    Parental investment and child health in a Yanomamö village suffering short-term food stress

    The 1998 El Niño significantly reduced garden productivity in the Upper Orinoco region in Venezuela. Consequently, parents were forced to allocate food carefully to their children. Nutrition data collected from village children combined with genealogical data allowed the determination of which children suffered most, and whether the patterns of food distribution accorded with predictions from parental investment theory. For boys, three social variables accounted for over 70% of the variance in subcutaneous fat after controlling for age: number of siblings, age of the mother’s youngest child, and whether the mother was the senior or junior co-wife, or was married monogamously. These results accord well with parental investment theory. Parents experiencing food stress faced a trade-off between quantity and quality, and between investing in younger versus older offspring. In addition, boys with access to more paternal investment (i.e. no stepmother) were better nourished. These variables did not account for any of the variance in female nutrition. Girls’ nutrition was associated with the size of their patrilineage and the number of non-relatives in the village, suggesting that lineage politics may have played a role. An apparent lack of relationship between orphan status and nutrition is also interesting, given that orphans suffered high rates of skin flea infections. The large number of orphans being cared for by only two grandparents suggests that grooming time may have been the resource in short supply. Introductio

    The Panchromatic Hubble Andromeda Treasury: Triangulum Extended Region (PHATTER) I. Ultraviolet to Infrared Photometry of 22 Million Stars in M33

    We present panchromatic resolved stellar photometry for 22 million stars in the Local Group dwarf spiral Triangulum (M33), derived from Hubble Space Telescope (HST) observations with the Advanced Camera for Surveys (ACS) in the optical (F475W, F814W), and the Wide Field Camera 3 (WFC3) in the near ultraviolet (F275W, F336W) and near-infrared (F110W, F160W) bands. The large, contiguous survey area covers \sim14 square kpc and extends to 3.5 kpc (14 arcmin, or 1.5-2 scale lengths) from the center of M33. The PHATTER observing strategy and photometry technique closely mimic those of the Panchromatic Hubble Andromeda Treasury (PHAT), but with updated photometry techniques that take full advantage of all overlapping pointings (aligned to within <<5-10 milliarcseconds) and improved treatment of spatially-varying point spread functions. The photometry reaches a completeness-limited depth of F475W\sim28.5 in the lowest surface density regions observed in M33 and F475W\sim26.5 in the most crowded regions found near the center of M33. We find the young populations trace several relatively tight arms, while the old populations show a clear, looser two-armed structure. We present extensive analysis of the data quality including artificial star tests to quantify completeness, photometric uncertainties, and flux biases.     Efficient Colonization and Therapy of Human Hepatocellular Carcinoma (HCC) Using the Oncolytic Vaccinia Virus Strain GLV-1h68

    Virotherapy using oncolytic vaccinia virus strains is one of the most promising new strategies for cancer therapy. In this study, we analyzed for the first time the therapeutic efficacy of the oncolytic vaccinia virus GLV-1h68 in two human hepatocellular carcinoma cell lines HuH7 and PLC/PRF/5 (PLC) in cell culture and in tumor xenograft models. By viral proliferation assays and cell survival tests, we demonstrated that GLV-1h68 efficiently colonized, replicated in, and did lyse these cancer cells in culture. Experiments with HuH7 and PLC xenografts have revealed that a single intravenous injection (i.v.) of mice with GLV-1h68 resulted in a significant reduction of primary tumor sizes compared to uninjected controls. In addition, replication of GLV-1h68 in tumor cells led to strong inflammatory and oncolytic effects resulting in intense infiltration of MHC class II-positive cells like neutrophils, macrophages, B cells and dendritic cells and in up-regulation of 13 pro-inflammatory cytokines. Furthermore, GLV-1h68 infection of PLC tumors inhibited the formation of hemorrhagic structures which occur naturally in PLC tumors. Interestingly, we found a strongly reduced vascular density in infected PLC tumors only, but not in the non-hemorrhagic HuH7 tumor model. These data demonstrate that the GLV-1h68 vaccinia virus may have an enormous potential for treatment of human hepatocellular carcinoma in man

    The effect of dietary nitrate supplementation on the spatial heterogeneity of quadriceps deoxygenation during heavy-intensity cycling.

    This study investigated the influence of dietary inorganic nitrate (NO3(-)) supplementation on pulmonary O2 uptake (V˙O2) and muscle deoxyhemoglobin/myoglobin (i.e. deoxy [Hb + Mb]) kinetics during submaximal cycling exercise. In a randomized, placebo-controlled, cross-over study, eight healthy and physically active male subjects completed two step cycle tests at a work rate equivalent to 50% of the difference between the gas exchange threshold and peak V˙O2 over separate 4-day supplementation periods with NO3(-)-rich (BR; providing 8.4 mmol NO3(-)∙day(-1)) and NO3(-)-depleted (placebo; PLA) beetroot juice. Pulmonary V˙O2 was measured breath-by-breath and time-resolved near-infrared spectroscopy was utilized to quantify absolute deoxy [Hb + Mb] and total [Hb + Mb] within the rectus femoris, vastus lateralis, and vastus medialis There were no significant differences (P > 0.05) in the primary deoxy [Hb + Mb] mean response time or amplitude between the PLA and BR trials at each muscle site. BR significantly increased the mean (three-site) end-exercise deoxy [Hb + Mb] (PLA: 91 ± 9 vs. BR: 95 ± 12 μmol/L, P < 0.05), with a tendency to increase the mean (three-site) area under the curve for total [Hb + Mb] responses (PLA: 3650 ± 1188 vs. BR: 4467 ± 1315 μmol/L sec(-1), P = 0.08). The V˙O2 slow component reduction after BR supplementation (PLA: 0.27 ± 0.07 vs. BR: 0.23 ± 0.08 L min(-1), P = 0.07) correlated inversely with the mean increases in deoxy [Hb + Mb] and total [Hb + Mb] across the three muscle regions (r(2) = 0.62 and 0.66, P < 0.05). Dietary NO3(-) supplementation increased O2 diffusive conductance across locomotor muscles in association with improved V˙O2 dynamics during heavy-intensity cycling transitions

    Mechanism of Oleogel-S10: A triterpene preparation for the treatment of epidermolysis bullosa

    Epidermolysis bullosa (EB) is a group of rare heterogeneous, genetic disorders. Currently, there is no effective pharmacological or genetic therapy for all EB subtypes. Dry extract from birch bark and betulin upregulate some pro-inflammatory mediators and downregulate others. The increase in pro-inflammatory cytokines is temporary and attenuated over long-term treatment. This inflammatory stimulus is thought to be prerequisite for a secondary anti-inflammatory response. Dry extract from birch bark and its active marker substances have also been shown to increase the migration of primary human keratinocytes, accelerate wound closure, and promote differentiation of keratinocytes in vitro and in vivo-processes that are essential for reepithelialization and maintenance of the skin barrier. Comprehensive clinical data are available to support the use of Oleogel-S10 in the treatment of partial thickness wounds of different etiologies, and a proof-of-concept Phase 2 study in patients with dystrophic EB has suggested the potential for faster reepithelialization of wounds treated with Oleogel-S10

    Teaching Practical Machine Learning Concepts to Professionals and Students: An Integrated and Interdisciplinary Qualification Project

    Machine learning (ML) has become highly relevant in applications across all industries, and specialists in the field are sought urgently. As it is a highly interdisciplinary field, requiring knowledge in computer science, statistics and the relevant application domain, experts are hard to find. Large corporations can sweep the job market by offering high salaries, which makes the situation for small and medium enterprises (SME) even worse, as they usually lack the capacities both for attracting specialists and for qualifying their own personnel. In order to meet the enormous demand in ML specialists, universities now teach ML in specifically designed degree programs as well as within established programs in science and engineering. While the teaching almost always uses practical examples, these are somewhat artificial or outdated, as real data from real companies is usually not available. The approach reported in this contribution aims to tackle the above challenges in an integrated course, combining three independent aspects: first, teaching key ML concepts to graduate students from a variety of existing degree programs; second, qualifying working professionals from SME for ML; and third, applying ML to real-world problems faced by those SME. The course was carried out in two trial periods within a government-funded project at a university of applied sciences in south-west Germany. The region is dominated by SME many of which are world leaders in their industries. Participants were students from different graduate programs as well as working professionals from several SME based in the region. The first phase of the course (one semester) consists of the fundamental concepts of ML, such as exploratory data analysis, regression, classification, clustering, and deep learning. In this phase, student participants and working professionals were taught in separate tracks. Students attended regular classes and lab sessions (but were also given access to e-learning materials), whereas the professionals learned exclusively in a flipped classroom scenario: they were given access to e-learning units (video lectures and accompanying quizzes) for preparation, while face-to-face sessions were dominated by lab experiments applying the concepts. Prior to the start of the second phase, participating companies were invited to submit real-world problems that they wanted to solve with the help of ML. The second phase consisted of practical ML projects, each tackling one of the problems and worked on by a mixed team of both students and professionals for the period of one semester. The teams were self-organized in the ways they preferred to work (e.g. remote vs. face-to-face collaboration), but also coached by one of the teaching staff. In several plenary meetings, the teams reported on their status as well as challenges and solutions. In both periods, the course was monitored and extensive surveys were carried out. We report on the findings as well as the lessons learned. For instance, while the program was very well-received, professional participants wished for more detailed coverage of theoretical concepts. A challenge faced by several teams during the second phase was a dropout of student members due to upcoming exams in other subjects

    Microgrid Systems: Towards a Technical Performance Assessment Frame

    A microgrid is an independent power system that can be connected to the grid or operated in an islanded mode. This single grid entity is widely used for furthering access to energy and ensuring reliable energy supply. However, if islanded, microgrids do not benefit from the high inertia of the main grid and can be subject to high variations in terms of voltage and frequency, which challenge their stability. In addition, operability and interoperability requirements, standards as well as directives have addressed main concerns regarding a microgrid’s reliability, use of distributed local resources and cybersecurity. Nevertheless, microgrid systems are quickly evolving through digitalization and have a large range of applications. Thus, a consensus over their testing must be further developed with the current technological development. Here, we describe existing technical requirements and assessment criteria for a microgrid’s main functionalities to foster harmonization of functionality-level testing and an international conception of system-level one. This framework is proposed as a reference document for assessment frame development serving both microgrid research and implementation for a comprehensive understanding of technical microgrid performance and its current assessment challenges, such as lack of standardization and evolving technology
