    La calcification testiculaire chez les boucs de centres d'insémination artificielle : étude clinique et répercussion sur la production de semence

    La calcification testiculaire a été étudiée chez 259 jeunes boucs de 2 centres d'insémination artificielle (CAPRI-IA et INRA-SEIA), en première saison sexuelle. A l'examen échographique des testicules, les lésions apparaissent comme des foyers hyperéchogènes plus ou moins nombreux dans le parenchyme testiculaire. Le pourcentage de boucs atteints augmente de 51% à 78% entre 1999 et 2001 et est plus élevé dans la race Alpine (71%) que dans la race Saanen (56%). Il n'y a pas d'influence de l'âge des animaux sur l'atteinte testiculaire. A l'histologie, 1 à 8% des tubes séminifères sont atteints et présentent une stase spermatique et une hypoplasie de l'épithélium germinal. Les spermatozoïdes accumulés meurent puis se calcifient progressivement. Ces lésions s'accompagnent de fibrose interstitielle et de dégénérescence testiculaire. Ces calcifications testiculaires pourraient être associées à une carence en zinc. En effet, les concentrations plasmatiques en zinc mesurées chez les boucs sont faibles et diminuent significativement au cours des 6 mois de la période d'étude. Elles ne différent pas suivant le statut pathologique des animaux. L'administration intramusculaire de zinc a permis de rétablir au moins partiellement les concentrations plasmatiques initiales, ce qui est en faveur d'un déficit en zinc. De plus, les apports alimentaires en calcium sont élevés et pourraient diminuer l'absorption du zinc. L'influence des pères et des grands-pères sur le statut pathologique de leurs descendants suggère qu'un facteur génétique pourrait jouer un rôle prédisposant dans le développement des lésions testiculaires. Les lésions ont peu de répercussions sur la quantité et la qualité de la semence récoltée, en raison probablement de la faible proportion de tubes séminifères lésés chez ces animaux

    Le musée comme forum de citoyenneté est une proposition qu'il convient de mettre en perspective. Cette posture intellectuelle et cet enjeu social sont couramment manipulés aujourd'hui par les musées y compris les musées de beaux-arts mais plus largement encore par d’autres domaines de la culture. La démocratisation culturelle tant attendue et rarement atteinte resurgirait sous cette forme spontanée et dynamique du forum où chaque citoyen trouverait un lieu d’expression. La participation citoyenne dont on aimerait pouvoir donner une définition et cerner les contours, devient un préalable à tout projet culturel. Le forum comme lieu public de débat n’est pas en reste. D’ailleurs les grands musées de société récemment inaugurés ont enfoui le terme de musée pour ériger la notion de cité (Cité de l’architecture, Cité de l’immigration) qui dans sa définition première n’est pas si loin que ça du forum. Il semblerait qu’en réunissant des activités différentes avec un souci de pluridisciplinarité dans un même lieu, on tende vers cet objectif de forum ou de cité, caractérisés par un souci d’ouverture au plus grand nombre

    HP-LT rocks exhumed during intra-oceanic subduction: the example of the Escambray massif (Cuba).

    High-Presssure metabasites embedded in a serpentinite or metasedimentary matrix from the Sancti Spiritus dome (Escambray massif, Central Cuba) have been studied in order to better understand the origine and the evolution of the Northern Carribean boundary plate during the Cretaceous, in a global subduction context. Geochemical analyses (major, trace elements and isotopes) of the high pressure rocks show that they could be partially derived from the Cretaceous calc-alkaline arc described in Central Cuba, these were probably incorporated in the subduction zone by tectonic erosion. The High-Pressure rocks record a prograde path from the epidote bearing amphibolite facies to the barroisite bearing eclogite facies (P = 19 ± 2 Kbar, T = 590 ± 90 °C). These metabasites show evidence of retrogression starting from the glaucophane bearing eclogite facies to the lawsonite bearing blueschist facies. Therefore, these HP/LT rocks are characterized by a counter-clockwise cooling P/T path, which can be explained by the exhumation of HP rocks while the subduction was still active. Concordant geochronological data (Rb/Sr and Ar/Ar) suggest that the main exhumation of HP/LT rocks from the Sancti Spiritus dome occurred 70 Ma ago by top to SW thrusting. The retrogressed trajectory of these rocks, means that the northeast subduction of the Farallon plate continued after 70Ma. The final exhumation can be correlated with the beginning of the collision between the Bahamas platform and the Cretaceous island arc that induced a change of the subduction kinematic

    Chronically Implanted, Nafion-Coated Ag/AgCl Reference Electrodes for Neurochemical Applications

    Fast-scan cyclic voltammetry at carbon-fiber microelectrodes can be used to measure behaviorally correlated dopamine changes in the extracellular fluid of the brain of freely moving rats. These experiments employ a chronically implanted Ag/AgCl reference electrode. When dopamine measurements are taken 4 days after implantation, there is often a potential shift, typically greater than +0.2 V, in the anodic and cathodic peaks in the cyclic voltammogram for dopamine. In this work, we optimized a method to coat sintered Ag/AgCl reference electrodes with the perfluorinated polymer, Nafion, to prevent this shift. We find that we can stabilize reference electrodes for up to 28 days. Immunohistochemistry of the tissue around the implant site shows extensive glial encapsulation around both bare and Nafion-coated devices. However, the lesion around bare electrodes has a rough texture implying that these cells are strongly adsorbed onto the bare reference electrode, while the lesion around a Nafion-coated electrode shows that cells are more intact implying that they adsorb less strongly. Energy dispersive X-ray spectroscopy and scanning electron microscopy analysis of the surface of the electrodes confirms this by visualizing a heavy buildup of plaques, organic in nature, only on bare electrodes. Impedance spectroscopy indicates no difference between the impedance of bare and Nafion-coated Ag/AgCl electrodes, indicating that glial encapsulation does not lead to an increase in uncompensated resistance between the working and reference electrodes. The electrochemical shift therefore must be due to the unique chemical microenvironment around the reference electrode that alters the chloride equilibrium, a process that the Nafion coating prevents

    SLC26A9 Gene Is Associated With Lung Function Response to Ivacaftor in Patients With Cystic Fibrosis

    Ivacaftor is a drug used to treat cystic fibrosis (CF) patients carrying specific gating CFTR mutations. Interpatient variability in the lung response has been shown to be partly explained by rs7512462 in the Solute Carrier Family 26 Member 9 (SLC26A9) gene. In an independent and larger cohort, we aimed to evaluate whether SLC26A9 variants contribute to the variability of the lung phenotype and if they influence the lung response to ivacaftor. We genotyped the French CF Gene Modifier Study cohort (n = 4,840) to investigate whether SLC26A9 variants were involved in the lung phenotype heterogeneity. Their influence in the response to ivacaftor was tested in the 30 treated patients who met the inclusion criteria: older than 6 years of age, percent-predicted forced expiratory volume measured in 1 s (FEV1pp) in the 3 months before treatment initiation ranging between 40 and 90%. Response to treatment was determined by the change in FEV1pp from baseline, averaged in 15–75 days, and the 1st-year post-treatment. We observed that SLC26A9 variants were not associated with lung function variability in untreated patients and that gain of lung function in patients treated with ivacaftor was similar to clinical trials. We confirmed that rs7512462 was associated with variability in ivacaftor-lung response, with a significant reduction in lung function improvement for patients with the C allele. Other SLC26A9 SNPs also contributed to the ivacaftor-response. Interindividual variability in lung response to ivacaftor is associated with SLC26A9 variants in French CF patients. Pharmacogenomics and personalized medicine will soon be part of CF patient care

    Enhance quality care performance: Determination of the variables for establishing a common database in French paediatric critical care units

    Abstract Selected variables for the French Paediatric Intensive Care registry. Rationale, aims, and objectives Providing quality care requires follow-up in regard to clinical and economic activities. Over the past decade, medical databases and patient registries have expanded considerably, particularly in paediatric critical care medicine (eg, the Paediatric Intensive Care Audit Network (PICANet) in the UK, the Australian and New Zealand Paediatric Intensive Care (ANZPIC) Registry in Australia and New Zealand, and the Virtual Paediatric Intensive Care Unit Performance System (VPS) in the USA). Such a registry is not yet available in France. The aim of this study was to determine variables that ought to be included in a French paediatric critical care registry. Methods Variables, items, and subitems from 3 foreign registries and 2 French local databases were used. Items described each variable, and subitems described items. The Delphi method was used to evaluate and rate 65 variables, 90 items, and 17 subitems taking into account importance or relevance based on input from 28 French physicians affiliated with the French Paediatric Critical Care Group. Two ratings were used between January and May 2013. Results Fifteen files from 10 paediatric intensive care units were included. Out of 65 potential variables, 48 (74%) were considered to be indispensable, 16 (25%) were considered to be optional, and 1 (2%) was considered to be irrelevant. Out of 90 potential items, 62 (69%) were considered to be relevant, 23 (26%) were considered to be of little relevance, and 5 (6%) were considered to be irrelevant. Out of 17 potential subitems, 9 (53%) were considered to be relevant, 6 (35%) were considered to be of little relevance, and 2 (12%) were considered to be irrelevant. Conclusions The necessary variables that ought to be included in a French paediatric critical care registry were identified. The challenge now is to develop the French registry for paediatric intensive care units

    Green Edge ice camp campaigns : understanding the processes controlling the under-ice Arctic phytoplankton spring bloom

    The Green Edge initiative was developed to investigate the processes controlling the primary productivity and fate of organic matter produced during the Arctic phytoplankton spring bloom (PSB) and to determine its role in the ecosystem. Two field campaigns were conducted in 2015 and 2016 at an ice camp located on landfast sea ice southeast of Qikiqtarjuaq Island in Baffin Bay (67.4797∘ N, 63.7895∘ W). During both expeditions, a large suite of physical, chemical and biological variables was measured beneath a consolidated sea-ice cover from the surface to the bottom (at 360 m depth) to better understand the factors driving the PSB. Key variables, such as conservative temperature, absolute salinity, radiance, irradiance, nutrient concentrations, chlorophyll a concentration, bacteria, phytoplankton and zooplankton abundance and taxonomy, and carbon stocks and fluxes were routinely measured at the ice camp. Meteorological and snow-relevant variables were also monitored. Here, we present the results of a joint effort to tidy and standardize the collected datasets, which will facilitate their reuse in other Arctic studies

    Bone Marrow Mononuclear Cells Up-Regulate Toll-Like Receptor Expression and Produce Inflammatory Mediators in Response to Cigarette Smoke Extract

    Several reports link cigarette smoking with leukemia. However, the effects of cigarette smoke extract (CSE) on bone marrow hematopoiesis remain unknown. The objective of this study was to elucidate the direct effects of cigarette smoke on human bone marrow hematopoiesis and characterize the inflammatory process known to result from cigarette smoking. Bone marrow mononuclear cells (BMCs) from healthy individuals when exposed to CSE had significantly diminished CFU-E, BFU-E and CFU-GM. We found increased nuclear translocation of the NF-κB p65 subunit and, independently, enhanced activation of AKT and ERK1/2. Exposure of BMCs to CSE induced IL-8 and TGF-β1 production, which was dependent on NF-κB and ERK1/2, but not on AKT. CSE treatment had no effect on the release of TNF-α, IL-10, or VEGF. Finally, CSE also had a significant induction of TLR2, TLR3 and TLR4, out of which, the up-regulation of TLR2 and TLR3 was found to be dependent on ERK1/2 and NF-κB activation, but not AKT. These results indicate that CSE profoundly inhibits the growth of erythroid and granulocyte-macrophage progenitors in the bone marrow. Further, CSE modulates NF-κB- and ERK1/2-dependent responses, suggesting that cigarette smoking may impair bone marrow hematopoiesis in vivo as well as induce inflammation, two processes that proceed malignant transformation
