    Fostering children's relationship with nature : exploring the potential of Forest school

    Forest School offers children opportunities to explore the natural environment regularly over an extended period of time. It is based on six key principles, one of which states Forest School fosters a relationship with nature and develops long-term pro-environment attitudes through regular personal experiences in nature. We consider to what extent this aim is achieved by exploring the published evidence. We identify that a relationship with nature could be demonstrated under six themes although there is little robust evidence to support this. We suggest further research is carried out directly linked to this aim of Forest School

    One is Not Enough: Multiple Exemplars Facilitate Infants' Generalizations of Novel Properties

    Across three experiments, we examined 9- and 11-month-oldsā€™ mappings of novel sound properties to novel animal categories. Infants were familiarized with novel animalā€“novel sound pairings (e.g., Animal A [red]ā€“Sound 1) and then tested on: (1) their acquisition of the original pairing and (2) their gen- eralization of the sound property to a new member of a familiarized cate- gory (e.g., Animal A [blue]ā€“Sound 1). When familiarized with a single exemplar of a category, 11-month-olds showed no evidence of acquiring or generalizing the animalā€“sound pairings. In contrast, 11-month-olds learnt the original animalā€“sound mappings and generalized the sound property to a novel member of that category when familiarized with multiple exemplars of a category. Finally, when familiarized with multiple exemplars, 9-month- old infants learnt the original animalā€“sound pairing, but did not extend the novel sound property. The results of these experiments provide evidence for developmental differences in the facilitative role of multiple exemplars in promoting the learning and generalization of information

    Using RNA-seq to characterize responses to 4 hydroxyphenylpyruvate dioxygenase (HPPD) inhibitor herbicide resistance in waterhemp (Amaranthus tuberculatus)

    Background: Waterhemp (Amaranthus tuberculatus (Moq.) J.D. Sauer) is a problem weed commonly found in the Midwestern United States that can cause crippling yield losses for both maize (Zea mays L.) and soybean (Glycine max L. Merr). In 2011, 4-hydroxyphenylpyruvate-dioxygenase (HPPD, EC 1.13.11.27) inhibitor herbicide resistance was first reported in two waterhemp populations. Since the discovery of HPPD-herbicide resistance, studies have identified the mechanism of resistance and described the inheritance of the herbicide resistance. However, no studies have examined genome-wide gene expression changes in response to herbicide treatment in herbicide resistant and susceptible waterhemp. Results: We conducted RNA-sequencing (RNA-seq) analyses of two waterhemp populations (HPPD-herbicide resistant and susceptible), from herbicide-treated and mock-treated leaf samples at three, six, twelve, and twenty-four hours after treatment (HAT). We performed a de novo transcriptome assembly using all sample sequences. Following assessments of our assembly, individual samples were mapped to the de novo transcriptome allowing us to identify transcripts specific to a genotype, herbicide treatment, or time point. Our results indicate that the response of HPPDherbicide resistant and susceptible waterhemp genotypes to HPPD-inhibiting herbicide is rapid, established as soon as 3 hours after herbicide treatment. Further, there was little overlap in gene expression between resistant and susceptible genotypes, highlighting dynamic differences in response to herbicide treatment. In addition, we used stringent analytical methods to identify candidate single nucleotide polymorphisms (SNPs) that distinguish the resistant and susceptible genotypes. Conclusions: The waterhemp transcriptome, herbicide-responsive genes, and SNPs generated in this study provide valuable tools for future studies by numerous plant science communities. This collection of resources is essential to study and understand herbicide effects on gene expression in resistant and susceptible weeds. Understanding how herbicides impact gene expression could allow us to develop novel approaches for future herbicide development. Additionally, an increased understanding of the prolific traits intrinsic in weed success could lead to crop improvement

    Progression of phosphine resistance in susceptible Tribolium castaneum (Herbst) populations under different immigration regimes and selection pressures

    Insecticide resistance is anĀ escalating global issue for a wide variety of agriculturally important pests. The genetic basis and biochemical mechanisms of resistance are well characterized in some systems, but little is known about the ecological aspects of insecticide resistance. We therefore designed a laboratory experiment to quantify the progressionĀ of phosphine resistance in Tribolium castaneum populations subject to different immigration regimes and selection pressures. Mated resistant females were added to originally susceptible populations under two distinct migration rates, and in addition, half of the populations in each migration treatment were exposed to selection pressures from phosphine fumigation.Ā The progression of phosphine resistance was assessed by screening beetles for the resistance allele at rph2. Phosphine resistance increased slowly in the low migration treatment and in the absence of selection, as expected. But at the higher migration rate, the increase in frequency of the resistance allele was lower than predicted. These outcomes result from the high levels of polyandry known in T.Ā castaneum females in the laboratory, because most of the Generation 1 offspring (86%) were heterozygous for theĀ rph2Ā allele, probably because resistant immigrant females mated again on arrival. Phosphine resistance was not fixed by fumigation as predicted, perhaps because susceptible gametes and eggs survived fumigation within resistant females. In terms of phosphine resistance progression in populations exposed to selection, the effect of fumigation negated the difference in migration rates. These results demonstrate how species-specific traits relating to the mating system may shape the progression of insecticide resistance within populations, and they have broad implications for the management of phosphine resistance inĀ T.Ā castaneum in the field. ā€‹We specify and discuss how these mating system attributes need to be accounted for when developing guidelines for resistance management

    Women with coronary artery disease report worse health-related quality of life outcomes compared to men

    BACKGROUND: Although there have been substantial medical advances that improve the outcomes following cardiac ischemic events, gender differences in the treatment and course of recovery for patients with coronary artery disease (CAD) continue to exist. There is a general paucity of data comparing the health related quality of life (HRQOL) in men and women undergoing treatment for CAD. The purpose of this study was to compare HRQOL outcomes of men and women in Alberta, at one-year following initial catheterization, after adjustment for known demographic, co-morbid, and disease severity predictors of outcome. METHOD: The HRQOL outcome data were collected by means of a self-reported questionnaire mailed to patients on or near the one-year anniversary of their initial cardiac catheterization. Using the Seattle Angina Questionnaire (SAQ), 5 dimensions of HRQOL were measured: exertional capacity, anginal stability, anginal frequency, quality of life and treatment satisfaction. Data from the APPROACH registry were used to risk-adjust the SAQ scale scores. Two analytical strategies were used including general least squares linear modeling, and proportional odds modeling sometimes referred to as the "ordinal logistic modeling". RESULTS: 3392 (78.1%) patients responded to the follow-up survey. The adjusted proportional odds ratios for men relative to women (PORs > 1 = better) indicated that men reported significantly better HRQOL on all 5 SAQ dimensions as compared to women. (PORs: Exertional Capacity 3.38 (2.75ā€“4.15), Anginal Stability 1.23 (1.03ā€“1.47), Anginal Frequency 1.70 (1.43ā€“2.01), Treatment Satisfaction 1.27 (1.07ā€“1.50), and QOL 1.74 (1.48ā€“2.04). CONCLUSIONS: Women with CAD consistently reported worse HRQOL at one year follow-up compared to men. These findings underline the fact that conclusions based on research performed on men with CAD may not be valid for women and that more gender-related research is needed. Future studies are needed to further examine gender differences in psychosocial adjustment following treatment for CAD, as adjustment for traditional clinical variables fails to explain sex differences in health related quality of life outcomes

    Can Forelā€“Ule Index Act as a Proxy of Water Quality in Temperate Waters? Application of Plume Mapping in Liverpool Bay, UK

    The use of ocean colour classification algorithms, linked to water quality gradients, can be a useful tool for mapping river plumes in both tropical and temperate systems. This approach has been applied in operational water quality programs in the Great Barrier Reef to map river plumes and assess trends in marine water composition and ecosystem health during flood periods. In this study, we used the Forelā€“Ule colour classification algorithm for Sentinel-3 OLCI imagery in an automated process to map monthly, annual and long-term plume movement in the temperate coastal system of Liverpool Bay (UK). We compared monthly river plume extent to the river flow and in situ water quality data between 2017ā€“2020. The results showed a strong positive correlation (Spearmanā€™s rho = 0.68) between the river plume extent and the river flow and a strong link between the FUI defined waterbodies and nutrients, SPM, turbidity and salinity, hence the potential of the Forelā€“Ule index to act as a proxy for water quality in the temperate Liverpool Bay water. The paper discusses how the Forelā€“Ule index could be used in operational water quality programs to better understand river plumes and the land-based inputs to the coastal zones in UK waters, drawing parallels with methods that have been developed in the GBR and Citclops project. Overall, this paper provides the first insight into the systematic long-term river plume mapping in UK coastal waters using a fast, cost-effective, and reproducible workflow. The study created a novel water assessment typology based on the common physical, chemical and biological ocean colour properties captured in the Forelā€“Ule index, which could replace the more traditional eutrophication assessment regions centred around strict geographic and political boundaries. Additionally, the Forelā€“Ule assessment typology is particularly important since it identifies areas of the greatest impact from the land-based loads into the marine environment, and thus potential risks to vulnerable ecosystems

    Riding the knowledge translation roundabout: lessons learned from the Canadian Institutes of Health Research Summer Institute in knowledge translation

    <p>Abstract</p> <p>Background</p> <p>Funding the education and training of the next generation of health researchers is a key mandate of the Canadian Institutes of Health Research (CIHR) knowledge translation (KT) portfolio. The field of KT is growing daily; thus, the training and development of a new generation of KT researchers is essential.</p> <p>Methods</p> <p>Using curriculum documents, participant evaluations, and self-reflection, this paper describes a unique Summer Institute hosted by the CIHR in Cornwall, Ontario, Canada. We outline the key aspects of a successful training initiative that could inform organizations and agencies worldwide with an interest in or who have a mandate for KT.</p> <p>Results</p> <p>This work provides potential funders, faculty, and students with an inside look into the purpose, process, and outcomes of such training initiatives.</p> <p>Conclusion</p> <p>National and international KT organizations, research institutions, and funding agencies are encouraged to consider replicating the training model employed here, as investment into KT personnel will foster the advancement of the field within and beyond local borders.</p> <p>'To the individual who devotes his/her life to science, nothing can give more happiness than when the results immediately find practical application.     Genetic background modulates behavioral impairments in R6/2 mice and suggests a role for dominant genetic modifiers in Huntingtonā€™s disease pathogenesis

    Variability and modification of the symptoms of Huntingtonā€™s disease (HD) are commonly observed in both patient populations and animal models of the disease. Utilizing a stable line of the R6/2 HD mouse model, the present study investigated the role of genetic background in the onset and severity of HD symptoms in a transgenic mouse. R6/2 congenic C57BL/6J and C57BL/6JĀ Ć—Ā DBA/2J F1 (B6D2F1) mice were evaluated for survival and a number of behavioral phenotypes. This study reports that the presence of the DBA/2J allele results in amelioration or exacerbation of several HD-like phenotypes characteristic of the R6/2 mouse model and indicates the presence of dominant genetic modifiers of HD symptoms. This study is the first step in identifying genes that confer natural genetic variation and modify the HD symptoms. This identification may lead to novel targets for treatment and help elucidate the molecular mechanisms of HD pathogenesis

    Cis-interactions between Notch and Delta generate mutually exclusive signalling states

    The Notchā€“Delta signalling pathway allows communication between neighbouring cells during development^1. It has a critical role in the formation of ā€˜fine-grainedā€™ patterns, generating distinct cell fates among groups of initially equivalent neighbouring cells and sharply delineating neighbouring regions in developing tissues. The Delta ligand has been shown to have two activities: it transactivates Notch in neighbouring cells and cis-inhibits Notch in its own cell. However, it remains unclear how Notch integrates these two activities and how the resulting system facilitates pattern formation. Here we report the development of a quantitative time-lapse microscopy platform for analysing Notchā€“Delta signalling dynamics in individual mammalian cells, with the aim of addressing these issues. By controlling both cis- and trans-Delta concentrations, and monitoring the dynamics of a Notch reporter, we measured the combined cisā€“trans inputā€“output relationship in the Notchā€“Delta system. The data revealed a striking difference between the responses of Notch to trans- and cis-Delta: whereas the response to trans-Delta is graded, the response to cis-Delta is sharp and occurs at a fixed threshold, independent of trans-Delta. We developed a simple mathematical model that shows how these behaviours emerge from the mutual inactivation of Notch and Delta proteins in the same cell. This interaction generates an ultrasensitive switch between mutually exclusive sending (high Delta/low Notch) and receiving (high Notch/low Delta) signalling states. At the multicellular level, this switch can amplify small differences between neighbouring cells even without transcription-mediated feedback. This Notchā€“Delta signalling switch facilitates the formation of sharp boundaries and lateral-inhibition patterns in models of development, and provides insight into previously unexplained mutant behaviours
