Conformationally-Locked C-Glycosides: Tuning Aglycone Interactions for Optimal Cheperone Behaviour in Gaucher Fibroblasts
A series of conformationally locked C-glycosides based on the 3-aminopyrano[3,2-b]pyrrol-2(1H)-one (APP) scaffold has been synthesized. The key step involved a totally stereocontrolled C-Michael addition of a serine-equivalent C-nucleophile to tri-O-benzyl-2-nitro-D-galactal, previously published by the authors. Stereoselective transformations of the Michael adduct allowed us the synthesis of compounds with mono- or diantennated aglycone moieties and different topologies. In vitro screening showed highly selective inhibition of bovine liver β-glucosidase/β-galactosidase and specific inhibition of human β-glucocerebrosidase among lysosomal glycosidases for compounds bearing palmitoyl chains in the aglycone, with a marked dependence of the inhibition potency upon their number and location. Molecular dynamics simulations highlighted the paramount importance of an optimal orientation of the hydrophobic substituent to warrant efficient non-glycone interactions, which are critical for the binding affinity. The results provide a rationale for the strong decrease of the inhibition potency of APP compounds on going from neutral to acidic pH. The best candidate was found to behave as pharmacological chaperone in Gaucher fibroblasts with homozygous N370S and F213I mutations, with enzyme activity enhancements similar to those encountered for the reference compound AmbroxolMinisterio de Economía y Competitividad CTQ2012-36365, SAF2013-44021-RJunta de Andalucía FQM-1467European Union Seventh Framework Programme FP7-People-2012-CI
Decoherence due to an excited-state quantum phase transition in a two-level boson model
10 págs.; 8 figs.; 2 tabs.; PACS number s : 03.65.Yz, 05.70.Fh, 64.70.TgThe decoherence induced on a single qubit by its interaction with the environment is studied. The environment is modeled as a scalar two-level boson system that can go through either first-order or continuous-excited-state quantum phase transitions, depending on the values of the control parameters. A mean-field method based on the Tamm-Damkoff approximation is worked out in order to understand the observed behavior of the decoherence. Only the continuous-excited-state phase transition produces a noticeable effect in the decoherence of the qubit. This is maximal when the system-environment coupling brings the environment to the critical point for the continuous phase transition. In this situation, the decoherence factor (or the fidelity) goes to zero with a finite-size scaling power law. © 2009 The American Physical Society. This work has been partially supported by the Spanish

Multi-technology battery storage system for optimal demand-side management
Demand-side management (DSM) provides additional system flexibility for the electric energy environment. To enable the spread of DSM policies the use of automatic systems combined with a local battery energy storage system (BESS) is proposed. Following this statement, it has been developed and tested a 50 kVA low-cost multi-technology energy storage system, with a novel converter topology that permits the integration of multiple battery types or aging. It consists of a 50 kVA bidirectional three-level AC/DC converter for grid connection which feeds five 10 kW isolated bi-directional DC/DC battery converters, based on SiC and Si MOSFETs. The development leads in a pick-and-place system, adding interoperability, ease of connection and modularity

Evaluation of optical techniques for characterising soil organic matter quality in agricultural soils
Soil organic matter (SOM) is one of the main global carbon pools. It is a measure of soil quality as its presence increases carbon sequestration and improves physical and chemical soil properties. The determination and characterisation of humic substances gives essential information of the maturity and stresses of soils as well as of their health. However, the determination of the exact nature and molecular structure of these substances has been proven difficult. Several complex techniques exist to characterise SOM and mineralisation and humification processes. One of the more widely accepted for its accuracy is nuclear magnetic resonance (NMR) spectroscopy. Despite its efficacy, NMR needs significant economic resources, equipment, material and time. Proxy measures like the fluorescence index (FI), cold and hot-water extractable carbon (CWC and HWC) and SUVA-254 have the potential to characterise SOM and, in combination, provide qualitative and quantitative data of SOM and its processes. Spanish and British agricultural cambisols were used to measure SOM quality and determine whether similarities were found between optical techniques and 1H NMR results in these two regions with contrasting climatic conditions. High correlations (p < 0.001) were found between the specific aromatic fraction measured with 1H NMR and SUVA-254 (Rs = 0.95) and HWC (Rs = 0.90), which could be described using a linear model. A high correlation between FI and the aromatics fraction measured with 1H NMR (Rs = −0.976) was also observed. In view of our results, optical measures have a potential, in combination, to predict the aromatic fraction of SOM without the need of expensive and time consuming techniques

Characterization of Implanted Stents through Neointimal Tissue Bioimpedance Simulations
This work describes how is possible the definition
of the light hole or lumen in implanted stents affected by
restenosis processes using the BioImpedance (BI) as biomarker.
The main approach is based on the fact that neointimal tissues
implied in restenosis can be detected and measured thanks to
their respective conductivity and dielectric properties. For this
goal, it is proposed a four-electrode setup for bioimpedance
measurement. The influence of the several involved tissues in
restenosis: fat, muscle, fiber, endothelium and blood, have been
studied at several frequencies, validating the setup and
illustrating the sensitivity of each one. Finally, a real example
using a standard stent, has been analyzed for stable and
vulnerable plaques in restenosis test cases, demonstrating that
the proposed method is useful for the stent obstruction test.
