IMPACT OF ASPARTAME CONSUMPTION ON NEUROTRANSMITTERS IN RAT BRAIN
Background: Aspartame (APM), a common artificial sweetener, has been used for diabetic subjects and body weight
control for a long time. The goal of the present study was to evaluate the impact of APM consumption on
neurotransmitters and oxidative stress in rat’s brain.
Materials and Methods: Four groups of male Wistar albino rats was used: Group1, control (Rats fed on normal diet).
Group 2: Rats were received aspartame (50 mg/kg b w). Group3: Rats were received aspartame (75 mg/kg b w). Group 4:
Rats were received aspartame (125 mg/kg b w). Five rats were decapitated after 10, 20, 30 and 40 days from start of
experiment. Blood and brain tissue were collected for biochemical analysis. Biochemical analysis of brain tissue includes
neurotransmitters (Acetylcholine, epinephrine, norepinephrine, γ-aminobutyric acid and serotonin). Serum for
determination of lipid peroxidation (MDA), reduced glutathione and superoxide dismutase (SOD).

Effects of air-abrasion pressure on the resin bond strength to zirconia: a combined cyclic loading and thermocycling aging study
Objectives
To determine the combined effect of fatigue cyclic loading and thermocycling (CLTC) on the shear bond strength (SBS) of a resin cement to zirconia surfaces that were previously air-abraded with aluminum oxide (Al2O3) particles at different pressures.
Materials and Methods
Seventy-two cuboid zirconia specimens were prepared and randomly assigned to 3 groups according to the air-abrasion pressures (1, 2, and 2.8 bar), and each group was further divided into 2 groups depending on aging parameters (n = 12). Panavia F 2.0 was placed on pre-conditioned zirconia surfaces, and SBS testing was performed either after 24 hours or 10,000 fatigue cycles (cyclic loading) and 5,000 thermocycles. Non-contact profilometry was used to measure surface roughness. Failure modes were evaluated under optical and scanning electron microscopy. The data were analyzed using 2-way analysis of variance and χ2 tests (α = 0.05).
Results
The 2.8 bar group showed significantly higher surface roughness compared to the 1 bar group (p < 0.05). The interaction between pressure and time/cycling was not significant on SBS, and pressure did not have a significant effect either. SBS was significantly higher (p = 0.006) for 24 hours storage compared to CLTC. The 2 bar-CLTC group presented significantly higher percentage of pre-test failure during fatigue compared to the other groups. Mixed-failure mode was more frequent than adhesive failure.
Conclusions
CLTC significantly decreased the SBS values regardless of the air-abrasion pressure used
MOLECULAR EPIDEMIOLOGICAL STUDIES ON TRYPANOSOMA EVANSI TYPE A AND TYPE B IN CAMELS (CAMELUS DROMEDARIES) FROM FIVE DIFFERENT REGIONS OF SAUDI ARABIA USING THE ITS1 RDNA AND ROTAT 1.2 VSG GENE
Trypanosoma evansi is the most widespread of the pathogenic salivarian trypanosomes and
cause a serious disease called (surra) that is affect the domestic animals such camels and horses in Tropical and subtropical countries and often leads to reduced productivity and economic
losses. Therefore, the objectives of the present study were to determine the prevalence rates of
trypanosomiasis using polymerase chain reaction (PCR) among camels from five different regions of Saudi Arabia and to sequence and characterized the T. evansi from these animals. In
the current study, 832 camel blood samples collected from five different regions of Saudi Arabia for detecting T. evansi. A generic ITS1-PCR and RoTat 1.2 VSG gene were applied in this
study to analyze camels’ blood samples. Molecular analysis was performed using ITS1-PCR
which showed that the highest prevalence of trypanosomes was observed in Al-Qaseem province (50.1%) followed Riyadh province (49%), whereas in Hail and the Northern Borders,
there were fewer infections with trypanosomes (28.4% &17.6%), respectively. PCR amplification was carried out targeting RoTat 1.2 VSG gene on TS1-positive samples and some of them
were negative for RoTat1.2. The test negative in RoTat 1.2 PCR but ITS1 PCR positive could
suggest T. evansi type B. Presence of T. evansi type B is interest to the international community, as this has a message to redesign the existing molecular and serological diagnostic markers.
However, to our knowledge this the first study demonstrating T. evansi type B out of Africa.

Quality of Type 2 Diabetes Management in the States of The Co-Operation Council for the Arab States of the Gulf: A Systematic Review
Type 2 diabetes mellitus is a growing, worldwide public health concern. Recent growth has been particularly dramatic in the states of The Co-operation Council for the Arab States of the Gulf (GCC), and these and other developing economies are at particular risk. We aimed to systematically review the quality of control of type 2 diabetes in the GCC, and the nature and efficacy of interventions. We identified 27 published studies for review. Studies were identified by systematic database searches. Medline and Embase were searched separately (via Dialog and Ovid, respectively; 1950 to July 2010 (Medline), and 1947 to July 2010 (Embase)) on 15/07/2009. The search was updated on 08/07/2010. Terms such as diabetes mellitus, non-insulin-dependent, hyperglycemia, hypertension, hyperlipidemia and Gulf States were used. Our search also included scanning reference lists, contacting experts and hand-searching key journals. Studies were judged against pre-determined inclusion/exclusion criteria, and where suitable for inclusion, data extraction/quality assessment was achieved using a specifically-designed tool. All studies wherein glycaemic-, blood pressure- and/or lipid- control were investigated (clinical and/or process outcomes) were eligible for inclusion. No limitations on publication type, publication status, study design or language of publication were imposed. We found the extent of control to be sub-optimal and relatively poor. Assessment of the efficacy of interventions was difficult due to lack of data, but suggestive that more widespread and controlled trial of secondary prevention strategies may have beneficial outcomes. We found no record of audited implementation of primary preventative strategies and anticipate that controlled trial of such strategies would also be useful
Novel spectrophotometric method for determination of cinacalcet hydrochloride in its tablets via derivatization with 1,2-naphthoquinone-4-sulphonate
This study represents the first report on the development of a novel spectrophotometric method for determination of cinacalcet hydrochloride (CIN) in its tablet dosage forms. Studies were carried out to investigate the reaction between CIN and 1,2-naphthoquinone-4-sulphonate (NQS) reagent. In alkaline medium (pH 8.5), an orange red-colored product exhibiting maximum absorption peak (λmax) at 490 nm was produced. The stoichiometry and kinetic of the reaction were investigated and the reaction mechanism was postulated. This color-developing reaction was employed in the development of a simple and rapid visible-spectrophotometric method for determination of CIN in its tablets. Under the optimized reaction conditions, Beer's law correlating the absorbance with CIN concentration was obeyed in the range of 3 - 100 μg/ml with good correlation coefficient (0.9993). The molar absorptivity (ε) was 4.2 × 105 l/mol/cm. The limits of detection and quantification were 1.9 and 5.7 μg/ml, respectively. The precision of the method was satisfactory; the values of relative standard deviations (RSD) did not exceed 2%. No interference was observed from the excipients that are present in the tablets. The proposed method was applied successfully for the determination of CIN in its pharmaceutical tablets with good accuracy and precisions; the label claim percentage was 100.80 - 102.23 ± 1.27 - 1.62%. The results were compared favorably with those of a reference pre-validated method. The method is practical and valuable in terms of its routine application in quality control laboratories
Metabolomics Profile in Depression:A Pooled Analysis of 230 Metabolic Markers in 5283 Cases With Depression and 10,145 Controls
Background: Depression has been associated with metabolic alterations, which adversely impact cardiometabolic health. Here, a comprehensive set of metabolic markers, predominantly lipids, was compared between depressed and nondepressed persons. Methods: Nine Dutch cohorts were included, comprising 10,145 control subjects and 5283 persons with depression, established with diagnostic interviews or questionnaires. A proton nuclear magnetic resonance metabolomics platform provided 230 metabolite measures: 51 lipids, fatty acids, and low-molecular-weight metabolites; 98 lipid composition and particle concentration measures of lipoprotein subclasses; and 81 lipid and fatty acids ratios. For each metabolite measure, logistic regression analyses adjusted for gender, age, smoking, fasting status, and lipid-modifying medication were performed within cohort, followed by random-effects meta-analyses. Results: Of the 51 lipids, fatty acids, and low-molecular-weight metabolites, 21 were significantly related to depression (false discovery rate q <.05). Higher levels of apolipoprotein B, very-low-density lipoprotein cholesterol, triglycerides, diglycerides, total and monounsaturated fatty acids, fatty acid chain length, glycoprotein acetyls, tyrosine, and isoleucine and lower levels of high-density lipoprotein cholesterol, acetate, and apolipoprotein A1 were associated with increased odds of depression. Analyses of lipid composition indicators confirmed a shift toward less high-density lipoprotein and more very-low-density lipoprotein and triglyceride particles in depression. Associations appeared generally consistent across gender, age, and body mass index strata and across cohorts with depressive diagnoses versus symptoms. Conclusions: This large-scale meta-analysis indicates a clear distinctive profile of circulating lipid metabolites associated with depression, potentially opening new prevention or treatment avenues for depression and its associated cardiometabolic comorbidity
