    Interplay of cell proliferation and tissue remodelling in colorectal cancer

    Human cancers can be classified based on gene expression signatures quantifying the degree of cell proliferation and a feature we term tissue remodelling. Yet, specific factors regulating the gene expression programs of cell proliferation and tissue remodelling are not well understood. In this work, I address this question using colorectal cancer as a case study. In an initial evaluation of clinical outcome prediction in the case study cohort, our classification based on cell proliferation and tissue remodelling signatures delivers a better patient stratification than a recently reported consensus molecular subtyping (CMS). Although the CMS scheme predicts a worse prognosis for patients with a mesenchymal signature (a feature that strongly overlaps with tissue remodelling), it cannot differentiate the remaining patients based on clinical outcome. I show that cell proliferation is the missing factor, which is associated with good prognosis. In the analyses of specific factors driving the tissue remodelling programme, I identify transcription factor KLF4 and microRNA mir-22 as putative regulators. Concordant with KLF4 role in immune cell regulation, KLF4 activity scores are significantly higher in colorectal tumours with predicted myeloid cells infiltration, suggesting its association with myeloid cell infiltration and poor prognosis in colorectal cancer. However, mir-22 is not associated with immune cell infiltration, suggesting it may be expressed in the tumour stroma as well. Lastly, I propose a network that may be regulated by IRF8, SPI1, KLF4 and mir-22 in monocyte differentiation and in colorectal cancer. The correlation analysis suggests a negative feedback loop, whereby mir-22 may repress Irf8 gene to control the rate of IRF8 transcription. Taken together, I have identified specific regulators driving remodelling processes and their association with immune cell infiltration. In the future, it will be beneficial to conduct further studies to understand the mechanisms driving myeloid cell infiltration in colorectal cancer, particularly to propose treatments for patients exhibiting poor prognosis

So that an alternative learning media is needed in order to improve understanding of the concept of chemical objects interactively. Machine learning technology is expected to provide a more realistic interaction and is an advancement of a promising technological method that can motivate users to engage in a more active learning system

    Consumer’s Response on E-mail advertisements from Tour and Travel Agency in Indonesia

    This study aims to determine and analyze consumer response to email advertising from online tour and travel agency in Indonesia. This research was a causal research with quantitative approach. This study used purposive sampling approach with an assessment of the researchers who can be made respondents in accordance with the characteristics of the population. The characteristics of the population were consumers who bought and used online tour and travel agency twice in the past year, lived in Indonesia, and the minimum of level education was bachelor. The number of respondents in this study were up to 125 people. The analysis in this study used a model of SEM (Structural Equation Modeling) through SPSS software version 22.0 for Windows and AMOS version 21.0 for Windows for testing Measurement and Structural Model. The result of this study indicated that with the intention of consumers to buy, build relationship, and trust with good service then consumers will read, open, take advantage of email advertising from tour and travel agencies in Indonesia and even consumers want to get email ads in the future

    In order to create machine learning, we need to build a model. The model is created from a process called training. The goal of training is to develop an accurate model that answers some questions and in order to train a model, we need to collect a dataset. The quality and quantity of the data gathered will determine how good the predictive model can be. Helping the model to understand datasets like humans do is one of the important processes of machine learning. Datasets need to be constructed and transformed correctly. In this research, we compare the difference between creating a COCO dataset manually and creating a synthetic COCO dataset. Creating datasets for chemistry apparatus is not as difficult as creating a human object. The apparatus has a specific shape and form, thus the dataset had to have a limited number. As a result, we create both a dataset both manually and synthetically. The synthetic dataset helps to gain more datasets by combining some objects with different backgrounds

    Antibacterial Effect Of Curcuma Domestica Ethanolic Extract On Group A Streptococcus Î’ Hemoyticus

    Introduction : Pharyngitis is one of the most common infection that general practitioners handled. It is estimated in one year there are 15 million people with pharyngitis will come to the doctor. American Society of Microbiology said that 94,3 % from 402 patient who were suspected pharyngitis received antibiotic therapy even without indication of antibiotic therapy. Antibiotic resistance is becoming a great threat for the world today. World Health Organization state that one of the plan to fight antibiotic resistance is to research and develop new drugs. Curcuma domestica is a plant that is widely used in Indonesia and some research said that it has anti-inflammatory, antioxidant, and antimicrobial effect. Aim: The aim of this study is to find the antibacterial effect of Curcuma domestica ethanolic extract on Group A Streptococcus β hemolyticus and find the Minimum Bactericidal Concentration (MBC) value. Methods : This was in vitro experimental study with broth microdilution method and inoculation on agar blood media to find the MBC. Treatment group consisting of, media (Mueller Hinton Blood Broth), Group A Streptococcus β hemolyticus bacteria, and 5 concentrations (312.5 μg/ml, 625 μg/ml, 1250 μg/ml, 2500 μg/ml, 5000 μg/ml) of Curcuma domestica extract. Result : There is no bacterial growth at 5000 μg/ml concentration on solid media, but there are less bacterial growth at 2500 μg/ml than at concentration 312.5 μg/ml, 625 μg/ml, 1250 μg/ml. Conclusion: There is an antibacterial effect of Curcuma domestica ethanolic extract on Group A Streptococcus β hemolyticus with MBC value in range 2500 – 5000 μg/ml

    The Comparison Of Antibacterial Effects On Cinnamomum Burmannii Water Extract With Penicillin Against Staphylococcus Aureus In Vitro

    Introduction: Along with the development of science, many varieties of microorganisms are resistant to antibiotics. To avoid cases of resistance grew severe, researchers developed alternative medicines as a substitute for antibiotics. One of the ingredients of herbs researched that has antibacterial activity is Cinnamon (Cinnamomum burmannii). Aim: This study aimed to compare antibacterial effects water extract of Cinnamomum burmannii with penicillin against Staphylococcus aureus Methods: This an experimental studies witd non-equivalent control group design. Research using the bacteria S. aureus are divided into two groups, the control group and experimental group. The concentration of Cinnamomum burmannii and penicillin are used that is 80- 150mg/L. Antibacterial test used dilution method and a microplate reader for reading absorbance. This research conducted in the laboratory of Microbiology, Faculty of Pharmacy Widya Mandala Catholic University Surabaya for 2 weeks. Result: Based on visual observations, Cinnamomum burmannii’s MIC is located at concentration 320- 600 mg/L while penicillin’s MIC is located at concentration 160- 300 mg/L. Based on the results of microdilution test, Cinnamomum burmannii’s MIC is located at concentration 160- 300 mg/L while penicillin’s MIC is located at concentration 640- 1200 mg/L. Conclusion: The conclusion of this research is giving Cinnamomum burmannii water extract in Staphylococcus aureus has efficacy or inhibitory that is not significantly different with penicillin

    ABSTRACT Garbage is the rest of human daily activities or natural processes in solid form. In general, the problem of waste is related to how to manage waste properly in the community and how to reduce waste generation and utilize all the potential generated. One of the right ways to manage waste from waste generation sources is the waste bank program. This study aims to examine the management system of the Dream Waste Bank and the characteristics of the Dream Waste Bank which is managed in the BCL 5 Housing in Jambi City. Research method with qualitative descriptive. The units analyzed were all residents' houses that became customers, namely 29 households and 8 informants. The results of the study found that the management of the Dream Waste Bank based on PERMEN LH No. 13 of 2012 has taken place as it should based on the standardization of the waste bank, the working mechanism of the waste bank, the advantages of the waste bank system, the implementation of the waste bank system, the stage of utilizing inorganic waste and the development of a waste bank. Meanwhile, the characteristics of the waste bank obtained in the study are the volume of inorganic waste, the composition of inorganic waste that enters the waste bank and the generation/amount of waste collected each month and the reduction in waste generation.   Keywords : Waste Bank, Waste Management, Characteristics   ABSTRAK Sampah merupakan sisa kegiatan sehari-hari manusia atau proses alam yang berbentuk padat. Secara umum permasalahan persampahan berkaitan dengan bagaimana cara pengelolaan sampah yang baik di masyarakat dan bagaimana cara mengurangi timbulan sampah serta memanfaatkan segala potensi yang mungkin dihasilkan. Salah satu cara yang tepat untuk pengelolaan sampah dari sumber timbulan sampah yaitu dengan program bank sampah. Penelitian ini bertujuan untuk mengkaji sistem pengelolaan Bank Sampah Dream dan karakteristik Bank Sampah Dream yang dikelola di Perumahan BCL 5 Kota Jambi. Metode Penelitian dengan deskriptif kualitatif. Unit yang dianalisis yaitu seluruh rumah warga yang menjadi nasabah yakni 29 rumah tangga dan 8 orang informan yang terbagi menjadi 2 informan utama dari pengurus Bank Sampah Dream dan 6 informan pendukung dari warga yang menjadi nasabah. Hasil Penelitian didapatkan pada pengelolaan Bank Sampah Dream berdasarkan Peraturan Menteri Lingkungan Hidup No.13 Tahun 2012 sudah berlangsung sebagaimana mestinya berdasarkan standarisasi bank sampah, mekanisme kerja bank sampah, keuntungan sistem bank sampah, pelaksanaan sistem bank sampah, tahap pemanfaatan sampah anorganik yang terkumpul dan pengembangan bank sampah. Sementara karakteristik bank sampah yang diperoleh dalam penelitian adalah volume sampah anorganik, komposisi sampah anorganik yang masuk ke bank sampah dan timbulan/jumlah sampah tiap bulan yang terkumpul serta adanya pengurangan timbulan sampah.   Kata kunci: Bank Sampah, Pengelolaan Sampah, Karakteristi
