Re-Os dating of pyrite confirms an early diagenetic onset and extended duration of mineralization in the Irish Zn-Pb ore field
0000-0002-7706-6003The Irish Midlands region contains one of the world’s largest hydrothermal Zn-Pb ore districts, but uncertainty exists in the timing of mineralization relative to host rock ages. Consequently, genetic models for ore formation are poorly constrained and remain controversial. Here we use Re-Os geochronology to show that ore-stage pyrite from the Lisheen deposit formed at 346.6 ± 3.0 Ma, shortly after host rock deposition. Pyrite from the Silvermines deposit returns an age of 334.0 ± 6.1 Ma, indicating that at least some mineralization occurred during later burial. These age determinations show that the much younger paleomagnetic ages reported for the Irish Zn-Pb deposits reflect remagnetization during the Variscan orogeny, a process that we suggest affects paleomagnetic dating more widely. The Re-Os ages overlap with the ages of lower Carboniferous volcanic rocks in the Midlands, which are the product of magmatism that has been invoked as the driving force for hydrothermal activity. The relatively low initial Os ratios for both Lisheen (0.253 ± 0.045) and Silvermines (0.453 ± 0.006) are compatible with derivation of Os from these magmas, or from the Caledonian basement that underlies the ore deposits.This journal is published under the terms of Green Open Access. Effect of Pseudomonas fluorescens proteases on the quality of Cheddar cheese
peer-reviewedThe objective of this study was to investigate the effect of adding different levels of a thermoresistant protease produced by a Pseudomonas fluorescens strain to milk on the manufacture and quality of Cheddar cheese. Fresh raw milk was collected, standardized, and pasteurized at 72°C for 15 s, and the enzyme was added to give a protease activity of 0.15 or 0.60 U/L (treatments P1 and P4, respectively), while one sample had no enzyme added (control). Milk was stored at 4°C for 48 h and Cheddar cheese was manufactured after 0 and 48 h of storage. Results indicated that the protease was active in milk during 48 h of storage; however, its effect on milk composition was minimal. The protein that was preferentially hydrolyzed by the protease over storage was β-casein, followed by κ-casein. The mean cheese yield and recovery of fat and protein obtained for all cheeses were not affected by protease activity. The protease showed low activity during cheese manufacture, possibly because of unfavorable conditions, including low pH. One of the factors that might have influenced protease activity was the pH of the curd (approximately 6.55 after acidification and 5.35 at milling), which was lower than that at which the enzyme would have optimum activity (pH 7 to 9). Consequently, the composition, pH, patterns of proteolysis, and hardness of all cheeses produced were similar and in accordance with values expected for that type of cheese, independently of the protease activity level. However, slight increases in proteolysis were observed in P4 cheeses and produced using milk stored for 48 h. Both the P1 and P4 cheeses had higher concentrations of free amino acids (FAA) compared with the control, whereas urea-PAGE electrophoretograms indicated a greater breakdown of caseins in the P4 cheese samples, which may be related to possible increases in numbers of proteolytic bacteria in milk during storage. Therefore, the thermoresistant psychrotrophic bacterial protease(s) tested in this study may affect the manufacture or quality of Cheddar cheese during ripening to a relatively limited extent. However, controlling initial levels of proteolytic bacteria in raw milk remains essential, because proteolysis affects the development of flavor and texture in cheese
Duodenal enteroglucagonoma revealed by differential comparison of serum and tissue glucagon reactivity with Siemens' Double Glucagon Antibody and DakoCytomation's Polyclonal Rabbit Anti-Human Glucagon: a case report
<p>Abstract</p> <p>Introduction</p> <p>This case report demonstrates that the differential immunohistochemical reactivities of Siemens' <it>Double Antibody Glucagon </it>compared to DakoCytomation's <it>Polyclonal Rabbit Anti-Human Glucagon </it>allow for pathologic distinction of enteral versus pancreatic glucagonoma.</p> <p>Case presentation</p> <p>A 64-year-old Caucasian man was diagnosed with a duodenal enteroglucagonoma following presentation with obstructive jaundice. He had a low serum glucagon level using Siemens' <it>Double Antibody Glucagon</it>, a clinical syndrome consistent with glucagon hypersecretion. A periampullary mass biopsy proved to be a neuroendocrine tumor, with positive immunohistochemical reactivity to DakoCytomation's <it>Polyclonal Rabbit Anti-Human Glucagon</it>.</p> <p>Conclusions</p> <p>Differential comparison of the immunohistochemical reactivities of Siemens' <it>Double Antibody Glucagon </it>and DakoCytomation's <it>Polyclonal Rabbit Anti-Human Glucagon </it>discerns enteroglucagon from pancreatic glucagon.</p
Walk well:a randomised controlled trial of a walking intervention for adults with intellectual disabilities: study protocol
Background - Walking interventions have been shown to have a positive impact on physical activity (PA) levels, health and wellbeing for adult and older adult populations. There has been very little work carried out to explore the effectiveness of walking interventions for adults with intellectual disabilities. This paper will provide details of the Walk Well intervention, designed for adults with intellectual disabilities, and a randomised controlled trial (RCT) to test its effectiveness. Methods/design - This study will adopt a RCT design, with participants allocated to the walking intervention group or a waiting list control group. The intervention consists of three PA consultations (baseline, six weeks and 12 weeks) and an individualised 12 week walking programme. A range of measures will be completed by participants at baseline, post intervention (three months from baseline) and at follow up (three months post intervention and six months from baseline). All outcome measures will be collected by a researcher who will be blinded to the study groups. The primary outcome will be steps walked per day, measured using accelerometers. Secondary outcome measures will include time spent in PA per day (across various intensity levels), time spent in sedentary behaviour per day, quality of life, self-efficacy and anthropometric measures to monitor weight change. Discussion - Since there are currently no published RCTs of walking interventions for adults with intellectual disabilities, this RCT will examine if a walking intervention can successfully increase PA, health and wellbeing of adults with intellectual disabilities

The influence of exercise training status on antigen-stimulated IL-10 production in whole blood culture and numbers of circulating regulatory T cells
The final publication is available at springerlink.com.Highly trained athletes are associated with high resting antigen-stimulated whole blood culture interleukin (IL)-10 production. The purpose of the present study was to examine the effects of training status on resting circulating T regulatory (T) cell counts and antigen-stimulated IL-10 production and the effect of acute bout of exercise on the T response. Forty participants volunteered to participate and were assigned to one of the four groups: sedentary (SED), recreationally active (REC), sprint-trained athletes and endurance-trained athletes (END). From the resting blood sample, CD4CD25CD127 T cells and in vitro antigen-stimulated IL-10 production were assessed. Ten REC subjects performed 60 min cycling at 70 % of maximal oxygen uptake and blood samples for T analysis were collected post- and 1 h post-exercise. IL-10 production was greater in END compared with the other groups (P < 0.05). END had a higher T percentage of total lymphocyte count compared with SED (P < 0.05). A smaller proportion of T CD4 cells were observed in SED compared with all other groups (P < 0.05). IL-10 production significantly correlated with the proportion of T within the total lymphocyte population (r = 0.51, P = 0.001). No effect of acute exercise was evident for T cell counts in the REC subjects (P > 0.05). Our results demonstrate that high training loads in END are associated with greater resting IL-10 production and T cell count and suggest a possible mechanism for depression of immunity commonly reported in athletes engaged in high training loads. © 2013 Springer-Verlag Berlin Heidelberg

An overview on the inconsistencies of approach in regulating the capital position of banks: Will the United Kingdom step out of line with Europe?
After the collapse of a number of banking institutions and bailouts of banks by governments, regulators have taken a different attitude and now appear keen to take regulation seriously when it comes to ensuring that banks have adequate capital and sufficient liquidity. Not only that, but in the United Kingdom, the Independent Commission on Banking Reform has made proposals with regard to the capital position of banks. This article, which is an overview, will look at matters from a UK perspective and at the proposals for reform. This article, after its introduction and summary, will look at a number of areas: first, the reforms made by Basel III; second, the regulation of Systemically Important Financial Institutions (Sifis) and the proposals for dealing with these; third, some matters in relation to lending that relate to capital and liquidity generally; fourth, increased stress testing of banks; fifth, derivatives and risk taking and the new proposed structure of regulation in the United Kingdom; sixth, the war of spin between regulators and banks; seventh, Shadow Banking; and eighth, The Independent Commission on Banking Reform and its proposals for reform. It will also be a theme that the various proposals lack consistency and that this could lead to regulatory arbitrage. It is already clear that there are inconsistencies between the various regulatory organisations, with proposals in the United Kingdom indicating that banks will be required to keep much higher levels of capital than those proposed by Basel and the European Community. Cosmic Ray Anomalies from the MSSM?
The recent positron excess in cosmic rays (CR) observed by the PAMELA
satellite may be a signal for dark matter (DM) annihilation. When these
measurements are combined with those from FERMI on the total () flux
and from PAMELA itself on the ratio, these and other results are
difficult to reconcile with traditional models of DM, including the
conventional mSUGRA version of Supersymmetry even if boosts as large as
are allowed. In this paper, we combine the results of a previously
obtained scan over a more general 19-parameter subspace of the MSSM with a
corresponding scan over astrophysical parameters that describe the propagation
of CR. We then ascertain whether or not a good fit to this CR data can be
obtained with relatively small boost factors while simultaneously satisfying
the additional constraints arising from gamma ray data. We find that a specific
subclass of MSSM models where the LSP is mostly pure bino and annihilates
almost exclusively into pairs comes very close to satisfying these
requirements. The lightest in this set of models is found to be
relatively close in mass to the LSP and is in some cases the nLSP. These models
lead to a significant improvement in the overall fit to the data by an amount
dof in comparison to the best fit without Supersymmetry
while employing boosts . The implications of these models for future

The plasma membrane carbonic anhydrase in murine hepatocytes identified as isozyme XIV
BACKGROUND: Biochemical and histochemical studies have both previously indicated plasma membrane-associated carbonic anhydrase (CA) activity in hepatocytes which has been assumed to be CA IV. However, immunohistochemical data did not support this assignment. Recent northern blotting results indicated the presence of mRNA for the most recently discovered membrane-bound CA isozyme, CA XIV, in the liver. The present study was designed to examine whether CA XIV could contribute to the CA activity described in the hepatocytes. METHODS: Tissue samples from mouse liver were subjected to immunohistochemical staining using the antibodies raised against recombinant mouse CA XIV and CA IV. RT-PCR and western blotting were also performed for CA XIV. RESULTS: A strong immunofluorescent signal was observed in the plasma membrane of mouse hepatocytes. Although CA XIV was expressed on both the apical and basolateral surfaces, the staining was more prominent at the apical (canalicular) membrane domain. The expression of CA XIV in the liver was confirmed by RT-PCR and western blotting. CONCLUSIONS: The presence of CA XIV in the hepatocyte plasma membrane places this novel enzyme at a strategic site to control pH regulation and ion transport between the hepatocytes, sinusoids and bile canaliculi
