    The Effect of Furosemide on Arterial Blood Gases and Performance in Quarter Horses Performing a Fatigue Test on a Treadmill

    Four Quarter Horses (1 filly age 2, 1 mare age 5 and 2 geldings ages 3 and 4; average weight 539 kg) were used in a 2 x 2 crossover design. The effects of furosemide (Lasix(Rx)) on arterial blood packed ceii voiume (PCV), hemogiobin (Hb), pH, pO2, pCO2, HCO-3 and base excess (BE) were measured. Plasma lactate, heart rate, and fatigue time were determined as indicators of perlormance while the horses performed a fatigue test on a high-speed treadmill. The left carotid artery was surgically elevated subcutaneously to facilitate collection of arterial blood samples. Horses were conditioned for 13 weeks with increasing intensity then randomly assigned furosemide (F) or physiological saline (C) as treatments. Treatments were administered 4 hours prior to the fatigue test in accordance with racing regulations. Arterial blood samples were collected prior to treatment dose, prior to exercise, at the 2nd, 4th, and 6th minute during the fatigue test, at fatigue, and at the 5th, 15th, 30th, and 45th minute post-exercise. Arterial blood samples were analyzed for blood gases, Hb, PCV, and plasma lactate. Heart rate and fatigue time were recorded. No difference between treatments (P \u3e 0.05) was observed for blood gases except for pCO2 at rest, and HCO-3 and BE at the 2 minute collection period. No difference between treatments (P \u3e 0.05) was observed for Hb, PCV, lactate and heart rate except at 15 minutes post-exercise for Hb and PCV, and 45 minutes postexercise for Hb. Fatigue times were 11 min 56 sec Ā± 5 min 30 sec for F horses and 11 min 35 sec--Ā± 2 min 6 sec for C horses. No difference (P \u3e 0.05) was observed in fatigue time. Based on our data, the trend indicated that all parameters measured returned to pre-exercise levels more rapidly for furosemide treated horses. However, furosemide did not enhance performance

    Coculture of Staphylococcus aureus with Pseudomonas aeruginosa Drives S. aureus towards Fermentative Metabolism and Reduced Viability in a Cystic Fibrosis Model

    The airways of patients with cystic fibrosis are colonized with diverse bacterial communities that change dynamically during pediatric years and early adulthood. Staphylococcus aureus is the most prevalent pathogen during early childhood, but during late teens and early adulthood, a shift in microbial composition occurs leading to Pseudomonas aeruginosa community predominance in āˆ¼50% of adults. We developed a robust dual-bacterial in vitro coculture system of P. aeruginosa and S. aureus on monolayers of human bronchial epithelial cells homozygous for the Ī”F508 cystic fibrosis transmembrane conductance regulator (CFTR) mutation to better model the mechanisms of this interaction. We show that P. aeruginosa drives the S. aureus expression profile from that of aerobic respiration to fermentation. This shift is dependent on the production of both 2-heptyl-4-hydroxyquinoline N-oxide (HQNO) and siderophores by P. aeruginosa. Furthermore, S. aureus-produced lactate is a carbon source that P. aeruginosa preferentially consumes over medium-supplied glucose. We find that initially S. aureus and P. aeruginosa coexist; however, over extended coculture P. aeruginosa reduces S. aureus viability, also in an HQNO- and P. aeruginosa siderophore-dependent manner. Interestingly, S. aureus small-colony-variant (SCV) genetic mutant strains, which have defects in their electron transport chain, experience reduced killing by P. aeruginosa compared to their wild-type parent strains; thus, SCVs may provide a mechanism for persistence of S. aureus in the presence of P. aeruginosa. We propose that the mechanism of P. aeruginosa-mediated killing of S. aureus is multifactorial, requiring HQNO and P. aeruginosa siderophores as well as additional genetic, environmental, and nutritional factors

    Evidence of a Large-Scale Functional Organization of Mammalian Chromosomes

    Evidence from inbred strains of mice indicates that a quarter or more of the mammalian genome consists of chromosome regions containing clusters of functionally related genes. The intense selection pressures during inbreeding favor the coinheritance of optimal sets of alleles among these genetically linked, functionally related genes, resulting in extensive domains of linkage disequilibrium (LD) among a set of 60 genetically diverse inbred strains. Recombination that disrupts the preferred combinations of alleles reduces the ability of offspring to survive further inbreeding. LD is also seen between markers on separate chromosomes, forming networks with scale-free architecture. Combining LD data with pathway and genome annotation databases, we have been able to identify the biological functions underlying several domains and networks. Given the strong conservation of gene order among mammals, the domains and networks we find in mice probably characterize all mammals, including humans

    Seafloor massive sulfide (SMS) deposits are of increasing economic interest in order to satisfy the relentless growth in worldwide metal demand. The Transā€Atlantic Geotraverse (TAG) hydrothermal field at 26Ā°N on the Midā€Atlantic Ridge hosts several such deposits. This study presents new controlled source electromagnetic, bathymetric, and magnetic results from the TAG field. Potential SMS targets were selected based on their surface expressions in highā€resolution bathymetric data. Highā€resolution reducedā€toā€theā€pole magnetic data show negative anomalies beneath and surrounding the SMS deposits, revealing large areas of hydrothermal alteration. Controlled source electromagnetic data, sensitive to the electrical conductivity of SMS mineralization, further reveal a maximum thickness of up to 80 m and conductivities of up to 5 S/m. SMS samples have conductivities of up to a few thousand Siemens per meter, suggesting that remotely inferred conductivities represent an average of metal sulfide ores combined with silicified and altered host basalt that likely dominates at greater depths

    Prioritising prevention strategies for patients in antiretroviral treatment programmes in resource-limited settings

    Expanded access to antiretroviral therapy (ART) offers opportunities to strengthen HIV prevention in resource-limited settings. We invited 27 ART programmes from urban settings in Africa, Asia and South America to participate in a survey, with the aim to examine what preventive services had been integrated in ART programmes. Twenty-two programmes participated; eight (36%) from South Africa, two from Brazil, two from Zambia and one each from Argentina, India, Thailand, Botswana, Ivory Coast, Malawi, Morocco, Uganda and Zimbabwe and one occupational programme of a brewery company included five countries (Nigeria, Republic of Congo, Democratic Republic of Congo, Rwanda and Burundi). Twenty-one sites (96%) provided health education and social support, and 18 (82%) provided HIV testing and counselling. All sites encouraged disclosure of HIV infection to spouses and partners, but only 11 (50%) had a protocol for partner notification. Twenty-one sites (96%) supplied male condoms, seven (32%) female condoms and 20 (91%) provided prophylactic ART for the prevention of mother-to child transmission. Seven sites (33%) regularly screened for sexually transmitted infections (STI). Twelve sites (55%) were involved in activities aimed at women or adolescents, and 10 sites (46%) in activities aimed at serodiscordant couples. Stigma and discrimination, gender roles and funding constraints were perceived as the main obstacles to effective prevention in ART programmes. We conclude that preventive services in ART programmes in lower income countries focus on health education and the provision of social support and male condoms. Strategies that might be equally or more important in this setting, including partner notification, prompt diagnosis and treatment of STI and reduction of stigma in the community, have not been implemented widely
