    The Complexity of Separating Points in the Plane

    We study the following separation problem: given n connected curves and two points s and t in the plane, compute the minimum number of curves one needs to retain so that any path connecting s to t intersects some of the retained curves. We give the first polynomial (O(n3)) time algorithm for the problem, assuming that the curves have reasonable computational properties. The algorithm is based on considering the intersection graph of the curves, defining an appropriate family of closed walks in the intersection graph that satisfies the 3-path-condition, and arguing that a shortest cycle in the family gives an optimal solution. The 3-path-condition has been used mainly in topological graph theory, and thus its use here makes the connection to topology clear. We also show that the generalized version, where several input points are to be separated, is NP-hard for natural families of curves, like segments in two directions or unit circles

    Optimising low acoustic impedance back-fill material wave barrier dimensions to shield structures from ground borne high speed rail vibrations

    Ground borne vibrations generated due to high speed train passage can cause undesirable effects on the nearby environment. One approach to mitigate vibration levels is to use “wave barriers”, however their installation cost may be very high. In an attempt to minimize construction costs, numerical modelling and physical experiments are performed to determine guidelines for the optimisation of trench dimensions. Substantial savings are found to be achievable through carefully chosen barrier dimensions. Firstly, for the purposes of testing a numerical model that can be used by railway wave barrier designers, a geophysical investigation is undertaken on a high speed railway line outside Edinburgh, Scotland. The testing schedule consists of vibration measurements adjacent to the line and a multi-channel analysis of surface waves technique to determine the underlying soil properties. An outline is then presented describing a newly developed three dimensional finite element railway model using commercially available software. The resulting model is modified to replicate the track and soil properties at the test site and then it is shown that the model results exhibit a strong correlation in comparison to those collected during the experimental stage. The numerical model is then used to assess the ability of a range of wave barrier configurations to protect nearby sites from railway vibration. It is found that depth and length have a strong influence on the mitigation of vibration levels but the effect of trench width is negligible. Lastly, for a specific example it is shown that cost savings of 95% are achievable if trench dimensions are carefully planned.     Effect of luteinized unruptured follicle cycles on clinical outcomes of frozen thawed embryo transfer in Chinese women

    Objective To determine the effect of luteinized unruptured follicle (LUF) cycles on frozen thawed embryo transfer (FET). Design A retrospective analysis comparing the clinical outcomes after FET among 144 cases of luteinized unruptured follicle (LUF) cycles and 866 cases of ovulation cycles. Setting Reproductive medical center, Beijing China. Patient(s) Chinese infertile women who underwent FET. Intervention(s) None. Main outcome measure(s) Clinical pregnancy rate (PR), implantation rate. Result(s) The implantation rate, clinical pregnancy rate, on-going pregnancy rate and live birth rate in LUF group were 12.76% (49/384), 27.78% (40/144), 24.31% (35/144) and 19.44% (28/144), respectively, and in ovulation group, 14.74% (332/2251), 31.29% (271/866), 28.29% (245/866) and 22.23% (193/866), respectively (p > 0.05). Conclusion(s) LUF does not affect the clinical outcomes of FET.     The CIMT-monitoring panel: a two-step approach to harmonize the enumeration of antigen-specific CD8(+) T lymphocytes by structural and functional assays

    The interpretation of the results obtained from immunomonitoring of clinical trials is a difficult task due to the variety of methods and protocols available to detect vaccine-specific T-cell responses. This heterogeneity as well as the lack of standards has led to significant scepticism towards published results. In February 2005, a working group was therefore founded under the aegis of the Association for Immunotherapy of Cancer ("CIMT") in order to compare techniques and protocols applied for the enumeration of antigen-specific T-cell responses. Here we present the results from two consecutive phases of an international inter-laboratory testing project referred to as the "CIMT monitoring panel". A total of 13 centers from six European countries participated in the study in which pre-tested PBMC samples, synthetic peptides and PE-conjugated HLA-tetramers were prepared centrally and distributed to participants. All were asked to determine the number of antigen-specific T-cells in each sample using tetramer staining and one functional assay. The results of the first testing round revealed that the total number of cells analyzed was the most important determinant for the sensitive detection of antigen-specific CD8(+) T-cells by tetramer staining. Analysis by ELISPOT was influenced by a combination of cell number and a resting phase after thawing of peripheral blood mononuclear cells. Therefore, the experiments were repeated in a second phase but now the participants were asked to change their protocols according to the new guidelines distilled from the results of the first phase. The recommendations improved the number of antigen-specific T-cell responses that were detected and decreased the variability between the laboratories. We conclude that a two-step approach in inter-laboratory testing allows the identification of distinct variables that influence the sensitivity of different T-cell assays and to formally show that a defined correction to the protocols successfully increases the sensitivity and reduces the inter-center variability.
