    A simple model of radiative emission in M87

    We present a simple physical model of the central source emission in the M87 galaxy. It is well known that the observed X-ray luminosity from this galactic nucleus is much lower than the predicted one, if a standard radiative efficiency is assumed. Up to now the main model invoked to explain such a luminosity is the ADAF (Advection-Dominated-Accretion-Flow) model. Our approach supposes only a simple axis-symmetric adiabatic accretion with a low angular momentum together with the bremsstrahlung emission process in the accreting gas. With no other special hypothesis on the dynamics of the system, this model agrees well enough with the luminosity value measured by Chandra.Comment: 11 pages, 6 figures, accepted for publication in The Astrophysical Journa

    Why Canonical Disks Cannot Produce Advection Dominated Flows

    Using simple arguments we show that the canonical thin keplerian accretion disks cannot smoothly match any plain advection dominated flow (ADAF) model. By 'plain' ADAF model we mean the ones with zero cooling. The existence of sonic points in exact solutions is critical and imposes constraints that cannot be surpassed adopting 'reasonable' physical conditions at the hypothetical match point. Only the occurrence of new critical physical phenomena may produce a transition. We propose that exact advection models are a class of solutions which don't necessarily involve the standard thin cool disks and suggest a different scenario in which good ADAF solutions could eventually occur.Comment: 12 pages, 2 figures, Accepted for publication in Ap.J.     Pazopanib and trametinib as a synergistic strategy against osteosarcoma: Preclinical activity and molecular insights

    Receptor tyrosine kinases (RTKs) inhibitors’ activity in advanced osteosarcoma is significant but short-lived. To prevent or at least delay drug resistance, we explored a vertical inhibition by combining drugs acting at different levels of the RTK pathways (pazopanib + trametinib). We studied pazopanib + trametinib antitumor activity both in vitro and in vivo (MNNG-HOS and KHOS xenografts in NOD/SCID mice) investigating the molecular mechanisms and potential escapes. The involvement of MAPK-PI3K pathways was validated by Nanostring technology, western blot and by silencing/overexpression experiments. Pazopanib targets were expressed on seven osteosarcoma cell lines and their pathways were activated. Pazopanib + trametinib exhibited synergistic antitumor activity by inducing apoptosis and inhibiting ERK1/2 and Akt. In vivo antitumor activity was shown in osteosarcoma-bearing mice. The drug combination significantly down-modulated RTK Ephrin Type-A Receptor 2 (EphA2) and Interleukin-7 Receptor (IL-7R), whereas induced mitogen-activated protein-kinase kinase (MAPKK) MEK6. EphA2 silencing significantly reduced osteosarcoma cell proliferation and migration, while impeding MEK6 up-regulation in the treated cells significantly increased the antitumor effect of the studied drugs. Moreover, the up-regulation of MEK6 reduced combination activity. Pazopanib + trametinib demonstrated synergistic antitumor effects in osteosarcoma models through ERK and Akt inhibition and EphA2 and IL-7R down-modulation.     Randomized, double-blind, placebo-controlled trial of rapamycin in amyotrophic lateral sclerosis

    In preclinical studies rapamycin was found to target neuroinflammation, by expanding regulatory T cells, and affecting autophagy, two pillars of amyotrophic lateral sclerosis (ALS) pathogenesis. Herein we report a multicenter, randomized, double-blind trial, in 63 ALS patients who were randomly assigned in a 1:1:1 ratio to receive rapamycin 2 mg/m2/day,1 mg/m2/day or placebo (EUDRACT 2016-002399-28; NCT03359538). The primary outcome, the number of patients exhibiting an increase >30% in regulatory T cells from baseline to treatment end, was not attained. Secondary outcomes were changes from baseline of T, B, NK cell subpopulations, inflammasome mRNA expression and activation status, S6-ribosomal protein phosphorylation, neurofilaments; clinical outcome measures of disease progression; survival; safety and quality of life. Of the secondary outcomes, rapamycin decreased mRNA relative expression of the pro-inflammatory cytokine IL-18, reduced plasmatic IL-18 protein, and increased the percentage of classical monocytes and memory switched B cells, although no corrections were applied for multiple tests. In conclusion, we show that rapamycin treatment is well tolerated and provides reassuring safety findings in ALS patients, but further trials are necessary to understand the biological and clinical effects of this drug in ALS

    Thermo-elastic induced phase noise in the LISA Pathfinder spacecraft

    During the On-Station Thermal Test campaign of the LISA Pathfinder the data and diagnostics subsystem was tested in nearly space conditions for the first time after integration in the satellite. The results showed the compliance of the temperature measurement system, obtaining temperature noise around 104KHz1/210^{-4}\,{\rm K}\, {\rm Hz}^{-1/2} in the frequency band of 130  mHz1-30\;{\rm mHz}. In addition, controlled injection of heat signals to the suspension struts anchoring the LISA Technology Package (LTP) Core Assembly to the satellite structure allowed to experimentally estimate for the first time the phase noise contribution through thermo-elastic distortion of the LTP interferometer, the satellite's main instrument. Such contribution was found to be at 1012mHz1/210^{-12}\,{\rm m}\, {\rm Hz}^{-1/2}, a factor of 30 below the measured noise at the lower end of the measurement bandwidth (1mHz1\,{\rm mHz})

    Enhancing methane production from lignocellulosic biomass by combined steam‑explosion pretreatment and bioaugmentation with cellulolytic bacterium Caldicellulosiruptor bescii

    Background: Biogas production from lignocellulosic biomass is generally considered to be challenging due to the recalcitrant nature of this biomass. In this study, the recalcitrance of birch was reduced by applying steam-explosion (SE) pretreatment (210 °C and 10 min). Moreover, bioaugmentation with the cellulolytic bacterium Caldicellulosiruptor bescii was applied to possibly enhance the methane production from steam-exploded birch in an anaerobic digestion (AD) process under thermophilic conditions (62 °C). Results: Overall, the combined SE and bioaugmentation enhanced the methane yield up to 140% compared to untreated birch, while SE alone contributed to the major share of methane enhancement by 118%. The best methane improvement of 140% on day 50 was observed in bottles fed with pretreated birch and bioaugmentation with lower dosages of C. bescii (2 and 5% of inoculum volume). The maximum methane production rate also increased from 4-mL CH4/ g VS (volatile solids)/day for untreated birch to 9-14-mL CH4/ g VS/day for steam-exploded birch with applied bioaugmentation. Bioaugmentation was particularly effective for increasing the initial methane production rate of the pretreated birch yielding 21-44% more methane than the pretreated birch without applied bioaugmentation. The extent of solubilization of the organic matter was increased by more than twofold when combined SE pretreatment and bioaugmentation was used in comparison with the methane production from untreated birch. The beneficial effects of SE and bioaugmentation on methane yield indicated that biomass recalcitrance and hydrolysis step are the limiting factors for efficient AD of lignocellulosic biomass. Microbial community analysis by 16S rRNA amplicon sequencing showed that the microbial community composition was altered by the pretreatment and bioaugmentation processes. Notably, the enhanced methane production by pretreatment and bioaugmentation was well correlated with the increase in abundance of key bacterial and archaeal communities, particularly the hydrolytic bacterium Caldicoprobacter, several members of syntrophic acetate oxidizing bacteria and the hydrogenotrophic Methanothermobacter.
