Dual Regulation of the lin-14 Target mRNA by the lin-4 miRNA
microRNAs (miRNAs) are ∼22 nt regulatory RNAs that in animals typically bind with partial complementarity to sequences in the 3′ untranslated (UTR) regions of target mRNAs, to induce a decrease in the production of the encoded protein. The relative contributions of translational inhibition of intact mRNAs and degradation of mRNAs caused by binding of the miRNA vary; for many genetically validated miRNA targets, translational repression has been implicated, whereas some analyses of other miRNA targets have revealed only modest translational repression and more significant mRNA destabilization. In Caenorhabditis elegans, the lin-4 miRNA accumulates during early larval development, binds to target elements in the lin-14 mRNA, and causes a sharp decrease in the abundance of LIN-14 protein. Here, we monitor the dynamics of lin-14 mRNA and protein as well as lin-4 miRNA levels in finely staged animals during early larval development. We find complex regulation of lin-14, with the abundance of lin-14 mRNA initially modestly declining followed by fluctuation but little further decline of lin-14 mRNA levels accompanied by continuing and more dramatic decline in LIN-14 protein abundance. We show that the translational inhibition of lin-14 is dependent on binding of the lin-4 miRNA to multiple lin-4 complementary sites in the lin-14 3′UTR. Our results point to the importance of translational inhibition in silencing of lin-14 by the lin-4 miRNA
Small Texas School Districts’ Response to State Funding Reductions
In response to a challenging state economy, the Texas Legislature implemented the Regular Program Adjustment Factor (RPAF) in 2011, effectively reducing state funding to all Texas school districts. This mixed methods study reveals the effect of the RPAF on a sample of the smallest Texas school districts and their response to decreased state funding – inclusive of reducing staff, implementing tax rollback and bond elections, and securing revenue from other, non-traditional, financing sources, which ultimately served as the largest revenue enhancement – 97 percent of which was comprised of the issuance of capital-related debt

Japanese Beef Policy and GATT Negotiations: Analysis of Reducing Assistance to Beef Producers
Since at least the mid-1970s, Japan and its beef import suppliers, the U.S. and Australia in particular, have engaged in heated negotiations on the level of the Japanese beef import quota. U.S. negotiators have demanded that Japan completely liberalize beef imports. The Japanese have responded in a piecemeal fashion, increasing the quota by comparatively small amounts in an apparent attempt both to appease U.S. interests and to minimize the opposition of the politically powerful domestic cattle producers. The most recent Japanese concession was an agreement in the fall of 1984 to expand the total beef import quota by 9,000 metric tons (mt) per year for four years, bringing total imports to 177,000 mt by early 1988
An opportunity analysis system for space surveillance experiments with the MSX
The Mid-Course Space Experiment consists of a set of payloads on a satellite being designed and built under the sponsorship of Ballistic Missile Defense Office. The MSX satellite will conduct a series of measurements of phenomenology of backgrounds, missile targets, plumes and resident space objects (RSO's); and will engage in functional demonstrations in support of detection, acquisition and tracking for ballistic missile defense and space-based space surveillance missions. A complex satellite like the MSX has several constraints imposed on its operation by the sensors, the supporting instrumentation, power resources, data recording capability, communications and the environment in which all these operate. This paper describes the implementation of an opportunity and feasibility analysis system, developed at Lincoln Laboratory, Massachusetts Institute of Technology, specifically to support the experiments of the Principal Investigator for space-based surveillance
Conclusion: Meditations on the Archaeology of Northern Plantations
A summary of the methods employed and the conclusions reached after nine seasons of archaeological fieldwork are presented. Emphasis is placed on the success and limitations of the methods employed in the investigations at Sylvester Manor and results of those investigations. Although excavations concentrated on the plantation core, additional areas examined produced little in the way of archaeological features. The results, although preliminary, point to a major role for Native Americans as laborers during the earliest phases of the plantation’s operation. Landscape evidence also suggests an evolving economy as the Manor transitions from a provisioning operation to a commercial farm/tenant run operation within a decade of Nathaniel Sylvester’s death in 1680. A third transition saw the commercial farm reconfigured once again as a Georgian-inspired country estate eclipsed it

Inducing hardening and healability in poly(ethylene-co-acrylic acid) via blending with complementary low molecular weight additives
The design and synthesis of low molecular weight additives based on self-assembling nitroarylurea units, and their compatibility with poly(ethylene-co-acrylic acid) copolymers are reported. The self-assembly properties of the low molecular weight additives have been demonstrated in a series of gelation studies. Upon blending at low percentage weights (≤ 5%) with poly(ethylene-co-acrylic acid) the additives were capable of increasing the stress and strain to failure when compared to the parent copolymer. By varying the percentage weight of the additive as well as the type of additive the mechanical properties of poly(ethylene-co-acrylic acid) could be tailored. Finally, the healability characteristics of the blends were improved when compared to the original polymer via the introduction of a supramolecular ‘network within a network’
