A influência do secretoma de adipócitos e resposta imune no crescimento bacteriano
Obesity is characterized by excessive fat in adipose tissue and continues to be described as a public health problem. In addition, it is associated with the development and worsening of comorbidities such as type II diabetes, cardiovascular diseases, hypertension and metabolic syndrome. Another public health problem focuses on antimicrobial resistance (AMR). Over time, they have been gaining alarming proportions worldwide, due to the ability to acquire new mechanisms of resistance threatening the treatment of infections acquired in the community and in hospitals, resulting in a prolonged illness, disability, and death. The state of obesity entails an imbalance in the production of proinflammatory and anti-inflammatory factors that contributes to infections’ susceptibility. This susceptibility translates into respiratory and skin barrier impairment, obesity-related comorbidities, slower healing and inefficient antimicrobial therapy. Bacteria’s alarming ability to gain resistance has raised concerning obstacles in antimicrobial therapies in patients with chronic inflammation, such as obese individuals. This study aimed to understand the influence of an inflammatory obesity-mimicking environment in the growth of Staphylococcus aureus, Mycobacterium smegmatis, Klebsiella pneumoniae, Escherichia coli and Pseudomonas aeruginosa strains with different antibiotic resistance. For this, the cell lines Raw 264.7 (macrophages) and 3T3-L1 (adipocytes) were collected to serve as a conditioned medium for bacterial growth. All strains were exposed to a variety of conditioned media (DMEM, DMEM enriched with 10% and 50% adipocyte secretome (SA), macrophage secretome (SM) and macrophage secretome enriched with 10% and 50% SA). Results showed Methicillin-resistant Staphylococcus aureus (MRSA) and Klebsiella pneumoniae Carbapenem/ESBL are of greater risk factor in a SM environment. On the other hand, S.aureus, M. smegmatis, wild-type K.pneumoniae, K.pneumoniae ESBL, K.pneumoniae Carbapenem, E. coli and P. aeruginosa only seems to depend more on SA environment. Additionally, bacterial strains with AmpC resistance showed high risk to bacterial growth in environment enriched with SA. Overall, this study demonstrates that the kind of resistance and the environment in which each bacteria strain is present can contribute to the state of chronic inflammation.A obesidade é caraterizada pelo excesso de gordura no tecido adiposo, continuando a ser descrita como um problema à saúde pública. Além disso, está associada ao desenvolvimento e agravamento de comorbidades como diabetes tipo II, doenças cardiovasculares, hipertensão e síndrome metabólica. Outro problema à saúde pública incide nas resistências antimicrobianas (AMR). Ao longo do tempo, têm vindo a ganhar proporções alarmantes à escala Mundial, devido à capacidade de adquirir novos mecanismos de resistência, ameaçando o tratamento de infeções adquiridas na comunidade e nos hospitais, do qual resulta numa doença prolongada, incapacidade e morte. O estado de obesidade acarreta um desequilíbrio na produção de fatores pró-inflamatórios e anti-inflamatórios que contribuem para a suscetibilidade das infeções. Essa suscetibilidade traduz-se em comprometimento respiratório e da barreira cutânea, comorbidades relacionadas à obesidade, cicatrização mais lenta e terapia antimicrobiana ineficiente. A capacidade alarmante das bactérias em adquirir resistência tem levantado obstáculos à terapia antimicrobiana em pacientes com inflamação crônica, como indivíduos obesos. Este estudo teve como objetivo compreender a influência de um ambiente inflamatório que mimetiza a obesidade, no crescimento de Staphylococcus aureus, Mycobacterium smegmatis, Klebsiella pneumoniae, Escherichia coli e Pseudomonas aeruginosa com diferentes resistências a antibióticos. Para tal, as linhagens celulares Raw 264.7 (macrófagos) e 3T3-L1 (adipócitos) foram escolhidas para servir como meio condicionado para o crescimento bacteriano. As estirpes bacterianas foram expostas a uma variedade de meios condicionados (DMEM, DMEM enriquecido com 10% e 50% de secretoma de adipócitos (SA), secretoma de macrófagos (SM) e secretoma de macrófagos enriquecido com 10% e 50% de SA). Os resultados mostraram que S.aureus com resistência a meticilina (MRSA) e K. pneumoniae Carbapenem/ESBL contribuem para um maior fator de risco num ambiente de SM. Por outro lado, S.aureus, M. smegmatis, K. pneumoniae “estirpe-selvagem”, K. pneumoniae ESBL, K. pneumoniae Carbapenem, E. coli e P. aeruginosa parecem depender mais de um ambiente enriquecido em SA. Ainda, as estirpes bacterianas com resistência a AmpC apresentam crescimento bacteriano elevado num ambiente enriquecido em SA. No geral, os resultados mostraram que a estirpe e painel de resistência, podem contribuir para o estado de inflamação crónico de forma diferenciada.Mestrado em Microbiologi
Oocyte secreted factors receptors in a granulosa cell line. An approach to oocyte quality assessment
Planting tradition and harvesting innovation : a case study of how a Portuguese family winery achieved international reputation
Although small family-owned businesses populate the wine industry, they encounter greater
barriers when trying to promote their wines, especially at an international level. The purpose of
this thesis is to investigate how small wineries can stand out from competitors. The level of
rivalry in the market is lessened when producers can differentiate their products. While an
innovation strategy is crucial for wine producers to compete, research on how they approach
such practices is scarce. This work seeks to complement the existing literature by illustrating
the analysis with a case study of Ervideira, a Portuguese family-run winery. The company’s
international success relies on the production of high-end wines and innovation both in the
production methods applied and the services provided. The company has invested in promoting
the wines through direct contact with clients via the wine club, its stores, and its website, as
well as attempting to attract potential customers through wine tourism and trade fair
participation. I conclude that small wineries can differentiate themselves in the market by
managing innovation and quality while nurturing tradition and adjusting to global consumer
trends.Embora os pequenos produtores familiares sejam comuns no setor vinícola, enfrentam maiores
obstáculos ao tentar promover os seus vinhos, especialmente ao nível internacional. O objetivo
desta tese é investigar como é que eles se podem demarcar dos seus concorrentes. O nível de
competitividade no mercado é minorado quando os produtores conseguem diferenciar os seus
produtos. Embora uma estratégia assente na inovação seja crucial para os produtores de vinho
competirem no mercado, a pesquisa existente explora pouco sobre como eles abordam estas
práticas. Este trabalho procura complementar a literatura existente ao ilustrar a análise com um
estudo de caso da Ervideira, uma empresa vitivinícola portuguesa. O sucesso internacional da
empresa passa pela produção de vinhos de alta qualidade e pela inovação tanto nos métodos de
produção aplicados como nos serviços prestados. A empresa tem apostado na promoção dos
vinhos através do contacto direto com os clientes por meio do clube de vinhos, das suas lojas e
do seu website, bem como na procura de potenciais clientes através do enoturismo e da
participação em feiras. Concluo que os pequenos produtores de vinhos podem diferenciar-se no
mercado aliando inovação à qualidade, enquanto cultivam a tradição e se adaptam às tendências
de consumo globais

The job market works under asymmetric information, making it hard for firms to know the real capabilities of the workers they hire. Workers can help overcome this asymmetry by signaling their productivity with investments in education. However, it’s fair to assume that workers sometimes miscalculate their own abilities, which disturbs the functioning of the signal. This study intends to analyze the effects of overconfidence amongst workers on the various parameters of the game by conducting an experiment. The results from the experiment suggest that the profits of both workers and firms will be greatly impaired as a consequence
Melanocortin signalling in fat cells
[resumo]Melanocortin peptides and their receptors have long been known to affect the central control of food intake and body weight, thus playing a critical role in the development of obesity [1]. The melanocortin 5 receptor (MC5R) has been directly linked with lipid metabolism in peripheral tissues: MC5R deletion in mouse generates defects in sebaceous gland secretion [1] and MC5R stimulation increases fatty acid oxidation in muscle cells [2]. In this study, we tackled the role of MC5R in lipolysis and adipocyte function using the well established model of differentiated mouse 3T3-L1 adipocytes. Cell-signalling based experiments were performed after adipocyte treatment with alpha-melanocyte-stimulating hormone (alpha-MSH) and compared with the MC5R specific activation previously described in HEK293 [3] and HeLa cells stably transfected with MC5R-GFP. Adipocyte response to alpha-MSH resulted in a loss of cytoplasmatic lipid content and release of glycerol into the cell medium by an ERK1/2 dependent mechanism. Moreover, downstream of ERK1/2, alpha-MSH induces p90RSK and CREB phosphorylation and c-Fos expression. A Casa do Futuro, hoje. [Re]visão contemporânea
A tensão entre dois tempos distintos como o passado e o futuro é
algo complicado de entender e percecionar. Esse tempo intermédio
que chamamos de presente é efémero e muito acelerado. Quando
damos por nós a pensar no que se passa no momento, já este é
passado e o tempo em que vivemos faz já parte do futuro. Com isto,
em que momento este futuro e passado podem viver em sintonia,
numa simbiose de tempos que faz esta tensão entre ambos ser
equilibrada?
É necessário entender que influência tem um no outro. Claramente,
o passado tem uma grande influência no futuro e, no caso da
arquitetura e na sua evolução ao longo dos anos, o passado está
sempre presente, podendo dizer-se que este está na base do futuro.
"A Tradição é um desafio à inovação".1
No caso concreto da Casa, como dispositivo do dia a dia do ser
humano, a sua mudança é inevitável, acompanhando os novos
hábitos que criamos. É, deste modo, necessário entender o que vai
ser este futuro, próximo ou não, e que alterações tem de sofrer a
casa, na sua disposição e composição entre espaços interiores e
exteriores, e de que forma, mesmo tendo por base uma estrutura
antiga, podemos chamá-la Casa do Futuro.
Assim, e pegando no caso concreto de uma casa de família,
pretende-se abordá-la e conseguir chegar a esse limite entre os
dois tempos, para que ambos vivam em harmonia e se adaptem às
necessidades pretendidas, passando, primeiramente, por uma fase
de caraterização da casa e, posteriormente, a um projeto concreto
de intervenção.
Mas como nos podemos agarrar a essa estrutura antiga da Casa e
transpô-la para o futuro? O que é uma Casa do Futuro?
Este pensamento traz outras questões essenciais que são: Quem
habita a casa? Qual o modo de vida de quem a habita? E que casa
é essa na qual queremos intervir?
Perceber um modo de intervenção invoca a importância que o tempo
tem e o que está entre o que permanece e o que se transforma,
uma nova dimensão entre as Coisas e as Pessoas. Cria-se, assim,
uma tensão, um jogo de contrários, de opostos e roturas, em que é
necessário encontrar no que está, o que precisa de ser feito.
É preciso 'tratar com respeito, mas sem receio'.
Para além disto, a influência dos arquitetos Alison e Peter Smithson
é preponderante para o desenvolvimento do projeto, já que refletem
sobre este assunto no Movimento Moderno, através do seu projeto
Casa do Futuro.The tension between two different times such as the past and the
future is rather complicated to realize and to perceive. That split
time, which we call the present is ephemeral and very fast. Moreover,
before we know it, the present is a fleeting moment, it is in the past
already and the time we live in is within the future. Herewith, how
can this future and this past be tuned, in a symbiosis of times that
balances this tension?
It is necessary to grasp how they influence each other. Clearly, the
past has a great influence in the future; regarding architecture and
how it has evolved throughout the years, the past is always present,
and it is true to say that this is a basis for the future.
"Tradition is a challenge to innovation".2
In the specific case of the House, as a day-by-day mechanism of the
human being, its change is inevitable, as it follows the new ways we
create. It is, therefore, necessary to understand what this future will
be (near future or not) and what changes the house must endure, as
far as indoor and outdoor spaces are made up and displayed, and
in what way, even after being based in an old structure, it can be
referred to as House of the Future.
Accordingly, and taking the specific case of a family house, the
objective is to approach it and to be able to reach that split time
between those two times, so that both can adapt to the intended
needs, the first of the steps being the profiling of the house and,
subsequently, the developing of a concrete intervention project.
However, how can one hold on to that House's old structure and
transpose it to the future? What is a House of the Future? This notion
raises other important issues, namely: Who inhabits the house? What
is the lifestyle of its inhabitants? And what is that house on which we
want to take action?
Understanding a means of intervention refers to the importance
of time and what remains between what continues and what is
transformed, a new dimension between Things and People. It makes
for a tension, a game of contraries, of opposites and ruptures, where
it is necessary to find out what needs to be done within that that
exists already. It is necessary to 'treat with respect, but with no fear'.
Furthermore, the influence by architects Alison and Peter Smithson
takes a leading role as regards the project's development since they
address this matter in the Modern Movement, in their project House
of the Future
