    Transport and kinase activities of CbrA of Pseudomonas putida KT2440

    The CbrA/CbrB system is a two-component signal transduction system known to participate in the regulation of the cellular carbon/nitrogen balance and to play a central role in carbon catabolite repression in Pseudomonas species. CbrA is composed of a domain with similarity to proteins of the solute/sodium symporter family (SLC5) and domains typically found in bacterial sensor kinases. Here, the functional properties of the sensor kinase CbrA and its domains are analyzed at the molecular level using the system of the soil bacterium P. putida KT2440 as a model. It is demonstrated that CbrA can bind and transport L-histidine. Transport is specific for L-histidine and probably driven by an electrochemical proton gradient. The kinase domain is not required for L-histidine uptake by the SLC5 domain of CbrA, and has no significant impact on transport kinetics. Furthermore, it is shown that the histidine kinase can autophosphorylate and transfer the phosphoryl group to the response regulator CbrB. The SLC5 domain is not essential for these activities but appears to modulate the autokinase activity. A phosphatase activity of CbrA is not detected. None of the activities is significantly affected by L-histidine. The results demonstrate that CbrA functions as a L-histidine transporter and sensor kinase

    A contribuição brasileira de Alcindo Flores Cabral à classificação periódica dos elementos

    This paper presents the contributions of Alcindo Flores Cabral (1907-1982), professor of chemistry at the Faculdade de Agronomia Eliseu Maciel, nowadays part of the Universidade Federal de Pelotas, to chemistry teaching, a contribution almost unknown to the Brazilian chemical community, although recognized as valuable by several renowned chemists abroad, such as W. Hückel, G. Charlot, F. Strong, E. Fessenden and others. Cabral's innovative helicoidal representation is presented not only in connection not only with contemporary representations, but it also includes an incursion into the first helicoidal systems proposed, those of Hinrichs and of Baumhauer. Some comments are made on Cabral's Classificação natural dos elementos, published in 1946, and also on other texts that he wrote to make chemistry teaching more effective.Este artigo apresenta a contribuição de Alcindo Flores Cabral (1907-1982) - professor de química da Faculdade de Agronomia Eliseu Maciel, hoje incorporada à Universidade Federal de Pelotas - ao ensino de química, uma contribuição quase desconhecida pela própria comunidade química brasileira, embora reconhecida como relevante por diversos químicos estrangeiros importantes, como W. Hückel, G. Charlot, F. Strong, E. Fessenden e outros. A inovadora representação helicoidal de Cabral é apresentada não só em conexão com representações contemporâneas, mas também inclui-se uma incursão pelos primeiros sistemas helicoidais propostos, os de Hinrichs e de Baumhauer.     Idade relativa em jogadores de futebol profissional do Brasil

    Durante o processo de formação os jogadores de Futebol são agrupados por categorias etárias de acordo com o seu ano de nascimento. Os jogadores nascidos nos primeiros meses do ano normalmente apresentam uma maturação biológica mais avançada, fato que lhes proporcionam uma vantagem no desempenho da prestação esportiva. Essa vantagem é denominada na literatura esportiva como o efeito da idade relativa.Com isso, o objetivo deste estudo é verificar a existência do efeito da idade relativa entre os jogadores de Futebol no Campeonato Brasileiro Série A. Foram analisados os dados referentes à data de nascimento de 1045 jogadores de Futebol participantes do Campeonato Brasileiro da Série A em 2012. Os dados foram agrupados em quartis, de acordo com o mês de nascimento dos participantes, sendo: 1º quartil – janeiro, fevereiro emarço; 2º quartil – abril, maio e junho; 3º quartil – julho, agosto e setembro; 4º quartil – outubro, novembro e dezembro. Foi utilizada estatística descritiva (frequência e percentual) e para comparação da frequência de jogadores por quartil foi utilizado o teste qui-quadrado (x2). A análise mostrou a seguinte distribuição dos dados nos quartis: Q1 (n = 326), Q2 (n = 296), Q3 (n = 235) e Q4 (n = 188). O teste qui-quadrado revelou diferenças estatisticamente significativas entre os quartis (x²(3) = 43,846). Com os resultados apresentados, pode-se concluir que o efeito da idade relativa está presente entre os jogadores do Campeonato Brasileiro Série A

    Residential exposure to motor vehicle emissions and the risk of wheezing among 7-8 year-old schoolchildren: a city-wide cross-sectional study in Nicosia, Cyprus

    <p>Abstract</p> <p>Background</p> <p>Several studies have reported associations between respiratory outcomes in children and a range of self-reported, administrative or geographical indicators of traffic pollution. First-time investigation into the frequency of asthmatic symptoms among 7-8 year-old Cypriot children in 1999-2000 showed increased prevalence in the capital Nicosia compared to other areas. Geographical differences on an island the size of Cyprus may reflect environmental and/or lifestyle factors. This study investigates the relationship between self-reported symptoms and residential exposure to motor vehicle emissions among Nicosia schoolchildren.</p> <p>Methods</p> <p>The addresses of children in the metropolitan area of Nicosia who participated in the original survey (N = 1,735) were geo-coded and the level of exposure of each child was assessed using distance- and emission-based indicators (i.e. estimated levels of particulate matter and nitrogen oxides emissions due to motor vehicles on main roads around the residence). Odds ratios of wheezing and asthma diagnosis in relation to levels of exposure were estimated in logistic regression models adjusting for person-based factors, co-morbidity and intra-school clustering.</p> <p>Results</p> <p>We found an increased risk of wheezing at distances less than 50 m from a main road and/or only among those experiencing the highest levels of exposure. The strongest effect estimates were observed when exposure was defined in terms of the cumulative burden at all roads around the residence. Adjusted odds ratios for current wheezing were 2.33 (95% CI 1.27, 4.30) amongst the quartile of participants exposed to the highest levels of PM at all roads 50 m of their residence and 2.14 (95% CI 1.05, 4.35) for NOx, with no effect at intermediate levels of exposure. While the direction of effect was apparent at longer distances, differences were generally not statistically significant.</p> <p>Conclusions</p> <p>Children experiencing the highest burden of emissions in Nicosia seem to be at a higher risk of reporting asthmatic symptoms. Due to the small number of children residing at close proximity to main roads and lack of evidence of risk at intermediate levels of exposure or longer distances, the observed pattern alone does not explain the generally higher prevalence observed in urban Nicosia compared to other areas.</p

    The Canadian Healthy Infant Longitudinal Development (CHILD) birth cohort study: Assessment of environmental exposures

    The Canadian Healthy Infant Longitudinal Development birth cohort was designed to elucidate interactions between environment and genetics underlying development of asthma and allergy. Over 3600 pregnant mothers were recruited from the general population in four provinces with diverse environments. The child is followed to age 5 years, with prospective characterization of diverse exposures during this critical period. Key exposure domains include indoor and outdoor air pollutants, inhalation, ingestion and dermal uptake of chemicals, mold, dampness, biological allergens, pets and pests, housing structure, and living behavior, together with infections, nutrition, psychosocial environment, and medications. Assessments of early life exposures are focused on those linked to inflammatory responses driven by the acquired and innate immune systems. Mothers complete extensive environmental questionnaires including time-activity behavior at recruitment and when the child is 3, 6, 12, 24, 30, 36, 48, and 60 months old. House dust collected during a thorough home assessment at 3–4 months, and biological specimens obtained for multiple exposure-related measurements, are archived for analyses. Geo-locations of homes and daycares and land-use regression for estimating traffic-related air pollution complement time-activity-behavior data to provide comprehensive individual exposure profiles. Several analytical frameworks are proposed to address the many interacting exposure variables and potential issues of co-linearity in this complex data set

    Mouse models to unravel the role of inhaled pollutants on allergic sensitization and airway inflammation

    Air pollutant exposure has been linked to a rise in wheezing illnesses. Clinical data highlight that exposure to mainstream tobacco smoke (MS) and environmental tobacco smoke (ETS) as well as exposure to diesel exhaust particles (DEP) could promote allergic sensitization or aggravate symptoms of asthma, suggesting a role for these inhaled pollutants in the pathogenesis of asthma. Mouse models are a valuable tool to study the potential effects of these pollutants in the pathogenesis of asthma, with the opportunity to investigate their impact during processes leading to sensitization, acute inflammation and chronic disease. Mice allow us to perform mechanistic studies and to evaluate the importance of specific cell types in asthma pathogenesis. In this review, the major clinical effects of tobacco smoke and diesel exhaust exposure regarding to asthma development and progression are described. Clinical data are compared with findings from murine models of asthma and inhalable pollutant exposure. Moreover, the potential mechanisms by which both pollutants could aggravate asthma are discussed
