Lysosomal enzyme precursors in coated vesicles derived from the exocytic and endocytic pathways.
The molecular forms of two lysosomal enzymes, cathepsin C and cathepsin D, have been examined in lysosomes and coated vesicles (CVs) of rat liver. In addition, the relative proportion of these lysosomal enzymes residing in functionally distinct CV subpopulations was quantitated. CVs contained newly synthesized precursor forms of the enzymes in contrast to lysosomes where only the mature forms were detected. Exocytic and endocytic CV subpopulations were prepared by two completely different protocols. One procedure, a density shift method, uses cholinesterase to alter the density of CVs derived from exocytic or endocytic pathways. The other relies on electrophoretic heterogeneity to accomplish the CV subfractionation. Subpopulations of CVs prepared by either procedure showed similar results, when examined for their relative proportion of cathepsin C and cathepsin D precursors. Within the starting CV preparation, exocytic CVs contained approximately 80-90% of the total steady-state levels of these enzymes while the level in the endocytic population was approximately 10-13%. The implications of these findings are discussed with regard to lysosome trafficking

Se emplearon 96 aves distribuidas en 4 tratamientos con 4 repeticiones y 6 aves en cada una, alimentadas con cromo a partir del d?a noveno de vida hasta el d?a 35. Los tratamientos 1, 2, 3, y 4 conten?an cromo en 0ppm, 0,5 ppm, 1 ppm, y 1,5 ppm en la dieta respectivamente. No se encontraron diferencias significativas (p0,05) en el consumo de alimento. Pese a lo anterior los mejores resultados obtenidos se presentaron en los tratamientos 2. Peso final (1983.3 g vs 2032,3 g), ganancia de peso (1811,9 g vs 1859,9 g ), conversi?n (1,666 vs 1,670). Con respecto al consumo el tratamiento control presento consumo m?s bajo (3305 g vs 3395 g), y rendimiento en canal el tratamiento control presento mejor rendimiento con respecto a los dem?s tratamientos (71,8 % vs 71,3 %). Con respecto a los cortes los resultados favorecen a los tratamientos con cromo, alas (172,9 g vs 181,5 g), pechugas (536,3 g vs 545,1 g), perniles (699,1 g vs 708,6 g). Se determin? que el tratamiento 2 presenta los mejores resultados econ?micos.ABSTRACT. This project aims to evaluate the effect of chromium in breeding chicken diets with different inclusion levels. The items evaluated were: initial weight, final weight, increase in weight, conversion, consumptions, carcass development, wings, breasts, and legs weight, cholesterol, blood sugar, and HDL. A sample of 96 birds was used distributed in 4 treatments with 4 repetitions and 6 birds each, fed with chromium since the 9th day of life until day 35. Treatments 1, 2, 3 and 4 had chromium concentrations of 0 ppm, 0,5 ppm, 1 ppm, and 1,5 ppm in their diet respectively. No significant differences were found (p0,05) was food consumption. Nevertheless, the best results were found in birds subjected to treatment 2: Final weight (1983,3 g vs. 2032,3 g), increase in weight (1811,9 g vs. 1859,9 g), and conversion (1.666 vs. 1.670). Regarding consumption, the control treatment presented the lowest rate (3305 g vs. 3395 g); and regarding yield in channel, the control treatment presented better outputs than the others (71,8 % vs 71,3 %). Examining other criteria, outputs favor chromium treatments: wings (172,9 g vs. 181,5 g), breasts (536,3 g vs. 545,1 g), and legs (669,1 g vs. 708,6 g). Evidence of Climate-Induced Range Contractions in Bull Trout \u3ci\u3eSalvelinus confluentus\u3c/i\u3e in a Rocky Mountain Watersehd, U.S.A.
Many freshwater fish species are considered vulnerable to stream temperature warming associated with climate change because they are ectothermic, yet there are surprisingly few studies documenting changes in distributions. Streams and rivers in the U.S. Rocky Mountains have been warming for several decades. At the same time these systems have been experiencing an increase in the severity and frequency of wildfires, which often results in habitat changes including increased water temperatures. We resampled 74 sites across a Rocky Mountain watershed 17 to 20 years after initial samples to determine whether there were trends in bull trout occurrence associated with temperature, wildfire, or other habitat variables. We found that site abandonment probabilities (0.36) were significantly higher than colonization probabilities (0.13), which indicated a reduction in the number of occupied sites. Site abandonment probabilities were greater at low elevations with warm temperatures. Other covariates, such as the presence of wildfire, nonnative brook trout, proximity to areas with many adults, and various stream habitat descriptors, were not associated with changes in probability of occupancy. Higher abandonment probabilities at low elevation for bull trout provide initial evidence validating the predictions made by bioclimatic models that bull trout populations will retreat to higher, cooler thermal refuges as water temperatures increase. The geographic breadth of these declines across the region is unknown but the approach of revisiting historical sites using an occupancy framework provides a useful template for additional assessments

A systematic review of cerebral microdialysis and outcomes in TBI: relationships to patient functional outcome, neurophysiologic measures, and tissue outcome.
OBJECTIVE: To perform a systematic review on commonly measured cerebral microdialysis (CMD) analytes and their association to: (A) patient functional outcome, (B) neurophysiologic measures, and (C) tissue outcome; after moderate/severe TBI. The aim was to provide a foundation for next-generation CMD studies and build on existing pragmatic expert guidelines for CMD. METHODS: We searched MEDLINE, BIOSIS, EMBASE, Global Health, Scopus, Cochrane Library (inception to October 2016). Strength of evidence was adjudicated using GRADE. RESULTS: (A) Functional Outcome: 55 articles were included, assessing outcome as mortality or Glasgow Outcome Scale (GOS) at 3-6Â months post-injury. Overall, there is GRADE C evidence to support an association between CMD glucose, glutamate, glycerol, lactate, and LPR to patient outcome at 3-6Â months. (B) Neurophysiologic Measures: 59 articles were included. Overall, there currently exists GRADE C level of evidence supporting an association between elevated CMD measured mean LPR, glutamate and glycerol with elevated ICP and/or decreased CPP. In addition, there currently exists GRADE C evidence to support an association between elevated mean lactate:pyruvate ratio (LPR) and low PbtO2. Remaining CMD measures and physiologic outcomes displayed GRADE D or no evidence to support a relationship. (C) Tissue Outcome: four studies were included. Given the conflicting literature, the only conclusion that can be drawn is acute/subacute phase elevation of CMD measured LPR is associated with frontal lobe atrophy at 6Â months. CONCLUSIONS: This systematic review replicates previously documented relationships between CMD and various outcome, which have driven clinical application of the technique. Evidence assessments do not address the application of CMD for exploring pathophysiology or titrating therapy in individual patients, and do not account for the modulatory effect of therapy on outcome, triggered at different CMD thresholds in individual centers. Our findings support clinical application of CMD and refinement of existing guidelines

Achieving Longevity in Wireless Body Area Network by Efficient Transmission Power Control for IoMT Applications
The application of tiny body sensors to collect, process, store, analyze, and retrieve medical information from a human body is a part of the Internet of Medical Things (IoMT). IoMT helps to monitor and track human vital health parameters, predict disease, notify the patients and the health care professionals with relevant data for analyzing the problems before they become severe and for earlier invention. By 2022, more than 60 % of IoT applications will be health-related. The convergence of biomedical sensors, wireless body area networks (WBAN), Information technology, and bioinformatics will help improve the efficiency of saving human lives. In a WBAN, network longevity is challenging because of the limited supply of low power battery energy in tiny body sensor nodes. Here, we proposed an energy-efficient transmission power control (TPC) algorithm to extend the network lifetime in IoMT networks for healthcare applications by eliminating the transceiver overhearing problem. In TPC, human tissue resistivity properties are considered to adjust the transmission power, which reduces the communication power and extends the network lifetime. The simulation results show that network power consumption is reduced by 35%

Benchmarking short-term postoperative mortality across neurosurgery units: is hospital administrative data good enough for risk-adjustment?
BACKGROUND: Surgical mortality indicators should be risk-adjusted when evaluating the performance of organisations. This study evaluated the performance of risk-adjustment models that used English hospital administrative data for 30-day mortality after neurosurgery. METHODS: This retrospective cohort study used Hospital Episode Statistics (HES) data from 1 April 2013 to 31 March 2018. Organisational-level 30-day mortality was calculated for selected subspecialties (neuro-oncology, neurovascular and trauma neurosurgery) and the overall cohort. Risk adjustment models were developed using multivariable logistic regression and incorporated various patient variables: age, sex, admission method, social deprivation, comorbidity and frailty indices. Performance was assessed in terms of discrimination and calibration. RESULTS: The cohort included 49,044 patients. Overall, 30-day mortality rate was 4.9%, with unadjusted organisational rates ranging from 3.2 to 9.3%. The variables in the best performing models varied for the subspecialties; for trauma neurosurgery, a model that included deprivation and frailty had the best calibration, while for neuro-oncology a model with these variables plus comorbidity performed best. For neurovascular surgery, a simple model of age, sex and admission method performed best. Levels of discrimination varied for the subspecialties (range: 0.583 for trauma and 0.740 for neurovascular). The models were generally well calibrated. Application of the models to the organisation figures produced an average (median) absolute change in mortality of 0.33% (interquartile range (IQR) 0.15-0.72) for the overall cohort model. Median changes for the subspecialty models were 0.29% (neuro-oncology, IQR 0.15-0.42), 0.40% (neurovascular, IQR 0.24-0.78) and 0.49% (trauma neurosurgery, IQR 0.23-1.68). CONCLUSIONS: Reasonable risk-adjustment models for 30-day mortality after neurosurgery procedures were possible using variables from HES, although the models for trauma neurosurgery performed less well.
