Increased mRNA expression levels of ERCC1, OGG1 and RAI in colorectal adenomas and carcinomas
BACKGROUND: The majority of colorectal cancer (CRC) cases develop through the adenoma-carcinoma pathway. If an increase in DNA repair expression is detected in both early adenomas and carcinomas it may indicate that low repair capacity in the normal mucosa is a risk factor for adenoma formation. METHODS: We have examined mRNA expression of two DNA repair genes, ERCC1 and OGG1 as well as the putative apoptosis controlling gene RAI, in normal tissues and lesions from 36 cases with adenomas (mild/moderat n = 21 and severe n = 15, dysplasia) and 9 with carcinomas. RESULTS: Comparing expression levels of ERCC1, OGG1 and RAI between normal tissue and all lesions combined yielded higher expression levels in lesions, 3.3-fold higher (P = 0.005), 5.6-fold higher(P < 3·10(-5)) and 7.7-fold higher (P = 0.0005), respectively. The levels of ERCC1, OGG1 and RAI expressions when comparing lesions, did not differ between adenomas and CRC cases, P = 0.836, P = 0.341 and P = 0.909, respectively. When comparing expression levels in normal tissue, the levels for OGG1 and RAI from CRC cases were significantly lower compared to the cases with adenomas, P = 0.012 and P = 0.011, respectively. CONCLUSION: Our results suggest that increased expression of defense genes is an early event in the progression of colorectal adenomas to carcinomas
Effect of Blend of Essential Oils on Growth Performance, Carcass Characteristics, Meat Quality, Intestinal Morphology, Serum Biochemistry, and Immune Response of Broiler Chickens
ABSTRACT The purpose of this study was to assess the impact of a blend of essential oils from eucalyptus, citrus, bromohexene HCl, thymole and camphor on the growth performance, carcass characteristics, meat quality, intestinal morphology, serum biochemistry, and immune response of broiler chickens. A total of 240 day-old chicks were divided into four groups, each with six replicates containing ten birds. The experiment was conducted under a completely randomized design (CRD). Different concentrations of the oil blend (0mL/kg, 0.15mL/kg, 0.30mL/kg and 0.45mL/kg) were added to the diet. The findings showed that, in comparison to the other groups, the birds that were given a blend of essential oils at concentrations of 0.30 and 0.45 mL/kg showed improved weight gain, feed efficiency, carcass yield, villus height, crypt depth, and greater immune response against Newcastle disease vaccination (p<0.05). Nonetheless, there was no statistically significant difference in the yields of the breast and thighs, feed consumption, mortality, weights of the liver, wing, heart, and gizzard, or abdominal fat between the treatments. The addition of a blend of essential oils at the doses of 0.30 and 0.45 mL/kg significantly lowered the pH of the meat in comparison to the other groups (p<0.05). All blood biochemical markers, including total serum protein, albumin, globulin, glucose, cholesterol, triglyceride, and uric acid, revealed no variations between the treatments. In conclusion, adding 0.30mL/kg of a blend of essential oils to broiler diets may be the optimum level to improve overall performance without adversely affecting the blood biochemical profile
Polymorphisms of the XRCC1, XRCC3 and XPD genes and risk of colorectal adenoma and carcinoma, in a Norwegian cohort: a case control study
BACKGROUND: Genetic polymorphisms in DNA repair genes may influence individual variation in DNA repair capacity, which may be associated with risk of developing cancer. For colorectal cancer the importance of mutations in mismatch repair genes has been extensively documented. Less is known about other DNA repair pathways in colorectal carcinogenesis. In this study we have focused on the XRCC1, XRCC3 and XPD genes, involved in base excision repair, homologous recombinational repair and nucleotide excision repair, respectively. METHODS: We used a case-control study design (157 carcinomas, 983 adenomas and 399 controls) to test the association between five polymorphisms in these DNA repair genes (XRCC1 Arg(194)Trp, Arg(280)His, Arg(399)Gln, XRCC3 Thr(241)Met and XPD Lys(751)Gln), and risk of colorectal adenomas and carcinomas in a Norwegian cohort. Odds ratio (OR) and 95% confidence interval (95% CI) were estimated by binary logistic regression model adjusting for age, gender, cigarette smoking and alcohol consumption. RESULTS: The XRCC1 280His allele was associated with an increased risk of adenomas (OR 2.30, 95% CI 1.19–4.46). The XRCC1 399Gln allele was associated with a reduction of risk of high-risk adenomas (OR 0.62, 95% CI 0.41–0.96). Carriers of the variant XPD 751Gln allele had an increased risk of low-risk adenomas (OR 1.40, 95% CI 1.03–1.89), while no association was found with risk of carcinomas. CONCLUSION: Our results suggest an increased risk for advanced colorectal neoplasia in individuals with the XRCC1 Arg(280)His polymorphism and a reduced risk associated with the XRCC1 Arg(399)Gln polymorphism. Interestingly, individuals with the XPD Lys(751)Gln polymorphism had an increased risk of low-risk adenomas. This may suggest a role in regression of adenomas
The synergetic effects of surface texturing and MoDDP additive applied to ball-on-disk friction subject to both flooded and starved lubrication conditions
This paper reports a novel work on the synergetic effects of microscale surface texturing and lubricant friction modifier additive of molybdenum dialkyldithiophosphate (MoDDP) subject to both flooded and starved lubrication conditions. The experiments were performed on reciprocating ball-on-disk friction in GTL8 base oil with and without MoDDP. In the flooded lubrication condition, the test results demonstrated that the presence of the MoDDP additive contributed to lower friction coefficients, and also more pronounced effect of surface textures on friction than in the case of the bare base oil. In the starved lubrication experiments, textured and texture-free surfaces in the oils with and without MoDDP additive were tested until an abrupt rising of friction coefficient was detected. The results showed that the magnitude of friction coefficient before terminating each test was the almost same for various tests, while the endurance time in different test conditions was significantly different. The textured surface exhibited longer endurance time than the texture-free surface, especially when the MoDDP additive was used. The mechanism of the synergetic effects of surface textures and MoDDP additive has been discussed based on the experimental observations in the following sections. This study provides a new idea for the application of surface texture in boundary lubrication when lubricant additive is contained in the lubricating oils
Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study
Background: Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. // Methods: We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung's disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. // Findings: We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung's disease) from 264 hospitals (89 in high-income countries, 166 in middle-income countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in low-income countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. // Interpretation: Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between low-income, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030
Pharmacological evaluation of poly(3-methylthiophene) and its titanium(IV)phosphate nanocomposite: DNA interaction, molecular docking, and cytotoxic activity
Cancer and pathogenic microbial diseases have terribly affected human health over a longer period of time. In response to the increasing casualties due to cancer and microbial diseases, unique poly(3-methylthiophene) and poly(3-methylthiophene)-titanium(IV)phosphate composite were prepared via in-situ oxidative chemical polymerization in this work. The poly(3-methylthiophene) and poly(3-methylthiophene)-titanium(IV)phosphate composite were well characterized by Fourier transform infrared spectroscopy and field emission scanning electron microscopy. DNA binding studies by UV–Visible and fluorescence spectroscopic investigations indicated strong binding affinities of poly(3-methylthiophene) and poly(3-methylthiophene)-titanium(IV)phosphate nanocomposite; leading to structural damage of DNA. Poly(3-methylthiophene)-titanium(IV)phosphate nanocomposite showed stronger interactions with DNA as compared to poly(3-methylthiophene) and from dye displacement assay it was confirmed that mode of binding of both the formulations was intercalative. The antimicrobial screening revealed that polymer and its composite displayed stronger antibacterial effects than ampicillin against Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella typhimurium. Besides, the poly(3-methylthiophene) and poly(3-methylthiophene)-titanium(IV)phosphate nanocomposite showed dose dependent effects towards estrogen receptor positive breast cancer (MCF-7) and estrogen receptor negative breast cancer (MDA-MB-231) cell lines; with poly(3-methylthiophene)-titanium(IV)phosphate nanocomposite showing better activities against both cell lines. In all in-vitro biological investigations, poly(3-methylthiophene)-titanium(IV)phosphate composite showed superior properties to that of the pure poly(3-methylthiophene), which encouraged us to suggest its potential as future therapeutic gear in drug delivery and other allied fields
Bioremediation and decontamination potential of flagellate Poteriospumella sp
Water pollution due to heavy metals and water borne pathogen is one of the significant public health issues for developing countries in particular where safe drinking water supply is depleting rapidly. Flagellates are aquatic microscopic organisms that have developed excellent survival strategies, which help them withstand environmental stresses and therefore can act as natural biocontrol in complex ecological systems. This study investigates the wastewater treatment and bioremediation potential of flagellate Poteriospumella sp. Poteriospumella sp. was isolated from northern areas of Pakistan and identified by using 18SrRNA analysis. Wastewater samples were collected from nullah (S1) and two industrial areas of Islamabad and Rawalpindi (S2 and S3) and physiochemical properties (Biological oxygen demand, dissolved oxygen, total nitrogen, ammonia, phosphate and sulfate) were analyzed initially and after treatment with Poteriospumella sp. alone and in combination with consortia of four already characterized bacterial strains i.e., Pseudomonas sp., Salmonella sp., Citrobacter sp., and Enterobacter sp. for 15 days. Furthermore, the bacterivores potential of flagellates was investigated against commonly found endemic waterborne pathogen i.e. Vibrio cholera as well as their ability to sequester or remove heavy metal, mercury (Hg), was also evaluated. The results clearly showed the excellent potential of flagellates in improving wastewater physiochemical properties of all the three wastewater samples i.e. the removal range of BOD (0–10.5%), DO (7.8–38.2%), TN (11.6–53.6%), ammonia (2.7–33.5%), phosphate (0–40%) and sulfate (5.1–12.5%) was recorded. In nutshell, Poteriospumella sp. showed excellent bioremediation potential against Hg (removal of 92–93% of mercuric ions within 2 weeks). In addition, it has significantly reduced the CFU’s of unique subtypes of Vibrio cholerae types circulating in Pakistan and hence can be employed as natural predator to control cholera transmission through water
Effects of polymorphisms in <it>ERCC1</it>, <it>ASE-1 </it>and <it>RAI </it>on the risk of colorectal carcinomas and adenomas: a case control study
<p>Abstract</p> <p>Background</p> <p>The risk of sporadic colorectal cancer is mainly associated with lifestyle factors and may be modulated by several genetic factors of low penetrance. Genetic variants represented by single nucleotide polymorphisms in genes encoding key players in the adenoma carcinoma sequence may contribute to variation in susceptibility to colorectal cancer. In this study, we aimed to evaluate whether the recently identified haplotype encompassing genes of DNA repair and apoptosis, is associated with increased risk of colorectal adenomas and carcinomas.</p> <p>Methods</p> <p>We used a case-control study design (156 carcinomas, 981 adenomas and 399 controls) to test the association between polymorphisms in the chromosomal region 19q13.2-3, encompassing the genes <it>ERCC1</it>, <it>ASE-1 </it>and <it>RAI</it>, and risk of colorectal adenomas and carcinomas in a Norwegian cohort. Odds ratio (OR) and 95% confidence interval (CI) were estimated by binary logistic regression model adjusting for age and gender.</p> <p>Results</p> <p>The <it>ASE-1 </it>polymorphism was associated with an increased risk of adenomas, OR of 1.39 (95% CI 1.06–1.81), which upon stratification was apparent among women only, OR of 1.66 (95% CI 1.15–2.39). The <it>RAI </it>polymorphism showed a trend towards risk reduction for both adenomas (OR of 0.70, 95% CI 0.49–1.01) and carcinomas (OR of 0.49, 95% CI 0.21–1.13) among women, although not significant. Women who were homozygous carriers of the high risk haplotype had an increased risk of colorectal cancer, OR of 2.19 (95% CI 0.95–5.04) compared to all non-carriers although the estimate was not statistically significant.</p> <p>Conclusion</p> <p>We found no evidence that the studied polymorphisms were associated with risk of adenomas or colorectal cancer among men, but we found weak indications that the chromosomal region may influence risk of colorectal cancer and adenoma development in women.</p
