Mixed Hodge polynomials of character varieties
We calculate the E-polynomials of certain twisted GL(n,C)-character varieties
M_n of Riemann surfaces by counting points over finite fields using the
character table of the finite group of Lie-type GL(n,F_q) and a theorem proved
in the appendix by N. Katz. We deduce from this calculation several geometric
results, for example, the value of the topological Euler characteristic of the
associated PGL(n,C)-character variety. The calculation also leads to several
conjectures about the cohomology of M_n: an explicit conjecture for its mixed
Hodge polynomial; a conjectured curious Hard Lefschetz theorem and a conjecture
relating the pure part to absolutely indecomposable representations of a
certain quiver. We prove these conjectures for n = 2.Comment: with an appendix by Nicholas M. Katz; 57 pages. revised version: New

Deregulation of the telomerase reverse transcriptase (TERT) gene by chromosomal translocations in B-cell malignancies
Sequence variants at the TERT-CLPTM1L locus in chromosome 5p have been recently associated with disposition for various cancers. Here we show that this locus including the gene encoding the telomerase reverse-transcriptase TERT at 5p13.33 is rarely but recurrently targeted by somatic chromosomal translocations to IGH and non-IG loci in B-cell neoplasms, including acute lymphoblastic leukemia, chronic lymphocytic leukemia, mantle cell lymphoma and splenic marginal zone lymphoma. In addition, cases with genomic amplification of TERT locus were identified. Tumors bearing chromosomal aberrations involving TERT showed higher TERT transcriptional expression and increased telomerase activity. These data suggest that deregulation of TERT gene by chromosomal abnormalities leading to increased telomerase activity might contribute to B-cell lymphomagenesis
GeneChip analyses point to novel pathogenetic mechanisms in mantle cell lymphoma
The translocation t(11;14)(q13;q32) is the genetic hallmark of mantle cell lymphoma (MCL) but is not sufficient for inducing lymphomagenesis. Here we performed genome-wide 100K GeneChip Mapping in 26 t(11;14)-positive MCL and six MCL cell lines. Partial uniparental disomy (pUPD) was shown to be a recurrent chromosomal event not only in MCL cell lines but also in primary MCL. Remarkably, pUPD affected recurrent targets of deletion like 11q, 13q and 17p. Moreover, we identified 12 novel regions of recurrent gain and loss as well as 12 high-level amplifications and eight homozygously deleted regions hitherto undescribed in MCL. Interestingly, GeneChip analyses identified different genes, encoding proteins involved in microtubule dynamics, such as MAP2, MAP6 and TP53, as targets for chromosomal aberration in MCL. Further investigation, including mutation analyses, fluorescence in situ hybridisation as well as epigenetic and expression studies, revealed additional aberrations frequently affecting these genes. In total, 19 of 20 MCL cases, which were subjected to genetic and epigenetic analyses, and five of six MCL cell lines harboured at least one aberration in MAP2, MAP6 or TP53. These findings provide evidence that alterations of microtubule dynamics might be one of the critical events in MCL lymphomagenesis contributing to chromosomal instability
Genetic Relationships of Crown Rust Resistance, Grain Yield, Test Weight, and Seed Weight in Oat
Integrating selection for agronomic performance and quantitative resistance to crown rust, caused by Puccinia coronata Corda var. avenae W.P. Fraser & Ledingham, in oat (Avena sativa L.) requires an understanding of their genetic relationships. This study was conducted to investigate the genetic relationships of crown rust resistance, grain yield, test weight, and seed weight under both inoculated and fungicide-treated conditions. A Design II mating was performed between 10 oat lines with putative partial resistance to crown rust and nine lines with superior grain yield and grain quality potential. Progenies from this mating were evaluated in both crown rust-inoculated and fungicide-treated plots in four Iowa environments to estimate genetic effects and phenotypic correlations between crown rust resistance and grain yield, seed weight, and test weight under either infection or fungicide-treated conditions. Lines from a random-mated population derived from the same parents were evaluated in three Iowa environments to estimate heritabilities of, and genetic correlations between, these traits. Resistance to crown rust, as measured by area under the disease progress curve (AUDPC), was highly heritable (H = 0.89 on an entry-mean basis), and was favorably correlated with grain yield, seed weight, and test weight measured in crown rust-inoculated plots. AUDPC was unfavorably correlated or uncorrelated with grain yield, test weight, and seed weight measured in fungicide-treated plots. To improve simultaneously crown rust resistance, grain yield, and seed weight under both lower and higher levels of crown rust infection, an optimum selection index can be developed with the genetic parameters estimated in this stud
Search for R-Parity Violating Decays of Scalar Fermions at LEP
A search for pair-produced scalar fermions under the assumption that R-parity
is not conserved has been performed using data collected with the OPAL detector
at LEP. The data samples analysed correspond to an integrated luminosity of
about 610 pb-1 collected at centre-of-mass energies of sqrt(s) 189-209 GeV. An
important consequence of R-parity violation is that the lightest supersymmetric
particle is expected to be unstable. Searches of R-parity violating decays of
charged sleptons, sneutrinos and squarks have been performed under the
assumptions that the lightest supersymmetric particle decays promptly and that
only one of the R-parity violating couplings is dominant for each of the decay
modes considered. Such processes would yield final states consisting of
leptons, jets, or both with or without missing energy. No significant
single-like excess of events has been observed with respect to the Standard
Model expectations. Limits on the production cross- section of scalar fermions
in R-parity violating scenarios are obtained. Constraints on the supersymmetric
particle masses are also presented in an R-parity violating framework analogous
to the Constrained Minimal Supersymmetric Standard Model.Comment: 51 pages, 24 figures, Submitted to Eur. Phys. J.
TFAP2B influences the effect of dietary fat on weight loss under energy restriction
BACKGROUND:
Numerous gene loci are related to single measures of body weight and shape. We investigated if 55 SNPs previously associated with BMI or waist measures, modify the effects of fat intake on weight loss and waist reduction under energy restriction.
METHODS AND FINDINGS:
Randomized controlled trial of 771 obese adults. (Registration: ISRCTN25867281.) One SNP was selected for replication in another weight loss intervention study of 934 obese adults. The original trial was a 10-week 600 kcal/d energy-deficient diet with energy percentage from fat (fat%) in range of 20-25 or 40-45. The replication study used an 8-weeks diet of 880 kcal/d and 20 fat%; change in fat% intake was used for estimation of interaction effects. The main outcomes were intervention weight loss and waist reduction. In the trial, mean change in fat% intake was -12/+4 in the low/high-fat groups. In the replication study, it was -23/-12 among those reducing fat% more/less than the median. TFAP2B-rs987237 genotype AA was associated with 1.0 kg (95% CI, 0.4; 1.6) greater weight loss on the low-fat, and GG genotype with 2.6 kg (1.1; 4.1) greater weight loss on the high-fat (interaction p-value; p = 0.00007). The replication study showed a similar (non-significant) interaction pattern. Waist reduction results generally were similar. Study-strengths include (i) the discovery study randomised trial design combined with the replication opportunity (ii) the strict dietary intake control in both studies (iii) the large sample sizes of both studies. Limitations are (i) the low minor allele frequency of the TFAP2B polymorphism, making it hard to investigate non-additive genetic effects (ii) the different interventions preventing identical replication-discovery study designs (iii) some missing data for non-completers and dietary intake. No adverse effects/outcomes or side-effects were observed.
CONCLUSIONS:
Under energy restriction, TFAP2B may modify the effect of dietary fat intake on weight loss and waist reduction

Measurement of the Hadronic Photon Structure Function F_2^gamma at LEP2
The hadronic structure function of the photon F_2^gamma is measured as a
function of Bjorken x and of the factorisation scale Q^2 using data taken by
the OPAL detector at LEP. Previous OPAL measurements of the x dependence of
F_2^gamma are extended to an average Q^2 of 767 GeV^2. The Q^2 evolution of
F_2^gamma is studied for average Q^2 between 11.9 and 1051 GeV^2. As predicted
by QCD, the data show positive scaling violations in F_2^gamma. Several
parameterisations of F_2^gamma are in agreement with the measurements whereas
the quark-parton model prediction fails to describe the data.Comment: 4 pages, 2 figures, to appear in the proceedings of Photon 2001,

A measurement of the tau mass and the first CPT test with tau leptons
We measure the mass of the tau lepton to be 1775.1+-1.6(stat)+-1.0(syst.) MeV
using tau pairs from Z0 decays. To test CPT invariance we compare the masses of
the positively and negatively charged tau leptons. The relative mass difference
is found to be smaller than 3.0 10^-3 at the 90% confidence level.Comment: 10 pages, 4 figures, Submitted to Phys. Letts.
Measurement of the B0 Lifetime and Oscillation Frequency using B0->D*+l-v decays
The lifetime and oscillation frequency of the B0 meson has been measured
using B0->D*+l-v decays recorded on the Z0 peak with the OPAL detector at LEP.
The D*+ -> D0pi+ decays were reconstructed using an inclusive technique and the
production flavour of the B0 mesons was determined using a combination of tags
from the rest of the event. The results t_B0 = 1.541 +- 0.028 +- 0.023 ps, Dm_d
= 0.497 +- 0.024 +- 0.025 ps-1 were obtained, where in each case the first
error is statistical and the second systematic.Comment: 17 pages, 4 figures, submitted to Phys. Lett.
