MARKETS FOR NORTHERN PLAINS AQUACULTURE--CASE STUDY OF TILAPIA
The purpose of this study is to identify and investigate alternative fresh and frozen fillet markets for tilapia within the region. The competition for this market is primarily an imported product from Asia and Central America. Total imports plus domestic production has increased from 16.95 million pounds in 1992 to 70.74 million pounds in 1997. Thirty-seven of the 79 respondents handled tilapia in their business. Thirty of these businesses handled and preferred fresh fillets while ten handled frozen tilapia. The tilapia businesses were clear in their preferences: 5 to 7 ounce fillets, quick delivery response time, constant supply, taste and size, and suppliers oriented toward customer service. Twenty-six of the 37 respondents were open to new suppliers. The responding businesses which did not handle tilapia gave their reasons: lack of demand due to customer unfamiliarity, name recognition and taste of tilapia. The need for an established market, i.e., consumer demand, was the major factor. The domestically produced tilapia did not test well in any of the three sensory perception taste tests. The results of these tests indicate both a quality issue and a variation in quality from test to test. Mental health-related communication in a virtual community: text mining analysis of a digital exchange platform during the Covid-19 pandemic.
BACKGROUND
Virtual communities played an important role in mental health and well-being during the Covid-19 pandemic by providing access to others and thereby preventing loneliness. The pandemic has accelerated the urge for digital solutions for people with pre-existing mental health problems. So far, it remains unclear how the people concerned communicate with each other and benefit from peer-to-peer support on a moderated digital platform.
OBJECTIVE
The aim of the project was to identify and describe the communication patterns and verbal expression of users on the inCLOUsiv platform during the first lockdown in 2020.
METHODS
Discussions in forums and live chats on inCLOUsiv were analysed using text mining, which included frequency, correlation, n-gram and sentiment analyses.
RESULTS
The communication behaviour of users on inCLOUsiv was benevolent and supportive; and 72% of the identified sentiments were positive. Users addressed the topics of 'corona', 'anxiety' and 'crisis' and shared coping strategies.
CONCLUSIONS
The benevolent interaction between users on inCLOUsiv is in line with other virtual communities for Covid-19 and the potential for peer-to-peer support. Users can benefit from each other's experiences and support each other. Virtual communities can be used as an adjuvant to existing therapy, particularly in times of reduced access to local health services
Cj0683 Is a Competence Protein Essential for Efficient Initialization of DNA Uptake in Campylobacter jejuni
C. jejuni is an important food-borne pathogen displaying high genetic diversity, substantially based on natural transformation. The mechanism of DNA uptake from the environment depends on a type II secretion/type IV pilus system, whose components are partially known. Here, we quantified DNA uptake in C. jejuni at the single cell level and observed median transport capacities of approximately 30 kb per uptake location. The process appeared to be limited by the initialization of DNA uptake, was finite, and, finalized within 30 min of contact to DNA. Mutants lacking either the outer membrane pore PilQ or the inner membrane channel ComEC were deficient in natural transformation. The periplasmic DNA binding protein ComE was negligible for DNA uptake, which is in contrast to its proposed function. Intriguingly, a mutant lacking the unique periplasmic protein Cj0683 displayed rare but fully functional DNA uptake events. We conclude that Cj0683 was essential for the efficient initialization of DNA uptake, consistent with the putative function as a competence pilus protein. Unravelling features important in natural transformation might lead to target identification, reducing the adaptive potential of pathogens
Can Frequency Domain Heart Rate Measures Detect Impaired Driver Performance?
An overnight driving simulation scenario with partial sleep deprivation was utilized to induce driver performance impairment. Heart rate (HR) was recorded over the entire experiment; frequency domain HR measures were derived and correlated to variation of lane deviation (VLD), a driving performance measure, and to the driver\u27s state, which was estimated by the Karolinska Sleepiness Scale (KSS). The aim of this study is to evaluate whether frequency domain heart rate measures can be used to detect impaired driver performance as well as reduced driver state. We generalize the concept of the conventional frequency domain HR measures – namely the very-low frequency (VLF), low frequency (LF) band and high frequency (HF) band – into finer-grained frequency bands of 0.02 Hz width. These newly defined frequency bands show a more detailed correlation to driving performance and to driver sleepiness state, taking subjectspecific differences into account
Electron effective mass in Sn-doped monoclinic single crystal -gallium oxide determined by mid-infrared optical Hall effect
The isotropic average conduction band minimum electron effective mass in
Sn-doped monoclinic single crystal -GaO is experimentally
determined by mid-infrared optical Hall effect to be
combining investigations on () and () surface cuts. This result
falls within the broad range of values predicted by theoretical calculations
for undoped -GaO. The result is also comparable to recent
density functional calculations using the
Gaussian-attenuation-Perdue-Burke-Ernzerhof hybrid density functional, which
predict an average effective mass of (arXiv:1704.06711
[cond-mat.mtrl-sci]). Within our uncertainty limits we detect no anisotropy for
the electron effective mass, which is consistent with most previous theoretical
calculations. We discuss upper limits for possible anisotropy of the electron
effective mass parameter from our experimental uncertainty limits, and we
compare our findings with recent theoretical results
Maize Transposable Elements Ac/Ds as Insertion Mutagenesis Tools in Candidaalbicans
In non-model systems genetic research is often limited by the lack of
techniques for the generation and identification of gene mutations. One
approach to overcome this bottleneck is the application of transposons for
gene tagging. We have established a two-element transposon tagging system,
based on the transposable elements Activator (Ac)/Dissociation (Ds) from
maize, for in vivo insertion mutagenesis in the fungal human pathogen Candida
albicans. A non-autonomous Ds transposon carrying a selectable marker was
constructed into the ADE2 promoter on chromosome 3 and a codon usage-adapted
Ac transposase gene was inserted into the neutral NEUT5L locus on chromosome
5. In C. albicans cells expressing the transposase the Ds element efficiently
excised and reintegrated elsewhere in the genome, which makes the Ac/Ds
transposons promising tools for saturating insertion mutagenesis in clinical
strains of C. albicans
Curcumin Mitigates Immune-Induced Epithelial Barrier Dysfunction by Campylobacter jejuni
Campylobacter jejuni (C. jejuni) is the most common cause of foodborne gastroenteritis worldwide. The bacteria induce diarrhea and inflammation by invading the intestinal epithelium. Curcumin is a natural polyphenol from turmeric rhizome of Curcuma longa, a medical plant, and is commonly used in curry powder. The aim of this study was the investigation of the protective effects of curcumin against immune-induced epithelial barrier dysfunction in C. jejuni infection. The indirect C. jejuni-induced barrier defects and its protection by curcumin were analyzed in co-cultures with HT-29/B6-GR/MR epithelial cells together with differentiated THP-1 immune cells. Electrophysiological measurements revealed a reduction in transepithelial electrical resistance (TER) in infected co-cultures. An increase in fluorescein (332 Da) permeability in co-cultures as well as in the germ-free IL-10-/- mouse model after C. jejuni infection was shown. Curcumin treatment attenuated the C. jejuni-induced increase in fluorescein permeability in both models. Moreover, apoptosis induction, tight junction redistribution, and an increased inflammatory response-represented by TNF-α, IL-1β, and IL-6 secretion-was observed in co-cultures after infection and reversed by curcumin. In conclusion, curcumin protects against indirect C. jejuni-triggered immune-induced barrier defects and might be a therapeutic and protective agent in patients
