    Protein Complexes are Central in the Yeast Genetic Landscape

    If perturbing two genes together has a stronger or weaker effect than expected, they are said to genetically interact. Genetic interactions are important because they help map gene function, and functionally related genes have similar genetic interaction patterns. Mapping quantitative (positive and negative) genetic interactions on a global scale has recently become possible. This data clearly shows groups of genes connected by predominantly positive or negative interactions, termed monochromatic groups. These groups often correspond to functional modules, like biological processes or complexes, or connections between modules. However it is not yet known how these patterns globally relate to known functional modules. Here we systematically study the monochromatic nature of known biological processes using the largest quantitative genetic interaction data set available, which includes fitness measurements for ∼5.4 million gene pairs in the yeast Saccharomyces cerevisiae. We find that only 10% of biological processes, as defined by Gene Ontology annotations, and less than 1% of inter-process connections are monochromatic. Further, we show that protein complexes are responsible for a surprisingly large fraction of these patterns. This suggests that complexes play a central role in shaping the monochromatic landscape of biological processes. Altogether this work shows that both positive and negative monochromatic patterns are found in known biological processes and in their connections and that protein complexes play an important role in these patterns. The monochromatic processes, complexes and connections we find chart a hierarchical and modular map of sensitive and redundant biological systems in the yeast cell that will be useful for gene function prediction and comparison across phenotypes and organisms. Furthermore the analysis methods we develop are applicable to other species for which genetic interactions will progressively become more available

    Bringing order to protein disorder through comparative genomics and genetic interactions

    Abstract Background Intrinsically disordered regions are widespread, especially in proteomes of higher eukaryotes. Recently, protein disorder has been associated with a wide variety of cellular processes and has been implicated in several human diseases. Despite its apparent functional importance, the sheer range of different roles played by protein disorder often makes its exact contribution difficult to interpret. Results We attempt to better understand the different roles of disorder using a novel analysis that leverages both comparative genomics and genetic interactions. Strikingly, we find that disorder can be partitioned into three biologically distinct phenomena: regions where disorder is conserved but with quickly evolving amino acid sequences (flexible disorder); regions of conserved disorder with also highly conserved amino acid sequences (constrained disorder); and, lastly, non-conserved disorder. Flexible disorder bears many of the characteristics commonly attributed to disorder and is associated with signaling pathways and multi-functionality. Conversely, constrained disorder has markedly different functional attributes and is involved in RNA binding and protein chaperones. Finally, non-conserved disorder lacks clear functional hallmarks based on our analysis. Conclusions Our new perspective on protein disorder clarifies a variety of previous results by putting them into a systematic framework. Moreover, the clear and distinct functional association of flexible and constrained disorder will allow for new approaches and more specific algorithms for disorder detection in a functional context. Finally, in flexible disordered regions, we demonstrate clear evolutionary selection of protein disorder with little selection on primary structure, which has important implications for sequence-based studies of protein structure and evolution

    In vitro toxicity and photodynamic properties of porphyrinoids bearing imidazolium salts and N-heterocyclic carbene gold(I) complexes

    Porphyrins bearing imidazolium salts were synthesized and used as N-heterocylic carbene (NHC) precursors for the preparation of gold(I) complexes. The dark toxicity and phototoxicity of the obtained compounds were investigated in vitro on MCF-7 breast cancer cells. The obtained data showed that porphyrins equipped with imidazolium salts are non-toxic in the dark and present interesting photodynamic properties. On the contrary, corresponding NHC-gold(I) complexes are not suitable photosensitizers for photodynamic therapy (PDT) applications. Their dark toxicity strongly depends on the nature of the linker between the porphyrin core and the NHC.     Cutaneous tactile allodynia associated with microvascular dysfunction in muscle

    <p>Abstract</p> <p>Background</p> <p>Cutaneous tactile allodynia, or painful hypersensitivity to mechanical stimulation of the skin, is typically associated with neuropathic pain, although also present in chronic pain patients who do not have evidence of nerve injury. We examine whether deep tissue microvascular dysfunction, a feature common in chronic non-neuropathic pain, contributes to allodynia.</p> <p>Results</p> <p>Persistent cutaneous allodynia is produced in rats following a hind paw ischemia-reperfusion injury that induces microvascular dysfunction, including arterial vasospasms and capillary slow flow/no-reflow, in muscle. Microvascular dysfunction leads to persistent muscle ischemia, a reduction of intraepidermal nerve fibers, and allodynia correlated with muscle ischemia, but not with skin nerve loss. The affected hind paw muscle shows lipid peroxidation, an upregulation of nuclear factor kappa B, and enhanced pro-inflammatory cytokines, while allodynia is relieved by agents that inhibit these alterations. Allodynia is increased, along with hind paw muscle lactate, when these rats exercise, and is reduced by an acid sensing ion channel antagonist.</p> <p>Conclusion</p> <p>Our results demonstrate how microvascular dysfunction and ischemia in muscle can play a critical role in the development of cutaneous allodynia, and encourage the study of how these mechanisms contribute to chronic pain. We anticipate that focus on the pain mechanisms associated with microvascular dysfunction in muscle will provide new effective treatments for chronic pain patients with cutaneous tactile allodynia.</p

    Prevalencia de las extracciones prematuras en niños menores a 8 años.

    Se considera pérdida prematura de un diente temporal cuando ocurre antes de su exfoliación natural siendo el único tratamiento viable la extracción de los dientes afectados, a pesar de los tratamientos preventivos odontológico, las extracciones dentales prematuras siguen siendo un procedimiento frecuente en la práctica odontológica. Objetivos: Determinar la prevalencia de las extracciones prematura en niños menores a 8 años, identificando causas y efectos que produces estos procedimientos quirúrgicos. Materiales y Métodos: Se realizó un estudio cuantitativo bibliográfico - documental, observacional, de campo, de corte transversal, tipo exploratoria, descriptiva. Que acudieron al subcentro esperanza del Carmen, de agosto del 2018 a marzo del 2019; se utilizó la técnica de observación y él instrumento que se aplico es la recolección de datos que se realizó mediante una tabla de observación, los resultados se les aplicó estadística descriptiva. Resultados: se estudiaron&nbsp;43 pacientes, 26 fueron hombres, 17 mujeres, se extrajeron 67 órganos dentales debido a que algunos pacientes se les realizo 3 a 4 extracciones dentales, siendo la caries dental la enfermedad que más afecto a los pacientes con un promedio de 18 niños seguida de las enfermedades periodontales con 13 niños y las fracturas dentales provocadas por diferentes enfermedades que fueron 12 niños mientras la edad más susceptible fue el rango de 6 años con 16 niños atendidos. Conclusiones: Se determinó mediante el estudio realizado que los niños de 6 años presentan con mayor frecuencia perdidas de sus piezas dentales temporales y Se identificó las causas más comunes de la pérdida prematura de las piezas dentarias que fueron las caries dentales seguidas de las enfermedades periodontales que si no son tratadas inmediatamente con el profesional conlleva a la perdida de la pieza dental.Premature loss of a temporary tooth is considered when it occurs before its natural exfoliation, being the only viable treatment the extraction of the affected teeth, despite preventive dental treatments, premature dental extractions remain a frequent procedure in dental practice. Objectives: To determine the prevalence of premature extractions in children under 8 years of age, identifying causes and effects that these surgical procedures produce. Materials and Methods: A quantitative bibliographic - documentary, observational, field, cross-sectional, exploratory, descriptive study was conducted. That they went to the Carmen sub-center, from August 2018 to March 2019; The observation technique was used and the instrument that was applied is the data collection that was carried out using an observation table, the results were applied descriptive statistics. Results: 43 patients were studied, 26 were men, 17 women, 67 dental organs were removed because some patients had 3 to 4 dental extractions, with dental caries being the disease that most affected patients with an average of 18 Children followed by periodontal diseases with 13 children and dental fractures caused by different diseases that were 12 children while the most susceptible age was the range of 6 years with 16 children attended. Conclusions: It was determined through the study that 6-year-old children present more frequently lost their temporary dental pieces and the most common causes of premature loss of dental pieces that were dental caries followed by periodontal diseases that were identified were identified. If they are not treated immediately with the professional, this leads to the loss of the dental piece

    Influence of the synthetic method on the properties of two-photon-sensitive mesoporous organosilica nanoparticles

    International audienceHerein we report the modulation of the properties of mesoporous silica nanoparticles (NPs) via various synthetic approaches. Three types of elaborations were compared, one in aqueous media at 25 °C, and the other two at 80 °C in water or in a water–ethanol mixture. For all these methods, an alkoxysilylated two-photon photosensitizer (2PS) was co-condensed with tetraethylorthosilicate (TEOS) in the presence of cetyltrimethylammonium bromide (CTAB), leading to five two-photon-sensitive mesoporous silica (M2PS) NPs. The M2PS NP porous structure could be tuned from radial to worm-like and MCM-41 types of organization. Besides, the 2PS precursor spatial dispersion was found to be highly dependent on both the 2PS initial concentration and the elaboration process. As a result, two-photon properties were modulated by the choice of the synthesis, the best results being found in aqueous media at 25 or 80 °C. Finally, the M2PS NPs were used for in vitro two-photon imaging of cancer cells

    Flt3(+) macrophage precursors commit sequentially to osteoclasts, dendritic cells and microglia

    BACKGROUND: Macrophages, osteoclasts, dendritic cells, and microglia are highly specialized cells that belong to the mononuclear phagocyte system. Functional and phenotypic heterogeneity within the mononuclear phagocyte system may reveal differentiation plasticity of a common progenitor, but developmental pathways leading to such diversity are still unclear. RESULTS: Mouse bone marrow cells were expanded in vitro in the presence of Flt3-ligand (FL), yielding high numbers of non-adherent cells exhibiting immature monocyte characteristics. Cells expanded for 6 days, 8 days, or 11 days (day 6-FL, day 8-FL, and day 11-FL cells, respectively) exhibited constitutive potential towards macrophage differentiation. In contrast, they showed time-dependent potential towards osteoclast, dendritic, and microglia differentiation that was detected in day 6-, day 8-, and day 11-FL cells, in response to M-CSF and receptor activator of NFκB ligand (RANKL), granulocyte-macrophage colony stimulating-factor (GM-CSF) and tumor necrosis factor-α (TNFα), and glial cell-conditioned medium (GCCM), respectively. Analysis of cell proliferation using the vital dye CFSE revealed homogenous growth in FL-stimulated cultures of bone marrow cells, demonstrating that changes in differential potential did not result from sequential outgrowth of specific precursors. CONCLUSIONS: We propose that macrophages, osteoclasts, dendritic cells, and microglia may arise from expansion of common progenitors undergoing sequential differentiation commitment. This study also emphasizes differentiation plasticity within the mononuclear phagocyte system. Furthermore, selective massive cell production, as shown here, would greatly facilitate investigation of the clinical potential of dendritic cells and microglia

    The Quiet Revolution and the Family: Gender Composition of Tertiary Education and Early Fertility Patterns

    It is well known that highly 'female' fields of study in tertiary education are characterized by higher fertility. However, existing work does not disentangle the selection-causality nexus. We use variation in gender composition of fields of study implied by the recent expansion of tertiary education in 19 European countries and a difference-in-differences research design, to show that the share of women on study peer groups affects early fertility levels only little. Early fertility by endogamous couples, i.e., by tertiary graduates from the same field of study, declines for women and increases for men with the share of women in the group, but non-endogamous fertility almost fully compensates for these effects, consistent with higher early fertility in highly 'female' fields of study being driven by selection of family-oriented students into these fields. We also show that the EU-wide level of gender segregation across fields of study has not changed since 2000

    2008 Inter-laboratory Comparison Study of a Reference Material for Nutrients in Seawater

    Autoclaved natural seawater collected in the North Pacific Ocean was used as a reference material for nutrients in seawater (RMNS) during an inter-laboratory comparison (I/C) study conducted in 2008. This study was a follow-up to previous studies conducted in 2003 and 2006. A set of six samples was distributed to each of 58 laboratories in 15 countries around the globe, and results were returned by 54 of those laboratories (15 countries). The homogeneities of samples used in the 2008 I/C study, based on analyses for three determinants, were improved compared to those of samples used in the 2003 and 2006 I/C studies. Results of these I/C studies indicate that most of the participating laboratories have an analytical technique for nutrients that is sufficient to provide data of high comparability. The differences between reported concentrations from the same laboratories in the 2006 and 2008 I/C studies for the same batch of RMNS indicate that most of the laboratories have been maintaining internal comparability for two years. Thus, with the current high level of performance in the participating laboratories, the use of a common reference material and the adaptation of an internationally accepted nutrient scale system would increase comparability among laboratories worldwide, and the use of a certified reference material would establish traceability. In the 2008 I/C study we observed a problem of non-linearity of the instruments of the participating laboratories similar to that observed among the laboratories in the 2006 I/C study. This problem of non-linearity should be investigated and discussed to improve comparability for the full range of nutrient concentrations. For silicate comparability in particular, we see relatively larger consensus standard deviations than those for nitrate and phosphate
