Abstract : All discourse presupposes an ethos, which are constituted as an important argumentative strategy, and, therefore, determinant and enlightener from the discursive effectiveness. The central purpose of this work is to investigate the linguistics mechanisms and the discursive procedures mobilized on the ethos journalistic construction. For that, undertakes a theoretical reflection located in the interface between the Pragmatics and the Discourse Analysis, resulting in a hybrid theoretical-methodological approach, which has revealed suitable for ethos apprehension, a notion of vocation clearly interdisciplinary. The discursive analysis of the elements manipulated under the construction of the journalistic ethos has as objects the texts carried in the editorial section of the CartaCapital magazine and prioritizes the concepts of the scenography and general scene and discursive heterogeneity expressions. The results have revealed that the editorialist call up consistent values with the previously constructed image of the magazine and associated stereotypes to the ethical world of the Journalism

Two Colors, One Species: The Case of Melissodes nigroaenea (Apidae: Eucerini), an Important Pollinator of Cotton Fields in Brazil

Accurate taxonomic delimitation in ecological research is absolutely critical as studies that seek to evaluate levels of biodiversity and qualify human effects on the environment are rapidly undertaken. Coloration is a widely used morphological character for species identification through dichotomous keys. However, taxonomic identification based upon coloration is often unreliable because this character can exhibit high degree of intraspecific variation. In this study, we use a DNA barcoding approach to investigate the interpretation of two color morphs (yellow or dark) in the eucerine bee Melissodes nigroaenea. Our hypothesis is that if significant genetic divergence exists between each morphotype of M. nigroaenea, coloration reflects two distinct evolutionary lineages within this species, which may require taxonomic revision. Our alternative hypothesis is that, if genetic divergence is low between each morphotype of M. nigroaenea, we can attribute this variation to color polymorphism. Our Bayesian phylogenetic reconstruction revealed that both yellow and black individuals clustered together in a highly supported phylogenetic group. Additionally, pairwise genetic distances between M. nigroaenea color morphotypes were lower than 3%. These results indicate that both mesosome color morphs correspond to intraspecific variability within the same evolutionary unit. Together, our results indicate that mesosome coloration is not a reliable character for taxonomic differentiation of these Melissodes species, and that the incorporation of DNA barcoding approaches to taxonomic classification can help resolve some of the problems that originate while relying on purely morphological taxonomy

The species is a native tree from the Atlantic Forest biome commonly used in forest restoration; it has medicinal potential and the wood is economically useful. Methods and Results: Twenty-eight microsatellite loci were identified from an enriched genomic library. Fifteen loci resulted in successful amplifications and were characterized in a natural population of 94 individuals. Twelve loci were polymorphic, with allele numbers ranging from three to 15 per locus, and expected and observed heterozygosities ranging from 0.2142 to 0.8325 and 0.190 to 0.769, respectively. Conclusions: The developed markers will be used in further studies of population genetics of P. gonoacantha, aimed at conservation and management of the species in natural populations and in forest restoration projects.32Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)FAPESP [Biota/FAPESP-11/50296-8]CNPq [CNPq-140036/2011-3

Introduction: The aim of this study was to evaluate the effectiveness of 2.5% sodium hypochlorite associated with 17% Ethylenediaminetetraacetic acid (NaOCl-EDTA), versus that of 1% peracetic acid (PA), in removing the smear layer, as assessed by scanning electron microscopy (SEM), and in exerting bactericidal action against Enterococcus faecalis (E. faecalis), as assessed by the real-time polymerase chain reaction (real-time PCR). Methods and Materials: Fifty-five extracted mandibular single-rooted premolars were selected, and divided into two experimental groups (NaOCl-EDTA and PA; n=25) and one control group (0.9% saline; n=5). Pre- and post-instrumentation samples were collected and assessed for the presence of E. faecalis using real-time PCR. The teeth were instrumented using hand files and the ProTaper Universal system (hybrid technique) for a standardized time of 7 min. A total of 20 mL of NaOCl followed by 5 mL of EDTA were applied during instrumentation in the NaOCl-EDTA group, whereas 20 mL of PA and 20 mL of saline were applied in the PA and control groups, respectively. An additional 5 mL of saline was applied in all the groups to neutralize the environment. A scoring system was used to conduct the SEM assessment. The results were submitted to the Kruskal-Wallis test, complemented by Dunn's test (SEM analysis) (P<0.05). Results: A significant microbial reduction was observed in both the PA and the NaOCl-EDTA groups (P<0.05). In the PA group, the presence of a smear layer in the apical third was significantly greater than in the cervical third (P<0.05); no significant differences were observed between the middle and cervical thirds, or between the middle and apical thirds (P>0.05). In the NaOCl-EDTA group, the smear layer scores were significantly higher in the apical third than in the cervical and middle thirds (P<0.05). Conclusion: This in vitro study showed that there was no significant difference between PA and NaOCl-EDTA irrigation regimens regarding either antimicrobial action against E. faecalis or removal of the smear layer, except for greater removal in the middle third by the NaOCl-EDTA group. Keywords: Endodontic Irrigation; Enterococcus faecalis; Peracetic Acid; Real-Time Polymerase Chain Reaction; Sodium Hypochlorite
