    Functional interplay between DEAD-box RNA helicases Ded1 and Dbp1 in preinitiation complex attachment and scanning on structured mRNAs in vivo

    RNA structures that impede ribosome binding or subsequent scanning of the 5′-untranslated region (5′-UTR) for the AUG initiation codon reduce translation efficiency. Yeast DEAD-box RNA helicase Ded1 appears to promote translation by resolving 5′-UTR structures, but whether its paralog, Dbp1, performs similar functions is unknown. Furthermore, direct in vivo evidence was lacking that Ded1 or Dbp1 resolves 5′-UTR structures that impede attachment of the 43S preinitiation complex (PIC) or scanning. Here, profiling of translating 80S ribosomes reveals that the translational efficiencies of many more mRNAs are reduced in a ded1-ts dbp1Δ double mutant versus either single mutant, becoming highly dependent on Dbp1 or Ded1 only when the other helicase is impaired. Such ‘conditionally hyperdependent’ mRNAs contain unusually long 5′-UTRs with heightened propensity for secondary structure and longer transcript lengths. Consistently, overexpressing Dbp1 in ded1 cells improves the translation of many such Ded1-hyperdependent mRNAs. Importantly, Dbp1 mimics Ded1 in conferring greater acceleration of 48S PIC assembly in a purified system on mRNAs harboring structured 5′-UTRs.     Seroprevalence of rubella in pregnant women

    Background: Rubella is a droplet infection characterized by self-limiting illness. However infection during pregnancy may result in miscarriage, congenital birth defects leading to long term morbidity. The aim of the study was to estimate the seroprevalence of rubella immunity in pregnant women.Methods: Antenatal patients, irrespective of period of gestation, fulfilling the inclusion criteria were tested for rubella IgG antibodies.Results: A total of 258 pregnant women were included in the study. The estimated seroprevalence of immunity against Rubella infection was 70.5% (n=182) whereas 29.5% (n=76) were seronegative and thus susceptible to rubella infection. The distribution of seroprevalence of rubella immunity based on age group and gravidity were also evaluated.Conclusions: The results reveal high level of rubella sero positivity, which indicates continued transmission of rubella infection in the community.

    Analysis procedure of the positronium lifetime spectra for the J-PET detector

    Positron Annihilation Lifetime Spectroscopy (PALS) has shown to be a powerful tool to study the nanostructures of porous materials. Positron Emissions Tomography (PET) are devices allowing imaging of metabolic processes e.g. in human bodies. A newly developed device, the J-PET (Jagiellonian PET), will allow PALS in addition to imaging, thus combining both analyses providing new methods for physics and medicine. In this contribution we present a computer program that is compatible with the J-PET software. We compare its performance with the standard program LT 9.0 by using PALS data from hexane measurements at different temperatures. Our program is based on an iterative procedure, and our fits prove that it performs as good as LT 9.0.Comment: 4 figures, 8 page

The smart-phone is used very commonly by everyone who is educated or non-educated. This application can provide different languages, which is Hindi and English this language commonly used and easy to use on the system.

    Generation, annotation, and analysis of ESTs from midgut tissue of adult female Anopheles stephensi mosquitoes

    <p>Abstract</p> <p>Background</p> <p>Malaria is a tropical disease caused by protozoan parasite, <it>Plasmodium</it>, which is transmitted to humans by various species of female anopheline mosquitoes. <it>Anopheles stephensi </it>is one such major malaria vector in urban parts of the Indian subcontinent. Unlike <it>Anopheles gambiae</it>, an African malaria vector, transcriptome of <it>A. stephensi </it>midgut tissue is less explored. We have therefore carried out generation, annotation, and analysis of expressed sequence tags from sugar-fed and <it>Plasmodium yoelii </it>infected blood-fed (post 24 h) adult female <it>A. stephensi </it>midgut tissue.</p> <p>Results</p> <p>We obtained 7061 and 8306 ESTs from the sugar-fed and <it>P. yoelii </it>infected mosquito midgut tissue libraries, respectively. ESTs from the combined dataset formed 1319 contigs and 2627 singlets, totaling to 3946 unique transcripts. Putative functions were assigned to 1615 (40.9%) transcripts using BLASTX against UniProtKB database. Amongst unannotated transcripts, we identified 1513 putative novel transcripts and 818 potential untranslated regions (UTRs). Statistical comparison of annotated and unannotated ESTs from the two libraries identified 119 differentially regulated genes. Out of 3946 unique transcripts, only 1387 transcripts were mapped on the <it>A. gambiae </it>genome. These also included 189 novel transcripts, which were mapped to the unannotated regions of the genome. The EST data is available as ESTDB at <url>http://mycompdb.bioinfo-portal.cdac.in/cgi-bin/est/index.cgi</url>.</p> <p>Conclusion</p> <p>3946 unique transcripts were successfully identified from the adult female <it>A. stephensi </it>midgut tissue. These data can be used for microarray development for better understanding of vector-parasite relationship and to study differences or similarities with other malaria vectors. Mapping of putative novel transcripts from <it>A. stephensi </it>on the <it>A. gambiae </it>genome proved fruitful in identification and annotation of several genes. Failure of some novel transcripts to map on the <it>A. gambiae </it>genome indicates existence of substantial genomic dissimilarities between these two potent malaria vectors.</p

    Feasibility study of the time reversal symmetry tests in decay of metastable positronium atoms with the J-PET detector

    This article reports on the feasibility of testing of the symmetry under reversal in time in a purely leptonic system constituted by positronium atoms using the J-PET detector. The present state of T symmetry tests is discussed with an emphasis on the scarcely explored sector of leptonic systems. Two possible strategies of searching for manifestations of T violation in nonvanishing angular correlations of final state observables in the decay of metastable triplet states of positronium available with J-PET are proposed and discussed. Results of a pilot measurement with J-PET and assessment of its performance in reconstruction of three-photon decays are shown along with an analysis of its impact on the sensitivity of the detector for the determination of T-violation sensitive observables

    Introduction of total variation regularization into filtered backprojection algorithm

    In this paper we extend the state-of-the-art filtered backprojection (FBP) method with application of the concept of Total Variation regularization. We compare the performance of the new algorithm with the most common form of regularizing in the FBP image reconstruction via apodizing functions. The methods are validated in terms of cross-correlation coefficient between reconstructed and real image of radioactive tracer distribution using standard Derenzo-type phantom. We demonstrate that the proposed approach results in higher cross-correlation values with respect to the standard FBP method
