalis, Fimbristylis miliacea, Cyperus iria sampai dengan 6 minggu setelah aplikasi (MSA), serta gulmaLeptochloa chinensissampai 3 MSA.Kombinasi herbisida penoxsulam+butachlor pada dosis7,5+300 sampai dengan 15,0+600g/ha tidak meracuni tanaman padi (Oryza sativa L.). Kombinasi herbisida penoxsulam+butachlor pada dosis 7,5+300 sampai dengan 15,0+600 g/ha tidak mempengaruhi pertumbuhan dan produksi tanaman padi sawah, tetapi keberadaan gulma menurunkan hasil Gabah Kering Giling (GKG) sehingga perlu dilakukan pengendalian

    Genetic and enzymatic characterisation of wood degrading strains of Phanerochaete species

    Thesis (PhD)--University of Stellenbosch, 2000.ENGLISH ABSTRACT: White rot fungi are of interest in the paper and pulp industry because of their removal of lignin from wood. In this study over 600 Basidiomycete fungi were isolated from indigenous forests as well as from commercial Eucalyptus spp. and Pinus spp. plantations in South Africa. One hundred isolates were identified to genus level. Biochemical tests were done to screen the fungal cultures for characteristics that are favourable for biopulping, e.g. low cellulase activity with concomitant high activity of ligninolytic enzymes. Various Phanerochaete isolates with potentially high ligninolytic activity were identified. Although Phanerochaete chrysosporium Burds. has previously been isolated from the indigenous forest at Knysna in South Africa, this study showed that P. chrysosporium was a natural coloniser of wood chip piles in South Africa, indicating potential for application in industry. A possible new species of Phanerochaete, viz. Phanerochaete pseudomagnoliae nom. provo (strain PP25) from decayed wood collected in Stellenbosch, South Africa, was described and illustrated. It differs from previously described Phanerochaete species in having smaller basidiospares, and in the formation of few chlamydospores on malt extract agar but more on xylose containing media. The potential of using internal transcribed spacer DNA sequences (ITS) to infer phylogenetic relationships among species of the genus Phanerochaete was investigated. Consensus phylogenetic trees could be presented, but the presence of ambiguous aligning sequences within the ITS made inferring of phylogenetic relationships within the whole genus difficult. Fifty-five South African strains of P. chrysosporium were screened for lignin peroxidase (liP), manganese peroxidase (MnP) and glyoxal oxidase (GLOX) enzymes. Manganese peroxidase activity was quantified on agar media. The liP and GLOX activities of 13 selected strains, including control strains and P. pseudomagnoliae (PP25), were also quantified on agar media. Differences in MnP and GLOX activities existed among the strains. Preliminary biochemical characterisation of strain PP25 indicated that the most important difference was the apparent unique regulation of ligninolytic enzymes. Under low nitrogen, liP activity of the selected strains showed no significant variation, whereas strain PP25 had significantly increased liP levels under high nitrogen conditions. Restriction fragment length polymorph isms of the lignin and manganese peroxidase gene DNA fragments showed variability among strains, whereas there was probably only allelic variation for the glox gene DNA fragments. Previous research has indicated xylose oxidation activity within P. chrysosporium. To investigate whether GLOX can oxidise xylose, a purified recombinant GLOX (rGLOX) from P. chrysosporium BKM-F-1767 Burds. was used in this study. This rGLOX oxidised D-xylose and D-glucose (D-xylose > D-glucose) to produce H202. Xylose was oxidised to xylono-1 ,4-lactone with a 1:1 stoichiometric relationship between H202 produced and xylose used. Xylono-1,4-lactone was converted non-enzymatically to xylonate. This suggested that the furanose form of xylose, rather than the pyranose form, is a substrate of GLOX. The production of H202 and the removal of inhibitory compounds by GLOX could enhance ligninolytic activity. . To conclude, unique strains of P. chrysosporium have been isolated from South Africa with potential biotechnological use in paper manufacturing. The relationship of P. pseudomagnoliae nom. provo to other Phanerochaete species was evaluated and light was shed on the possible role of GLOX in lignin degradation.AFRIKAANSE OPSOMMING: Witvrot swamme is van belang vir die papier en pulp industrie omdat hulle lignin vanaf hout kan verwyder. Meer as 600 Sasidiomiseet fungi, afkomstig vanaf inheemse woude asook kommersiële Eucalyptus spp. en Pinus spp. plantasies, IS geïsoleer. Een honderd isolate is tot op genusvlak geïdentifiseer. Die isolate is biochemies vir eienskappe wat voordelig vir "bioverpulping" kon wees, bv. die gelyktydige produksie van lae sellulosemaar hoë ligninolitiese ensiemaktiwiteit, getoets. Verskeie isolate met potensieel hoë vlakke van ligninolitiese aktiwiteit is verkry. Alhoewel Phanerochaete chrysosporium Surds. vantevore in die Knysna inheemse woud in Suid-Afrika geïsoleer is, het hierdie studie gewys dat P. chrysosporium natuurlik op hope houtblokkies voorgekom, met moontlike toepasing in die industrie. Isolaat PP25, geisoleer vannaf verrottende hout te Stellenbosch, Suid Afrika, is as 'n potensieel nuwe spesie van die genus Phanerochaete beskryf en as Phanerochaete pseudomagnoliae nom. provo benoem. Hierdie isolaat verskil van ander Phanerochaete-spesies daarin dat dit kleiner basidiospore vorm en nie klamydospore op moutekstrakagar produseer nie, maar wel op media wat xilose bevat. Die potensiaal van intern getranskribeerde spasieerder ONS opeenvolging vir die aflei van filogenetiese verhoudings tussen spesies van die genus Phanerochaete is ondersoek. Konsensus filogenetiese bome kon bepaal word, maar die teenwoordigheid van varieerbare areas het die afleiding van filogenetiese verwantskappe vir die hele genus bemoeilik. Vyf-en-vyftig Suid-Afrikaanse isolate van P. chrysosporium is vir die teenwoordigheid van lignienperoksidase- (liP), mangaanperoksidase- (MnP) en glioksaaloksidase (GLOX)-aktiwiteit getoets. Vlakke van MnP-aktiwiteit is op agarplate gekwantifiseer. Vlakke van LiP- en GLOX-ensieme op agarplate is vir 13 geselekteerde isolate, insluitend kontroles en ras PP25, gekwantifiseer. Aktiwiteit van MnP en GLOX het statisties betekenisvol tussen isolate verskil. Lignienperoksidase-aktiwiteit onder lae stikstof toestande het nie statisties betekenisvol van mekaar verskil nie. Onder hoë stikstof toestande het isolaat PP25 wel verhoogde liP-aktiwiteit getoon. Restriksie fragment polimorfismes van die lignien- en mangaanperoxidase-gene het variasie getoon, terwyl waarskynlik slegs alleliese variasie vir die glox geen waargeneem IS. Rekombinante GLOX (GLOX vanaf P. chrysosporium BKM-F-1767) het xilose en glukose (D-xilose > D-glukose) geoksideer met meegaande produksie van H202. Xilose is na xilono-1,4-laktoon geoksideer met 'n 1:1 stoigiometrie tussen H202-produksie en xilose verbruik. Xilono-1,4-laktoon is nieensiematies na xilonaat omgeskakel. Bogenoemde resultaat dui aan dat die furanose vorm van xilose die werklike substraat vir GLOX is. Deur die meegaande produksie van H202 en die verwydering van inhiberende produkte word lignoliese aangehelp.     Coordinated Fc-effector and neutralization functions in HIV-infected children define a window of opportunity for HIV vaccination

    OBJECTIVES: Antibody function has been extensively studied in HIV-infected adults but is relatively understudied in children. Emerging data suggests enhanced development of broadly neutralizing antibodies (bNAbs) in children but Fc effector functions in this group are less well defined. Here, we profiled overall antibody function in HIV-infected children. DESIGN: Plasma samples from a cross-sectional study of 50 antiretroviral therapy-naive children (aged 1-11 years) vertically infected with HIV-1 clade A were screened for HIV-specific binding antibody levels and neutralizing and Fc-mediated functions. METHODS: Neutralization breadth was determined against a globally representative panel of 12 viruses. HIV-specific antibody levels were determined using a multiplex assay. Fc-mediated antibody functions measured were antibody-dependent: cellular phagocytosis (ADCP); neutrophil phagocytosis (ADNP); complement deposition (ADCD) and natural killer function (ADNK). RESULTS: All children had HIV gp120-specific antibodies, largely of the IgG1 subtype. Fifty-four percent of the children exhibited more than 50% neutralization breadth, with older children showing significantly broader neutralization activity. Apart from ADCC, observed only in 16% children, other Fc-mediated functions were common (>58% children). Neutralization breadth correlated with Fc-mediated functions suggesting shared determinants of enhanced antibody function exist. CONCLUSIONS: These results are consistent with previous observations that children may develop high levels of neutralization breadth. Furthermore, the striking association between neutralization breadth and Fc effector function suggests that HIV vaccination in children could yield multifunctional antibodies. Paediatric populations may therefore provide an ideal window of opportunity for HIV vaccination strategies

    Cost-effectiveness of endovascular revascularization compared to supervised hospital-based exercise training in patients with intermittent claudication: A randomized controlled trial

    BackgroundThe optimal first-line treatment for intermittent claudication is currently unclear.ObjectiveTo compare the cost-effectiveness of endovascular revascularization vs supervised hospital-based exercise in patients with intermittent claudication during a 12-month follow-up period.DesignRandomized controlled trial with patient recruitment between September 2002-September 2006 and a 12-month follow-up per patient.SettingA large community hospital.ParticipantsPatients with symptoms of intermittent claudication due to an iliac or femoro-popliteal arterial lesion (293) who fulfilled the inclusion criteria (151) were recruited. Excluded were, for example, patients with lesions unsuitable for revascularization (iliac or femoropopliteal TASC-type D and some TASC type-B/C.InterventionParticipants were randomly assigned to endovascular revascularization (76 patients) or supervised hospital-based exercise (75 patients).MeasurementsMean improvement of health-related quality-of-life and functional capacity over a 12-month period, cumulative 12-month costs, and incremental costs per quality-adjusted life year (QALY) were assessed from the societal perspective.ResultsIn the endovascular revascularization group, 73% (55 patients) had iliac disease vs 27% (20 patients) femoral disease. Stents were used in 46/71 iliac lesions (34 patients) and in 20/40 femoral lesions (16 patients). In the supervised hospital-based exercise group, 68% (51 patients) had iliac disease vs 32% (24 patients) with femoral disease. There was a non-significant difference in the adjusted 6- and 12-month EuroQol, rating scale, and SF36-physical functioning values between the treatment groups. The gain in total mean QALYs accumulated during 12 months, adjusted for baseline values, was not statistically different between the groups (mean difference revascularization versus exercise 0.01; 99% CI −0.05, 0.07; P = .73). The total mean cumulative costs per patient was significantly higher in the revascularization group (mean difference €2318; 99% CI €2130, € 2506; P < .001) and the incremental cost per QALY was 231 800 €/QALY adjusted for the baseline variables. One-way sensitivity analysis demonstrated improved effectiveness after revascularization (mean difference 0.03; CI 0.02, 0.05; P < .001), making the incremental costs 75 208 €/QALY.ConclusionIn conclusion, there was no significant difference in effectiveness between endovascular revascularization compared to supervised hospital-based exercise during 12-months follow-up, any gains with endovascular revascularization found were non-significant, and endovascular revascularization costs more than the generally accepted threshold willingness-to-pay value, which favors exercise

    Effect of position feedback during task-oriented upper-limb training after stroke: Five-case pilot study

    Feedback is an important element in motor learning during rehabilitation therapy following stroke. The objective of this pilot study was to better understand the effect of position feedback during task-oriented reach training of the upper limb in people with chronic stroke. Five subjects participated in the training for 30 minutes three times a week for 6 weeks. During training, subjects performed reaching movements over a predefined path. When deviation from this path occurred, shoulder and elbow joints received position feedback using restraining forces. We recorded the amount of position feedback used by each subject. During pre- and posttraining assessments, we collected data from clinical scales, isometric strength, and workspace of the arm. All subjects showed improvement on one or several kinematic variables during a circular motion task after training. One subject showed improvement on all clinical scales. Subjects required position feedback between 7.4% and 14.7% of training time. Although augmented feedback use was limited, kinematic outcome measures and movement performance during training increased in all subjects, which was comparable with other studies. Emphasis on movement errors at the moment they occur may possibly stimulate motor learning when movement tasks with sufficiently high levels of difficulty are applied

    Migraine-associated vertigo and dizziness as presenting complaint in a private general medical practice

    Migraine-associated vertigo (MV) remains a developing entity because accepted diagnostic criteria are unavailable. Patients present with debilitating dizziness without experiencing headache, and are often misdiagnosed as anxious. The condition is manageable in primary care without the need for neurological referral. The aim of this study was to investigate the prevalence of MV and migraine-associated dizziness (MD) as presenting complaints. Methods: Patients presented with dizziness probably or definitely associated with migraine history based on the criteria of the International Headache Society. Patients with other vestibulopathies and medical conditions were excluded. Patients were evaluated over a period of nine months. Seven hundred and seventeen patients were examined. The numbers of patients were recorded as a percentage of the population visiting a general practitioner. Response to migraine prophylactic medications was regarded as supporting evidence of the diagnosis. Response was regarded as a complete resolution of symptoms. Results: Of the 717 patients seen, 12 were identified as having probable or definite MV. Five patients were treated with migraine prophylactic medications, namely amitriptyline 25 mg nocte and/or sodium valproate CR 300 mg bd, and all showed a response to the treatment. Conclusions: We conclude that the prevalence of MV as presenting complaint may be as high as 1.67%. This figure does however not reflect the total patient population that suffers from the condition – this figure may be much higher. Of those patients treated for MV the response was 100%, further supporting the diagnosis. MV is a relevant complaint that is often misdiagnosed as psychogenic in origin

    Moderate-to-deep sedation technique, using propofol and ketamine, allowing synchronised breathing for magnetic resonance high-intensity focused ultrasound (MR-HIFU) treatment for uterine fibroids: a pilot study

    BACKGROUND: Magnetic resonance high-intensity focused ultrasound (MR-HIFU) treatment for uterine fibroids is rapidly gaining popularity as a treatment modality. This procedure is generally uncomfortable, painful, and requires minimal or absence of movement and an MR-HIFU synchronised breathing pattern of the patient. Procedural sedation and analgesia protocols have become the standard practice in interventional radiology departments worldwide. The aim of this study was to explore if a sedation regimen with low-dose propofol and ketamine performed by trained non-medical sedation practitioners could result in relief of discomfort for the patient and in adequate working conditions for MR-HIFU treatment for uterine fibroids. METHODS: In this study, conducted from August 2013 until November 2014, 20 patients were subjected to MR-HIFU treatment of uterine fibroids. Patients were deeply sedated using intravenous propofol and esketamine according to a standardised hospital protocol to allow synchronisation of the breathing pattern to the MR-HIFU. The quality of sedation for MR-HIFU and complications were recorded and analysed. The side effects of the sedation technique, the propofol and esketamine consumption rate, the duration of recovery, and patient satisfaction after 24 h were examined. RESULTS: A total of 20 female patients (mean age 42.4 [range 32-53] years) were enrolled. Mean propofol/esketamine dose was 1309 mg/39.5 mg (range 692-1970 mg/ 23.6-87.9 mg). Mean procedure time was 269 min (range 140-295 min). Application of the sedation protocol resulted in a regular breathing pattern, which could be synchronised with the MR-HIFU procedures without delay. The required treatment was completed in all cases. There were no major adverse events. Hypoxemia (oxygen desaturation <92%) and hallucinations were not observed. CONCLUSIONS: The use of a specific combination of IV propofol and esketamine for procedural sedation and analgesia reduced the discomfort and pain during MR-guided HIFU treatments of uterine fibroids. The resulting regular breathing pattern allowed for easy synchronisation of the MR-HIFU procedure. Based on our results, esketamine and propofol sedation performed by trained non-medical sedation practitioners is feasible and safe, has a low risk of major adverse events, and has a short recovery time, avoiding a session of general anaesthesia

    We investigate Body Area Networks for ambulant patient monitoring. As well as sensing physiological parameters, BAN applications may provide feedback to patients. Automating formulation of feedback requires realtime analysis and interpretation of streaming biosignals and other context and knowledge sources. We illustrate with two prototype applications: the first is designed to detect epileptic seizures and support appropriate intervention. The second is a decision support application aiding weight management; the goal is to promote health and prevent chronic illnesses associated with overweight/obesity. We begin to explore extending these and other m-health applications with generic AI-based decision support and machine learning. Monitoring success of different behavioural change strategies could provide a basis for machine learning, enabling adaptive clinical decision support by personalising and adapting strategies to individuals and their changing needs. Data mining applied to BAN data aggregated from large numbers of patients opens up possibilities for discovery of new clinical knowledge
