    Increasing oil accumulation in olive mesocarp: Cloning and characterization of phospholipid: diacylglycerol acyltransferase (PDAT) genes from olive fruit

    Motivation: To identify, characterize and study the regulation of genes and enzymes involved in the biosynthesis of triacylglycerols, in order to increase oil accumulation in olive mesocarp. In particular, to investigate the contribution of phospholipid:diacylglycerol acyltransferase (PDAT) genes (Dahlqvist et al., 2000).   Methods: Isolation of full-length cDNA clones by PCR, sequencing of amplified fragments, and sequence analysis using bioinformatic tools. Gene expression analysis using quantitative real-time PCR (qRT-PCR). Functional expression in a mutant of Saccharomyces cerevisiae (Sandager et al., 2002), using a yeast vector containing an inducible GAL1 promoter. Results: A cDNA sequence, designated OepPDAT2, encoding phospholipid:diacylglycerol acyltransferase (PDAT) has been isolated from olive. Gene expression analysis has been performed in Picual and Arbequina cultivars using different olive tissues and in olive mesocarp under different abiotic stresses such as draught, low and high temperature, darkness and wounding. Overexpression in yeast is being carried out in a quadruple mutant of S. cerevisiae that cannot synthesize triacylglycerols to confirm gene identity. Conclusions: Sequence analysis suggests that the OepPDAT2 gene encodes a PDAT enzyme. Gene expression analysis in olive seed and mesocarp tissues indicates that OePDAT2 participates in triacylglycerol synthesis during development and ripening of olive fruit, and it is transcriptionally regulated by different environmental factor

    Induced martensitic transformation during tensile test in nanostructured bainitic steels

    Retained austenite in nanostructured bainite is able to undergo mechanically induced martensitic transformation. However, the link between transformation and deformation mechanisms involved makes difficult the understanding of the process. In this work, a model has been developed to assess the effect of the external stress itself on the martensite phase transformation. In addition, after a detailed initial microstructural characterization, the martensite fraction evolution during tensile deformation has been obtained by means of X-ray diffraction analyses after interrupted tensile tests in several nanostructured bainitic steels. Experimental results have been compared to the outputs of the model, as a reference.     The Bose–Hubbard model with squeezed dissipation

    The stationary properties of the Bose–Hubbard model under squeezed dissipation are investigated. The dissipative model does not possess aU (1) symmetry but conserves parity. We find that 〈a j 〉 = 0 always holds, so no symmetry breaking occurs. Without the onsite repulsion, the linear case is known to be critical. At the critical point the system freezes to an EPR state with infinite two mode entanglement. We show here that the correlations are rapidly destroyed whenever the repulsion is switched on. As we increase the latter, the system approaches a thermal state with an effective temperature defined in terms of the squeezing parameter in the dissipators. We characterize this transition by means of a Gutzwiller ansatz and the Gaussian Hartree–Fock–Bogoliubov approximation

    Changes in volatile compounds of pork loin (fresh and marinated) with different irradiation and packaging during storage

    The analysis of volatile compounds by gas chromatography- mass spectrometry after extraction by purge and trap has been used to investigate the volatile compounds of fresh and marinated pork loin after E-beam treatmet as a function of packaging type (air, vacuum and modified atmosphere), radiation dose (1 and 2 kGy) and storage temperature (4 and 8 °C). Major differences were found between fresh and marinated samples but, in general, only minor differences were found in the volatile compounds of both types of loin due to storage temperature, packaging method and doses of irradiation. It is concluded that the application of E-beam is a very useful way to extend the shelflife of fresh and marinated pork loin with no changes in the odor of the products.<br><br>Se ha utilizado la cromatografía de gases/espectrometría de masas, la extracción mediante purga y trampa para estudiar los compuestos volátiles de lomo de cerdo fresco y adobado, tratados con electrones acelerados (1 y 2 kGy) y almacenado en refrigeración (4 y 8 °C) bajo diferentes atmósferas (aire, vacío y atmósfera modificada). Se observaron diferencias importantes entre las muestras de lomo fresco y adobado pero, en general, solo pequeñas diferencias fueron observadas en algunos compuestos volátiles de ambos tipos de lomo debidas al efecto de la temperatura, tiempo de almacenamiento, tipo de atmósfera o dosis de radiación. Se ha concluido que la aplicación de electrones acelerados es una tecnología muy eficaz para ampliar la vida útil del lomo de cerdo fresco y adobado sin que se detecten cambios en el olor de los productos

    [EN] Today there is an urgent need to find new ways to satisfy the current and growing food demand and to maintain crop protection and food safety. One of the most promising changes is the replacement of chemical fertilizers with biofertilizers, which include plant root-associated beneficial bacteria. This work describes and shows the use of B. halotolerans SCCPVE07 and R. laguerreae PEPV40 strains as efficient biofertilizers for escarole crops, horticultural species that are widely cultivated. An in silico genome study was performed where coding genes related to plant growth promoting (PGP) mechanisms or different enzymes implicated in the metabolism of phenolic compounds were identified. An efficient bacterial root colonization process was also analyzed through fluorescence microscopy. SCCPVE07 and PEPV40 promote plant development under normal conditions and saline stress. Moreover, inoculated escarole plants showed not only an increase in potassium, iron and magnesium content but also a significant improvement in protocatechuic acid, caffeic acid or kaempferol 3-O-glucuronide plant content. Our results show for the first time the beneficial effects in plant development and the food quality of escarole crops and highlight a potential and hopeful change in the current agricultural system even under saline stress, one of the major non-biological stresses.[ES] Hoy en día existe una necesidad urgente de encontrar nuevas formas de satisfacer la actual y creciente demanda de alimentos y mantener la protección de los cultivos y la seguridad alimentaria. Uno de los cambios más prometedores es la de los fertilizantes químicos por biofertilizantes, que incluyen bacterias beneficiosas asociadas a las raíces de las plantas. Este trabajo describe y muestra el uso de las cepas B. halotolerans SCCPVE07 y R. laguerreae PEPV40 como biofertilizantes eficientes para los cultivos de escarola, especies hortícolas ampliamente cultivadas. Se realizó un estudio genómico in silico en el que se codificaron genes relacionados con los mecanismos de promoción del crecimiento vegetal (PGP) o con diferentes enzimas implicadas en el metabolismo de compuestos fenólicos. También se analizó el proceso de colonización bacteriana de las raíces mediante microscopía de fluorescencia. SCCPVE07 y PEPV40 promueven el desarrollo de la planta en condiciones normales y estrés salino. Además, las plantas de escarola inoculadas mostraron no sólo un aumento en el contenido de potasio, hierro y magnesio, sino también una mejora significativa en el contenido de ácido protocatechuico, ácido cafeico o kaempferol 3-O-glucurónido. Nuestros resultados muestran por primera vez los efectos beneficiosos en desarrollo de la planta y la calidad alimentaria de los cultivos de escarola y ponen de manifiesto un cambio potencial y esperanzador en el sistema agrícola actual, incluso bajo estrés salino, uno de los principales estreses no biológicos.     Late-onset thymidine kinase 2 deficiency: a review of 18 cases

    BACKGROUND: TK2 gene encodes for mitochondrial thymidine kinase, which phosphorylates the pyrimidine nucleosides thymidine and deoxycytidine. Recessive mutations in the TK2 gene are responsible for the 'myopathic form' of the mitochondrial depletion/multiple deletions syndrome, with a wide spectrum of severity. METHODS: We describe 18 patients with mitochondrial myopathy due to mutations in the TK2 gene with absence of clinical symptoms until the age of 12. RESULTS: The mean age of onset was 31 years. The first symptom was muscle limb weakness in 10/18, eyelid ptosis in 6/18, and respiratory insufficiency in 2/18. All patients developed variable muscle weakness during the evolution of the disease. Half of patients presented difficulty in swallowing. All patients showed evidence of respiratory muscle weakness, with need for non-invasive Mechanical Ventilation in 12/18. Four patients had deceased, all of them due to respiratory insufficiency. We identified common radiological features in muscle magnetic resonance, where the most severely affected muscles were the gluteus maximus, semitendinosus and sartorius. On muscle biopsies typical signs of mitochondrial dysfunction were associated with dystrophic changes. All mutations identified were previously reported, being the most frequent the in-frame deletion p.Lys202del. All cases showed multiple mtDNA deletions but mtDNA depletion was present only in two patients. CONCLUSIONS: The late-onset is the less frequent form of presentation of the TK2 deficiency and its natural history is not well known.
