    Population Structure And Hybridization Of Alaskan Caribou And Reindeer: Integrating Genetics And Local Knowledge

    Thesis (Ph.D.) University of Alaska Fairbanks, 2012Alaskan caribou (Rangifer tarandus granti) are a valued game species and a key grazer in Alaska's terrestrial ecosystem. Caribou herds, defined by female fidelity to calving grounds, are management units. However, the extent to which herds constitute genetic populations is unknown. Historical fluctuations in herd size, range, and distribution suggest periods of contact and isolation between herds. Likewise, historical contact between caribou and introduced domestic reindeer (R.t. tarandus) created opportunities for hybridization, but its extent is not known. I conducted an interdisciplinary study to understand how historical processes influence genetic identity and population structure of caribou and reindeer. Interviews with herders and hunters in Barrow, Alaska, revealed that many reindeer migrated away with caribou in the 1940s despite herder efforts to prevent mixing. Local observations of reindeer-like animals in caribou herds today suggest feral reindeer may survive and interbreed. Using genetic analysis of North Slope caribou and Seward Peninsula reindeer (n = 312) at 19 microsatellite loci, I detected individuals with hybrid ancestry in all four caribou herds and in reindeer. Selective hunting of reindeer-like animals, along with herd size and natural selection, may remove reindeer from caribou herds over time. I used genetics as well to describe caribou population structure and determine how it is influenced by geography, historical demography, and ecotypes. I found that Alaskan caribou from 20 herds (n = 655) are subdivided into two genetic clusters: the Alaska Peninsula and the mainland. Alaska Peninsula herds are genetically distinct, while many mainland herds are not. I hypothesize that Alaska Peninsula herds have diverged due to post-glacial founder effects and recent bottlenecks driven by constraints to population size from marginal habitat and reduced gene flow across a habitat barrier at the nexus of the peninsula. I hypothesize that mainland herds have maintained genetic connectivity and large effective population size via range expansions and shifts over time. However, I find evidence that herds of different ecotypes (migratory, sedentary) can remain differentiated despite range overlap. Genetic evidence provides information for herd-based management, while also demonstrating the importance of spatial connectivity of herds and their habitats over the long-term

    From Earth to Orbit: An assessment of transportation options

    The report assesses the requirements, benefits, technological feasibility, and roles of Earth-to-Orbit transportation systems and options that could be developed in support of future national space programs. Transportation requirements, including those for Mission-to-Planet Earth, Space Station Freedom assembly and operation, human exploration of space, space science missions, and other major civil space missions are examined. These requirements are compared with existing, planned, and potential launch capabilities, including expendable launch vehicles (ELV's), the Space Shuttle, the National Launch System (NLS), and new launch options. In addition, the report examines propulsion systems in the context of various launch vehicles. These include the Advanced Solid Rocket Motor (ASRM), the Redesigned Solid Rocket Motor (RSRM), the Solid Rocket Motor Upgrade (SRMU), the Space Shuttle Main Engine (SSME), the Space Transportation Main Engine (STME), existing expendable launch vehicle engines, and liquid-oxygen/hydrocarbon engines. Consideration is given to systems that have been proposed to accomplish the national interests in relatively cost effective ways, with the recognition that safety and reliability contribute to cost-effectiveness. Related resources, including technology, propulsion test facilities, and manufacturing capabilities are also discussed

    Wall Microstructures of High Aspect Ratio Enabled by Near‐Field Electrospinning

    Near-field electrospinning (NFES) holds the potential to develop into a versatile additive nanomanufacturing platform. However, the impact of a variety of processing variables remains unresolved.Herein, the effect of solvents used to prepare suitable solutions for 3D microstructuring by electrospinning is studied. 3D straight walls of stacked fibers are fabricated using a layer-by-layer fiber deposition approach. The effect of the choice of substrate material is also explored. The results show that a high vapor pressure, and a low dielectric constant of the solvent, as well as a high substrate conductivity facilitate improved stacking of fiber layers. Utilizing these conditions, 3D stacked walls of polyethylene oxide are fabricated, and a maximum aspect ratio of 191.7 ± 52.6, while using a chromium/gold substrate and dichloromethane/methanol as the solvent is achieved

    Improving co-design methods implies that we need to understand those methods, paying attention to not only the effect of method choices on design outcomes, but also how methods affect the people involved in co-design. In this article, we explore participants' experiences from a year-long participatory health service design project to develop ‘Better Outpatient Services for Older People’. The project followed a defined method called experience-based design (EBD), which represented the state of the art in participatory service design within the UK National Health Service. A sample of participants in the project took part in semi-structured interviews reflecting on their involvement in and their feelings about the project. Our findings suggest that the EBD method that we employed was successful in establishing positive working relationships among the different groups of stakeholders (staff, patients, carers, advocates and design researchers), although conflicts remained throughout the project. Participants' experiences highlighted issues of wider relevance in such participatory design: cost versus benefit, sense of project momentum, locus of control, and assumptions about how change takes place in a complex environment. We propose tactics for dealing with these issues that inform the future development of techniques in user-centred healthcare design
