    Leaf-to-branch scaling of C-gain in field-grown almond trees under different soil moisture regimes

    Branch/tree-level measurements of carbon (C)-acquisition provide an integration of the physical and biological processes driving the C gain of all individual leaves. Most research dealing with the interacting effects of high-irradiance environments and soil-induced water stress on the C-gain of fruit tree species has focused on leaf-level measurements. The C-gain of both sun-exposed leaves and branches of adult almond trees growing in a semi-arid climate was investigated to determine the respective costs of structural and biochemical/physiological protective mechanisms involved in the behaviour at branch scale. Measurements were performed on well-watered (fully irrigated, FI) and drought-stressed (deficit irrigated, DI) trees. Leaf-to-branch scaling for net CO2 assimilation was quantified by a global scaling factor (fg), defined as the product of two specific scaling factors: (i) a structural scaling factor (fs), determined under well-watered conditions, mainly involving leaf mutual shading; and (ii) a water stress scaling factor (fws,b) involving the limitations in C-acquisition due to soil water deficit. The contribution of structural mechanisms to limiting branch net C-gain was high (mean fs ∼0.33) and close to the projected-to-total leaf area ratio of almond branches (ε = 0.31), while the contribution of water stress mechanisms was moderate (mean fws,b ∼0.85), thus supplying an fg ranging between 0.25 and 0.33 with slightly higher values for FI trees with respect to DI trees. These results suggest that the almond tree (a drought-tolerant species) has acquired mechanisms of defensive strategy (survival) mainly based on a specific branch architectural design. This strategy allows the potential for C-gain to be preserved at branch scale under a large range of soil water deficits. In other words, almond tree branches exhibit an architecture that is suboptimal for C-acquisition under well-watered conditions, but remarkably efficient to counteract the impact of DI and drought events

    Synthesis and characterization of titanium-vanadium ternary nitride (TixV1-xN).

    En el presente trabajo se aborda la síntesis del nitruro ternario de titanio y vanadio por reducción carbotérmica de una mezcla de óxidos o bien por nitruración directa de la mezcla de metales, en todo el rango de composiciones empleando el método de análisis térmico a velocidad controlada (ATVC). La caracterización de los productos obtenidos se ha llevado a cabo mediante difracción de rayos-X y microscopia electrónica. El empleo de la técnica EELS ha mostrado su capacidad para la cuantificación de este tipo de materiales, así como la utilización de la técnica espectroscópica XAS nos ha servido para corroborar los datos obtenidos a partir de los diagramas de difracción de rayos-X sobre distancias de enlace, número de coordinación y grado de desorden, esto último no detectado por las otras técnicas usadas en la caracterización de los productos. Los valores de microdureza registrados para las muestras sintetizadas se encuentran dentro del rango de valores reportados para este tipo de materiales.Titanium-Vanadium nitride (TiVN) has been prepared from carbothermal reduction of corresponding oxides and also by direct nitridation of a mix of two metals employing the ATVC method. The characterization of the final product by X-ray diffraction, scanning electron microscopy, electron energy loss (EELS), and X-ray absorption spectroscopy (XAS) is presented.The synthesis of the ternary nitride has been possible in all range of composition and the final product is obtained with nanometric particle size and a high microhardness after sintering

    El arqueado de tallos en cultivo de rosas: su papel en los intercambios gaseosos a escala del cultivo

    Se ha estudiado la influencia del arqueado de tallos (técnica del “pulmón”) en la tasa de transpiración (E) y en la capacidad fotosintética (Alm) de un cultivo de rosas (Rosa hybrida cv. Dallas) sin suelo bajo invernadero. E se evaluó por medio de una balanza electrónica en plantas desarrolladas y en plantas con tallos arqueados. Alm se midió, en hojas adultas, a saturación de luz en diferentes estratos foliares del tallo floral y del pulmón. Los resultados muestran que el pulmón juega un papel preponderante en la tasa de transpiración del cultivo. La contribución del tallo floral y del pulmón a la transpiración del cultivo era muy similar (*50%). Se ha observado una disminución de Alm desde las hojas de la cima del tallo floral (16.4 mmolco2m-2s-1) hasta las hojas del pulmón, llegando a valores muy bajos en los tallos arqueados más viejos (6 mmolco2m-2s-1). El papel que juega la superficie foliar del pulmón en la fotosíntesis del cultivo (contribución 30 %) es de menor alcance que el que ejerce en la tasa transpiración, lo que indica que posee una menor eficiencia de uso del agua que el tallo floral. Por otro lado, los resultados sugieren que los tallos del pulmón deben renovarse con frecuencia, debido a una disminución drástica de Alm con la edad y con el sombreo mutuo entre las hojas

    Oxidative stress is tightly regulated by cytochrome c phosphorylation and respirasome factors in mitochondria

    Respiratory cytochrome c has been found to be phosphorylated at tyrosine 97 in the postischemic brain upon neuroprotective insulin treatment, but how such posttranslational modification affects mitochondrial metabolism is unclear. Here, we report the structural features and functional behavior of a phosphomimetic cytochrome c mutant, which was generated by site-specific incorporation at position 97 of p-carboxymethyl-l-phenylalanine using the evolved tRNA synthetase method. We found that the point mutation does not alter the overall folding and heme environment of cytochrome c, but significantly affects the entire oxidative phosphorylation process. In fact, the electron donation rate of the mutant heme protein to cytochrome c oxidase, or complex IV, within respiratory supercomplexes was higher than that of the wild-type species, in agreement with the observed decrease in reactive oxygen species production. Direct contact of cytochrome c with the respiratory supercomplex factor HIGD1A (hypoxia-inducible domain family member 1A) is reported here, with the mutant heme protein exhibiting a lower affinity than the wild-type species. Interestingly, phosphomimetic cytochrome c also exhibited a lower caspase-3 activation activity. Altogether, these findings yield a better understanding of the molecular basis for mitochondrial metabolism in acute diseases, such as brain ischemia, and thus could allow the use of phosphomimetic cytochrome c as a neuroprotector with therapeutic applications.España, Junta de Andalucía BIO-198España MINECO BFU2015-71017/BM

    ObjetivoConocer la prevalencia de úlceras por presión (UPP) en pacientes incapacitados de una Zona Básica de Salud (ZBS) y caracterizar los factores de riesgo asociados a su aparición.DiseñoEstudio transversal observacional.EmplazamientoZBS Rincón de la Victoria (Málaga).ParticipantesPacientes incluidos en el Programa de Incapacitados (n = 178), de la ZBS, residentes en su domicilio o en una residencia geriátrica.MedicionesCuestionario específico con variables sociodemográficas y de salud. Para la valoración de factores de riesgo asociados con la formación de UPP, se utilizó la Escala de Norton Modificada (ENM). La variable resultado de interés fue la presencia o ausencia de UPP, localización y grado. El denominador para el cálculo de la prevalencia ha sido el total de pacientes valorados.ResultadosLa prevalencia fue del 12,9%. La característica sociodemográfica más significativa asociada con la presencia de úlceras en estos pacientes fue el nivel de instrucción del cuidador. The denominator used for the calculation of the prevalence was the total of evaluated patients.ResultsPrevalence of pressure ulcers in our Basic Health Area was 12.9%. The most important sociodemographic characteristic associated with the presence of pressure ulcers in these patients was the educational attainment of the caregiver. Health varaibles of the patient were also associated with the risk of developing ulcers.ConclusionsResults indicate the need of diagnostic protocols with standardised instruments and prevention plans to reduce pressure ulcers in the community. Longitudinal studies are needed to evaluate interventions in this area

    Integrating magnetic capabilities to intracellular chips for cell trapping

    Current microtechnologies have shown plenty of room inside a living cell for silicon chips. Microchips as barcodes, biochemical sensors, mechanical sensors and even electrical devices have been internalized into living cells without interfering their cell viability. However, these technologies lack from the ability to trap and preconcentrate cells in a specific region, which are prerequisites for cell separation, purification and posterior studies with enhanced sensitivity. Magnetic manipulation of microobjects, which allows a non-contacting method, has become an attractive and promising technique at small scales. Here, we show intracellular Ni-based chips with magnetic capabilities to allow cell enrichment. As a proof of concept of the potential to integrate multiple functionalities on a single device of this technique, we combine coding and magnetic manipulation capabilities in a single device. Devices were found to be internalized by HeLa cells without interfering in their viability. We demonstrated the tagging of a subpopulation of cells and their subsequent magnetic trapping with internalized barcodes subjected to a force up to 2.57 pN (for magnet-cells distance of 4.9 mm). The work opens the venue for future intracellular chips that integrate multiple functionalities with the magnetic manipulation of cells

    Inverse relation between FASN expression in human adipose tissue and the insulin resistance level

    <p>Abstract</p> <p>Background</p> <p>Adipose tissue is a key regulator of energy balance playing an active role in lipid storage and may be a dynamic buffer to control fatty acid flux. Just like PPARγ, fatty acid synthesis enzymes such as FASN have been implicated in almost all aspects of human metabolic alterations such as obesity, insulin resistance or dyslipemia. The aim of this work is to investigate how FASN and PPARγ expression in human adipose tissue is related to carbohydrate metabolism dysfunction and obesity.</p> <p>Methods</p> <p>The study included eighty-seven patients which were classified according to their BMI and to their glycaemia levels in order to study FASN and PPARγ gene expression levels, anthropometric and biochemical variables.</p> <p>Results</p> <p>The main result of this work is the close relation between FASN expression level and the factors that lead to hyperglycemic state (increased values of glucose levels, HOMA-IR, HbA1c, BMI and triglycerides). The correlation of the enzyme with these parameters is inversely proportional. On the other hand, PPARγ is not related to carbohydrate metabolism.</p> <p>Conclusions</p> <p>We can demonstrate that FASN expression is a good candidate to study the pathophysiology of type II diabetes and obesity in humans.</p

    Similar incidence of coronavirus disease 2019 (COVID-19) in patients with rheumatic diseases with and without hydroxychloroquine therapy

    Background Hydroxychloroquine is not efficacious as post-exposure prophylaxis against coronavirus disease 2019 (COVID-19). It is not known whether as pre-exposure prophylaxis it may prevent COVID-19. Objective To compare the incidence of COVID-19 in Spanish patients with autoimmune rheumatic diseases treated with and without hydroxychloroquine. Patients and methods Retrospective electronic record review, from February 27th to June 21st, 2020, of patients with autoimmune inflammatory diseases followed at two academic tertiary care hospitals in Seville, Spain. The cumulative incidence of confirmed COVID-19, by PCR or serology, was compared between patients with and without hydroxychloroquine as part of their treatment of autoimmune inflammatory diseases. Results Among 722 included patients, 290 (40%) were receiving hydroxychloroquine. During the seventeen-week study period, 10 (3.4% [95% CI: 1.7%-6.7%] cases of COVID-19 were registered among patients with hydroxychloroquine and 13 (3.0% [1.6%-5.1%]) (p = 0.565) in those without hydroxychloroquine. COVID-19 was diagnosed by PCR in four (1.4%, 95% CI 0.38%-3.5%) subject with hydroxychloroquine and six (1.4%, 95% CI 0.5%-3.0%) without hydroxychloroquine (p = 0.697). Three patients on hydroxychloroquine and four patients without hydroxychloroquine were admitted to the hospital, none of them required to be transferred to the intensive care unit and no patient died during the episode. Conclusions The incidence and severity of COVID-19 among patients with autoimmune rheumatic diseases with and without hydroxychloroquine was not significantly different.Instituto de Salud Carlos III I3SNSMinisterio de Ciencia, Innovación y Universidades CP18/0014

    PP2A is activated by cytochrome c upon formation of a diffuse encounter complex with SET/TAF-Iß

    Intrinsic protein flexibility is of overwhelming relevance for intermolecular recognition and adaptability of highly dynamic ensemble of complexes, and the phenomenon is essential for the understanding of numerous biological processes. These conformational ensembles—encounter complexes—lack a unique organization, which prevents the determination of well-defined high resolution structures. This is the case for complexes involving the oncoprotein SET/template-activating factor-Iß (SET/TAF-Iß), a histone chaperone whose functions and interactions are significantly affected by its intrinsic structural plasticity. Besides its role in chromatin remodeling, SET/TAF-Iß is an inhibitor of protein phosphatase 2A (PP2A), which is a key phosphatase counteracting transcription and signaling events controlling the activity of DNA damage response (DDR) mediators. During DDR, SET/TAF-Iß is sequestered by cytochrome c (Cc) upon migration of the hemeprotein from mitochondria to the cell nucleus. Here, we report that the nuclear SET/TAF-Iß:Cc polyconformational ensemble is able to activate PP2A. In particular, the N-end folded, globular region of SET/TAF-Iß (a.k.a. SET/TAF-Iß ¿C)—which exhibits an unexpected, intrinsically highly dynamic behavior—is sufficient to be recognized by Cc in a diffuse encounter manner. Cc-mediated blocking of PP2A inhibition is deciphered using an integrated structural and computational approach, combining small-angle X-ray scattering, electron paramagnetic resonance, nuclear magnetic resonance, calorimetry and molecular dynamics simulations

    Cytochrome c speeds up caspase cascade activation by blocking 14-3-3¿-dependent Apaf-1 inhibition article

    Apoptosis is a highly regulated form of programmed cell death, essential to the development and homeostasis of multicellular organisms. Cytochrome c is a central figure in the activation of the apoptotic intrinsic pathway, thereby activating the caspase cascade through its interaction with Apaf-1. Our recent studies have revealed 14-3-3€ (a direct inhibitor of Apaf-1) as a cytosolic cytochrome c target. Here we explore the cytochrome c / 14-3-3€ interaction and show the ability of cytochrome c to block 14-3-3€-mediated Apaf-1 inhibition, thereby unveiling a novel function for cytochrome c as an indirect activator of caspase-9/3. We have used calorimetry, NMR spectroscopy, site mutagenesis and computational calculations to provide an insight into the structural features of the cytochrome c / 14-3-3€ complex. Overall, these findings suggest an additional cytochrome c-mediated mechanism to modulate apoptosome formation, shedding light onto the rigorous apoptotic regulation network
