    Amphibians and Reptiles of Luzon Island, Philippines: the Herpetofauna of Pantabangan-Carranglan Watershed, Nueva Ecija Province, Caraballo Mountain Range

    We present detailed species accounts for fifty-nine (59) species of amphibians and reptiles (17 frogs, 14 skinks, 3 agamids, 6 gekkonid lizards, 2 varanids, and 17 snakes) from Pantabangan-Carranglan Watershed, which lies within the Caraballo Mountain Range, whose biota is poorly-known. This was also the first extensive survey of herpetofauna within the watershed. Together with data from previous literature reviews, our records bring the total number of species of amphibians and reptiles for the Caraballo Mountain Range to 66. Forty-two (42) species from the area were Philippine endemics, with 25 species recorded only from Luzon faunal region. Seven species of herpetofauna are associated with unresolved taxonomic issues (new species and species complexes needing taxonomic partitioning, e.g. splitting of species groups). Two species recorded from the area were rarely represented in museum collections. Major distributional and elevational range extensions were recorded for several species. Comparison with Luzon’s other mountain ranges showed that the Caraballo Mountain Range is similar, in terms of species composition, to the northern Sierra Madre and Cordillera Mountain Ranges. The result of this survey showed the Caraballo Mountain Range and its mountains as a possible new center of herpetofaunal diversity and endemicity within Luzon. The importance of the Caraballo Mountain Range as an important biogeographic link merits further study

    Global monitoring of soil multifunctionality in drylands using satellite imagery and field data

    Models derived from satellite image data are needed to monitor the status of terrestrial ecosystems across large spatial scales. However, a remote sensing-based approach to quantify soil multifunctionality at the global scale is missing despite significant research efforts on this topic. A major constraint for doing so is the availability of suitable global-scale field data to calibrate remote sensing indicators (RSI) and, to a lesser extent, the sensitivity of spectral data of available satellite sensors to soil background and atmospheric conditions. Here, we aimed to develop a soil multifunctionality model to monitor global drylands coupling ground data on 14 soil functions of 222 dryland areas from six continents to 18 RSI derived from a time series (2006–2013) Landsat dataset. Among the RSI evaluated, the chlorophyll absorption ratio index was the best predictor of soil multifunctionality in single-variable-based models (r = 0.66, P < 0.01, NMRSE = 0.17). However, a multi-variable RSI model combining the chlorophyll absorption ratio index, the global environment monitoring index and the canopy-air temperature difference improved the accuracy of quantifying soil multifunctionality (r = 0.73, P < 0.01, NMRSE = 0.15). Furthermore, the correlation between RSI and soil variables shows a wide range of accuracy with upper and lower values obtained for AMI (r = 0.889, NMRSE = 0.05) and BGL (r = 0.685, NMRSE = 0.18) respectively. Our results provide new insights on assessing soil multifunctionality using RSI that may help to monitor temporal changes in the functioning of global drylands effectively.Field data were obtained with the support of the European Research Council (ERC) grant agreement 242658 (BIOCOM).     Regulation of DMD pathology by an ankyrin-encoded miRNA

    <p>Abstract</p> <p>Background</p> <p>Duchenne muscular dystrophy (DMD) is an X-linked myopathy resulting from the production of a nonfunctional dystrophin protein. MicroRNA (miRNA) are small 21- to 24-nucleotide RNA that can regulate both individual genes and entire cell signaling pathways. Previously, we identified several mRNA, both muscle-enriched and inflammation-induced, that are dysregulated in the skeletal muscles of DMD patients. One particularly muscle-enriched miRNA, miR-486, is significantly downregulated in dystrophin-deficient mouse and human skeletal muscles. miR-486 is embedded within the <it>ANKYRIN1(ANK1) </it>gene locus, which is transcribed as either a long (erythroid-enriched) or a short (heart muscle- and skeletal muscle-enriched) isoform, depending on the cell and tissue types.</p> <p>Results</p> <p>Inhibition of miR-486 in normal muscle myoblasts results in inhibited migration and failure to repair a wound in primary myoblast cell cultures. Conversely, overexpression of miR-486 in primary myoblast cell cultures results in increased proliferation with no changes in cellular apoptosis. Using bioinformatics and miRNA reporter assays, we have identified platelet-derived growth factor receptor β, along with several other downstream targets of the phosphatase and tensin homolog deleted on chromosome 10/AKT (PTEN/AKT) pathway, as being modulated by miR-486. The generation of muscle-specific transgenic mice that overexpress miR-486 revealed that miR-486 alters the cell cycle kinetics of regenerated myofibers <it>in vivo</it>, as these mice had impaired muscle regeneration.</p> <p>Conclusions</p> <p>These studies demonstrate a link for miR-486 as a regulator of the PTEN/AKT pathway in dystrophin-deficient muscle and an important factor in the regulation of DMD muscle pathology.</p

    SOX2 Expression and Transcriptional Activity Identifies a Subpopulation of Cancer Stem Cells in Sarcoma with Prognostic Implications

    Stemness in sarcomas is coordinated by the expression of pluripotency factors, like SOX2, in cancer stem cells (CSC). The role of SOX2 in tumor initiation and progression has been well characterized in osteosarcoma. However, the pro-tumorigenic features of SOX2 have been scarcely investigated in other sarcoma subtypes. Here, we show that SOX2 depletion dramatically reduced the ability of undifferentiated pleomorphic sarcoma (UPS) cells to form tumorspheres and to initiate tumor growth. Conversely, SOX2 overexpression resulted in increased in vivo tumorigenicity. Moreover, using a reporter system (SORE6) which allows to monitor viable cells expressing SOX2 and/or OCT4, we found that SORE6+ cells were significantly more tumorigenic than the SORE6- subpopulation. In agreement with this findings, SOX2 expression in sarcoma patients was associated to tumor grade, differentiation, invasive potential and lower patient survival. Finally, we studied the effect of a panel of anti-tumor drugs on the SORE6+ cells of the UPS model and patient-derived chondrosarcoma lines. We found that the mithramycin analogue EC-8042 was the most efficient in reducing SORE6+ cells in vitro and in vivo. Overall, this study demonstrates that SOX2 is a pro-tumorigenic factor with prognostic potential in sarcoma.     Amphibian and reptile diversity along a ridge-to-reef elevational gradient on a small isolated oceanic island of the central Philippines

    Despite multiple recent field studies, herpetological species diversity of the Romblon Island Group in the central Philippines&amp;mdash;particularly Sibuyan Island&amp;mdash;has remained underestimated. Recently, we investigated the diversity of the herpetofauna of Mount Guiting-Guiting Natural Park, based on an elevational transect (10&amp;ndash;1557 m a.s.l.). Our surveys resulted in a total of 47 species of amphibians and reptiles, including 14 new island records and one atypical occurrence of a snake species recorded for the first time from a high elevation (939 m a.s.l). These new records constitute a notable increase (21%) in Sibuyan&amp;rsquo;s herpetological species diversity as compared to surveys from a decade ago. We also provide updates of the taxonomy and identification of species endemic to this island (e.g., members of the genera Platymantis G&amp;uuml;nther, 1858, Brachymeles Dum&amp;eacute;ril &amp;amp; Gibron, 1839, and Pseudogekko Taylor, 1922), and discuss the importance of continued surveys and field-derived data to inform conservation status assessments of Sibuyan&amp;rsquo;s unique assemblage of amphibians and reptiles

    Introducció i objectius La tendinopatia és una lesió freqüent durant la pràctica esportiva que es manifesta amb una alteració estructural del tendó. L’objectiu d’aquest estudi fou avaluar l’eficàcia i la seguretat d’un complement alimentari a base de mucopolisacàrids, col·lagen tipus i i vitamina C (Tendoactive®) sobre l’evolució clínica i estructural de les tendinopaties del tendó d’Aquil·les, del rotular i de l’epicòndil lateral del colze. Material i mètodes Es realitzà un estudi multicèntric prospectiu, de tipus exploratori en fase iv, obert i no comparatiu. S’inclogueren un total de 98 pacients amb tendinopaties (32 d’Aquil·les, 32 de rotular i 34 de l’epicòndil lateral) que reberen una dosi diària de 435 mg de mucopolisacàrids, 75 mg de col·lagen tipus i i 60 mg de vitamina C (equivalent a 3 càpsules al dia de Tendoactive®) durant 90 dies consecutius. Mensualment s’avaluà el dolor en repòs i en activitat mitjançant una escala visual analògica (EVA), la funció articular mitjançant els qüestionaris VISA-A, VISA-P i PRTEE, i el tendó afectat es caracteritzà ecogràficament. Resultats En els 3 tipus de tendinopatia es registrà una reducció significativa del dolor, tant en repòs com en activitat, des de la primera visita de control (dia 30) fins al final de l’estudi (dia 90). Així mateix, el dia 90 es detectà una millora del 38% en VISA-A, del 46% en VISA-P i del 77% en PRTEE (p < 0,001). Simultàniament es registrà una reducció del 12% en el gruix del tendó d’Aquil·les, del 10% en el rotular i del 20% en el tendó de l’epicòndil lateral (p < 0,05). Conclusions Els resultats de l’estudi indiquen que l’administració de Tendoactive® és segura i eficaç per millorar els símptomes clínics i l’evolució estructural de les tendinopaties del tendó d’Aquil·les, del tendó rotular i del tendó de l’epicòndil lateral

    The efficacy and safety of oral mucopolysaccharide, type I collagen and vitamin C treatment in tendinopathy patients

    Introduction and objectives Tendinopathy, which is accompanied by structural changes to the tendon, is a common sporting injury. The aim of this study was to evaluate the efficacy and safety of a nutritional supplement containing mucopolysaccharides, type I collagen and vitamin C (TendoactiveTM) on the clinical and structural evolution of tendinopathies of the Achilles tendon, patellar tendon and lateral epicondyle tendon in the elbow. Materials and methods A multicenter, open-label, non-comparative, prospective, exploratory phase iv study was performed. A total of 98 tendinopathy patients (32 Achilles, 32 patellar and 34 lateral epicondylar), who received a daily dose of 435 mg mucopolysaccharides, 75 mg type i collagen and 60 mg vitamin C (equivalent to three capsules of TendoactiveTM per day) for 90 consecutive days, were included. Every month, pain at rest and when active was assessed using a visual analogue scale (VAS), joint function was assessed using the VISA-A, VISA-P and PRTEE questionnaires, and the tendon affected was characterized by ultrasound. Results A significant reduction in pain both at rest and when active was observed between the first control visit (day 30) and the end of the study (day 90) for all three types of tendinopathy. Thus, a 38% improvement in VISA-A, 46% in VISA-P and 77% in PRTEE was observed on day 90 (P < .001). Similarly, a 12% decrease in the thickness of the Achilles tendon, a 10% decrease in the patellar tendon and a 20% decrease in the lateral epicondyle tendon was observed (P < .05). Conclusions The results of this study show that the administration of TendoactiveTM is safe and effective for improving the clinical symptoms and structural evolution of tendinopathies of the Achilles, patella and lateral epicondyle tendons

    Introducción y objetivos La tendinopatía es una lesión frecuente durante la práctica deportiva que cursa con una alteración estructural del tendón. El objetivo de este estudio fue evaluar la eficacia y la seguridad de un complemento alimentario a base de mucopolisacáridos, colágeno tipo i y vitamina C (Tendoactive®) sobre la evolución clínica y estructural de las tendinopatías del tendón de Aquiles, rotuliano y del epicóndilo lateral del codo. Material y métodos Se realizó un estudio multicéntrico prospectivo, de tipo exploratorio en fase iv , abierto y no comparativo. Se incluyeron un total de 98 pacientes con tendinopatías (32 de Aquiles, 32 de rotuliano y 34 del epicóndilo lateral) que recibieron una dosis diaria de 435 mg de mucopolisacáridos, 75 mg de colágeno tipo i y 60 mg de vitamina C (equivalente a 3 cápsulas al día de Tendoactive®) durante 90 días consecutivos. Mensualmente se evaluó el dolor en reposo y en actividad mediante una escala visual analógica (EVA), la función articular mediante los cuestionarios VISA-A, VISA-P y PRTEE, y se caracterizó ecográficamente el tendón afectado. Resultados En los 3 tipos de tendinopatía se registró una reducción significativa del dolor tanto en reposo como en actividad desde la primera visita de control (día 30) hasta el final del estudio (día 90). Asimismo el día 90 se detectó una mejora del 38% en VISA-A, del 46% en VISA-P y del 77% en PRTEE (p < 0,001). Simultáneamente se registró una reducción del 12% en el grosor del tendón de Aquiles, del 10% en el rotuliano y del 20% en el tendón del epicóndilo lateral (p < 0,05). Conclusiones Los resultados del estudio indican que la administración de Tendoactive® es segura y eficaz para mejorar los síntomas clínicos y la evolución estructural de las tendinopatías del tendón de Aquiles, tendón rotuliano y tendón del epicóndilo lateral
