    In 1872, Isabella Valancy Crawford answered a call printed in George-Édouard Desbarats’s weekly story paper the Hearthstone seeking: “narratives, novels, sketches penned by vigorous Canadian hands, welling out from fresh and fertile Canadian brains, thrilling with the adventures by sea and land, of Canadian heroes” (Early and Peterman 25). Crawford’s winning submission to the Hearthstone's call, Winona; or, The Foster-Sisters, reaps the materials for its narrative from “inexhaustible fields” of both “fact and fancy” of a burgeoning Canadian national imagination (25). This paper is interested in exploring the specifically Canadian anxieties expressed by the novel, as this paper examines the manner in which the displaced occupants of the novel’s Howard lodge act as uncanny avatars of the natural world and of a wilderness as they resist (or, are denied) a place in the domestic space established by the “national family” (167).  In this paper, I argue that Crawford’s Winona, with its attention to both domestic and natural spaces, provides a productive site through which to interrogate the vexed relationship of a newly Confederated country with its own “native materials” (Johnson 7; Early and Peterman 10)

    Preservice Elementary Teachers’ Beliefs about the Role of Definition in the Learning of Mathematics

    Answering a call to emphasize the act of defining over the learning of definitions, we have shifted the content of a geometry course for preservice elementary teachers (PSETs) away from comparing and applying pre-written classification structures to classroom episodes centered on authoring definitions for special quadrilaterals. PSETs complete activities using geometry software, and collaboratively create definitions for specific quadrilaterals. In order to fully understand the potential of this curricular shift, we asked preservice elementary mathematics teachers’ to share their perceptions of the process of writing mathematical definitions. Data from participant reflections were analyzed for themes related to mathematical definition and the act of defining. The framework that resulted from iterative discussions by the researchers examined beliefs about the nature of definition and mathematical empathy (Araki, 2015). Findings also suggest that beliefs about authority and positioning students as authors of mathematics are associated with mathematical empathy. Experiences related to the process of defining enabled PSETs to see far greater subjectivity in the discipline of mathematics and to consider, perhaps for the first time, that they, too, were both able and deserving of becoming authors of mathematical ideas

    This study examined a random stratified sample (n=62) of prospective teachers\u27 work across eight institutions on three tasks that utilized dynamic statistical software. Our work was guided by considering how teachers may utilize their statistical knowledge and technological statistical knowledge to engage in cycles of investigation. Although teachers did not tend to take full advantage of dynamic linking capabilities, they utilized a large variety of graphical representations and often added statistical measures or other augmentations to graphs as part of their analysis

    Characterization of the yeast ionome: a genome-wide analysis of nutrient mineral and trace element homeostasis in Saccharomyces cerevisiae

    BACKGROUND: Nutrient minerals are essential yet potentially toxic, and homeostatic mechanisms are required to regulate their intracellular levels. We describe here a genome-wide screen for genes involved in the homeostasis of minerals in Saccharomyces cerevisiae. Using inductively coupled plasma-atomic emission spectroscopy (ICP-AES), we assayed 4,385 mutant strains for the accumulation of 13 elements (calcium, cobalt, copper, iron, potassium, magnesium, manganese, nickel, phosphorus, selenium, sodium, sulfur, and zinc). We refer to the resulting accumulation profile as the yeast 'ionome'. RESULTS: We identified 212 strains that showed altered ionome profiles when grown on a rich growth medium. Surprisingly few of these mutants (four strains) were affected for only one element. Rather, levels of multiple elements were altered in most mutants. It was also remarkable that only six genes previously shown to be involved in the uptake and utilization of minerals were identified here, indicating that homeostasis is robust under these replete conditions. Many mutants identified affected either mitochondrial or vacuolar function and these groups showed similar effects on the accumulation of many different elements. In addition, intriguing positive and negative correlations among different elements were observed. Finally, ionome profile data allowed us to correctly predict a function for a previously uncharacterized gene, YDR065W. We show that this gene is required for vacuolar acidification. CONCLUSION: Our results indicate the power of ionomics to identify new aspects of mineral homeostasis and how these data can be used to develop hypotheses regarding the functions of previously uncharacterized genes

    Minocycline 200 mg or 400 mg versus placebo for mild Alzheimer's disease: the MADE Phase II, three-arm RCT

    Background: Minocycline is an anti-inflammatory drug and protects against the toxic effects of ÎČ-amyloid in vitro and in animal models of Alzheimer’s disease. To the best of our knowledge, no randomised placebo-controlled clinical trials in patients with Alzheimer’s disease looking at the efficacy and tolerability of minocycline have been carried out. Objectives: The trial investigated whether or not minocycline was superior to placebo in slowing down the rate of decline in cognitive and functional ability over 2 years. The safety and tolerability of minocycline were also assessed. Design: A Phase II, three-arm, randomised, double-blind, multicentre trial with a semifactorial design. Participants continued on trial treatment for up to 24 months. Setting: Patients were identified from memory services, both within the 32 participating NHS trusts and within the network of memory services supported by the Dementias and Neurodegenerative Diseases Research Network (also known as DeNDRoN). Participants: Patients with standardised Mini Mental State Examination scores of > 23 points and with Alzheimer’s disease assessed by the National Institute on Aging–Alzheimer’s Association’s criteria were identified from memory services. Intervention: Patients with mild Alzheimer’s disease were randomly allocated 1 : 1 : 1 to receive one of three treatments: arm 1 – 400 mg per day of minocycline; arm 2 – 200 mg per day of minocycline; or arm 3 – placebo. Patients continued treatment for 24 months. Participants, investigators and outcome assessors were blind to treatment allocation. Main outcome measures: Primary outcome measures were decline in standardised Mini Mental State Examination and Bristol Activities of Daily Living Scale scores of combined minocycline treatment arms versus placebo, as analysed by intention-to-treat repeated measures regression. Results: Between 23 May 2014 and 14 April 2016, 554 participants were randomised. Of the 544 eligible participants, the mean age was 74.3 years and the average standardised Mini Mental State Examination score was 26.4 points. A total of 252 serious adverse events were reported, with the most common categories being neuropsychiatric and cardiocirculatory. Significantly fewer participants completed treatment with 400 mg of minocycline [29% (53/184)] than 200 mg [62% (112/181)] or placebo [64% (114/179)] (p < 0.0001), mainly because of gastrointestinal symptoms (p = 0.0008), dermatological side effects (p = 0.02) and dizziness (p = 0.01). Assessment rates were also lower in the 400-mg treatment arm: 68% (119 of 174 expected) for standardised Mini Mental State Examination scores at 24 months, compared with 82% (144/176) for the 200-mg treatment arm and 84% (140/167) for the placebo arm. Decline in standardised Mini Mental State Examination scores over the 24-month study period in the combined minocycline arms was similar to that in the placebo arm (4.1- vs. 4.3-point reduction; p = 0.9), as was the decline in the 400- and 200-mg treatment arms (3.3 vs. 4.7 points; p = 0.08). Likewise, worsening of Bristol Activities of Daily Living Scale scores over 24 months was similar in all trial arms (5.7, 6.6 and 6.2 points in the 400-mg treatment arm, 200-mg treatment arm and placebo arm, respectively; a p-value of 0.57 for minocycline vs. placebo and a p-value of 0.77 for 400 vs. 200 mg of minocycline). Results were similar in different patient subgroups and in sensitivity analyses adjusting for missing data. Limitations: Potential limitations of the study include that biomarkers were not used to confirm the diagnosis of Alzheimer’s disease, as these and apolipoprotein E (APOE) genotyping are not routinely available within the NHS. Compliance was also worse than expected and differential follow-up rates were observed, with fewer assessments obtained for the 400-mg treatment arm than for the 200-mg treatment and placebo arms. Conclusions: Minocycline does not delay the progress of cognitive or functional impairment in people with mild Alzheimer’s disease over a 2-year period. Minocycline at a dose of 400 mg is poorly tolerated in this population. Future work: The Minocycline in mild Alzheimer’s DiseasE (MADE) study provides a framework for a streamlined trial design that can be usefully applied to test other disease-modifying therapies

    Admixed Portrait: Design to Understand Facebook Portrayals in New Parenthood

    We report on a design-led study of the photographic representation of self and family on Facebook during and after becoming parents for the first time. Our experience-centered, research-through-design study engaged eight participants across five UK homes, in a month-long deployment of a prototype technology -- a design research artifact, Admixed Portrait, that served to prompt participant reflection on first-time parenthood. In addition to pre- and post-deployment interviews, participants kept diaries capturing personal reflections during the deployment, on daily social media use and interactions with Admixed. Our qualitative insights on social media representations of transitional experience and identity for new parents, reveal how their online 'photowork' related to self-expression and social functioning. We contribute design considerations for developing tools to support photographic expression in social media use, and methodological insights about design-led inquiry for understanding transitional experiences

    #DryByChristmas: A patient and public involvement study on women's engagement with humorous pelvic floor muscle training digital nudges on social media

    Introduction: Patient and public involvement (PPI) is essential for women's health research. Little is known about how women engage with humorous social media and behavioural health messaging targeting pelvic floor muscle training (PFMT). This PPI aimed to understand how women engage with a humorous social media campaign encouraging PFMT. The study findings will influence the co-design of a digital intervention to support women's adherence to PFMT. Methods: The Guidance for Reporting Involvement of Patients and the Public Version 2 short form was used to report the study's findings. The study examined public engagement with a humorous social media campaign encouraging PFMT in women. A healthcare professional and comedian ran the campaign following the national guidelines for engagement in PFMT. Instagram analytics gave insight into the demographics of the public who engaged, how they engaged and the most popular content. The behaviour change techniques (BCTs) used in the digital nudges that generated the highest levels of engagement were analysed using the Capability Opportunity Motivation Behaviour Change Wheel. Results: The majority (96%) of the population showing the highest levels of engagement were women aged 35–44 years and were based in the United Kingdom (77%). The Instagram account saw an increase in engagement by 12% over the 3-month campaign, with 22,032 users seeing digital nudges and 2645 engaging with the digital nudges. The preferred way of engaging was using Likes (9723). The common themes in the digital nudges that generated the highest levels of engagement were BCTs associated with the ‘social influences’ theoretical domain framework that targeted the core behaviour opportunity. Conclusion: The study findings suggest humour may improve women's engagement with online PFMT programmes; however, more rigorous research is required to better understand diverse women's experiences of humorous online PFMT nudges. Future studies may use PFMT mobile apps instead of social media to capture true user engagement and adherence to PFMT more accurately. The insights gained from the study will be taken forward to co-design a digital behavioural intervention as part of a larger study. Public Contribution: Members of the public were involved in the co-design of a digital health intervention that will be trialled as part of a larger research study. The public was involved using the social media platform Instagram.     Left in the cold? Evolutionary origin of Laternula elliptica a keystone bivalve species of Antarctic benthos

    The large, burrowing bivalve Laternula elliptica is an abundant component of shallow-water soft-substrate communities around Antarctica but its congeners are temperate and tropical in distribution and their phylogenetic relationships are obscure. A new molecular analysis of Laternulidae species shows that there are two distinct clades, one of Exolaternula species, E. spengleri and E. liautaudi, possessing a ligamental lithodesma and a larger clade of species lacking the lithodesma. Of the latter, Laternula elliptica is a sister taxon to temperate and tropical species, including those that live around the coasts of Australia from Tasmania to Darwin. It is suggested that L. elliptica was left isolated around Antarctica following the opening of the Tasman Gateway and initiation of the Circum-Antarctic Current as Australia drifted northwards following the final breakup of Gondwana. A further scenario is that as Australia moved closer to Asia, species spread into tropical habitats and more widely to the Red Sea and Japan. Exolaternula species have a likely Tethyan origin and the present-day range is from the Arabian Gulf, around southern Asia and as far north as southern Russia

    The interface between assisted reproductive technologies and genetics: technical, social, ethical and legal issues

    The interface between assisted reproductive technologies (ART) and genetics comprises several sensitive and important issues that affect infertile couples, families with severe genetic diseases, potential children, professionals in ART and genetics, health care, researchers and the society in general. Genetic causes have a considerable involvement in infertility. Genetic conditions may also be transmitted to the offspring and hence create transgenerational infertility or other serious health problems. Several studies also suggest a slightly elevated risk of birth defects in children born following ART. Preimplantation genetic diagnosis (PGD) has become widely practiced throughout the world for various medical indications, but its limits are being debated. The attitudes towards ART and PGD vary substantially within Europe. The purpose of the present paper was to outline a framework for development of guidelines to be issued jointly by European Society of Human Genetics and European Society of Human Reproduction and Embryology for the interface between genetics and ART.
