    Human Cancer Cell Radiation Response Investigated through Topological Analysis of 2D Cell Networks

    Clonogenic assays are routinely used to evaluate the response of cancer cells to external radiation fields, assess their radioresistance and radiosensitivity, estimate the performance of radiotherapy. However, classic clonogenic tests focus on the number of colonies forming on a substrate upon exposure to ionizing radiation, and disregard other important characteristics of cells such their ability to generate structures with a certain shape. The radioresistance and radiosensitivity of cancer cells may depend less on the number of cells in a colony and more on the way cells interact to form complex networks. In this study, we have examined whether the topology of 2D cancer-cell graphs is influenced by ionizing radiation. We subjected different cancer cell lines, i.e. H4 epithelial neuroglioma cells, H460 lung cancer cells, PC3 bone metastasis of grade IV of prostate cancer and T24 urinary bladder cancer cells, cultured on planar surfaces, to increasing photon radiation levels up to 6 Gy. Fluorescence images of samples were then processed to determine the topological parameters of the cell-graphs developing over time. We found that the larger the dose, the less uniform the distribution of cells on the substrate—evidenced by high values of small-world coefficient (cc), high values of clustering coefficient (cc), and small values of characteristic path length (cpl). For all considered cell lines, >1 for doses higher or equal to 4 Gy, while the sensitivity to the dose varied for different cell lines: T24 cells seem more distinctly affected by the radiation, followed by the H4, H460 and PC3 cells. Results of the work reinforce the view that the characteristics of cancer cells and their response to radiotherapy can be determined by examining their collective behavior—encoded in a few topological parameters—as an alternative to classical clonogenic assays

However, the successful application of AI is dependent on the availability of high-quality data, the addressing of ethical concerns, and the recognition of the limitations of AI-based approaches. In this article, the benefits, challenges and drawbacks of AI in this field are reviewed, and possible strategies and approaches for overcoming the present obstacles are proposed. The use of data augmentation, explainable AI, and the integration of AI with traditional experimental methods, as well as the potential advantages of AI in pharmaceutical research are also discussed. Overall, this review highlights the potential of AI in drug discovery and provides insights into the challenges and opportunities for realizing its potential in this field.     Selection of New Probiotics for Endometrial Health

    Microbiota is a crucial player in gynecologic health, in which bacteria can shift to a dysbiotic state triggering a pathogenic process. Based on an ecological understanding of the problem, the aim of this study is to select a potential probiotic strain to improve female reproductive tract based on its capacity to initially lower pH and to promote the reduction of pathogenic bacteria. Based on this rationale, strain Lactobacillus rhamnosus BPL005 was initially selected for its capacity to reduce in vitro pH levels and produce organic acids. Subsequently, strain L. rhamnosus BPL005 (CECT 8800) was demonstrated to have a protective role on endometrial infections in an in vitro model of bacterial colonization of primary endometrial epithelial cells with Atopobium vaginae, Gardnerella vaginalis, Propionibacterium acnes, and Streptococcus agalactiae. In this model, BPL005 when co-cultured with those pathogens was shown to lower pH and to produce organic acids, being lactic acid the most relevant. The co-cultivation of strain L. rhamnosus BPL005 with tested reference pathogens produced a significant reduction in P. acnes and St. agalactiae levels and a non-significant reduction in A. vaginae and G. vaginalis. The colonization of L. rhamnosus BPL005 in the culture decreased IL-6, IL-8, and MCP-1, heightened in the presence of pathogens, and increased IL-1RA and IL-1 beta. Finally, safety was evaluated showing no signs of cytotoxicity, irritation in vaginal tests, or allergic contact dermatitis potential through the Local Lymph Node Assay. Overall, these results show the potential of L. rhamnosus BPL005 strain as a probiotic in gynecological health

    Maximizing resource recovery from urban wastewater through an innovative facility layout

    [EN] This research work proposes an innovative layout for urban wastewater treatment based on anaerobic technology, microalgal cultivation and membrane technology. The proposed Water Resource Recovery Facility (WRRF) system can treat urban wastewater efficiently, complying with legal discharge limits and allowing for resource recovery, i.e. energy, nutrients and reclaimed water. In addition, the proposed layout produces less solid wastes than a conventional wastewater treatment plant (WWTP) and it is possible to recover energy as biogas, not only from the original wastewater sources but also from the biomass generated in the WRRF system

    ImageCLEF 2020: Multimedia Retrieval in Lifelogging, Medical, Nature, and Security Applications

    This paper presents an overview of the 2020 ImageCLEF lab that will be organized as part of the Conference and Labs of the Evaluation Forum - CLEF Labs 2020 in Thessaloniki, Greece. ImageCLEF is an ongoing evaluation initiative (run since 2003) that promotes the evaluation of technologies for annotation, indexing and retrieval of visual data with the aim of providing information access to large collections of images in various usage scenarios and domains. In 2020, the 18th edition of ImageCLEF will organize four main tasks: (i) a Lifelog task (videos, images and other sources) about daily activity understanding, retrieval and summarization, (ii) a Medical task that groups three previous tasks (caption analysis, tuberculosis prediction, and medical visual question answering) with new data and adapted tasks, (iii) a Coral task about segmenting and labeling collections of coral images for 3D modeling, and a new (iv) Web user interface task addressing the problems of detecting and recognizing hand drawn website UIs (User Interfaces) for generating automatic code. The strong participation, with over 235 research groups registering and 63 submitting over 359 runs for the tasks in 2019 shows an important interest in this benchmarking campaign. We expect the new tasks to attract at least as many researchers for 2020

    ImageCLEF 2022: Multimedia Retrieval in Medical, Nature, Fusion, and Internet Applications

    ImageCLEF is part of the Conference and Labs of the Evaluation Forum (CLEF) since 2003. CLEF 2022 will take place in Bologna, Italy. ImageCLEF is an ongoing evaluation initiative which promotes the evaluation of technologies for annotation, indexing, and retrieval of visual data with the aim of providing information access to large collections of images in various usage scenarios and domains. In its 20th edition, ImageCLEF will have four main tasks: (i) a Medical task addressing concept annotation, caption prediction, and tuberculosis detection; (ii) a Coral task addressing the annotation and localisation of substrates in coral reef images; (iii) an Aware task addressing the prediction of real-life consequences of online photo sharing; and (iv) a new Fusion task addressing late fusion techniques based on the expertise of the pool of classifiers. In 2021, over 100 research groups registered at ImageCLEF with 42 groups submitting more than 250 runs. These numbers show that, despite the COVID-19 pandemic, there is strong interest in the evaluation campaign

    This paper presents the ideas for the 2021 ImageCLEF lab that will be organized as part of the Conference and Labs of the Evaluation Forum — CLEF Labs 2021 in Bucharest, Romania. ImageCLEF is an ongoing evaluation initiative (active since 2003) that promotes the evaluation of technologies for annotation, indexing and retrieval of visual data with the aim of providing information access to large collections of images in various usage scenarios and domains. In 2021, the 19th edition of ImageCLEF will organize four main tasks: (i) a Medical task addressing visual question answering, a concept annotation and a tuberculosis classification task, (ii) a Coral task addressing the annotation and localisation of substrates in coral reef images, (iii) a DrawnUI task addressing the creation of websites from either a drawing or a screenshot by detecting the different elements present on the design and a new (iv) Aware task addressing the prediction of real-life consequences of online photo sharing. The strong participation in 2020, despite the COVID pandemic, with over 115 research groups registering and 40 submitting over 295 runs for the tasks shows an important interest in this benchmarking campaign. We expect the new tasks to attract at least as many researchers for 2021
