    Circumpolar Diversity and Geographic Differentiation of mtDNA in the Critically Endangered Antarctic Blue Whale (Balaenoptera musculus intermedia)

The published article is copyrighted by the Public Library of Science and can be found at: http://www.plosone.org/home.action.The Antarctic blue whale (Balaenoptera musculus intermedia) was hunted to near extinction between 1904 and 1972, declining from an estimated initial abundance of more than 250,000 to fewer than 400. Here, we describe mtDNA control region diversity and geographic differentiation in the surviving population of the Antarctic blue whale, using 218 biopsy samples collected under the auspices of the International Whaling Commission (IWC) during research cruises from 1990-2009. Microsatellite genotypes and mtDNA sequences identified 166 individuals among the 218 samples and documented movement of a small number of individuals, including a female that traveled at least 6,650 km or 131 degrees longitude over four years. mtDNA sequences from the 166 individuals were aligned with published sequences from 17 additional individuals, resolving 52 unique haplotypes from a consensus length of 410 bp. From this minimum census, a rarefaction analysis predicted that only 72 haplotypes (95% CL, 64, 86) have survived in the contemporary population of Antarctic blue whales. However, haplotype diversity was relatively high (0.968 +/- 0.004), perhaps as a result of the longevity of blue whales and the relatively recent timing of the bottleneck. Despite the potential for circumpolar dispersal, we found significant differentiation in mtDNA diversity (F-ST = 0.032, p<0.005) and microsatellite alleles (F-ST = 0.005, p<0.05) among the six Antarctic Areas historically used by the IWC for management of blue whales

    Endothelial cell cytosolic free calcium regulates neutrophil migration across monolayers of endothelial cells

    Polymorphonuclear leukocytes (PMN) traverse an endothelial cell (EC) barrier by crawling between neighboring EC. Whether EC regulate the integrity of their intercellular adhesive and junctional contacts in response to chemotaxing PMN is unresolved. EC respond to the binding of soluble mediators such as histamine by increasing their cytosolic free calcium concentration ([Ca++]i) (Rotrosen, D., and J.I. Gallin. 1986. J. Cell Biol. 103:2379-2387) and undergoing shape changes (Majno, G., S. M. Shea, and M. Leventhal. 1969. J. Cell Biol. 42:617-672). Substances such as leukotriene C4 (LTC4) and thrombin, which increased the permeability of EC monolayers to ions, as measured by the electrical resistance of the monolayers, transiently increased EC [Ca++]i. To determine whether chemotaxing PMN cause similar changes in EC [Ca++]i, human umbilical vein endothelial cells (HUVEC) maintained as monolayers were loaded with fura-2. [Ca++]i was measured in single EC during PMN adhesion to and migration across these monolayers. PMN-EC adhesion and transendothelial PMN migration in response to formyl-methionyl-leucyl-phenylalanine (fMLP) as well as to interleukin 1 (IL-1) treated EC induced a transient increase in EC [Ca++]i which temporally corresponded with the time course of PMN-EC interactions. When EC [Ca++]i was clamped at resting levels with a cell permeant calcium buffer, PMN migration across EC monolayers and PMN induced changes in EC monolayer permeability were inhibited. However, clamping of EC [Ca++]i did not inhibit PMN-EC adhesion. These studies provide evidence that EC respond to stimulated PMN by increasing their [Ca++]i and that this increase in [Ca++]i causes an increase in EC monolayer permeability. Such [Ca++]i increases are required for PMN transit across an EC barrier. We suggest EC [Ca++]i regulates transendothelial migration of PMN by participating in a signal cascade which stimulates EC to open their intercellular junctions to allow transendothelial passage of leukocytes

    Inpatient Massage Therapy Versus Music Therapy Versus Usual Care: A Mixed-methods Feasibility Randomized Controlled Trial.

    BACKGROUND: Little is known about the feasibility of providing massage or music therapy to medical inpatients at urban safety-net hospitals or the impact these treatments may have on patient experience. OBJECTIVE: To determine the feasibility of providing massage and music therapy to medical inpatients and to assess the impact of these interventions on patient experience. DESIGN: Single-center 3-arm feasibility randomized controlled trial. SETTING: Urban academic safety-net hospital. PATIENTS: Adult inpatients on the Family Medicine ward. INTERVENTIONS: Massage therapy consisted of a standardized protocol adapted from a previous perioperative study. Music therapy involved a preference assessment, personalized compact disc, music-facilitated coping, singing/playing music, and/or songwriting. Credentialed therapists provided the interventions. MEASUREMENTS: Patient experience was measured with the Hospital Consumer Assessment of Healthcare Providers and Systems (HCAHPS) within 7 days of discharge. We compared the proportion of patients in each study arm reporting "top box" scores for the following a priori HCAHPS domains: pain management, recommendation of hospital, and overall hospital rating. Responses to additional open-ended postdischarge questions were transcribed, coded independently, and analyzed for common themes. RESULTS: From July to December 2014, 90 medical inpatients were enrolled; postdischarge data were collected on 68 (76%) medical inpatients. Participants were 70% females, 43% non-Hispanic black, and 23% Hispanic. No differences between groups were observed on HCAHPS. The qualitative analysis found that massage and music therapy were associated with improved overall hospital experience, pain management, and connectedness to the massage or music therapist. CONCLUSIONS: Providing music and massage therapy in an urban safety-net inpatient setting was feasible. There was no quantitative impact on HCAHPS. Qualitative findings suggest benefits related to an improved hospital experience, pain management, and connectedness to the massage or music therapist

    Unexpected patterns of global population structure in melon-headed whales Peponocephala electra

    Foraging specialization, environmental barriers, and social structure have driven the development of strong genetic differentiation within many marine species, including most of the large dolphin species commonly referred to as ‘blackfish’ (subfamily Globicephalinae). We used mitochondrial sequence data (mtDNA) and genotypes from 14 nuclear microsatellite loci (nDNA) to examine patterns of genetic population structure in melon-headed whales Peponocephala electra (MHWs), poorly known members of the blackfish family for which genetic structuring is unknown. MHWs are globally distributed in tropical and subtropical waters, and have formed resident populations around oceanic islands. They frequently mass strand, suggesting strong social cohesion within groups. Based on these characteristics, we hypothesized that MHWs would exhibit strong regional genetic differentiation, similar to that observed in other members of the Globicephalinae subfamily. Instead we found only moderate differentiation (median mtDNA ΩST = 0.204, median nDNA FST = 0.012) among populations both within and between ocean basins. Our results suggest that populations of MHWs that are resident to oceanic islands maintain a higher level of genetic connectivity than is seen in most other blackfish. MHWs may be more behaviorally similar to delphinids from the Delphininae subfamily (particularly the spinner dolphin Stenella longirostris), which are known to form coastal and island-associated resident populations that maintain genetic connectivity either through occasional long-distance dispersal or gene flow with larger pelagic populations.     Galaxy cluster mass bias from projected mass maps: The Three Hundred-NIKA2 LPSZ twin samples

    The determination of the mass of galaxy clusters from observations is subject to systematic uncertainties. Beyond the errors due to instrumental and observational systematic effects, in this work we investigate the bias introduced by modelling assumptions. In particular, we consider the reconstruction of the mass of galaxy clusters from convergence maps employing spherical mass density models. We make use of The Three Hundred simulations, selecting clusters in the same redshift and mass range as the NIKA2 Sunyaev-Zel'dovich Large Program sample: 3≀M500/1014M⊙≀103 \leq M_{500}/ 10^{14} \mathrm{M}_{\odot} \leq 10 and 0.5≀z≀0.90.5 \leq z \leq 0.9. We study different modelling and intrinsic uncertainties that should be accounted for when using the single cluster mass estimates for scaling relations. We confirm that the orientation of clusters and the radial ranges considered for the fit have an important impact on the mass bias. The effect of the projection adds uncertainties to the order of 10%10\% to 14%14\% to the mass estimates. We also find that the scatter from cluster to cluster in the mass bias when using spherical mass models is less than 20%20\% of the true mass of the clusters

    Home-based music therapy - a systematic overview of settings and conditions for an innovative service in healthcare

    <p>Abstract</p> <p>Background</p> <p>Almost every Western healthcare system is changing to make their services more centered around out-patient care. In particular, long-term or geriatric patients who have been discharged from the hospital often require home-based care and therapy. Therefore, several programs have been developed to continue the therapeutic process and manage the special needs of patients after discharge from hospital. Music therapy has also moved into this field of healthcare service by providing home-based music therapy (HBMT) programs. This article reviews and summarizes the settings and conditions of HBMT for the first time.</p> <p>Methods</p> <p>The following databases were used to find articles on home-based music therapy: AMED, CAIRSS, EMBASE, MEDLINE, PsychINFO, and PSYNDEX. The search terms were "home-based music therapy" and "mobile music therapy". Included articles were analyzed with respect to participants as well as conditions and settings of HBMT. Furthermore, the date of publication, main outcomes, and the design and quality of the studies were investigated.</p> <p>Results</p> <p>A total of 20 international publications, 11 clinical studies and nine reports from practice, mainly from the United States (n = 8), were finally included in the qualitative synthesis. Six studies had a randomized controlled design and included a total of 507 patients. The vast majority of clients of HBMT are elderly patients living at home and people who need hospice and palliative care. Although settings were heterogeneous, music listening programs played a predominant role with the aim to reduce symptoms like depression and pain, or to improve quality of life and the relationship between patients and caregivers as primary endpoints.</p> <p>Conclusions</p> <p>We were able to show that HBMT is an innovative service for future healthcare delivery. It fits with the changing healthcare system and its conditions but also meets the therapeutic needs of the increasing number of elderly and severely impaired people. Apart from music therapists, patients and their families HBMT is also interesting as a blueprint for home based care for other groups of caregivers.</p
