    Energy Dynamics in the Brain: Contributions of Astrocytes to Metabolism and pH Homeostasis

    Regulation of metabolism is complex and involves enzymes and membrane transporters, which form networks to support energy dynamics. Lactate, as a metabolic intermediate from glucose or glycogen breakdown, appears to play a major role as additional energetic substrate, which is shuttled between glycolytic and oxidative cells, both under hypoxic and normoxic conditions. Transport of lactate across the cell membrane is mediated by monocarboxylate transporters (MCTs) in cotransport with H+, which is a substrate, a signal and a modulator of metabolic processes. MCTs form a "transport metabolon" with carbonic anhydrases (CAs), which not only provide a rapid equilibrium between CO2, HCO3- and H+, but, in addition, enhances lactate transport, as found in Xenopus oocytes, employed as heterologous expression system, as well as in astrocytes and cancer cells. Functional interactions between different CA isoforms and MCTs have been found to be isoform-specific, independent of the enzyme's catalytic activity, and they require physical interaction between the proteins. CAs mediate between different states of metabolic acidosis, induced by glycolysis and oxidative phosphorylation, and play a relay function in coupling pH regulation and metabolism. In the brain, metabolic processes in astrocytes appear to be linked to bicarbonate transport and to neuronal activity. Here, we focus on physiological processes of energy dynamics in astrocytes as well as on the transfer of energetic substrates to neurons

    Effect of sildenafil citrate on the body weight, blood glucose and white blood cell count during wound healing process in diabetic rats

    Diabetes mellitus is a chronic metabolic disorder with major complication of delayed wound healing. Objectives: The purpose of this study was to evaluate the effect of sildenafil on blood glucose level, body weight and leucocytecount during wound healing in diabetic rats. Method: Forty-two Albino rats randomly divided into 7 groups of 6 rats per group were treatedorally with sildenafil (50 mg/kg) for 21 days and/or single dose of intraperitoneal injection of insulin (10 I.U.). Before treatments, diabetes was induced in groups A, B, C and D by a singleintraperitoneal injection of alloxan (130mg/kg) and a square-shaped wound measuring 1.5cm was created under anesthesia on the dorsum of rats in all groups except group G. The rats ingroup A were treated with sildenafil citrate orally at the dose rate of 50 mg/kg body weight for 21 days and the rats in group B were treated with insulin injection of 10 international units once and sildenafil citrate as in Group A. The rats in group C were treated with only insulin injection of 10 international units once and the rats in group D were treated with only distilled water. Groups E and F were non diabetic rats with wounds similar to those of diabetic rats in the previous groups and each rat in group E was also treated with sildenafil citrate as in Group A and those in group F were treated with only distilled water. The rats in Group G were normal rats without wound treated with sildenafil citrate as in Group A. Data collected were analysed using SPSS version 20. Results: The results indicated that sildenafil causes a non significant (P>0.05) decrease in the blood glucose of diabetic rats, a significant (P<0.05) increase in the leucocyte count and a significant decrease(p<0.05) in the rate of percentage decrease in bodyweight. Conclusion: Sildenafil may have the potential of reducing the rate of body weight loss in diabetic rats (with or without wound) receiving insulin treatment

    Evaluation of treatment descriptions and alignment with clinical guidance of apps for depression on app stores : systematic search and content analysis

    Background: The use of apps for the treatment of depression shows great promise. However, there is uncertainty regarding the alignment of publicly available apps for depression with clinical guidance, their treatment fidelity and evidence base, and their overall safety.Objective: Built on previous analyses and reviews, this study aims to explore the treatment and safety issues of publicly available apps for depression.Methods: We conducted a content analysis of apps for depression in the 2 main UK app stores (Google Play and Apple App Store). App store listings were analyzed for intervention content, treatment fidelity, and fit with the National Institute for Health and Care Excellence (NICE) guidelines for the treatment of depression in adults.Results: A total of 353 apps for depression were included in the review. App descriptions reported the use of 20 treatment approaches and 37 treatment strategies. Many apps used transdiagnostic (155/353, 43.9%) and multitheoretical interventions to treat multiple disorders including depression. Although many interventions appeared to be evidence-informed, there were issues with treatment fidelity, research evidence, and fit with clinical guidelines. None of the apps fully aligned with the NICE guidelines for depression.Conclusions: App developers have adopted many evidence-informed treatments in their interventions; however, more work is needed to improve clinical validity, treatment fidelity, and the safety of apps. We urge developers to consult relevant guidelines and standards, and to engage in reflective questioning on treatment and safety to address these issues and to improve treatment content and intervention design

    Electromagnetic wave diffraction by periodic planar metamaterials with nonlinear constituents

    We present a theory which explains how to achieve an enhancement of nonlinear effects in a thin layer of nonlinear medium by involving a planar periodic structure specially designed to bear a trapped-mode resonant regime. In particular, the possibility of a nonlinear thin metamaterial to produce the bistable response at a relatively low input intensity due to a large quality factor of the trapped-mode resonance is shown. Also a simple design of an all-dielectric low-loss silicon-based planar metamaterial which can provide an extremely sharp resonant reflection and transmission is proposed. The designed metamaterial is envisioned for aggregating with a pumped active medium to achieve an enhancement of quantum dots luminescence and to produce an all-dielectric analog of a 'lasing spaser'.Comment: 18 pages, 13 figure

    Hydrogen peroxide bleaching of cellulose pulps obtained from brewer’s spent grain

    Brewer’s spent grain (BSG) was evaluated for bleached pulp production. Two cellulose pulps with different chemical compositionswere produced by soda pulping: one from the original raw material and the other from material pretreated by dilute acid. Both of them were bleached by a totally chlorine-free sequence performed in three stages, using 5% hydrogen peroxide in the two initial, and a 0.25 NNaOHsolution in the last one. Chemical composition, kappa number, viscosity, brightness and yield of bleached and unbleached pulps were evaluated. The high hemicellulose (28.4% w/w) and extractives (5.8% w/w) contents in original BSG affected the pulping and bleaching processes.However, soda pulping of acid pretreated BSG gave a celluloserich pulp (90.4% w/w) with low hemicellulose and extractives contents (7.9% w/w and <3.4% w/w, respectively), which was easily bleached achieving a kappa number of 11.21, viscosity of 3.12 cp, brightness of 71.3%, cellulose content of 95.7% w/w, and residual lignin of 3.4% w/w.     Serious scientific games are games whose purpose is not only fun. In the field of science, the serious goals include crucial activities for scientists: outreach, teaching and research. The number of serious games is increasing rapidly, in particular citizen science games, games that allow people to produce and/or analyze scientific data. Interestingly, it is possible to build a set of rules providing a guideline to create or improve serious games. We present arguments gathered from our own experience ( Phylo , DocMolecules , HiRE-RNA contest and Pangu) as well as examples from the growing literature on scientific serious games

    Observation of bright polariton solitons in a semiconductor microcavity

    Microcavity polaritons are composite half-light half-matter quasi-particles, which have recently been demonstrated to exhibit rich physical properties, such as non-equilibrium Bose-Einstein condensation, parametric scattering and superfluidity. At the same time, polaritons have some important advantages over photons for information processing applications, since their excitonic component leads to weaker diffraction and stronger inter-particle interactions, implying, respectively, tighter localization and lower powers for nonlinear functionality. Here we present the first experimental observations of bright polariton solitons in a strongly coupled semiconductor microcavity. The polariton solitons are shown to be non-diffracting high density wavepackets, that are strongly localised in real space with a corresponding broad spectrum in momentum space. Unlike solitons known in other matter-wave systems such as Bose condensed ultracold atomic gases, they are non-equilibrium and rely on a balance between losses and external pumping. Microcavity polariton solitons are excited on picosecond timescales, and thus have significant benefits for ultrafast switching and transfer of information over their light only counterparts, semiconductor cavity lasers (VCSELs), which have only nanosecond response time
