Negative regulation of urokinase receptor activity by a GPI-specific phospholipase C in breast cancer cells.
The urokinase receptor (uPAR) is a glycosylphosphatidylinositol (GPI)-anchored protein that promotes tissue remodeling, tumor cell adhesion, migration and invasion. uPAR mediates degradation of the extracellular matrix through protease recruitment and enhances cell adhesion, migration and signaling through vitronectin binding and interactions with integrins. Full-length uPAR is released from the cell surface, but the mechanism and significance of uPAR shedding remain obscure. Here we identify transmembrane glycerophosphodiesterase GDE3 as a GPI-specific phospholipase C that cleaves and releases uPAR with consequent loss of function, whereas its homologue GDE2 fails to attack uPAR. GDE3 overexpression depletes uPAR from distinct basolateral membrane domains in breast cancer cells, resulting in a less transformed phenotype, it slows tumor growth in a xenograft model and correlates with prolonged survival in patients. Our results establish GDE3 as a negative regulator of the uPAR signaling network and, furthermore, highlight GPI-anchor hydrolysis as a cell-intrinsic mechanism to alter cell behavior
Quantifying the nanomachinery of the nanoparticle-biomolecule interface
A study is presented of the nanomechanical phenomena experienced by nanoparticle-conjugated biomolecules. A thermodynamic framework is developed to describe the binding of thrombin-binding aptamer (TBA) to thrombin when the TBA is conjugated to nanorods. Binding results in nanorod aggregation (viz. directed self-assembly), which is detectable by absorption spectroscopy. The analysis introduces the energy of aggregation, separating it into TBA–thrombin recognition and surface-work contributions. Consequently, it is demonstrated that self-assembly is driven by the interplay of surface work and thrombin-TBA recognition. It is shown that the work at the surface is about −10 kJ mol−1 [mol superscript -1] and results from the accumulation of in-plane molecular forces of pN magnitude and with a lifetime of <1 s, which arises from TBA nanoscale rearrangements fuelled by thrombin-directed nanorod aggregation. The obtained surface work can map aggregation regimes as a function of different nanoparticle surface conditions. Also, the thermodynamic treatment can be used to obtain quantitative information on surface effects impacting biomolecules on nanoparticle surfaces.MIT-IQS Exchange Progra
Long-term efficacy and safety of brodalumab in the treatment of psoriasis : 120-week results from the randomized, double-blind, placebo- and active comparator-controlled phase 3 AMAGINE-2 trial
Randomized controlled trials have shown the efficacy and safety of brodalumab in patients with moderate to severe plaque psoriasis. To evaluate the efficacy and safety of brodalumab through 120 weeks of treatment in the AMAGINE-2 trial. Patients received ustekinumab through week 52 followed by brodalumab 210 mg every 2 weeks, continuous brodalumab 210 mg every 2 weeks, or any dose of brodalumab. Efficacy data were reported through 120 weeks by using observed data, last observation carried forward, and nonresponder imputation analyses. Of patients who received brodalumab 210 mg every 2 weeks, 84.4%, 75.6%, and 61.1% achieved 75%, 90%, and 100% improvement from baseline in Psoriasis Area and Severity Index at 120 weeks (observed data analysis), respectively. Patients who received brodalumab 210 mg every 2 weeks after receiving ustekinumab through 52 weeks achieved a similar skin clearance response as patients who received continuous brodalumab 210 mg every 2 weeks. Safety through 120 weeks was comparable to that of the blinded study periods. A large number of discontinuations toward the end of the study (31% in the final 6 months) were due to early termination and led to differences between observed data and nonresponder imputation results. Brodalumab is well tolerated and showed robust efficacy for more than 2 years
Giant vortex state in perforated aluminum microsquares
We investigate the nucleation of superconductivity in a uniform perpendicular
magnetic field H in aluminum microsquares containing a few (2 and 4) submicron
holes (antidots). The normal/superconducting phase boundary T_c(H) of these
structures shows a quite different behavior in low and high fields. In the low
magnetic field regime fluxoid quantization around each antidot leads to
oscillations in T_c(H), expected from the specific sample geometry, and
reminiscent of the network behavior. In high magnetic fields, the T_c(H)
boundaries of the perforated and a reference non-perforated microsquare reveal
cusps at the same values of Phi/Phi_0 (where Phi is the applied flux threading
the total square area and Phi_0 is the superconducting flux quantum), while the
background on T_c(H) becomes quasi-linear, indicating that a giant vortex state
is established. The influence of the actual geometries on T_c(H) is analyzed in
the framework of the linearized Ginzburg-Landau theory.Comment: 14 pages, 6 PS figures, RevTex, accepted for publication in Phys.

A Nonperturbative Eliasson's Reducibility Theorem
This paper is concerned with discrete, one-dimensional Schr\"odinger
operators with real analytic potentials and one Diophantine frequency. Using
localization and duality we show that almost every point in the spectrum admits
a quasi-periodic Bloch wave if the potential is smaller than a certain constant
which does not depend on the precise Diophantine conditions. The associated
first-order system, a quasi-periodic skew-product, is shown to be reducible for
almost all values of the energy. This is a partial nonperturbative
generalization of a reducibility theorem by Eliasson. We also extend
nonperturbatively the genericity of Cantor spectrum for these Schr\"odinger
operators. Finally we prove that in our setting, Cantor spectrum implies the
existence of a -set of energies whose Schr\"odinger cocycle is not
reducible to constant coefficients
Paving the Way for a Real-Time Context-Aware Predictive Architecture
Internet of Things society generates and needs to consume huge amounts of data in a demanding context-aware scenario. Such exponentially growing data sources require the use of novel processing methodologies, technologies and tools to facilitate data processing in order to detect and prevent situations of interest for the users in their particular context. To solve this issue, we propose an architecture which making use of emerging technologies and cloud platforms can process huge amounts of heterogeneous data and promptly alert users of relevant situations for a particular domain according to their context. Last, but not least, we will provide a graphical tool for domain experts to easily model, automatically generate code and deploy the situations to be detected and the actions to be taken in consequence. The proposal will be evaluated through a real case study related to air quality monitoring and lung diseases in collaboration with a doctor specialist on lung diseases of a public hospital
2-Amino-5-nitropyridinium tetraoxidorhenate(VII) monohydrate
All the residues of the title compound, (C5H6N3O2)[ReO4]·H2O, are located on general crystallographic positions. The 2-amino-5-nitropyridinium cation has a typical planar conformation with one of the nitro O atoms −0.058 (5) Å out of plane; the amine H atoms are also a little out of the main ring plane towards the opposite side of the aforementioned O atom [by 0.02 (4) and 0.04 (4) Å]. The perrhenate anion is nearly ideally tetrahedral. Three distinct N—H⋯O hydrogen bonds give rise to C(8) zigzag chains running along [100]. R
