    "A new process for obtaining dibenzo[c,f][1,2,5]thiadiazepines (DBTDs) and their effects on GABAA receptors of guinea pig myenteric neurons are described. Synthesis of DBTD derivatives began with two commercial aromatic compounds. An azide group was obtained after two sequential reactions, and the central ring was closed via a nitrene to obtain the tricyclic sulfonamides (DBTDs). Whole-cell recordings showed that DBTDs application did not affect the holding current but inhibited the currents induced by GABA (IGABA), which are mediated by GABAA receptors. These DBTDs effects reached their maximum 3 min after application and were: (i) reversible, (ii) concentration-dependent (with a rank order of potency of 2c = 2d > 2b), (iii) mediated by a non-competitive antagonism, and (iv) only observed when applied extracellularly. Picrotoxin (which binds in the channel mouth) and DBTDs effects were not modified when both substances were simultaneous applied. Our results indicate that DBTD acted on the extracellular domain of GABAA channels but independent of the picrotoxin, benzodiazepine, and GABA binding sites. DBTDs used here could be the initial model for synthesizing new GABAA receptor inhibitors with a potential to be used as antidotes for positive modulators of these receptors or to induce experimental epilepsy.

    In this study, we sequenced the first complete chloroplast (cp) genome of a Peruvian chili pepper landrace, “arnacucho” (Capsicum chinense). This cp genome has a 156,931 bp in length with typical quadripartite structure, containing a large single copy (LSC) region (87,325 bp) and a 17,912 bp small single-copy (SSC) region, separated by two inverted repeat (IR) regions (25,847 bp); and the percentage of GC content was 37.71%. Arnaucho chili pepper chloroplast genome possesses 133 genes that consists of 86 protein-coding genes, 37 tRNA, eight rRNA, and two pseudogenes. Phylogenetic analysis revealed that this Peruvian chili pepper landrace is closely related to the undomesticated species C. galapagoense; all belong to the Capsiceae tribe

    Unlocking the Complete Chloroplast Genome of a Native Tree Species from the Amazon Basin, Capirona (Calycophyllum spruceanum Benth., Rubiaceae), and Its Comparative Analysis with Other Ixoroideae Species

    Capirona (Calycophyllum spruceanum Benth.) belongs to subfamily Ixoroideae, one of de major lineages in the Rubiaceae family, and is an important timber tree, with origin in the Amazon Basin and has widespread distribution in Bolivia, Peru, Colombia, and Brazil. In this study, we obtained the first complete chloroplast (cp) genome of capirona from department of Madre de Dios located in the Peruvian Amazon. High-quality genomic DNA was used to construct librar-ies. Pair-end clean reads were obtained by PE 150 library and the Illumina HiSeq 2500 platform. The complete cp genome of C. spruceanum has a 154,480 bp in length with typical quadripartite structure, containing a large single copy (LSC) region (84,813 bp) and a small single-copy (SSC) region (18,101 bp), separated by two inverted repeat (IR) regions (25,783 bp). The annotation of C. spruceanum cp genome predicted 87 protein-coding genes (CDS), 8 ribosomal RNA (rRNA) genes, 37 transfer RNA (tRNA) genes and 01 pseudogene. A total of 41 simple sequence repeats (SSR) of this cp genome were divided into mononucleotides (29), dinucleotides (5), trinucleotides (3), and tetranucleotide (4). Most of these repeats were distributed in the noncoding regions. Whole chloroplast genome comparison with the other six Ixoroideae species revealed that the small single copy and large single copy regions showed more divergence than invert regions. Finally, phylogenetic analysis resolved that C. spruceanum is a sister species to Emmenopterys henryi, and confirms its position within the subfamily Ixoroideae. This study reports for the first time the genome organization, gene content, and structural features of the chloroplast genome of C. spruceanum, providing valuable information for genetic and evolutionary studies in the genus Calycophyllum and beyond

    Revealing the Complete Chloroplast Genome of an Andean Horticultural Crop, Sweet Cucumber (Solanum muricatum), and Its Comparison with Other Solanaceae Species

    Sweet cucumber (Solanum muricatum) sect. Basarthrum is a neglected horticultural crop native of the Andean region. It is naturally distributed very close to other two Solanum crops of high importance, potatoes and tomatoes. To date, molecular tools for this crop are still undetermined. In this study, the complete sweet cucumber chloroplast (cp) genome was obtained and compared with seven Solanaceae species. The cp genome of S. muricatum had a 155,681 bp in length with included a large single copy (LSC) region of 86,182 bp and a small single-copy (SSC) region of 18,360 bp, separated by a pair of inverted repeats (IR) regions of 25,568 bp. The cp genome possessed 88 protein-coding genes (CDS), 37 transfer RNA (tRNA) genes, eight ribosomal RNA (rRNA) genes, and one pseudogene. Furthermore, 48 perfect microsatellites were identified, divided in mononucleotide repeats (32), followed by tetranucleotide (6) and dinucleotides (5). Microsatellites with trinucleotides repeats (3), pentanucleotide (1) and hexanucleotide (1) repeats motifs in these genomes were also identified, but in lower quantity. These repeats were mainly located in the noncoding regions. Whole cp genome comparative analysis revealed that the SSC and LSC regions showed more divergence than IR regions. Our phylogenetic analysis showed that S. muricatum is a sister species to members of sections Petota + Lycopersicum + Etuberosum. We expect to provide useful molecular data to shed light on the genetic diversity within sweet cucumber landrace, and also to determine the evolutionary processes in S. muricatum

    La actividad edificadora en Lima Metropolitana se ha visto afectada por el decrecimiento en su actividad, entre los años 2014 al 2017, teniendo en el año 2018 un crecimiento de 1.38 millones de m2 comparados al año 2017, pero siempre dentro de un proceso de incertidumbre que no brinda la seguridad y confianza a nuevos inversionistas. Siendo el mercado más competitivo, las empresas deben aplicar un sistema de gestión que estandarice el control de recursos y la mejora continua. Nuestra propuesta está orientado al uso y aplicación de las buenas prácticas que la Guía de los fundamentos para la dirección de proyecto (Guía del PMBOK®), 6ª ed., y Construction Extension to the PMBOK® Guide del 2016. El trabajo está basado en una obra adjudicada a A. Chávez Constructores E.I.R.L. en el distrito de Barranco cuya característica es un edificio multifamiliar de 6 pisos más azotea, con semisótano y tres sótanos de estacionamiento, el cual se está gestionado con un staff básico propuesto, con la finalidad de asegurar que el negocio sea sostenible en el tiempo y capaz de soportar las caídas del sector y sobre todo que su inversión tenga los resultados esperados, para ello proponemos un documento de gestión a la medida del proyecto. Estos procesos permitirán a la organización adquirir nuevos conocimientos y buenas prácticas para optimizar los recursos y lograr mejores resultados para el alcance de sus objetivos, y con ello alinearse con la misión y visión de la empresa y poder implementarlo en sus proyectos futuros.Building activity in Metropolitan Lima has been affected by the decrease in its activity, between 2014 and 2017, with a growth of 1.38 million m2 in 2018 compared to 2017, but always within a process of uncertainty that it does not provide security and confidence to new investors. Being the most competitive market, companies must apply a management system that standardizes the control of resources and continuous improvement. Our proposal is oriented to the use and application of the good practices that the Guide of the fundamentals for project management (PMBOK® Guide), Sixth Edition, and Construction Extension to the PMBOK® Guide of 2016. The work is based on a work awarded to A. Chávez Constructores E.I.R.L. in the Barranco district whose characteristic is a 6-story multifamily building plus a roof terrace, with a semi-basement and three parking basements, which is being managed with a proposed basic staff, in order to ensure that the business is sustainable over time and capable of withstanding the downturns of the sector and above all that your investment has the expected results, for this we propose a management document tailored to the project. These processes will allow the organization to acquire new knowledge and good practices to optimize resources and achieve better results for the achievement of its objectives, and thereby align with the mission and vision of the company and be able to implement it in its future projects.Trabajo de investigació

    Compromising between European and US allergen immunotherapy schools: Discussions from GUIMIT, the Mexican immunotherapy guidelines

    Background: Allergen immunotherapy (AIT) has a longstanding history and still remains the only disease-changing treatment for allergic rhinitis and asthma. Over the years 2 different schools have developed their strategies: the United States (US) and the European. Allergen extracts available in these regions are adapted to local practice. In other parts of the world, extracts from both regions and local ones are commercialized, as in Mexico. Here, local experts developed a national AIT guideline (GUIMIT 2019) searching for compromises between both schools. Methods: Using ADAPTE methodology for transculturizing guidelines and AGREE-II for evaluating guideline quality, GUIMIT selected 3 high-quality Main Reference Guidelines (MRGs): the European Academy of Allergy, Asthma and Immunology (EAACI) guideines, the S2k guideline of various German-speaking medical societies (2014), and the US Practice Parameters on Allergen Immunotherapy 2011. We formulated clinical questions and based responses on the fused evidence available in the MRGs, combined with local possibilities, patient's preference, and costs. We came across several issues on which the MRGs disagreed. These are presented here along with arguments of GUIMIT members to resolve them. GUIMIT (for a complete English version, see Supplementary data) concluded the following: Results: Related to the diagnosis of IgE-mediated respiratory allergy, apart from skin prick testing complementary tests (challenges, in vitro testing and molecular such as species-specific allergens) might be useful in selected cases to inform AIT composition. AIT is indicated in allergic rhinitis and suggested in allergic asthma (once controlled) and IgE-mediated atopic dermatitis. Concerning the correct subcutaneous AIT dose for compounding vials according to the US school: dosing tables and formula are given; up to 4 non-related allergens can be mixed, refraining from mixing high with low protease extracts. When using European extracts: the manufacturer's indications should be followed; in multi-allergic patients 2 simultaneous injections can be given (100% consensus); mixing is discouraged. In Mexico only allergoid tablets are available; based on doses used in all sublingual immunotherapy (SLIT) publications referenced in MRGs, GUIMIT suggests a probable effective dose related to subcutaneous immunotherapy (SCIT) might be: 50–200% of the monthly SCIT dose given daily, maximum mixing 4 allergens. Also, a table with practical suggestions on non-evidence-existing issues, developed with a simplified Delphi method, is added. Finally, dissemination and implementation of guidelines is briefly discussed, explaining how we used online tools for this in Mexico. Conclusions: Countries where European and American AIT extracts are available should adjust AIT according to which school is followed

    El presente trabajo tiene por objetivo determinar la importancia del Órgano de Solución de Diferencias de la OMC dentro de las negociaciones sobre el acceso a los mercados internacionales. El área que se está investigando es el funcionamiento de la OMC especialmente, cómo esta resuelve las diferencias entre países, especialmente teniendo en cuenta el análisis de costo-beneficio que se incurre. La investigación se lleva a cabo a través de la revisión de fuentes primarias y secundarias con un enfoque cualitativo y comparativo así como perspectiva analítica y descriptiva. El resultado que se obtuvo después de estudiar dos casos específicamente en Ecuador, nos muestra cómo el organismo proporciona buenas oportunidades de desarrollo comercial a nivel mundial con ahorro de recursos. Se recomienda que los países hagan conciencia y permitan que estos organismos les asesoren al momento de tomar decisiones y resolver conflictos en búsqueda de un comercio más eficaz y eficiente
