    Molecular and biochemical characterization of extracellular tannin acyl hydrolase activity from a Mexican isolate of Aspergillus niger

    "Microbial tannase, a hydrolysable tannin-degrading enzyme, is extensively used in manufacture of instant tea, beer, wine, and gallic acid. Aspergillus niger strain, obtained from a Mexican tannery wastewaters rich in gallic acid [Quebracho Phenolics-rich Tannery Wastewaters, (QPTW)], displayed a good growth and tannase activity in a minimal medium added with 1% (w/v) QPTW (Kr= 0.451 mm.h-1). Using PCR and RACE 3´ and 5´methodologies, a complete cDNA of a tannase was cloned from this isolate.Nucleotide sequence of complete cDNA was of 4690 bp with a complete ORF of 1833 bp encoding 611 amino acids. Transcriptionalinduction was observed in mineral medium added with carbon sources as tannic acid alone (1 and 10 g/l), as well as mix of glucose(1 and 10 g/l) and tannic acid (1 g/l) in the media. However, neither glucose (1 and 10 g/l) and sucrose (1 and 10 g/l) nor (+)-catechin(1 and 10 g/l) as sole carbon sources displayed gene induction in in vitro assays. A. niger-GTO is a new strain with interesting characteristics for industrial tannase production purposes.

    Biostimulant and Elicitor Responses to Cricket Frass (Acheta domesticus) in Tomato (Solanum lycopersicum L.) under Protected Conditions

    Agriculture in the current century is seeking sustainable tools in order to generate plant production systems with minimal negative environmental impact. In recent years it has been shown that the use of insect frass is an option to be used for this purpose. The present work studied the effect of low doses (0.1, 0.5, and 1.0% w/w) of cricket frass (Acheta domesticus) in the substrate during the cultivation of tomatos under greenhouse conditions. Plant performance and antioxidant enzymatic activities were measured in the study as explicative variables related to plant stress responses in order to determine possible biostimulant or elicitor effects of cricket frass treatments during tomato cultivation under greenhouse conditions. The main findings of this study indicated that tomato plants responded in a dose dependent manner to cricket frass treatments, recalling the hormesis phenomenon. On the one hand, a 0.1% (w/w) cricket frass treatment showed typical biostimulant features, while on the other hand, 0.5 and 1.0% treatments displayed elicitor effects in tomato plants under evaluated conditions in the present study. These results support the possibility that low doses of cricket frass might be used in tomato cultivation (and perhaps in other crops) for biostimulant/elicitor input into sustainable production systems

    Focusing on new food production methods and sustainable pillars’ accomplishments has changed the definition of sustainable pillars themselves. Moreover, some general characteristics of the main pillars can be redefined in separate dimensions to better explain their positive sustainable impacts. Therefore, the main objective of this research is to redefine the sustainable pillars linked to food production and review the most important cultural and technological sustainability impacts they have, in addition to the three classic pillars: economic, social, and environmental sustainability. Cultural and technological sustainability are increasingly important complements to the traditional sustainability concept. Furthermore, new food production technologies and systems are influenced by ancient production methods, as well as by profitable crop selection. Traditional agricultural and aquaculture production in relation to more recent aquaponic production concepts are still a major part of global food security, but the better usage of waste materials or residues generates a more favorable agroecological impact. In conclusion, constantly redefining the sustainable pillars in the context of sustainable food production methods and proving the viability of their general production impacts is important

    The quarantined bacterium Clavibacter michiganensis subsp. michiganensis (Cmm) causes significant losses on tomato plants worldwide, therefore efforts to reduce its damage are imperative. The alternatives offered by nanotechnology in the near future to be applied for the control of Cmm should be studied. Hence, this study aimed to determine the antibacterial activity of silver nanoparticles (AgNPs) biosynthesized on Moringa (Mo) leaves against Cmm in tomato plants (Solanum lycopersicum L.) and the possible protective mechanisms associated. UV–vis, TEM and FTIR, elucidated some structural and physicochemical characteristics of biosynthesized AgNPs. The preventive and curative effect of Mo extract and Mo-AgNPs at 50 ppm against Cmm in tomato plants was evaluated. Cmm was inoculated in the stems of tomato plants with 1 × 109 UFC/mL. Biometric parameters, disease severity, PAL, CAT, SOD activities, H2O2 levels, as well as chalcone synthase and pr1a gene expression were measured in greenhouse during 29 days. The UV–vis spectrum revealed the maximum absorption at 330 nm. Mo-AgNPs (32 nm; quasi-spherical shape) exhibited antibacterial potential against Cmm decreasing the disease severity by 86.29 % at 29 days after the treatments were applied. Besides, the Mo extract decreased the disease severity in 72 %, both compared to the control, being these two treatments the most promising for controlling Cmm. The Mo extract in preventive and curative treatments had the highest numbers of leaves, 33.31 %. The enzymatic activity exhibited no discernible pattern when comparing healthy and diseased plants, measured 29 days after the confrontations. Gene expression suggested a role of SAR pathway in the protective treatment by Mo Ag-NPs. In conclusion, the application of AgNPs derived from Mo leaf extracts effectively reduces the disease severity caused by Cmm on tomato plants under greenhouse conditions

    Precision agriculture has the objective of improving agricultural yields and minimizing costs by assisting management with the use of sensors, remote sensing, and information technologies. There are several approaches to improving crop yields where remote sensing has proven to be an important methodology to determine agricultural maps to show surface differences which may be associated with many phenomena. Remote sensing utilizes a wide variety of image sensors that range from common RGB cameras to sophisticated, hyper-spectral image cameras which acquire images from outside the visible electromagnetic spectrum. The NDVI and NGBVI are computer vision vegetation index algorithms that perform operations from color masks such as red, green, and blue from RGB cameras and hyper-spectral masks such as near-infrared (NIR) to highlight surface differences in the image to detect crop anomalies. The aim of the present study was to determine the relationship of NDVI and NGBVI as plant health indicators in tomato plants (Solanum lycopersicum) treated with the beneficial bacteria Bacillus cereus-Amazcala (B. c-A) as a protective agent to cope with Clavibacter michiganensis subsp. michiganensis (Cmm) infections. The results showed that in the presence of B. c-A after infection with Cmm, NDVI and NGBVI can be used as markers of plant weight and the activation of the enzymatic activities related to plant defense induction
