    Genetic Analysis of the Achaete-Scute System of DROSOPHILA MELANOGASTER

    Several mutations in the achaete-scute region of Drosophila have been analyzed phenotypically and cytologically. One group of them corresponds to point mutations, another to rearrangements with one breakpoint in this region. Trans heterozygotes of the different point mutations or of the different rearrangements show poor complementation or fail to complement; therefore, they could be interpreted as mutations affecting the same gene product. However, left-right inversion recombinants and duplication-deficiency combinations between rearrangements with different cytological breakpoints uncover a complex organization of the achaete-scute region. This region seems to contain several independent achaete and scute functions, as well as a lethal function, arranged as a tandem reverse repeat at both sides of a lethal locus. Since all of the mutants show the same phenotype qualitatively, though different quantitatively, we suggest that these functions are of a reiterative nature. The achaete-scute wild-type condition may well be dependent on a multimeric gene product made of several evolutionary related monomers.Peer reviewe

    Comunicación cartel con defensa oralWe aim to evaluate the impact of the individual and in-group analysis of OSCEs (filmed by other students) looking for correct and incorrect behaviours and contents in their learning of pharmacology. Summary of work and outcomes: A 5-year prospective study in which students of Pharmacology course from the Podiatry Degree analysed filmed OSCEs individually and in a group in a flipped classroom. Each group (max 5 students) analysed along 1 week a filmed OSCE, prepare a summary of correct and incorrect items related to clinical events, medicines uses, and people behaviours. The students presented their analysis results to the rest of the class. After each presentation, the other students of the class were encouraged to ask questions and after that, the students voluntarily answered a satisfaction survey. Result and Discussion: 405 students, 65.2% female, 20±5.3 years old were included. Students spend 13.4±5.4 h on making the filmed-OSCE analysis. The percentage of students satisfied with this way of studying pharmacology was 96.5%. OSCEs analysis by students increased their percentage of success in the final assessment in both OSCEs-related and OSCEs-non-related questions (+18.5% and +10.1%). Conclusion: Filmed Objective Structured Clinical Examinations analysis by undergraduate students on the Pharmacology course of Podiatry Degree improved their knowledge about medicines use and their communication skills during the assessment.Universidad de Málaga.     Functional characterization of two enhancers located downstream FOXP2

    Background: Mutations in the coding region of FOXP2 are known to cause speech and language impairment. However, it is not clear how dysregulation of the gene contributes to language deficit. Interestingly, microdeletions of the region downstream the gene have been associated with cognitive deficits. Methods: Here, we investigate changes in FOXP2 expression in the SK-N-MC neuroblastoma human cell line after deletion by CRISPR-Cas9 of two enhancers located downstream of the gene. Results: Deletion of any of these two functional enhancers downregulates FOXP2, but also upregulates the closest 3′ gene MDFIC. Because this effect is not statistically significant in a HEK 293 cell line, derived from the human kidney, both enhancers might confer a tissue specific regulation to both genes. We have also found that the deletion of any of these enhancers downregulates six well-known FOXP2 target genes in the SK-N-MC cell line. Conclusions: We expect these findings contribute to a deeper understanding of how FOXP2 and MDFIC are regulated to pace neuronal development supporting cognition, speech and language.Spanish National Research and Development Plan PI14/01884Instituto de Salud Carlos III PI14/01884FEDER PI14/0188

    Assessing Machine Learning Models for Gap Filling Daily Rainfall Series in a Semiarid Region of Spain

    The presence of missing data in hydrometeorological datasets is a common problem, usually due to sensor malfunction, deficiencies in records storage and transmission, or other recovery procedures issues. These missing values are the primary source of problems when analyzing and modeling their spatial and temporal variability. Thus, accurate gap-filling techniques for rainfall time series are necessary to have complete datasets, which is crucial in studying climate change evolution. In this work, several machine learning models have been assessed to gap-fill rainfall data, using different approaches and locations in the semiarid region of Andalusia (Southern Spain). Based on the obtained results, the use of neighbor data, located within a 50 km radius, highly outperformed the rest of the assessed approaches, with RMSE (root mean squared error) values up to 1.246 mm/day, MBE (mean bias error) values up to −0.001 mm/day, and R2 values up to 0.898. Besides, inland area results outperformed coastal area in most locations, arising the efficiency effects based on the distance to the sea (up to an improvement of 63.89% in terms of RMSE). Finally, machine learning (ML) models (especially MLP (multilayer perceptron)) notably outperformed simple linear regression estimations in the coastal sites, whereas in inland locations, the improvements were not such significant

    Filmed clinical cases improved communication skills of student of Pharmacology in Podiatry degree

    Solving clinical case by writing-report lacks of students sympathies and interest in many cases. Make a short film is a novelty that really like to the students and may improve their arguing and planning capacities and their communication skills. Aim: To evaluate the impact of presenting and resolving clinical cases through filming a short-film in the communication skills development and in the learning of medicines in Podiatry undergraduate students. Methods: A two-year study in which students were invited to voluntarily form groups (3 students maximum). Each group has to design and film a short-film (8 min maximum) showing a clinical case in which medicines’ use was needed to treat feet pathology. A camera, a mobile-phone's video editor or whatever they may use was allowed. The job of each group was supervised and helped by a teacher. The students were invited to present their work to the rest of the class. After each short-film projection the students were encouraged to ask questions if they wanted to do it. After all the projections the students voluntarily answered a satisfaction survey. Results: Students of Pharmacology of Podiatry Degree, N=101, 55.6% female, 20±1.3 years old were enrolled. 37 short-films showing a clinical case were made. The average time spent by students in making the film was 12.4±8 h. The percentage of students which were satisfied with this way of presentation of the clinical cases was 75.2%. Conclusion: Filmed clinical cases performed by student of Pharmacology of the Podiatry Degree improved their communication skills.Universidad de Málaga.     Giant pancreatic cystic lymphangioma

    Presentamos una paciente de 41 años diagnosticada de linfangioma quístico gigante en cuerpo-cola pancreático con aumento progresivo en el último año, de 20 cm de diámetro, como hallazgo incidental ecográfico. Se realiza punción-aspiración con aguja fina (PAAF), con resultado de lesión quística benigna. Ante el aumento de tamaño, se decide intervención quirúrgica. Se objetiva una tumoración quística retroperitoneal dependiente de cara posterior pancreática y se realiza resección completa laparoscópica hasta cápsula pancreática con preservación de páncreas y bazo. En la descripción anatomopatológica se confirma el diagnóstico de linfangioma quístico benigno. No se produjeron incidencias durante el postoperatorio inmediato y la paciente fue dada de alta al quinto día. Tras un año de seguimiento, la paciente permanece asintomática.A giant cystic lymphangioma in the pancreatic body-tail was diagnosed as an incidental ultrasound mass in a 41-year-old patient, with a progressive size that had increased in the last year by about 20 cm size. An ultrasound guided fine needle puncture was performed and the result was a benign cystic lesion. Given the increase in size, a surgical intervention was decided. A retroperitoneal cystic tumor dependent on the posterior pancreatic wall was identified and a full laparoscopic resection with pancreas and spleen preservation was performed. The pathological report confirmed the diagnosis of benign cystic lymphangioma. The patient was discharged on the fifth postoperative day without any remarkable complications. After one year of follow-up, the patient remains asymptomatic

    AgroML: An Open-Source Repository to Forecast Reference Evapotranspiration in Different Geo-Climatic Conditions Using Machine Learning and Transformer-Based Models

    Accurately forecasting reference evapotranspiration (ET0) values is crucial to improve crop irrigation scheduling, allowing anticipated planning decisions and optimized water resource management and agricultural production. In this work, a recent state-of-the-art architecture has been adapted and deployed for multivariate input time series forecasting (transformers) using past values of ET0 and temperature-based parameters (28 input configurations) to forecast daily ET0 up to a week (1 to 7 days). Additionally, it has been compared to standard machine learning models such as multilayer perceptron (MLP), random forest (RF), support vector machine (SVM), extreme learning machine (ELM), convolutional neural network (CNN), long short-term memory (LSTM), and two baselines (historical monthly mean value and a moving average of the previous seven days) in five locations with different geo-climatic characteristics in the Andalusian region, Southern Spain. In general, machine learning models significantly outperformed the baselines. Furthermore, the accuracy dramatically dropped when forecasting ET0 for any horizon longer than three days. SVM, ELM, and RF using configurations I, III, IV, and IX outperformed, on average, the rest of the configurations in most cases. The best NSE values ranged from 0.934 in Córdoba to 0.869 in Tabernas, using SVM. The best RMSE, on average, ranged from 0.704 mm/day for Málaga to 0.883 mm/day for Conil using RF. In terms of MBE, most models and cases performed very accurately, with a total average performance of 0.011 mm/day. We found a relationship in performance regarding the aridity index and the distance to the sea. The higher the aridity index at inland locations, the better results were obtained in forecasts. On the other hand, for coastal sites, the higher the aridity index, the higher the error. Due to the good performance and the availability as an open-source repository of these models, they can be used to accurately forecast ET0 in different geo-climatic conditions, helping to increase efficiency in tasks of great agronomic importance, especially in areas with low rainfall or where water resources are limiting for the development of crops

    Chern-Simons production during preheating in hybrid inflation models

    We study the onset of symmetry breaking after hybrid inflation in a model having the field content of the SU(2) gauge-scalar sector of the standard model, coupled to a singlet inflaton. This process is studied in (3+1)-dimensions in a fully non-perturbative way with the help of lattice techniques within the classical approximation. We focus on the role played by gauge fields and, in particular, on the generation of Chern-Simons number. Our results are shown to be insensitive to the various cut-offs introduced in our numerical approach. The spectra preserves a large hierarchy between long and short-wavelength modes during the whole period of symmetry breaking and Chern-Simons generation, confirming that the dynamics is driven by the low momentum sector of the theory. We establish that the Chern-Simons production mechanism is associated with local sphaleron-like structures. The corresponding sphaleron rates are of order 10^{-5} m^4, which, within certain scenarios of electroweak baryogenesis and a (not unnaturally large) additional source of CP violation, could explain the present baryon asymmetry of the universe.Comment: 28 pages, 15 figures, ReVTeX.     Reformulating Pro-Oxidant Microglia in Neurodegeneration

    In neurodegenerative diseases, microglia-mediated neuroinflammation and oxidative stress are central events. Recent genome-wide transcriptomic analyses of microglial cells under different disease conditions have uncovered a new subpopulation named disease-associated microglia (DAM). These studies have challenged the classical view of the microglia polarization state's proinflammatory M1 (classical activation) and immunosuppressive M2 (alternative activation). Molecular signatures of DAM and proinflammatory microglia (highly pro-oxidant) have shown clear differences, yet a partial overlapping gene profile is evident between both phenotypes. The switch activation of homeostatic microglia into reactive microglia relies on the selective activation of key surface receptors involved in the maintenance of brain homeostasis (a.k.a. pattern recognition receptors, PRRs). Two relevant PRRs are toll-like receptors (TLRs) and triggering receptors expressed on myeloid cells-2 (TREM2), whose selective activation is believed to generate either a proinflammatory or a DAM phenotype, respectively. However, the recent identification of endogenous disease-related ligands, which bind to and activate both TLRs and TREM2, anticipates the existence of rather complex microglia responses. Examples of potential endogenous dual ligands include amyloid β, galectin-3, and apolipoprotein E. These pleiotropic ligands induce a microglia polarization that is more complicated than initially expected, suggesting the possibility that different microglia subtypes may coexist.
