    Obsidian Source Use in Tongan Prehistory: New Results and Implications

    The article presents results of an obsidian sourcing study on artifacts from Tonga and Fiji. New LA-ICPMS data on obsidian source locations on Tafahi in northern Tonga are discussed in relation to inter-island mobility during two important phases in the Central Pacific: the late-Lapita phase in Fiji-West Polynesia at 2700-2600 cal. BP and during the time of the rise of Polynesian chiefdoms at ~1000-400 cal. BP. The sourcing results indicate that two sources of obsidian were exploited during Tongan prehistory. It is suggested that different modes of interaction were responsible for obsidian movement during the early and late phases of Tongan prehistory

    RASSF1A and the Taxol Response in Ovarian Cancer

    The RASSF1A tumor suppressor gene is frequently inactivated by promoter methylation in human tumors. The RASSF1A protein forms an endogenous complex with tubulin and promotes the stabilization of microtubules. Loss of RASSF1A expression sensitizes cells to microtubule destabilizing stimuli. We have observed a strong correlation between the loss of RASSF1A expression and the development of Taxol resistance in primary ovarian cancer samples. Thus, we sought to determine if RASSF1A levels could dictate the response to Taxol and whether an epigenetic therapy approach might be able to reverse the Taxol resistant phenotype of RASSF1A negative ovarian tumor cells. We found that knocking down RASSF1A expression in an ovarian cancer cell line inhibited Taxol-mediated apoptosis and promoted cell survival during Taxol treatment. Moreover, using a combination of small molecule inhibitors of DNA Methyl Transferase enzymes, we were able restore RASSF1A expression and Taxol sensitivity. This identifies a role for RASSF1A in modulating the tumor response to Taxol and provides proof of principal for the use of epigenetic therapy to overcome Taxol resistance

    All Transients, All the Time: Real-Time Radio Transient Detection with Interferometric Closure Quantities

    We demonstrate a new technique for detecting radio transients based on interferometric closure quantities. The technique uses the bispectrum, the product of visibilities around a closed-loop of baselines of an interferometer. The bispectrum is calibration independent, resistant to interference, and computationally efficient, so it can be built into correlators for real-time transient detection. Our technique could find celestial transients anywhere in the field of view and localize them to arcsecond precision. At the Karl G. Jansky Very Large Array (VLA), such a system would have a high survey speed and a 5-sigma sensitivity of 38 mJy on 10 ms timescales with 1 GHz of bandwidth. The ability to localize dispersed millisecond pulses to arcsecond precision in large volumes of interferometer data has several unique science applications. Localizing individual pulses from Galactic pulsars will help find X-ray counterparts that define their physical properties, while finding host galaxies of extragalactic transients will measure the electron density of the intergalactic medium with a single dispersed pulse. Exoplanets and active stars have distinct millisecond variability that can be used to identify them and probe their magnetospheres. We use millisecond time scale visibilities from the Allen Telescope Array (ATA) and VLA to show that the bispectrum can detect dispersed pulses and reject local interference. The computational and data efficiency of the bispectrum will help find transients on a range of time scales with next-generation radio interferometers.Comment: Accepted to ApJ. 8 pages, 5 figures, 2 tables.     Comparison of diet quality, physical activity and biochemical values of older adults either reporting or not reporting use of lipid-lowering medication

    Objectives: The aim of this study was to compare standard lipid profile, reported dietary intake, and physical activity in older adults who reported taking or not taking a lipid-lowering medication, namely statins. Design: Cross-sectional study utilizing baseline data collected from a subsample of a larger randomized clinical trial, The Study of Exercise and Nutrition in Older Rhode Islanders (SENIOR) Project. Participants and Setting: A total of 115 participants, 33 males and 82 females, over the age of 60, community-dwelling, primarily retired, and from East Providence, Rhode Island and surrounding communities in Rhode Island and Massachusetts. Measurements: Height and weight were measured and used to calculate body mass index. Waist circumference was measured. Medical history and medication use surveys were completed. Dietary assessment was done via three 24 hour recalls using NDS-R. The Yale Physical Activity Survey was used to determine energy expenditure during exercise and a physical activity summary score. Fasting blood samples were obtained to determine lipid profile. Results: 37 participants (32.2%) reported taking lipid-lowering medication, statins exclusively, and 78 (67.8%) reported not taking any lipid-lowering medication. Participants who reported taking statins had better lipid profiles than those participants who reported not taking statins but had significantly lower intakes of vitamin B12, vitamin K, calcium, and potassium. There were no differences between groups on reported physical activity. However, the mean intakes for both groups did not meet the Dietary Reference Intakes for multiple nutrients. Conclusion: Older adults need additional education on the importance of lifestyle changes in reducing CHD risk, whether taking lipid-lowering medications or not

    [ABRIDGED] We report the results of a 944-epoch survey for transient sources with archival data from the Very Large Array spanning 22 years with a typical epoch separation of 7 days. Observations were obtained at 5 or 8.4 GHz for a single field of view with a full-width at half-maximum of 8.6' and 5.1', respectively, and achieved a typical point-source detection threshold at the beam center of ~300 microJy per epoch. Ten transient sources were detected with a significance threshold such that only one false positive would be expected. Of these transients, eight were detected in only a single epoch. Two transients were too faint to be detected in individual epochs but were detected in two-month averages. None of the ten transients was detected in longer-term averages or associated with persistent emission in the deep image produced from the combination of all epochs. The cumulative rate for the short timescale radio transients above 370 microJy at 5 and 8.4 GHz is 0.07 < R < 40 deg^-2 yr^-1, where the uncertainty is due to the unknown duration of the transients, 20 min < t_char < 7 days. A two-epoch survey for transients will detect 1.5 +/- 0.4 transient per square degrees above a flux density of 370 microJy. Two transients are associated with galaxies at z=0.040 and z=0.249. These may be similar to the peculiar Type Ib/c radio supernova SN 1998bw associated with GRB 980428. Six transients have no counterparts in the optical or infrared (R=27, Ks=18).     Bone loss markers in the earliest Pacific Islanders

    International audienceAbstract Kingdom of Tonga in Polynesia is one of the most obese nations where metabolic conditions, sedentary lifestyles, and poor quality diet are widespread. These factors can lead to poor musculoskeletal health. However, whether metabolic abnormalities such as osteoporosis occurred in archaeological populations of Tonga is unknown. We employed a microscopic investigation of femur samples to establish whether bone loss afflicted humans in this Pacific region approximately 3000 years ago. Histology, laser confocal microscopy, and synchrotron Fourier-transform infrared microspectroscopy were used to measure bone vascular canal densities, bone porosity, and carbonate and phosphate content of bone composition in eight samples extracted from adult Talasiu males and females dated to 2650 BP. Compared to males, samples from females had fewer vascular canals, lower carbonate and phosphate content, and higher bone porosity. Although both sexes showed evidence of trabecularised cortical bone, it was more widespread in females (35.5%) than males (15.8%). Our data suggest experiences of advanced bone resorption, possibly as a result of osteoporosis. This provides first evidence for microscopic bone loss in a sample of archaeological humans from a Pacific population widely afflicted by metabolic conditions today

    p120 GAP Modulates Ras Activation of Jun Kinases and Transformation

    Although recent evidence demonstrates that Ras causes transformation by activation of multiple downstream pathways, the specific role of non-Raf effector pathways is presently unknown. Although Ras causes activation of the Jun NH2-terminal kinases (JNKs) via a Raf-independent pathway, the contribution of JNK activation to Ras transformation and the effector that mediates JNK activation have not been established. We observed that a dominant negative mutant of SEK1/JNKK, an activator of JNKs, selectively inhibited oncogenic Ras activation of JNK and Ras transformation, but not Ras activation of the p42 mitogen-activated protein kinase. In contrast, overexpression of wild type SEK1 enhanced Ras activation of JNK and transforming activity. Thus, JNK activation promotes Ras transformation. Furthermore, a dominant negative mutant of p120 GAP (designated N-GAP), a candidate Ras effector, blocked Ras, but not Raf, transformation and blocked Ras, but not Rac, activation of JNK. Since N-GAP overexpression reduced the association of p190 Rac/Rho GAP with endogenous p120 GAP, N-GAP may form nonproductive complexes with components critical for p120 GAP function. In summary, p120 GAP may function as an effector for Ras activation of JNK and Ras transformation

    A porcine model system of BRCA1 driven breast cancer

    BRCA1 is a breast and ovarian tumor suppressor. Hereditary mutations in BRCA1 result in a predisposition to breast cancer, and BRCA1 expression is down-regulated in ~30% of sporadic cases. The function of BRCA1 remains poorly understood, but it appears to play an important role in DNA repair and the maintenance of genetic stability. Mouse models of BRCA1 deficiency have been developed in an attempt to understand the role of the gene in vivo. However, the subtle nature of BRCA1 function and the well-known discrepancies between human and murine breast cancer biology and genetics may limit the utility of mouse systems in defining the function of BRCA1 in cancer and validating the development of novel therapeutics for breast cancer. In contrast to mice, pig biological systems and cancer genetics appear to more closely resemble their human counterparts. To determine if BRCA1 inactivation in pig cells promotes their transformation and may serve as a model for the human disease, we developed an immortalized porcine breast cell line and stably inactivated BRCA1 using miRNA. The cell line developed characteristics of breast cancer stem cells and exhibited a transformed phenotype. These results validate the concept of using pigs as a model to study BRCA1 defects in breast cancer and establish the first porcine breast tumor cell line

    A habitat-use model to determine essential fish habitat for juvenile brown shrimp (Farfantepenaeus aztecus) in Galveston Bay, Texas

    A density prediction model for juvenile brown shrimp (Farfantepenaeus aztecus) was developed by using three bottom types, five salinity zones, and four seasons to quantify patterns of habitat use in Galveston Bay, Texas. Sixteen years of quantitative density data were used. Bottom types were vegetated marsh edge, submerged aquatic vegetation, and shallow nonvegetated bottom. Multiple regression was used to develop density estimates, and the resultant formula was then coupled with a geographical information system (GIS) to provide a spatial mosaic (map) of predicted habitat use. These results indicate that this model may have a broader geographic application and is a plausible approach in refining current EFH designations for all Gulf of Mexico estuaries with similar geomorphological and hydrological characteristics
