    RESUMEN Este trabajo versa sobre el álbum ilustrado y su aplicación a la Educación Infantil. El álbum ilustrado es una herramienta reciente en la enseñanza de los niños en sus primeras etapas de aprendizaje. Se trata de un híbrido literario en el que el texto e imagen tienen la misma entidad. Es un instrumento literario, artístico y social que conecta al lector del mundo imaginario con la vida real. El trabajo consta de dos partes. La primera es un breve marco teórico que trata de explicar qué es el álbum ilustrado, de detallar la importancia y la necesidad de animar a leerlo en la escuela, así como sus elementos más característicos. En segundo lugar, se ha realizado una investigación acerca del uso de dicho instrumento en las aulas de Educación Infantil, centros infantiles y guarderías de Cantabria.ABSTRACT This work deals with the picture book and its application to Early Childhood Education. The illustrated album is a recent tool in teaching children in their early learning stages. It is a literary hybrid in which text and image have the same entity. It is a literary, artistic and social tool that connects the reader of the imaginary world with real life. The work consists of two parts. The first is a brief theoretical framework that tries to explain what the picture book is, to detail the importance and the need to encourage reading in school, as well as its most characteristic elements.     Preservation of the antioxidant capacity of resveratrol via encapsulation in niosomes

    Resveratrol (RSV) is a natural polyphenol which produces several benefits to human health, being the trans-isomer the most bioactive. However, its systemic absorption is limited due to its low water solubility, that reduces the oral bioavailability, and its chemical instability (owing to the trans-cis RSV isomer conversion upon light irradiation). Thus, encapsulation of this bioactive compound is required to protect it from destructive environmental conditions. Here, trans-RSV was encapsulated in food grade nanovesicles formed by Tween 80 and Span 80, with or without the addition of dodecanol (Dod) as membrane stabilizer. The size and shape of niosomes were evaluated by microscopy (TEM) and light scattering. RSV was successfully encapsulated in the vesicular systems (49– 57%). The effect of Dod in the membrane bilayer was evaluated on the RSV in vitro release experiments under simulated gastrointestinal conditions. The total antioxidant capacity of the encapsulated polyphenol was measured using radicals’ assays (DPPH and ABTS). The niosomes were able to maintain almost the total antioxidant capacity of encapsulated RSV, also preserved the ~85% of trans-RSV, thus offering considerable protection against high energy irradiation. These results make these systems suitable for different applications, particularly for photosensitive compounds.Fil: Machado, Noelia Daiana.     Personality in patients with eating disorders depending on the presence/absence of comorbidity with borderline personality disorder / Personalidad en pacientes con trastorno alimentario en función de la presencia/ausencia de comorbilidad con trastorno límite de la personalidad

    Abstract  Studies on comorbidity between eating disorders (ED) and personality disorders yield rates of 20-80%, a condition that makes difficult the diagnosis and complicates prognosis, especially regarding their comorbidity with borderline personality disorder (BPD). The objective of this study was to assess whether pathological personality alterations make it possible to distinguish not only between different types of ED, but also in terms of their comorbidity with BPD. Participants included 29 patients with ED, 10 with comorbid ED with BPD (EDc), 27 with BPD and 22 healthy controls, who completed the Millon Multiaxial Clinical Inventory (MCMI-II), and two other measures, one on impulsivity and other aimed at assess symptoms of bulimia nervosa (BN). The ED group was characterized by greater compulsivity, mainly in the subgroup with anorexia nervosa vs. BPD and EDc groups; however, these last two groups shared many features that show emotional instability, although less that the subgroup with BN. These findings support a continuum of severity in terms of compulsivity-impulsivity, with important etiopathogenic, diagnostic and psychotherapeutic implications.Resumen   Los estudios sobre comorbilidad entre los trastornos de la conducta alimentaria (TCA) y los trastornos de la personalidad reportan tasas del 20-80%, condición que dificulta el diagnóstico y ensombrece el pronóstico, especialmente respecto a su comorbilidad con el trastorno límite de la personalidad (TLP). El objetivo de este estudio fue valorar si las alteraciones patológicas de la personalidad permiten distinguir no solo entre distintos tipos de TCA, sino además en función de su comorbilidad con el TLP. Participaron 29 pacientes con TCA, 10 con TCA comórbido con TLP [TCAc], 27 con TLP y 22 controles sanas, quienes completaron el Inventario Clínico Multiaxial de Millon (MCMI-II) y otras dos medidas, una sobre impulsividad y otra dirigida a evaluar síntomas de bulimia nerviosa (BN). El grupo TCA se caracterizó por mayor compulsividad, principalmente en el caso del subgrupo con anorexia nerviosa vs. los grupos TLP y TCAc; no obstante, estos dos últimos compartieron un amplio número de rasgos indicativos de inestabilidad emocional, y también –aunque en menor medida− con el subgrupo con BN. Los hallazgos apoyan un continuum de gravedad en función de la compulsividad-impulsividad, con importantes implicaciones etiopatogénicas, diagnósticas y psicoterapéuticas.

    Signaling through the leukocyte integrin LFA-1 in T cells induces a transient activation of Rac-1 that is regulated by Vav and PI3K/Akt-1

    12 p.-8 fig.Integrin LFA-1 is a receptor that is able to transmit multiple intracellular signals in leukocytes. Herein we show that LFA-1 induces a potent and transient increase in the activity of the small GTPase Rac-1 in T cells. Maximal Rac-1 activity peaked 10-15 min after LFA-1 stimulation and rapidly declined to basal levels at longer times. We have identified Vav, a guanine nucleotide exchange factor for Rac-1, and PI3K/Akt, as regulators of the activation and inactivation phases of the activity of Rac-1, respectively, in the context of LFA-1 signaling based on the following experimental evidence: (i) LFA-1 induced activation of Vav and PI3K/Akt with kinetics consistent with a regulatory role for these molecules on Rac-1, (ii) overexpression of a constitutively active Vav mutant induces activation of Rac independently of LFA-1 stimulation whereas overexpression of a dominant-negative Vav mutant blocks LFA-1-mediated Rac activation, (iii) pharmacological inhibition of PI3K/Akt prevented the fall in the activity of Rac-1 after its initial activation but had no effect on Vav activity, and (iv) overexpression of a dominant-negative or a constitutively active Akt-1 induced or inhibited, respectively, Rac-1 activity. Finally, we show that T cells with a sustained Rac activity have impaired capacity to elongate onto ICAM-1.     Physiological lentiviral vectors for the generation of improved CAR-T cells

    Anti-CD19 chimeric antigen receptor (CAR)-T cells have achieved impressive outcomes for the treatment of relapsed and refractory B-lineage neoplasms.However, important limitations still remain due to severe adverse events (i.e., cytokine release syndrome and neuroinflammation) and relapse of 40%–50%of the treated patients.MostCAR-Tcells are generated using retroviral vectors with strong promoters that lead to high CAR expression levels, tonic signaling, premature exhaustion, and overstimulation, reducing efficacy and increasing side effects. Here, we show that lentiviral vectors (LVs) expressing the transgene through a WAS gene promoter (AW-LVs) closely mimic the T cell receptor (TCR)/CD3 expression kinetic upon stimulation. These AW-LVs can generate improved CAR-T cells as a consequence of theirmoderate andTCR-like expression profile. Compared with CAR-T cells generated with human elongation factor a (EF1a)-driven-LVs, AW-CAR-T cells exhibited lower tonic signaling, higher proportion of naive and stem cell memory T cells, less exhausted phenotype, and milder secretion of tumor necrosis factor alpha (TNF-a) and interferon (IFN)-ɣ after efficient destruction of CD19+ lymphoma cells, both in vitro and in vivo.Moreover, we also showed their improved efficiency using an in vitro CD19+ pancreatic tumor model. We finally demonstrated the feasibility of large-scale manufacturing ofAW-CAR-T cells in good manufacturing practice (GMP)-like conditions.     A Telehealth-Based Cognitive-Adaptive Training (e-OTCAT) to Prevent Cancer and Chemotherapy-Related Cognitive Impairment in Women with Breast Cancer: Protocol for a Randomized Controlled Trial

    Background: Many women with breast cancer experience a great number of side effects, such as cognitive impairment, during and after chemotherapy that reduces their quality of life. Currently, research focusing on the use of non-pharmacological, and specifically telehealth interventions to prevent or mitigate them has been insufficient. Methods: This protocol describes a randomized controlled trial aimed at studying the preventive effects of a videoconferenced cognitive-adaptive training (e-OTCAT) program (ClinicalTrials.gov NCT04783402). A number of 98 eligible participants will be randomized to one of the following groups: (a) the experimental group receiving the e-OTCAT program during 12 consecutive weeks since the beginning of chemotherapy; and (b) the control group receiving and educational handbook and usual care. The primary outcome will be the cognitive function. Secondary measures will be psychological distress, fatigue, sleep disturbance, quality of life and occupational performance. The time-points for these measures will be placed at baseline, after 12 weeks and six months of post-randomization. Conclusion: This trial may support the inclusion of multidimensional interventions through a telehealth approach in a worldwide growing population suffering from breast cancer, emphasizing the prevention of cognitive impairment as one of the side effects of cancer and its treatments.Spanish Government FPU17/00939 FPU18/03575Instituto de Salud Carlos III FI19/0023

    Lentiviral vectors for inducible, transactivator-free advanced therapy medicinal products: Application to CAR-T cells

    Controlling transgene expression through an externally administered inductor is envisioned as a potent strategy to improve safety and efficacy of gene therapy approaches. Generally, inducible ON systems require a chimeric transcription factor (transactivator) that becomes activated by an inductor, which is not optimal for clinical translation due to their toxicity. We generated previously the first all-in-one, transactivator-free, doxycycline (Dox)-responsive (Lent-On-Plus or LOP) lentiviral vectors (LVs) able to control transgene expression in human stem cells. Here, we have generated new versions of the LOP LVs and have analyzed their applicability for the generation of inducible advanced therapy medicinal products (ATMPs) with special focus on primary human T cells. We have shown that, contrary to all other cell types analyzed, an Is2 insulator must be inserted into the 30 long terminal repeat of the LOP LVs in order to control transgene expression in human primary T cells. Importantly, inducible primary T cells generated by the LOPIs2 LVs are responsive to ultralow doses of Dox and have no changes in phenotype or function compared with untransduced T cells. We validated the LOPIs2 system by generating inducible CAR-T cells that selectively kill CD19+ cells in the presence of Dox.     Perception of the educational environment by dental students undergoing a curricular transition in a Peruvian sample

    El objetivo de ésta investigación fue comparar la percepción del clima educativo de los estudiantes de odontología de la Universidad Nacional Mayor de San Marcos, en un período aproximado de seis meses posterior a la implementación del nuevo proyecto curricular en pregrado, con la percepción de los estudiantes que todavía se encuentran en el plan curricular anterior o tradicional, tomando en consideración el semestre o año de estudio y el género. Metodología: se encuestaron 169 estudiantes que representan el 40 % del total de estudiantes matriculados, de los cuales 129 pertenecen al nuevo currículo con régimen de estudios mixto: semestral en la mayoría de asignaturas y anual en las asignaturas clínicas integradas; y 40 que pertenecen al currículo anterior con régimen de estudio anual para todas las asignaturas. El 53.3 % de estudiantes eran del sexo masculino y el 46.7% del sexo femenino distribuidos equitativamente según semestre o año de estudio y seleccionados en forma aleatoria. Se utilizó el cuestionario Dundee Ready Education Environment Measure DREEM que evalúa el clima educacional en las escuelas de profesiones de salud versión en español. Resultados y conclusiones: se encontró un clima educativo más positivo que negativo con puntaje promedio en la muestra total de 136.83 ± 19.00. No se encontraron diferencias significativas entre la percepción del clima educacional del estudiante del nuevo plan curricular y el tradicional, tampoco entre años de estudios, ni género.The objective of this study was to compare the perception of the educational climate of San Marcos University dental students, in a period of approximately six months after implementation of the new undergraduate curriculum project, with the perception of students still found in the previous or traditional curriculum, taking into consideration the semester or year of study and gender. Methodology: it were surveyed 169 students representing 40% of all students enrolled, 129 of them belong to the new curriculum with mixed regime studies: semester in most asignatures and anual in integrated clinical asignatures; 40 belong to the previous regime curriculum with annual study for all asignatures. 53.3% of students were male and 46.7% female equitably distributed by semester or year of study and randomly chosen. The spanish version of Questionnaire Dundee Ready Education Environment Measure DREEM that evaluates the educational climate in schools of health professions, was used. Results and conclusions: a more positive than negative educational climate was found, with average score in the total sample of 136.83 ± 19.00.
