    Anti-Proliferative Activity of Nigella sativa Chloroform Extract on 7,12-Dimethylbenz[a]anthracene Induced Female Rats Splenocyte

    Previous study reported that Nigella sativa has in vitro and in vivo cancer activity.This study was conducted to observe the effect of chloroform extract of Nigella sativa seed (NCE) on 7,12-Dimethylbenz[a]anthracene (DMBA)-induced female rats’ splenocyte. The experiment consisted of five groups, corn oil solvent control group, DMBA group, DMBA+250 mg/kgBW NCE, DMBA+500 mg/kgBW NCE and DMBA+750 mg/kgBW NCE. Extract was dissolved in corn oil and oral administered daily for 2 weeks before and during the DMBA induction. Observation of cell proliferation was performed using haematoxylin and eosin (H&E) and AgNOR stainings. H&E staining showed decreased necrocis activity extract groups compared to DMBA group. From AgNOR staining results, mean AgNOR (mAgNOR) of extract groups was less in number compared to DMBA group. The mAgNOR in corn oil solvent control group, DMBA group, DMBA+250 mg/kgBW NCE, DMBA+500 mg/kgBW NCE and DMBA+750 mg/kgBW NCE were 1.22, 1.91, 1.29, 1.36 and 1.33, respectively. Our current results showed that NCE reduces the proliferation of DMBA-induced rat spleenocytes  Thus, NCE has potency to be developed as a chemopreventive agent.Keywords: Nigella sativa, spleen, DMBA, anti-proliferativ

    ANTICANCER ACTIVITYOF CALANONE ON HeLa CELL LINE

    ABSTRACT Calanone (coumarin derivate compound), isolated from Calophyl/um sp. had been shown to have cytotoxic activityon leukemia L1210 eel/line with IC50 =59.40 pg/mL. Calanone presumed have anticancer activity on HeLa cervical carcinoma cel/. This study was conducted to investigate the cytotoxic and apoptotic activity of calanone and its effect to p53 and p21 expression on HeLa cervical carcinqma cel/. Cytotoxic assay of calanone was performed on HeLa eel/line, using MTT assay. Apoptotic assay was performed on HeLa eel/line incubated with calanone for 24 h, by immunofluororescence method, using fIuorochromes ethidium bromide and acridine orange. Expression of p53 was examined on HeLa eel/line, by PCR with p53 wild-type primer. Expression of p21 was examined on HeLa eel/ line, by immunohistochemistry method. 5-fIuorourasil was used as positive control in cytotoxic, apoptotic assay, and p53 expression. The result showed that calanone has cytotoxic activity on HeLa eel/line, with ICw = 22.887 pg/mL, caused cytotoxicity through apoptotic mechanism, increase p53 tumor suppressor gene expression, while the p21 expression test showed a negative result.     Antiproliferative Activity of Black Seed (Nigella Sativa) on 7,12-dimethylbenz-[a]antracene (DMBA) Induced Mice Lung Cell

    Lung cancer is the leading cause of death in the world. Nigella sativa is a plant that has an anticancer activity. Previous research showed that the chloroform extract of N. sativahave cytotoxic effect on T47D cells. This study aimed to observe the effect of cloroform extract of N. sativaseed (NSS) on mice lung cell after initiation of 7,12-dimethylbenz [a] anthracene. Sprague Dawleystrain female rats were divided intofive groups. Each group consisted of 12 rats. The experiment consisted of five mice groups, corn oil as a solvent control group, the DMBA dose 20 mg/kgBW p.o. twice a week during five weeks, DMBA+NSS dose 250 mg/kgBW, DMBA+NSS dose 500 mg/kgBW, and DMBA+NSS dose 750 mg/kgBW. Extract which was dissolved into corn oil was administered daily by the oral route 1 week before and during the DMBA induction. At the end of the study, the experimental mice were sacrified and colon organs were collected and then stained with Haematoxylin and Eosin (H&E) and AgNOR method. H&E staining showed there was a decrease damage inthe treatment group compare with DMBA group. In AgNOR staining result showed mAgNOR value in DMBA group was 1.47±0.558, in extract group (250, 500 and 750mg/kgBB) was 0.44±0.172, 1.38±0.140 and 1.25±0.164 respectively and in corn oil group was 0.65±0.050. The results showed that N. sativareduced the damage of colon cells and inhibit colon cell proliferation on mice induced DMBA. This study indicated that N. sativacan be developed into a chemopreventive agent for lung cancer

    PENINGKATAN LITERASI KEAMANAN OBAT PADA SWAMEDIKASI BAYI DAN BALITA DI DESA SUDIMARA CILONGOK

    Masalah keamanan obat pada swamedikasi menjadi perhatian penting bagi banyak orang terutama apabila diberikan kepada anak anak. Penggunaan obat pada anak terutama bayi dan balita tanpa pengetahuan dan informasi memadai dapat menyebabkan masalah kesehatan baru. Maka diperlukan pemberdayaan masyarakat dalam meningkatkan kualitas kesehatan masyarakat dan mendukung program gerakan masyarakat cerdas menggunakan obat. Edukasi dilakukan pada masyarakat ibu ibu peserta posyandu terkait dengan swamedikasi pada bayi dan balita. Kegiatan pengabdian kepada masyarakat yang dilakukan di desa sudimara kecamatan cilongok kabupaten banyumas mendapat antusias dari para peserta. Hal ini ditunjukkan dengan partisipasi aktif peserta ketika dilakukan diskusi. Berdasarkan kegiatan ini tingkat pengetahuan peserta kegiatan edukasi kesehatan tentang swamedikasi meningkat. Kegiatan ini diharapkan dapat dilaksanakan secara berkelanjutan dengan harapan peningkatan kesadaran dan pengetahuan masyarakat di Desa Sudimara kecamatan cilongok kabupaten banyumas mengenai penggunaan obat yang aman pada bayi dan balita.Masalah keamanan obat pada swamedikasi menjadi perhatian penting bagi banyak orang terutama apabila diberikan kepada anak anak.     FAKTOR-FAKTOR YANG BERHUBUNGAN DENGAN KEHADIRAN IBU BAYI DAN IBU BALITA DI POSYANDU TANJUNG SEJAHTERA SEMARANG UTARA TAHUN 2008

    Wilayah kerja di Posyandu Tanjung Sejahtera terdapat masalah pada menurunnya tingkat kehadiran ibu bayi dan balita di Posyandu tersebut. Berdasarkan hasil survey petugas kesehatan Puskesmas Bandarharjo terhadap Posyandu Tanjung Sejahtera pada tahun 2007, didapatkan data yang menunjukkan kunjungan ibu bayi dan balita pada bulan Oktober 65%, November 45% dan desember 40%. dari populasi ibu bayi dan ibu balita yang berjumlah 75 orang. Menurunnya tingkat kehadiran ibu bayi dan balita ke Posyandu Tanjung Sejahtera dikarenakan masyarakat lebih memilih berkunjung ke Puskesmas Bandarharjo daripada melakukan kunjungan ke Posyandu Tanjung Sejahtera. Tujuan penelitian ini adalah mengetahui faktor - faktor yang berhubungan dengan kehadiran ibu bayi dan ibu balita di Posyandu Tanjung Sejahtera Kecamatan Semarang Utara. Jenis penelitian ini explanatory research. Metode penelitian yang digunakan adalah survey yaitu penelitian yang mengambil sample dari satu populasi dan menggunakan kuisioner sebagai alat pengumpulan data, dengan pendekatan cross sectional dimana pengamatan variabelnya diakukan secara bersama-sama Ada hubungan antara pengetahuan ibu bayi dan ibu balita tentang Posyandu dengan kehadiran ibu bayi dan ibu balita di Posyandu Tanjung Sejahtera, dengan rho = 0,475, p value = 0,001, tingkat signifikansi = 0,05, p = 0,001 < 0,005 (Ho ditolak = ada hubungan). Ada hubungan antara sikap ibu bayi dan ibu balita tentang Posyandu dengan kehadiran ibu bayi dan ibu balita di Posyandu Tanjung Sejahtera, dengan rho = 0,410, p value = 0,006, tingkat signifikansi = 0,05, p = 0,006 < 0,005 (Ho ditolak = ada hubungan). Ada hubungan antara dukungan keluarga ibu bayi dan ibu balita tentang Posyandu dengan kehadiran ibu bayi dan ibu balita di Posyandu Tanjung Sejahtera, dengan rho = 0,489, p value = 0,001, tingkat signifikansi = 0,05, p = 0,001 < 0,005 (Ho ditolak = ada hubungan). Berdasarkan hasil diatas disarankan agar kepada kader Posyandu diharapkan meningkatkan penyebaran informasi tentang pentingnya Posyandu kepada masyarakat melalui memberi penyuluhan pada saat adanya perkumpulan PKK. Bagi masyarakat, diharapkan terus meningkatkan kunjungan ke Posyandu dengan sering berkonsultasi dengan kader Posyandu ketika diadakan kegiatan Posyand

    THE ACTIVE FRACTION FROM <i>Nigella sativa</i> AND ITS ACTIVITY AGAINST T47D CELL LINE

    Breast cancer is one of the main causes of death in women. Cancer treatment with surgery, chemotherapy, and radiology often cause undesirable side effects. Therefore, alternative cancer treatment by using plants as traditional medicine was expected to reduce side effects. Nigella sativa is one of the plants used as anticancer empirically. This study conducted to examine the cytotoxic activity of Nigella sativa seeds and identify its components on T47D breast cancer cells. Petroleum ether, chloroform, ethyl acetate, and ethanol were used to extract N. sativa seeds. The extracts were tested their cytotoxic activity on T47D cell line using MTT method. The active compound was separated using column chromatography. Cytotoxic test on T47D cell line was perform for extracts of each separation stage. Data were analyzed by probit analysis to obtain IC50 values. Components identification was performed using GC-MS analysis. The results showed that chloroform extract has cytotoxic activity better than other extracts with IC50 of 124.206 µg/mL. The third fraction has cytotoxic activity better than other fractions with IC50 of 68.568 µg/mL. The GC-MS analysis showed that in the third fraction of the chloroform extract contain linoleat acid, the major compound and tryptamine

    Combination of three species of Zingiberaceae prevents doxorubicin-induced hepatotoxicity

    BACKGROUND Doxorubicin as an anticancer drug has hepatotoxic side effects. Curcuma xanthorrhiza, Curcuma longa and Zingiber officinale are commonly used as herbals in Indonesia and around the world. Several compounds in these plants have antioxidant activities and are known to exhibit protection against doxorubicin-induced toxicities. This study aimed to observe the hepatoprotective effect of a combination of C. xanthorriza, C. longa, and Z. officinale extract on doxorubicin-induced hepatotoxicity in rats. METHODS A total of 28 Wistar male rats were divided into four groups: 1) control group (0.9% NaCl); 2) doxorubicin 5 mg/kg intraperitoneally (ip) four times in 14 days (days 1, 5, 9, 13); 3) doxorubicin + combination of C. xanthorriza, C. longa, and Z. officinale (temulawak, kunyit, and jahe merah, designated as Tekuja) 250 mg/kg/day orally for 14 days; and 4) doxorubicin + Tekuja extract 500 mg/kg/day orally for 14 days. Measurements of parameters based on liver histopathology and the parameters of serum alanine amino transferase (ALT) and aspartate amino tranferase (AST). RESULTS Doxorubicin caused significant elevation in serum ALT and AST enzymes after 14 days of treatment. Rats treated with doxorubicin + Tekuja extract 250 mg/kg/ day showed no histological changes, but had decreased levels of ALT and AST.
