    Psychoneuroimmunology and Chronic Pain

    There is increasing evidence linking chronic pain to altered levels of central nervous system (CNS) inflammation and increased levels of perceived pain, anxiety, depression and sleep disturbance. However the inflammatory molecules responsible for physiologic and psychological components of chronic pain still warrant identification and exploration. Using central inflammation as a paramount factor in the creation and maintenance of chronic pain, this study aimed to investigate and describe the physical and psychological aspects of chronic pain associated with CNS inflammation. Using a cross-sectional correlational design, cerebrospinal fluid (CSF) inflammatory cytokine patterns present in 8 chronic pain participants were compared to inflammatory cytokine patterns present in 30 control CSF samples using multivariate analysis of variance (MANOVA), with analysis of variance (ANOVA) analysis as a follow-up. Levels of depression, anxiety, sleep disturbance and pain were measured in approximately 8 chronic pain patients and correlated to CSF levels of inflammatory cytokines using Pearson’s r, or Spearman’s Rho correlations. Demographic information was also explored for relationships to central inflammation and descriptive statistics were examined for responses. To our knowledge, this is the first study to describe increased CSF levels of Interleukin-8 (IL-8) in a population of majority Failed Back Surgery Syndrome chronic pain patients (F (1, 36) = 14.89, p \u3c 0.001, partial η2 = 0.293). Gender (F (1,6) = 7.782, p = 0.565), socioeconomic status (r = -0.823, p = 0.012) and educational level (r = 0.727, p = 0.041) were also correlated with central levels of inflammation, indicating that central physiologic changes may be related to host sex and psychosocial factors. All participants reported poor sleep quality and took at least one opioid medication, indicating that sleep and opioids may scaffold a portion of the chronic pain paradigm. This study richly describes the dynamic experience of chronic pain through the lens of Psychoneuroimmunology. Incorporating both physiologic and psychological aspects of the disease, this study describes an association between chronic pain, central inflammation, gender, socioeconomic status, opioid medications and poor sleep quality. Thus, the future of pain treatment must consider these aspects when treating patients and look to future studies for possible new treatment options which target these factors

    Modelación del efecto de escenarios de cobertura sobre la migración de nutrientes (N, P2O5) en la cuenca alta del río Magdalena (Huila, Colombia)*

    The combination of deforestation due to agricultural frontier expansion and the increased use of fertilizers due to agricultural intensification has lead to an increase in the load of sediments and pollutants that negatively affect water quality in rural areas. This problem is particularly serious in the Colombian Andean region. This study applies a hydrological simulation model to compare the effect of four vegetable coverage management scenarios on nutrient migration from agricultural practices in the Upper Magdalena river basin. The simulation of water movement over hillsides and channels, which was run using the MIKE SHE and MIKE 11 models, showed an acceptable performance (S/σ =0.84, R=0.82, R2=0.67). However, pollutant migration showed a very low sensitivity to vegetable coverage in the configuration of the different model simulations (p > 0.05), with reductions in pollutant loads ranging only between 2 and 4%. Such reductions have to do with the intrinsic properties of vegetation (which act like a barrier to sediment flow,) with its ability to increase soil infiltration (therefore reducing runoff,) and with nutrient absorption by plants

    Background: Inflammatory Bowel Diseases (IBD), are considered a growing global disease, with about ten million people being affected worldwide. Maintenance of intestinal barrier integrity is crucial for preventing IBD onset and exacerbations. Some recent patents regarding oily formulations containing probiotics (WO2010122107A1 and WO2010103374A9) and the use of probiotics for gastrointestinal complaints (US20110110905A1 and US9057112B2) exist, or are pending application. Objective: In this work, we studied the effect of a fixed combination of registered Lactobacillus reuteri and Lactobacillus acidophilus strains and herbal extracts in an in vitro inflammation experimental model. Methods: Caco-2 cell monolayer was exposed to INF-\u3b3+TNF-\u3b1 or to LPS; Trans Epithelial Electrical Resistance (TEER) and paracellular permeability were investigated. ZO-1 and occludin Tight Junctions (TJs) were also investigated by mean of immunofluorescence. Results: Pre-treatment with the fixed combination of probiotics and herbal extracts prevented the inflammation-induced TEER decrease, paracellular permeability increase and TJs translocation. Conclusions: In summary, the fixed combination of probiotics and herbal extracts investigated in this research was found to be an interesting candidate for targeting the re-establishment of intestinal barrier function in IBD conditions

    Antiveneno de serpiente coral producido en gallinas (Gallus domesticus)

    The production of anti-snake venom from large mammal's blood has been found to be low-yielding and arduous, consequently, antivenom immunoglobulins for treatment are achieved regularly as polyvalent serum. We have standardized an undemanding technique for making purified immunoglobulin IgY antivenom consisting of polyclonal antibodies against coral snake venom in the egg yolk of immunized hens. We have adapted a reported process of antibody purification from egg yolks, and achieved 90% antibody purity. The customized technique consisted of the removal of lipids from distilled water-diluted egg yolks by a freeze–thaw sequence. The specific immunoglobulins were present in the egg yolk for up to 180 days postimmunization. Therefore, by means of small venom quantities, a significant amount of immunoglobulins were found in an adequately purified state (The obtained material contained about 90% pure IgY). The antigen binding of the immunoglobulins was detected by a double immunodiffusion test. Titers of antibodies in the yolk were estimated with a serum protection assay (Median effective dose = ED50) (ED50= 477 mg/kg). Given that breeding hens is economically feasible, egg gathering is noninvasive and the purification of IgY antibodies is quick and easy, chicken immunization is an excellent alternative for the production of polyclonal antibodies. To the best of our knowledge, this is the first coral snake antivenom prepared in birds.La producción de antiveneno de serpiente usando sangre de grandes mamíferos se ha encontrado que es de bajo rendimiento y de trabajo arduo, en consecuencia, las inmunoglobulinas antiveneno para el tratamiento se obtienen generalmente, como suero polivalente. Hemos estandarizado una técnica poco exigente para la fabricación de inmunoglobulina purificada IgY, que consistió en generar anticuerpos policlonales contra el veneno de la serpiente coral en huevos de gallinas inmunizadas. La técnica consistió en la eliminación de lípidos de las yemas del huevo, diluidas en agua y en una secuencia de congelación-descongelación. Las inmunoglobulinas específicas estuvieron presentes en la yema de huevo hasta 180 días después de la inmunización. La unión del antígeno a las inmunoglobulinas se detectó mediante un ensayo de inmunodifusión doble. Los títulos de anticuerpos en la yema fueron estimados con un ensayo de protección (dosis efectiva media = ED50). Dado que las gallinas reproductoras son económicamente viables, la recolección de huevos es no invasiva y la purificación de anticuerpos IgY es rápida y fácil, la inmunización de la gallina es una excelente alternativa para la producción de anticuerpos policlonales.     Influence of Microbial Symbionts on Plant–Insect Interactions

    International audienceThere is growing evidence that microorganisms are important ‘hidden players’ in insect–plant interactions. Insect symbionts can directly affect these interactions by providing insects with key nutrients or by interfering with the plant to modulate food provisioning to insects and plant defences. Insect symbionts can also have indirect cascading ecological consequences at the community level through insect- and plant-mediated effects that include their impact on insect reproduction, on natural enemies of herbivores or on plant-associated microorganisms. Identification of symbiotic communities associated with insects, characterization of transmission and acquisition patterns as well as understanding of molecular mechanisms underlying these plant–insect–microbe interactions have important ecological and evolutionary consequences. This review highlights the excitement that surrounds these investigations and the promise they hold for a better understanding of the functional, ecological and evolutionary impacts of symbionts on plant–insect interactions, with implications and relevance for both applied and fundamental researches

    Mechanical durability of hydrophobic surfaces fabricated by injection moulding of laser-induced textures

    YesThe paper reports an investigation on the mechanical durability of textured thermoplastic surfaces together with their respective wetting properties. A range of laser-induced topographies with different aspect ratios from micro to nanoscale were fabricated on tool steel inserts using an ultrashort pulsed near infrared laser. Then, through micro-injection moulding the topographies were replicated onto polypropylene surfaces and their durability was studied systematically. In particular, the evolution of topographies on textured thermoplastic surfaces together with their wetting properties were investigated after undergoing a controlled mechanical abrasion, i.e. reciprocating dry and wet cleaning cycles. The obtained empirical data was used both to study the effects of cleaning cycles and also to identify cleaning procedures with a minimal impact on textured thermoplastic surfaces and their respective wetting properties.
