    Massive encapsulation of larval Anguillicoloides crassus in the intestinal wall of Japanese eels

    <p>Abstract</p> <p>Background</p> <p>Within the last 25 years, after the introduction of the swimbladder nematode <it>Anguillicoloides </it>crassus from East-Asia to Europe, a body of work has aggregated on the host parasite interactions in the acquired host <it>Anguilla anguilla</it>. Despite the emerging evolutionary interest there is still a lack of knowledge about host parasite relations of <it>A. crassus </it>in its natural host <it>Anguilla japonica</it>. We examined the <it>Anguillicoloides </it>infections of wild-caught Japanese eels as well as from aquacultured specimens in Taiwan with respect to the fate of migratory L3 larvae and performed infection experiments with Japanese eels.</p> <p>Results</p> <p>Inside the intestinal wall of cultured eels, where the infective pressure was higher than among wild eels, we found large numbers of granuloma-like cysts. In a few eels these cysts contained nematodes still recognizable as L3 larvae of <it>A. crassus</it>, while in most cases the content of these capsules was degraded to amorphous matter. Occurrence of these objects was correlated with the number of encapsulated larvae in the swimbladder wall. We were able to show, that the cysts contained disintegrated L3 larvae by amplification and subsequent sequencing of large subunit ribosomal rRNA. Furthermore we identified repeated infections with high doses of larvae as prerequisites for the processes of encapsulation in infection experiments.</p> <p>Conclusion</p> <p>Under high infective pressure a large percentage of L3 larvae of <it>A. crassus </it>coming from the gut lumen are eliminated by the natural host within its intestinal tissue. It is possible to reproduce this condition in infection experiments. We provide a fast, easy and reliable PCR-based method for identification of encapsulated swimbladder parasites.</p

    Evolutionary divergence of the swim bladder nematode Anguillicola crassus after colonization of a novel host, Anguilla anguilla

    BACKGROUND: Anguillicola crassus, a swim bladder nematode naturally parasitizing the Japanese eel, was introduced about 30 years ago from East Asia into Europe where it colonized almost all populations of the European eel. We conducted a common garden experiment under a reciprocal transfer design infecting both European and Japanese eels with populations of A. crassus from Germany, Poland and Taiwan. We tested, whether differences in infectivity, developmental dynamics and reproductive output between the European and Asian parasite populations occur while harboured in the specimens of native and colonized eel host, and if these differences are genetically based or are plastic responses to the new environment. RESULTS: Under common garden conditions an evolutionary change in the both European parasite populations of A. crassus compared with their Taiwanese conspecifics was observed for infectivity and developmental dynamics, but not for reproductive output. When infecting the European eel, current European populations of the parasite were less infective and developed faster than their Taiwanese conspecifics. In the reciprocally infected Japanese eel the genetically induced differences between the parasite strains were less apparent than in the European eel but higher infectivity, faster development and higher larval mortality of the European parasite populations could be inferred. CONCLUSIONS: The differences in infectivity and developmental dynamics between European and Taiwanese populations of A. crassus found in our study suggest rapid genetic divergence of this parasite after a successful host switch in Europe

    A Facile Route to Construct SiCO Nanospheres with Tunable Sizes

The thermal degradation of the F127/ PVSZ mixed micelles leads to the formation of SiCO nanospheres. The size of the resultant SiCO nanospheres can be tuned in the range from 25 nm to 75 nm by controlling annealing time at 70 degrees C

    DFT Study of Oxygen Reduction Reaction on Os/Pt Core−Shell Catalysts Validated by Electrochemical Experiment

    Proton exchange membrane fuel cells (PEMFCs) have attracted much attention as an alternative source of energy with a number of advantages, including high efficiency, sustainability, and environmentally friendly operation. However, the low kinetics of the oxygen reduction reaction (ORR) restricts the performance of PEMFCs. Various types of catalysts have been developed to improve the ORR efficiency, but this problem still needs further investigations and improvements. In this paper, we propose advanced Os/Pt core–shell catalysts based on our previous study on segregation of both bare surfaces and surfaces exposed to ORR adsorbates, and we evaluate the catalytic activity of the proposed materials by density functional theory (DFT). Quantum mechanics was applied to calculate binding energies of ORR species and reaction energy barriers on Os/Pt core–shell catalysts. Our calculations predict a much better catalytic activity of the Os/Pt system than that of pure Pt. We find that the ligand effect of the Os substrate is more important than the lattice compression strain effect. To validate our DFT prediction, we demonstrate the fabrication of Os/Pt core–shell nanoparticles using the underpotential deposition (UPD) technique and succeeding galvanic displacement reaction between the Pt ions and Cu-coated Os nanoparticles. The Os/Pt/C samples were evaluated for electrocatalytic activities toward the ORR in acidic electrolytes. The samples with two consecutive UPD-displacement reaction cycles show 3.5 to 5 times better ORR activities as compared to those of commercially available Pt/C. Our results show good agreement between the computational predictions and electrochemical experimental data for the Os/Pt core–shell ORR catalysts

    Arsenic and Fluoride Exposure in Drinking Water: Children’s IQ and Growth in Shanyin County, Shanxi Province, China

    BACKGROUND: Recently, in a cross-sectional study of 201 children in Araihazar, Bangladesh, exposure to arsenic (As) in drinking water has been shown to lower the scores on tests that measure children’s intellectual function before and after adjustment for sociodemographic features. OBJECTIVES: We investigated the effects of As and fluoride exposure on children’s intelligence and growth. METHODS: We report the results of a study of 720 children between 8 and 12 years of age in rural villages in Shanyin county, Shanxi province, China. The children were exposed to As at concentrations of 142 ± 106 μg/L (medium-As group) and 190 ± 183 μg/L (high-As group) in drinking water compared with the control group that was exposed to low concentrations of As (2 ± 3 μg/L) and low concentrations of fluoride (0.5 ± 0.2 mg/L). A study group of children exposed to high concentrations of fluoride (8.3 ± 1.9 mg/L) but low concentrations of As (3 ± 3 μg/L) was also included because of the common occurrence of elevated concentrations of fluoride in groundwater in our study area. A standardized IQ (intelligence quotient) test was modified for children in rural China and was based on the classic Raven’s test used to determine the effects of these exposures on children’s intelligence. A standardized measurement procedure for weight, height, chest circumference, and lung capacity was used to determine the effects of these exposures on children’s growth. RESULTS: The mean IQ scores decreased from 105 ± 15 for the control group, to 101 ± 16 for the medium-As group (p < 0.05), and to 95 ± 17 for the high-As group (p < 0.01). The mean IQ score for the high-fluoride group was 101 ± 16 and significantly different from that of the control group (p < 0.05). Children in the control group were taller than those in the high-fluoride group (p < 0.05); weighed more than the those in the high-As group (p < 0.05); and had higher lung capacity than those in the medium-As group (p < 0.05). CONCLUSIONS: Children’s intelligence and growth can be affected by high concentrations of As or fluoride. The IQ scores of the children in the high-As group were the lowest among the four groups we investigated. It is more significant that high concentrations of As affect children’s intelligence. It indicates that arsenic exposure can affect children’s intelligence and growth

    Sympatric Spawning but Allopatric Distribution of Anguilla japonica and Anguilla marmorata: Temperature- and Oceanic Current-Dependent Sieving

    Anguilla japonica and Anguilla marmorata share overlapping spawning sites, similar drifting routes, and comparable larval durations. However, they exhibit allopatric geographical distributions in East Asia. To clarify this ecological discrepancy, glass eels from estuaries in Taiwan, the Philippines, Indonesia, and China were collected monthly, and the survival rate of A. marmorata under varying water salinities and temperatures was examined. The composition ratio of these 2 eel species showed a significant latitude cline, matching the 24°C sea surface temperature isotherm in winter. Both species had opposing temperature preferences for recruitment. A. marmorata prefer high water temperatures and die at low water temperatures. In contrast, A. japonica can endure low water temperatures, but their recruitment is inhibited by high water temperatures. Thus, A. japonica glass eels, which mainly spawn in summer, are preferably recruited to Taiwan, China, Korea, and Japan by the Kuroshio and its branch waters in winter. Meanwhile, A. marmorata glass eels, which spawn throughout the year, are mostly screened out in East Asia in areas with low-temperature coastal waters in winter. During summer, the strong northward currents from the South China Sea and Changjiang River discharge markedly block the Kuroshio invasion and thus restrict the approach of A. marmorata glass eels to the coasts of China and Korea. The differences in the preferences of the recruitment temperature for glass eels combined with the availability of oceanic currents shape the real geographic distribution of Anguilla japonica and Anguilla marmorata, making them “temperate” and “tropical” eels, respectively
