    We study problems that arise in the context of covering certain geometric objects (so-called seeds, e.g., points or disks) by a set of other geometric objects (a so-called cover, e.g., a set of disks or homothetic triangles). We insist that the interiors of the seeds and the cover elements are pair wise disjoint, but they can touch. We call the contact graph of a cover a cover contact graph (CCG). We are interested in two types of tasks: (a) deciding whether a given seed set has a connected CCG, and (b) deciding whether a given graph has a realization as a CCG on a given seed set. Concerning task (a) we give efficient algorithms for the case that seeds are points and covers are disks or triangles. We show that the problem becomes NP-hard if seeds and covers are disks. Concerning task (b) we show that it is even NP-hard for point seeds and disk covers (given a fixed correspondence between vertices and seeds)

    Use of archival versus newly collected tumor samples for assessing PD-L1 expression and overall survival : an updated analysis of KEYNOTE-010 trial

    Background: In KEYNOTE-010, pembrolizumab versus docetaxel improved overall survival (OS) in patients with programmed death-1 protein (PD)-L1-positive advanced non-small-cell lung cancer (NSCLC). A prespecified exploratory analysis compared outcomes in patients based on PD-L1 expression in archival versus newly collected tumor samples using recently updated survival data. Patients and methods: PD-L1 was assessed centrally by immunohistochemistry (22C3 antibody) in archival or newly collected tumor samples. Patients received pembrolizumab 2 or 10 mg/kg Q3W or docetaxel 75 mg/m2 Q3W for 24 months or until progression/intolerable toxicity/other reason. Response was assessed by RECIST v1.1 every 9 weeks, survival every 2 months. Primary end points were OS and progression-free survival (PFS) in tumor proportion score (TPS) 50% and 1%; pembrolizumab doses were pooled in this analysis. Results: At date cut-off of 24 March 2017, median follow-up was 31 months (range 23-41) representing 18 additional months of follow-up from the primary analysis. Pembrolizumab versus docetaxel continued to improve OS in patients with previously treated, PD-L1-expressing advanced NSCLC; hazard ratio (HR) was 0.66 [95% confidence interval (CI): 0.57, 0.77]. Of 1033 patients analyzed, 455(44%) were enrolled based on archival samples and 578 (56%) on newly collected tumor samples. Approximately 40% of archival samples and 45% of newly collected tumor samples were PD-L1 TPS 50%. For TPS 50%, the OS HRs were 0.64 (95% CI: 0.45, 0.91) and 0.40 (95% CI: 0.28, 0.56) for archival and newly collected samples, respectively. In patients with TPS 1%, OS HRs were 0.74 (95% CI: 0.59, 0.93) and 0.59 (95% CI: 0.48, 0.73) for archival and newly collected samples, respectively. In TPS 50%, PFS HRs were similar across archival [0.63 (95% CI: 0.45, 0.89)] and newly collected samples [0.53 (95% CI: 0.38, 0.72)]. In patients with TPS 1%, PFS HRs were similar across archival [0.82 (95% CI: 0.66, 1.02)] and newly collected samples [0.83 (95% CI: 0.68, 1.02)]. Conclusion: Pembrolizumab continued to improve OS over docetaxel in intention to treat population and in subsets of patients with newly collected and archival samples

    Aplicación del método de la madurez para la estimación del plazo de descimbrado de forjados construidos con sistemas industrializados

    El presente artículo propone una metodología para la estimación del plazo de descimbrado de forjados construidos con sistemas industrializados. Para esto, se realiza un estudio de madurez de hormigones usados en este tipo de construcción. Con el fin de construir la curva de madurez en laboratorio se implementa la metodología descrita en la ASTM C1074, calculando la energía de activación por los tres métodos propuestos y midiendo el historial térmico de las mezclas a una temperatura controlada. Por último, se realiza la instrumentación en obra de una placa donde se estima la edad de descimbrado. Se ha observado que el tiempo de fraguado final no es un parámetro recomendable para descimbrar las placas, como lo recomiendan las productoras de hormigón en Colombia.     Broadcast spawning coral <i>Mussismilia hispida</i> can vertically transfer its associated bacterial core

    The hologenome theory of evolution (HTE), which is under fierce debate, presupposes that parts of the microbiome are transmitted from one generation to the next [vertical transmission (VT)], which may also influence the evolution of the holobiont. Even though bacteria have previously been described in early life stages of corals, these early life stages (larvae) could have been inoculated in the water and not inside the parental colony (through gametes) carrying the parental microbiome. How Symbiodinium is transmitted to offspring is also not clear, as only one study has described this mechanism in spawners. All other studies refer to incubators. To explore the VT hypothesis and the key components being transferred, colonies of the broadcast spawner species Mussismilia hispida were kept in nurseries until spawning. Gamete bundles, larvae and adult corals were analyzed to identify their associated microbiota with respect to composition and location. Symbiodinium and bacteria were detected by sequencing in gametes and coral planula larvae. However, no cells were detected using microscopy at the gamete stage, which could be related to the absence of those cells inside the oocytes/dispersed in the mucus or to a low resolution of our approach. A preliminary survey of Symbiodinium diversity indicated that parental colonies harbored Symbiodinium clades B, C and G, whereas only clade B was found in oocytes and planula larvae [5 days after fertilization (a.f.)]. The core bacterial populations found in the bundles, planula larvae and parental colonies were identified as members of the genera Burkholderia, Pseudomonas, Acinetobacter, Ralstonia, Inquilinus and Bacillus, suggesting that these populations could be vertically transferred through the mucus. The collective data suggest that spawner corals, such as M. hispida, can transmit Symbiodinium cells and the bacterial core to their offspring by a coral gamete (and that this gamete, with its bacterial load, is released into the water), supporting the HTE. However, more data are required to indicate the stability of the transmitted populations to indicate whether the holobiont can be considered a unit of natural selection or a symbiotic assemblage of independently evolving organisms

    Origin and fate of a bloom of Skeletonema costatum during a winter upwelling/downwelling sequence in the Ría de Vigo (NW Spain)

    Original research paperThe onset, development and decay of a winter bloom of the marine diatom Skeletonema costatum was monitored during a 10 d period in the coastal upwelling system of the Rı´a de Vigo (NW Spain). The succession of upwelling, relaxation and downwelling-favorable coastal winds with a frequency of 10 –20 d is a common feature of the NW Iberian shelf. The onset of the bloom occurred during an upwelling-favorable 1⁄2 wk period under winter thermal inversion conditions. The subsequent 1⁄2 wk coastal wind relaxation period allowed development of the bloom (gross primary production reached 8gCm–2 d–1) utilizing nutrients upwelled during the previous period.
