    Synergetic effect of aged garlic extract and methotrexate on rheumatoid arthritis induced by collagen in male albino rats

    33-38Aged garlic extract (AGE) exhibit anti-inflammatory effect in many diseases, and methotrexate (MTX) as rheumatoid arthritis (RA) treatment drug shows adverse hepatotoxicity effect. Therefore, in this study, we evaluated the antioxidant and anti-inflammatory effects of AGE treatment alone or with MTX in collagen-induced arthritis (CIA) rats to diminish the hepatotoxicity. The study used eight groups of rats as one control non treated group and seven treated groups with CIA, AGE (200 mg/kg/PO), MTX (1.5 mg/kg/2 days/subcutaneous), CIA-AGE, CIA-MTX, AGE-MTX and CIA-MTX-AGE. All treatments started from day 21 after the symptoms of arthritis appeared to day 50. The CIA-AGE and CIA-MTX-AGE groups showed significantly decreased serum liver function markers ASAT, ALAT and ALP enzymes activities. In line with the significantly increased antioxidants, total glutathione and SOD and CAT enzymes activities and decreased MDA levels as compared to CIA and CIA-MTX treated groups' values. In addition, the CIA-AGE and CIA-MTX-AGE groups recorded significant decrease in the measured cytokines (CRP and TNF) and interleukins (IL-17, IL-6, and IL-1) values as compared to the corresponding values in CIA and CIA-MTX groups. Results suggested the safety of AGE for achieving a better control in treatment of RA with the conventional drug MTX to diminish its hepatotoxicity

    Rutin ameliorates carbon tetrachloride (CCl4)-induced hepatorenal toxicity and hypogonadism in male rats

    Rutin, a food derived-polyphenolic bioflavonoid, has been acknowledged for several health benefits. This study aims to explore the ameliorative effects of rutin against carbon tetrachloride (CCl4) toxicity in male rats. Adult male rats were given either CCl4 (30% in olive oil, 3 ml/kg b.w. intraperitoneally) alone or in combination with rutin (70 mg/kg intragastrically) twice a week for 4 weeks. Our data showed that rutin mitigated CCl4 hepatorenal damage, as indicated by diagnostic markers (i.e., transaminases, alkaline phosphatase, total bilirubin, total protein, albumin, urea, uric acid and creatinine), and histopathological findings. In addition, CCl4 induced profound elevation of free radical generation and oxidative stress, as evidenced by increasing lipid peroxidation and reducing catalase, superoxide dismutase and glutathione peroxidase activities in liver, kidney and testicular tissues; these effects were suppressed by coexposure with rutin. Moreover, the increase in the levels of serum triglycerides, cholesterol, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol induced by CCl4 was effectively counteracted by rutin. The decrease in the level of high-density lipoprotein cholesterol in the CCl4 group was also counteracted by rutin treatment. Interestingly, the decreased levels of hormonal mediators associated with sperm production, including serum testosterone, luteinizing hormone and follicle-stimulating hormone, and the impaired sperm quality induced by CCl4 were reversed by rutin. Data from the current study clearly demonstrated that rutin supplementation could at least partly overcome CCl4-induced hepatotoxicity, nephrotoxicity and reproductive toxicity by antioxidant and antidyslipidemic effects

    Effect of Closed Versus Open Suction System on Cardiopulmonary Parameters of Ventilated Neonates

    Abstract: Removal of airway secretion is required in many neonates in the intensive care setting, and the process is most critical with respiratory problems. Clearance of secretions is essential in the mechanically ventilated neonates, because these neonates breathe slowly through an artificial airway. So, accumulation of secretions can lead to airway occlusion, serious physiological abnormalities and even death. Therefore, suctioning is essential for removing secretions and maintaining airway patency. This study aimed to determine the effect of closed versus open suction system on the cardiopulmonary parameters of ventilated neonates. The study was conducted at the Neonatal Intensive Care Unit at El-Shatby Maternity University Hospital in Alexandria. A Convenient sample of 60 neonates was randomly assigned into two groups. Thirty neonates (group A) were suctioned by closed suction system, and the other 30 neonates (group B) were suctioned by open suction system. The results revealed that that the closed suction system was more effective in maintaining the oxygen saturation, capillary refill and has less negative impact on the occurrence of cardiac arrhythmia as cardiopulmonary parameters. Other physiological parameters were also better maintained with closed than open suction system
