Exploring the culture of Greek children’s musical games in the school playground: An ethnographic study
This article reports findings from an ethnographic study investigating the culture of children’s musical games played in school playgrounds. The research took place in nine primary schools in Greece and lasted for 6 months. Data collection methods included open observation of children aged 6 to 11 years, focused small-group observation, semi-structured interviews of 53 children (aged 8–11 years), and video recordings of the children’s musical games. Analyses of the research data, in light of the relevant literature,
revealed the physical and human geographies of musical games, gender preferences, transmission sources and processes, learning and teaching practices, improvisations and variations, and communication among participants. The article concludes with implications for music education research and practice
Time, timing, talking and training : findings from an exploratory action research study to improve quality of end of life care for minority ethnic kidney patients
Background. With an ageing and increasingly diverse population at risk from rising levels of obesity, diabetes and cardiovascular disease, including kidney complications, there is a need to provide quality care at all stages in the care pathway including at the end of life and to all patients. Aim. This study purposively explored South Asian patients' experiences of kidney end of life care to understand how services can be delivered in a way that meets diverse patient needs. Methods. Within an action research design 14 focus groups (45 care providers) of kidney care providers discussed the recruitment and analysis of individual interviews with 16 South Asian kidney patients (eight men, eight women). Emergent themes from the focus groups were analysed thematically. The research took place at four UK centres providing kidney care to diverse populations: West London, Luton, Leicester and Bradford. Results. Key themes related to time and the timing of discussions about end of life care and the factors that place limitations on patients and providers in talking about end of life care. Lack of time and confidence of nurses in areas of kidney care, individual attitudes and workforce composition influence whether and how patients have access to end of life care through kidney services. Conclusion. Training, team work and time to discuss overarching issues (including timing and communication about end of life) with colleagues could support service providers to facilitate access and delivery of end of life care to this group of patients.Peer reviewedFinal Published versio
Population genetic structure of the parasite Anisakis simplex (s. s.) collected in Clupea harengus L. from North East Atlantic fishing grounds
The Atlantic herring is a schooling, pelagic species that inhabits both sides of the North Atlantic Ocean. Herring
stock identification is usually based on several approaches, including fish meristic characters, population genetic
analysis and the use of parasite species composition. A total of 654 Anisakis spp. larvae collected from herring of
four fishing grounds in the Norwegian Sea, Baltic Sea, North Sea, and the English Channel off the French coast,
was identified to species level using diagnostic allozymes and sequence analysis of EF1 α−1 nDNA and the
mtDNA cox2 genes. Population genetic differentiation of Anisakis simplex (s. s.) among the different fishing areas
was estimated, at the intraspecific level, on the basis of mtDNA cox2 sequences analysis. Spatial comparison
based on molecular variance analysis and Fst values was performed for the collected specimens (among regions).
Haplotype network construction showed relevant differences in haplotype frequencies between samples of A.
simplex (s. s.) from the different geographical areas. Results indicate a genetic sub-structuring of A. simplex (s. s.)
obtained from herring in different areas, with the population from the Norwegian Sea being the most differentiated one, and with North Sea and Baltic Sea populations being most similar. The population genetic structure
of A. simplex (s. s.) was in accordance with the herring population genetic structure throughout the host’s
geographical range in the NE Atlantic. Results suggest that mtDNA cox2 is a suitable genetic marker for A.
simplex (s. s.) population genetic structure analysis and a valuable tool to elucidate the herring stock structure in
the NE Atlantic Ocean
Resveratrol augments ER stress and the cytotoxic effects of glycolytic inhibition in neuroblastoma by downregulating Akt in a mechanism independent of SIRT1
Cancer cells typically display increased rates of aerobic glycolysis that are correlated with tumor aggressiveness and a poor prognosis. Targeting the glycolytic pathway has emerged as an attractive therapeutic route mainly because it should spare normal cells. Here, we evaluate the effects of combining the inhibition of glycolysis with application of the polyphenolic compound resveratrol (RSV) in neuroblastoma (NB) cancer cell lines. Inhibiting glycolysis with 2-deoxy-D-glucose (2-DG) significantly reduced NB cell viability and was associated with increased endoplasmic reticulum (ER) stress and Akt activity. Administration of 2-DG increased the expression of the ER molecular chaperones GRP78 and GRP94, the prodeath protein C/EBP homology protein (CHOP) and the phosphorylation of Akt at S473, T450 and T308. Combined treatment with both RSV and 2-DG reduced GRP78, GRP94 and Akt phosphorylation but increased CHOP and NB cell death when compared with the administration of 2-DG alone. The selective inhibition of Akt activity also decreased 2-DG-induced GRP78 and GRP94 expression and increased CHOP expression, suggesting that Akt can modulate ER stress. Protein phosphatase 1 alpha (PP1 alpha) was activated by RSV, as indicated by a reduction in PP1 alpha phosphorylation at T320. Pretreatment of cells with tautomycin, a selective PP1 alpha inhibitor, prevented the RSV-mediated decrease in Akt phosphorylation, suggesting that RSV enhances 2-DG-induced cell death by activating PP1 and downregulating Akt. The RSV-mediated inhibition of Akt in the presence of 2-DG was not prevented by the selective inhibition of SIRT1, a known target of RSV, indicating that the effects of RSV on this pathway are independent of SIRT1. We propose that RSV inhibits Akt activity by increasing PP1 alpha activity, thereby potentiating 2-DG-induced ER stress and NB cell death
Characterisation of the natural environment: quantitative indicators across Europe
BACKGROUND: The World Health Organization recognises the
importance of natural environments for human health. Evidence
for natural environment-health associations comes largely from
single countries or regions, with varied approaches to measuring
natural environment exposure. We present a standardised approach
to measuring neighbourhood natural environment exposure in
cities in different regions of Europe. METHODS: The Positive
Health Effects of the Natural Outdoor environment in TYPical
populations of different regions in Europe (PHENOTYPE) study
aimed to explore the mechanisms linking natural environment
exposure and health in four European cities (Barcelona, Spain;
Doetinchem, the Netherlands; Kaunas, Lithuania; and
Stoke-on-Trent, UK). Common GIS protocols were used to develop a
hierarchy of natural environment measures, from simple measures
(e.g., NDVI, Urban Atlas) using Europe-wide data sources, to
detailed measures derived from local data that were specific to
mechanisms thought to underpin natural environment-health
associations (physical activity, social interaction, stress
reduction/restoration). Indicators were created around
residential addresses for a range of straight line and network
buffers (100 m-1 km). RESULTS: For simple indicators derived
from Europe-wide data, we observed differences between cities,
which varied with different indicators (e.g., Kaunas and
Doetinchem had equal highest mean NDVI within 100 m buffer, but
mean distance to nearest natural environment in Kaunas was more
twice that in Doetinchem). Mean distance to nearest natural
environment for all cities suggested that most participants
lived close to some kind of natural environments (64 +/- 58-363
+/- 281 m; mean 180 +/- 204 m). The detailed classification
highlighted marked between-city differences in terms of
prominent types of natural environment. Indicators specific to
mechanisms derived from this classification also captured more
variation than the simple indicators. Distance to nearest and
count indicators showed clear differences between cities, and
those specific to the mechanisms showed within-city differences
for Barcelona and Doetinchem. CONCLUSIONS: This paper
demonstrates the feasibility and challenges of creating
comparable GIS-derived natural environment exposure indicators
across diverse European cities. Mechanism-specific indicators
showed within- and between-city variability that supports their
utility for ecological studies, which could inform more specific
policy recommendations than the traditional proxies for natural
environment access
Magnetic separation of peripheral nerve-resident cells underscores key molecular features of human Schwann cells and fibroblasts: an immunochemical and transcriptomics approach
Nerve-derived human Schwann cell (SC) cultures are irreplaceable models for basic and translational research but their use can be limited due to the risk of fibroblast overgrowth. Fibroblasts are an ill-defined population consisting of highly proliferative cells that, contrary to human SCs, do not undergo senescence in culture. We initiated this study by performing an exhaustive immunological and functional characterization of adult nerve-derived human SCs and fibroblasts to reveal their properties and optimize a protocol of magnetic-activated cell sorting (MACS) to separate them effectively both as viable and biologically competent cells. We next used immunofluorescence microscopy imaging, flow cytometry analysis and next generation RNA sequencing (RNA-seq) to unambiguously characterize the post-MACS cell products. High resolution transcriptome profiling revealed the identity of key lineage-specific transcripts and the clearly distinct neural crest and mesenchymal origin of human SCs and fibroblasts, respectively. Our analysis underscored a progenitor- or stem cell-like molecular phenotype in SCs and fibroblasts and the heterogeneity of the fibroblast populations. In addition, pathway analysis of RNA-seq data highlighted putative bidirectional networks of fibroblast-to-SC signaling that predict a complementary, yet seemingly independent contribution of SCs and fibroblasts to nerve regeneration. In sum, combining MACS with immunochemical and transcriptomics approaches provides an ideal workflow to exhaustively assess the identity, the stage of differentiation and functional features of highly purified cells from human peripheral nerve tissues.Fil: Peng, Kaiwen. Exploring access to end of life care for ethnic minorities with end stage kidney disease through recruitment in action research
BACKGROUND: Variation in provision of palliative care in kidney services and practitioner concerns to provide equitable access led to the development of this study which focussed on the perspectives of South Asian patients and their care providers. As people with a South Asian background experience a higher risk of Type 2 Diabetes (T2DM) and end stage kidney failure (ESKF) compared to the majority population but wait longer for a transplant, there is a need for end of life care to be accessible for this group of patients. Furthermore because non English speakers and people at end of life are often excluded from research there is a dearth of research evidence with which to inform service improvement. This paper aims to explore issues relating to the process of recruitment of patients for a research project which contribute to our understanding of access to end of life care for ethnic minority patients in the kidney setting. METHODS: The study employed an action research methodology with interviews and focus groups to capture and reflect on the process of engaging with South Asian patients about end of life care. Researchers and kidney care clinicians on four NHS sites in the UK recruited South Asian patients with ESKF who were requiring end of life care to take part in individual interviews; and other clinicians who provided care to South Asian kidney patients at end of life to take part in focus groups exploring end of life care issues. In action research planning, action and evaluation are interlinked and data were analysed with emergent themes fed back to care providers through the research cycle. Reflections on the process of patient recruitment generated focus group discussions about access which were analysed thematically and reported here. RESULTS: Sixteen patients were recruited to interview and 45 different care providers took part in 14 focus groups across the sites. The process of recruiting patients to interview and subsequent focus group data highlighted some of the key issues concerning access to end of life care. These were: the identification of patients approaching end of life; and their awareness of end of life care; language barriers and informal carers' roles in mediating communication; and contrasting cultures in end of life kidney care. CONCLUSIONS: Reflection on the process of recruitment in this action research study provided insight into the complex scenario of end of life in kidney care. Some of the emerging issues such as the difficulty identifying patients are likely to be common across all patient groups, whilst others concerning language barriers and third party communication are more specific to ethnic minorities. A focus on South Asian ethnicity contributes to better understanding of patient perspectives and generic concepts as well as access to end of life kidney care for this group of patients in the UK. "On the Spot": travelling artists and Abolitionism, 1770-1830
Until recently the visual culture of Atlantic slavery has rarely been critically scrutinised. Yet in the first decades of the nineteenth century slavery was frequently represented by European travelling artists, often in the most graphic, sometimes voyeuristic, detail. This paper examines the work of several itinerant artists, in particular Augustus Earle (1793-1838) and Agostino Brunias (1730–1796), whose very mobility along the edges of empire was part of a much larger circulatory system of exchange (people, goods and ideas) and diplomacy that characterised Europe’s Age of Expansion. It focuses on the role of the travelling artist, and visual culture more generally, in the development of British abolitionism between 1770 and 1830. It discusses the broad circulation of slave imagery within European culture and argues for greater recognition of the role of such imagery in the abolitionist debates that divided Britain. Furthermore, it suggests that the epistemological authority conferred on the travelling artist—the quintessential eyewitness—was key to the rhetorical power of his (rarely her) images.
Artists such as Earle viewed the New World as a boundless source of fresh material that could potentially propel them to fame and fortune. Johann Moritz Rugendas (1802-1858), on the other hand, was conscious of contributing to a global scientific mission, a Humboldtian imperative that by the 1820s propelled him and others to travel beyond the traditional itinerary of the Grand Tour. Some artists were implicated in the very fabric of slavery itself, particularly those in the British West Indies such as William Clark (working 1820s) and Richard Bridgens (1785-1846); others, particularly those in Brazil, expressed strong abolitionist sentiments. Fuelled by evangelical zeal to record all aspects of the New World, these artists recognised the importance of representing the harsh realities of slave life. Unlike those in the metropole who depicted slavery (most often in caustic satirical drawings), many travelling artists believed strongly in the evidential value of their images, a value attributed to their global mobility. Protocol for the development of guidance for stakeholder engagement in health and healthcare guideline development and implementation
Stakeholder engagement has become widely accepted as a necessary component of guideline development and implementation. While frameworks for developing guidelines express the need for those potentially affected by guideline recommendations to be involved in their development, there is a lack of consensus on how this should be done in practice. Further, there is a lack of guidance on how to equitably and meaningfully engage multiple stakeholders. We aim to develop guidance for the meaningful and equitable engagement of multiple stakeholders in guideline development and implementation.
METHODS:
This will be a multi-stage project. The first stage is to conduct a series of four systematic reviews. These will (1) describe existing guidance and methods for stakeholder engagement in guideline development and implementation, (2) characterize barriers and facilitators to stakeholder engagement in guideline development and implementation, (3) explore the impact of stakeholder engagement on guideline development and implementation, and (4) identify issues related to conflicts of interest when engaging multiple stakeholders in guideline development and implementation.
DISCUSSION:
We will collaborate with our multiple and diverse stakeholders to develop guidance for multi-stakeholder engagement in guideline development and implementation. We will use the results of the systematic reviews to develop a candidate list of draft guidance recommendations and will seek broad feedback on the draft guidance via an online survey of guideline developers and external stakeholders. An invited group of representatives from all stakeholder groups will discuss the results of the survey at a consensus meeting which will inform the development of the final guidance papers. Our overall goal is to improve the development of guidelines through meaningful and equitable multi-stakeholder engagement, and subsequently to improve health outcomes and reduce inequities in health
Dominance is common in mammals and is associated with trans-acting gene expression and alternative splicing
Background:
Dominance and other non-additive genetic effects arise from the interaction between alleles, and historically these phenomena play a major role in quantitative genetics. However, most genome-wide association studies (GWAS) assume alleles act additively.

Results:
We systematically investigate both dominance—here representing any non-additive within-locus interaction—and additivity across 574 physiological and gene expression traits in three mammalian stocks: F2 intercross pigs, rat heterogeneous stock, and mice heterogeneous stock. Dominance accounts for about one quarter of heritable variance across all physiological traits in all species. Hematological and immunological traits exhibit the highest dominance variance, possibly reflecting balancing selection in response to pathogens. Although most quantitative trait loci (QTLs) are detectable as additive QTLs, we identify 154, 64, and 62 novel dominance QTLs in pigs, rats, and mice respectively that are undetectable as additive QTLs. Similarly, even though most cis-acting expression QTLs are additive, gene expression exhibits a large fraction of dominance variance, and trans-acting eQTLs are enriched for dominance. Genes causal for dominance physiological QTLs are less likely to be physically linked to their QTLs but instead act via trans-acting dominance eQTLs. In addition, thousands of eQTLs are associated with alternatively spliced isoforms with complex additive and dominant architectures in heterogeneous stock rats, suggesting a possible mechanism for dominance.

Conclusions:
Although heritability is predominantly additive, many mammalian genetic effects are dominant and likely arise through distinct mechanisms. It is therefore advantageous to consider both additive and dominance effects in GWAS to improve power and uncover causality
