    Using organic phosphorus to sustain pasture productivity: A perspective

    Organic phosphorus (P) in grazed pastures/grasslands could sustain production systems that historically relied on inorganic P fertiliser. Interactions between inorganic P, plants and soils have been studied extensively. However, less is known about the transformation of organic P to inorganic orthophosphate. This paper investigates what is known about organic P in pasture/grassland soils used for agriculture, as well as the research needed to utilise organic P for sustainable plant production. Organic P comprises > 50% of total soil P in agricultural systems depending on location, soil type and land use. Organic P hydrolysis and release of orthophosphate by phosphatase enzymatic activity is affected by a range of factors including: (a) the chemical nature of the organic P and its ability to interact with the soil matrix; (b) microorganisms that facilitate mineralisation; (c) soil mineralogy; (d) soil water electrolytes; and (e) soil physicochemical properties. Current biogeochemical knowledge of organic P processing in soil limits our ability to develop management strategies that promote the use of organic P in plant production. Information is particularly needed on the types and sources of organic P in grassland systems and the factors affecting the activity of enzymes that mineralise organic P. Integrated approaches analysing the soil matrix, soil water and soil biology are suggested to address this knowledge gap

    RNA sequencing and proteomics approaches reveal novel deficits in the cortex of Mecp2-deficient mice, a model for Rett syndrome

    BACKGROUND: Rett syndrome (RTT) is an X-linked neurodevelopmental disorder caused by mutations in the transcriptional regulator MeCP2. Much of our understanding of MeCP2 function is derived from transcriptomic studies with the general assumption that alterations in the transcriptome correlate with proteomic changes. Advances in mass spectrometry-based proteomics have facilitated recent interest in the examination of global protein expression to better understand the biology between transcriptional and translational regulation. METHODS: We therefore performed the first comprehensive transcriptome-proteome comparison in a RTT mouse model to elucidate RTT pathophysiology, identify potential therapeutic targets, and further our understanding of MeCP2 function. The whole cortex of wild-type and symptomatic RTT male littermates (n = 4 per genotype) were analyzed using RNA-sequencing and data-independent acquisition liquid chromatography tandem mass spectrometry. Ingenuity(R) Pathway Analysis was used to identify significantly affected pathways in the transcriptomic and proteomic data sets. RESULTS: Our results indicate these two omics data sets supplement one another. In addition to confirming previous works regarding mRNA expression in Mecp2-deficient animals, the current study identified hundreds of novel protein targets. Several selected protein targets were validated by Western blot analysis. These data indicate RNA metabolism, proteostasis, monoamine metabolism, and cholesterol synthesis are disrupted in the RTT proteome. Hits common to both data sets indicate disrupted cellular metabolism, calcium signaling, protein stability, DNA binding, and cytoskeletal cell structure. Finally, in addition to confirming disrupted pathways and identifying novel hits in neuronal structure and synaptic transmission, our data indicate aberrant myelination, inflammation, and vascular disruption. Intriguingly, there is no evidence of reactive gliosis, but instead, gene, protein, and pathway analysis suggest astrocytic maturation and morphological deficits. CONCLUSIONS: This comparative omics analysis supports previous works indicating widespread CNS dysfunction and may serve as a valuable resource for those interested in cellular dysfunction in RTT

    Aims: To determine the proportion of people with diabetes who have Hb A1c measured, what proportion achieve an HbA1c level of < 58 mmol/mol (7.5%), the frequency of testing and if there was any change in HbA1c level in the year before and the year after an incident stroke. Methods: This study used the Secure Anonymised Information Linkage (SAIL) databank, which stores hospital data for the whole of Wales and ~65% of Welsh general practice records, to identify cases of stroke in patients with diabetes between 2000 and 2010. These were matched against patients with diabetes but without stroke disease. We assessed the frequency of HbA1c testing and change in HbA1c in the first year after stroke. Estimation was made of the proportion of patients achieving an HbA1c measurement ≤ 58 mmol/mol (7.5%). Results: There were 1741 patients with diabetes and stroke. Of these, 1173 (67.4%) had their HbA1c checked before their stroke and 1137 (65.3%) after their stroke. In the control group of 16 838 patients with diabetes but no stroke, 8413 (49.9%) and 9288 (55.1%) had their HbA1c checked before and after the case-matched stroke date, respectively. In patients with diabetes and stroke, HbA1c fell from 7.7 to 7.3% after their stroke ( P<0.001). Before the study, 55.0% of patients with stroke had an HbA1c ≥58 mmol/mol compared with 65.2% of control patients, these figures were 62.5% and 65.3% after the stroke. Conclusions: The frequency of diabetes testing was higher in patients who had experienced a stroke before and after their incident stroke compared with control patients but did not increase after their stroke.     Anaerobic digestion of whole-crop winter wheat silage for renewable energy production

    With biogas production expanding across Europe in response to renewable energy incentives, a wider variety of crops need to be considered as feedstock. Maize, the most commonly used crop at present, is not ideal in cooler, wetter regions, where higher energy yields per hectare might be achieved with other cereals. Winter wheat is a possible candidate because, under these conditions, it has a good biomass yield, can be ensiled, and can be used as a whole crop material. The results showed that, when harvested at the medium milk stage, the specific methane yield was 0.32 m3 CH4 kg–1 volatile solids added, equal to 73% of the measured calorific value. Using crop yield values for the north of England, a net energy yield of 146–155 GJ ha–1 year–1 could be achieved after taking into account both direct and indirect energy consumption in cultivation, processing through anaerobic digestion, and spreading digestate back to the land. The process showed some limitations, however: the relatively low density of the substrate made it difficult to mix the digester, and there was a buildup of soluble chemical oxygen demand, which represented a loss in methane potential and may also have led to biofoaming. The high nitrogen content of the wheat initially caused problems, but these could be overcome by acclimatization. A combination of these factors is likely to limit the loading that can be applied to the digester when using winter wheat as a substrat

    A QM/MM approach for the study of monolayer-protected gold clusters

    We report the development and implementation of hybrid methods that combine quantum mechanics (QM) with molecular mechanics (MM) to theoretically characterize thiolated gold clusters. We use, as training systems, structures such as Au25(SCH2-R)18 and Au38(SCH2-R)24, which can be readily compared with recent crystallographic data. We envision that such an approach will lead to an accurate description of key structural and electronic signatures at a fraction of the cost of a full quantum chemical treatment. As an example, we demonstrate that calculations of the 1H and 13C NMR shielding constants with our proposed QM/MM model maintain the qualitative features of a full DFT calculation, with an order-of-magnitude increase in computational efficiency.Comment: Journal of Materials Science, 201

    The politics of health services research: health professionals as hired hands in a commissioned research project in England

    Previous health services research has failed to account for the role played by clinical staff in the collection of data. In this paper we use the work of Roth on hired hand research to examine the politics of evidence production within health services research. Sociologies of work predict lack of engagement in the research tasks by subordinated groups of workers. We examine the role of midwives in researching ante-natal screening for sickle cell and thalassaemia in England, and construct three ideal types: repairers, refractors, and resisters to account for the variable engagement of health staff with research. We find some features of the hired hand phenomenon predicted by Roth to be in evidence, and suggest that the context of our project is similar to much health services research. We conclude that without concerted attempts (1) to change the social relations of research production; (2) to mitigate hired hand effects; (3) to assess the impact of the hired hand effect on the validity and reliability of findings, and (4) to report on these limitations, that health services research involving large teams of subordinated clinical staff as data collectors will be prone to produce evidence that is of limited trustworthiness.     Scattering of Stark-decelerated OH radicals with rare-gas atoms

    We present a combined experimental and theoretical study on the rotationally inelastic scattering of OH (X\,^2\Pi_{3/2}, J=3/2, f) radicals with the collision partners He, Ne, Ar, Kr, Xe, and D2_2 as a function of the collision energy between ∼70\sim 70 cm−1^{-1} and 400~cm−1^{-1}. The OH radicals are state selected and velocity tuned prior to the collision using a Stark decelerator, and field-free parity-resolved state-to-state inelastic relative scattering cross sections are measured in a crossed molecular beam configuration. For all OH-rare gas atom systems excellent agreement is obtained with the cross sections predicted by close-coupling scattering calculations based on accurate \emph{ab initio} potential energy surfaces. This series of experiments complements recent studies on the scattering of OH radicals with Xe [Gilijamse \emph{et al.}, Science {\bf 313}, 1617 (2006)], Ar [Scharfenberg \emph{et al.}, Phys. Chem. Chem. Phys. {\bf 12}, 10660 (2010)], He, and D2_2 [Kirste \emph{et al.}, Phys. Rev. A {\bf 82}, 042717 (2010)]. A comparison of the relative scattering cross sections for this set of collision partners reveals interesting trends in the scattering behavior.Comment: 10 pages, 5 figure

    Electronic Structure and Bonding of Icosahedral Core-Shell Gold-Silver Nanoalloy Clusters Au_(144-x)Ag_x(SR)_60

    Atomically precise thiolate-stabilized gold nanoclusters are currently of interest for many cross-disciplinary applications in chemistry, physics and molecular biology. Very recently, synthesis and electronic properties of "nanoalloy" clusters Au_(144-x)Ag_x(SR)_60 were reported. Here, density functional theory is used for electronic structure and bonding in Au_(144-x)Ag_x(SR)_60 based on a structural model of the icosahedral Au_144(SR)_60 that features a 114-atom metal core with 60 symmetry-equivalent surface sites, and a protecting layer of 30 RSAuSR units. In the optimal configuration the 60 surface sites of the core are occupied by silver in Au_84Ag_60(SR)_60. Silver enhances the electron shell structure around the Fermi level in the metal core, which predicts a structured absorption spectrum around the onset (about 0.8 eV) of electronic metal-to-metal transitions. The calculations also imply element-dependent absorption edges for Au(5d) \rightarrow Au(6sp) and Ag(4d) \rightarrow Ag(5sp) interband transitions in the "plasmonic" region, with their relative intensities controlled by the Ag/Au mixing ratio.Comment: 4 figure

    The politics of evidence-based policy in Europe’s ‘migration crisis’

    Significant increases in the number of refugees and other migrants arriving across the Mediterranean to Europe during 2015 were associated with an increased emphasis on gathering research evidence to enable policymakers to better understand the complexities of migration and improve the policy response. In the UK, the emphasis on evidence-based policy was reflected in funding by the Economic and Social Research Council for a Mediterranean Migration Research Programme. Drawing on the evidence gathered through this programme, the articles in the volume explore the nature of Europe’s ‘migration crisis’ and the extent to which the development of new migration management policies was grounded in evidence about the causes, drivers and consequences of migration to Europe. The authors conclude that there is a substantial ‘gap’ between the now significant body of evidence examining migration processes and European Union policy responses. This gap can be attributed to three main factors: the long-standing ‘paradigm war’ in social research between positivist, interpretivist and critical approaches which means that what counts as ‘evidence’ is contested; competing knowledge claims associated with research and other forms of evidence including the management data to construct and/or support particular policy narratives; and, perhaps most importantly, the political context within which migration policymaking takes place. The politics of policymaking, perhaps nowhere more evident than in the area of migration, has resulted in policies based on underlying assumptions and vested political interests rather than the evidence, even where this evidence has been funded directly by European governments

    Clinician acquisition and retention of Motivational Interviewing skills: a two-and-a-half-year exploratory study

    <p>Abstract</p> <p>Background</p> <p>Motivational interviewing (MI) is a collaborative, client-centred counselling style aimed at eliciting and strengthening clients' intrinsic motivation to change. There is strong research evidence supporting the efficacy of MI, notably in its application among alcohol and drug abuse populations. MI interventions in smoking cessation may yield modest but significant increases in quitting. The present study sought to assess the acquisition and retention of MI skills in counsellors at the Swedish National Tobacco Quitline.</p> <p>Methods</p> <p>Three audio-recorded sessions from each of three counsellors were assessed using the Motivational Interviewing Treatment Integrity (MITI) Code Version 3.0 over 11 assessment periods at fixed intervals in a two-and-a-half year period during which counsellors received ongoing supervision.</p> <p>Results</p> <p>The mean skill for all counsellors improved throughout the study period in most MITI variables. However, great variations in MI skill between counsellors were observed, as well as fluctuations in performance in counsellors over time.</p> <p>Conclusion</p> <p>The present exploratory study covers a longer time period than most evaluations of MI training, and has several advantages with regard to study design. It may provide a basis for (larger sample) replication to test MI skill (as measured by the MITI) in relation to behaviour change in clients, to evaluate MI training, and to assess the acquisition and retention of MI skill over time. Difficulties in acquiring and retaining MI skill may raise the issue of a selection policy for MI training. Moreover, fluctuations in MI skill over time emphasise the greater importance of continuous feedback and supervision over initial MI training, and the need for the use of validated treatment integrity assessment instruments in ordinary clinical implementations of MI.</p
