    NNLO QCD corrections to event orientation in e+e- annihilation

    We present a new implementation of the NNLO QCD corrections to three-jet final states and related event-shape observables in electron–positron annihilation. Our implementation is based on the antenna subtraction method, and is performed in the NNLOjet framework. The calculation improves upon earlier results by taking into account the full kinematical information on the initial state momenta, thereby allowing the event orientation to be computed to NNLO accuracy. We find the event-orientation distributions at LEP and SLC to be very robust under higher order QCD corrections

    Benchmarking Deep Learning Models for Tooth Structure Segmentation.

    A wide range of deep learning (DL) architectures with varying depths are available, with developers usually choosing one or a few of them for their specific task in a nonsystematic way. Benchmarking (i.e., the systematic comparison of state-of-the art architectures on a specific task) may provide guidance in the model development process and may allow developers to make better decisions. However, comprehensive benchmarking has not been performed in dentistry yet. We aimed to benchmark a range of architecture designs for 1 specific, exemplary case: tooth structure segmentation on dental bitewing radiographs. We built 72 models for tooth structure (enamel, dentin, pulp, fillings, crowns) segmentation by combining 6 different DL network architectures (U-Net, U-Net++, Feature Pyramid Networks, LinkNet, Pyramid Scene Parsing Network, Mask Attention Network) with 12 encoders from 3 different encoder families (ResNet, VGG, DenseNet) of varying depth (e.g., VGG13, VGG16, VGG19). On each model design, 3 initialization strategies (ImageNet, CheXpert, random initialization) were applied, resulting overall into 216 trained models, which were trained up to 200 epochs with the Adam optimizer (learning rate = 0.0001) and a batch size of 32. Our data set consisted of 1,625 human-annotated dental bitewing radiographs. We used a 5-fold cross-validation scheme and quantified model performances primarily by the F1-score. Initialization with ImageNet or CheXpert weights significantly outperformed random initialization (P < 0.05). Deeper and more complex models did not necessarily perform better than less complex alternatives. VGG-based models were more robust across model configurations, while more complex models (e.g., from the ResNet family) achieved peak performances. In conclusion, initializing models with pretrained weights may be recommended when training models for dental radiographic analysis. Less complex model architectures may be competitive alternatives if computational resources and training time are restricting factors. Models developed and found superior on nondental data sets may not show this behavior for dental domain-specific tasks

    Male Macrophages and Fibroblasts from C57/BL6J Mice Are More Susceptible to Inflammatory Stimuli

    Mounting evidence argues for the significant impact of sex in numerous cardiac pathologies, including myocarditis. Macrophage polarization and activation of cardiac fibroblasts play a key role in myocardial inflammation and remodeling. However, the role of sex in these processes is still poorly understood. In this study, we investigated sex-specific alterations in the polarization of murine bone marrow-derived macrophages (BMMs) and the polarization-related changes in fibroblast activation. Cultured male and female murine BMMs from C57/BL6J mice were polarized into M1 (LPS) and M2 (IL-4/IL-13) macrophages. Furthermore, male and female cardiac fibroblasts from C57/BL6J mice were activated with TNF-α, TGF-β, or conditioned medium from M1 BMMs. We found a significant overexpression of M1 markers (c-fos, NFκB, TNF-α, and IL-1β) and M2 markers (MCP-1 and YM1) in male but not female activated macrophages. In addition, the ROS levels were higher in M1 male BMMs, indicating a stronger polarization. Similarly, the pro-fibrotic markers TGF-β and IL-1β were expressed in activated cardiac male fibroblasts at a significantly higher level than in female fibroblasts. In conclusion, the present study provides strong evidence for the male-specific polarization of BMMs and activation of cardiac fibroblasts in an inflammatory environment. The data show an increased inflammatory response and tissue remodeling in male mice

    Prevalence of antibodies against influenza A and B viruses in children in Germany, 2008 to 2010

    The prevalence of influenza A and B virus-specific IgG was determined in sera taken between 2008 and 2010 from 1,665 children aged 0-17 years and 400 blood donors in Germany. ELISA on the basis of whole virus antigens was applied. Nearly all children aged nine years and older had antibodies against influenza A. In contrast, 40% of children aged 0-4 years did not have any influenza A virus-specific IgG antibodies. Eighty-six percent of 0-6 year-olds, 47% of 7-12 year-olds and 20% of 13-17 year-olds were serologically naive to influenza B viruses. By the age of 18 years, influenza B seroprevalence reached approximately 90%. There were obvious regional differences in the seroprevalence of influenza B in Germany. In conclusion, seroprevalences of influenza A and influenza B increase gradually during childhood. The majority of children older than eight years have basal immunity to influenza A, while comparable immunity against influenza B is only acquired at the age of 18 years. Children aged 0-6 years, showing an overall seroprevalence of 67% for influenza A and of 14% for influenza B, are especially at risk for primary infections during influenza B seasons

    Prospective Newborn Screening for SCID in Germany: A First Analysis by the Pediatric Immunology Working Group (API)

    Background: T-cell receptor excision circle (TREC)-based newborn screening (NBS) for severe combined immunodeficiencies (SCID) was introduced in Germany in August 2019. / Methods: Children with abnormal TREC-NBS were referred to a newly established network of Combined Immunodeficiency (CID) Clinics and Centers. The Working Group for Pediatric Immunology (API) and German Society for Newborn Screening (DGNS) performed 6-monthly surveys to assess the TREC-NBS process after 2.5 years. / Results: Among 1.9 million screened newborns, 88 patients with congenital T-cell lymphocytopenia were identified (25 SCID, 17 leaky SCID/Omenn syndrome (OS)/idiopathic T-cell lymphocytopenia, and 46 syndromic disorders). A genetic diagnosis was established in 88%. Twenty-six patients underwent hematopoietic stem cell transplantation (HSCT), 23/26 within 4 months of life. Of these, 25/26 (96%) were alive at last follow-up. Two patients presented with in utero onset OS and died after birth. Five patients with syndromic disorders underwent thymus transplantation. Eight syndromic patients deceased, all from non-immunological complications. TREC-NBS missed one patient, who later presented clinically, and one tracking failure occurred after an inconclusive screening result. / Conclusion: The German TREC-NBS represents the largest European SCID screening at this point. The incidence of SCID/leaky SCID/OS in Germany is approximately 1:54,000, very similar to previous observations from North American and European regions and countries where TREC-NBS was implemented. The newly founded API-CID network facilitates tracking and treatment of identified patients. Short-term HSCT outcome was excellent, but NBS and transplant registries will remain essential to evaluate the long-term outcome and to compare results across the rising numbers of TREC-NBS programs across Europe
