Cephalosporin-3’-diazeniumdiolate NO-donor prodrug PYRRO-C3D enhances azithromycin susceptibility of non-typeable Haemophilus influenzae biofilms

The Publisher's final version can be found by following the DOI link.Objectives: PYRRO-C3D is a cephalosporin-3-diazeniumdiolate nitric oxide (NO)-donor prodrug designed to selectively deliver NO to bacterial infection sites. The objective of this study was to assess the activity of PYRRO-C3D against non-typeable Haemophilus influenzae (NTHi) biofilms and examine the role of NO in reducing biofilm-associated antibiotic tolerance.
Methods: The activity of PYRRO-C3D on in vitro NTHi biofilms was assessed through CFU enumeration and confocal microscopy. NO release measurements were performed using an ISO-NO probe. NTHi biofilms grown on primary ciliated respiratory epithelia at an air-liquid interface were used to investigate the effects of PYRRO-C3D in the presence of host tissue. Label-free LC/MS proteomic analyses were performed to identify differentially expressed proteins following NO treatment.
Results: PYRRO-C3D specifically released NO in the presence of NTHi, while no evidence of spontaneous NO release was observed when the compound was exposed to primary epithelial cells. NTHi lacking β-lactamase activity failed to trigger NO release. Treatment significantly increased the susceptibility of in vitro NTHi biofilms to azithromycin, causing a log-fold reduction in viability (p<0.05) relative to azithromycin alone. The response was more pronounced for biofilms grown on primary respiratory epithelia, where a 2-log reduction was observed (p<0.01). Label-free proteomics showed that NO increased expression of sixteen proteins involved in metabolic and transcriptional/translational functions.
Conclusions: NO release from PYRRO-C3D enhances the efficacy of azithromycin against NTHi biofilms, putatively via modulation of NTHi metabolic activity. Adjunctive therapy with NO mediated through PYRRO-C3D represents a promising approach for reducing biofilm associated antibiotic tolerance
An approach to build an event set of European wind storms based on ECMWF EPS
The properties of European wind storms under present climate conditions are estimated on the basis of surface wind forecasts from the European Center of Medium-Range Weather Forecast (ECMWF) Ensemble Prediction System (EPS). While the EPS is designed to provide forecast information of the range of possible weather developments starting from the observed state of weather, we use its archive in a climatological context. It provides a large number of modifications of observed storm events, and includes storms that did not occur in reality. Thus it is possible to create a large sample of storm events, which entirely originate from a physically consistent model, whose ensemble spread represents feasible alternative storm realizations of the covered period. This paper shows that the huge amount of identifiable events in the EPS is applicable to reduce uncertainties in a wide range of fields of research focusing on winter storms. Wind storms are identified and tracked in this study over their lifetime using an algorithm, based on the local exceedance of the 98th percentile of instantaneous 10 m wind speed, calculating a storm severity measure. After removing inhomogeneities in the dataset arising from major modifications of the operational system, the distributions of storm severity, storm size and storm duration are computed. The overall principal properties of the homogenized EPS storm data set are in good agreement with storms from the ERA-Interim dataset, making it suitable for climatological investigations of these extreme events. A demonstrated benefit in the climatological context by the EPS is presented. It gives a clear evidence of a linear increase of maximum storm intensity and wind field size with storm duration. This relation is not recognizable from a sparse ERA-Interim sample for long lasting events, as the number of events in the reanalysis is not sufficient to represent these characteristics
Multi-performance optimisation framework for the selection of structural alternatives based on sustainable qualities
In recent years, the increasing demand for innovative sustainable policies in building engineering has shifted the decision rationale from traditional performance-based systems towards systems augmented by life-cycle sustainability notions. This paper investigates a novel optimisation framework, which supports the selection of buildings’ structural alternatives at concept stage by applying multiple performance, sustainable requirements. The established model explores ways to effectively compute and process expert knowledge across different stakeholders groups into a consolidated decision-making platform supported by Lean Theory. A systematic procedure based on the Quality Function Deployment is utilised to successfully translate 16 sustainability requirements into 27 corresponding engineering design requirements. The theoretical and mathematical principles of Analytic Network Process are applied on a pilot study to build general decision clusters, identify feedback links amongst the various engineering criteria and determine their inner dependences
High density LHRF experiments in Alcator C-Mod and implications for reactor scale devices
Parametric decay instabilities (PDI) appear to be an ubiquitous feature of lower hybrid current drive (LHCD) experiments at high density. In density ramp experiments in Alcator C-Mod and other machines the onset of PDI activity has been well correlated with a decrease in current drive efficiency and production of fast electron bremsstrahlung. However whether PDI is the primary cause of the 'density limit', and if so by exactly what mechanism (beyond the obvious one of pump depletion) has not been clearly established. In order to further understand the connection, the frequency spectrum of PDI activity occurring during Alcator C-Mod LHCD experiments has been explored in detail by means of a number of RF probes distributed around the periphery of the C-Mod tokamak including a probe imbedded in the inner wall. The results show that (i) the excited spectra consists mainly of a few discrete ion cyclotron (IC) quasi-modes, which have higher growth than the ion sound branch; (ii) PDI activity can begin either at the inner or outer wall, depending on magnetic configuration; (iii) the frequencies of the IC quasi-modes correspond to the magnetic field strength close to the low-field side (LFS) or high-field side separatrix; and (iv) although PDI activity may initiate near the inner separatrix, the loss in fast electron bremsstrahlung is best correlated with the appearance of IC quasi-modes characteristic of the magnetic field strength near the LFS separatrix. These data, supported by growth rate calculations, point to the importance of the LFS scrape-off layer (SOL) density in determining PDI onset and degradation in current drive efficiency. By minimizing the SOL density it is possible to extend the core density regime over which PDI can be avoided, thus potentially maximizing the effectiveness of LHCD at high density. Increased current drive efficiency at high density has been achieved in FTU and EAST through lithium coating and special fuelling methods, and in recent C-Mod experiments by operating at higher plasma current. Another approach would be to locate the launcher in the inner wall with double null operation. Casimir forces on a silicon micromechanical chip
Quantum fluctuations give rise to van der Waals and Casimir forces that
dominate the interaction between electrically neutral objects at sub-micron
separations. Under the trend of miniaturization, such quantum electrodynamical
effects are expected to play an important role in micro- and nano-mechanical
devices. Nevertheless, utilization of Casimir forces on the chip level remains
a major challenge because all experiments so far require an external object to
be manually positioned close to the mechanical element. Here, by integrating a
force-sensing micromechanical beam and an electrostatic actuator on a single
chip, we demonstrate the Casimir effect between two micromachined silicon
components on the same substrate. A high degree of parallelism between the two
near-planar interacting surfaces can be achieved because they are defined in a
single lithographic step. Apart from providing a compact platform for Casimir
force measurements, this scheme also opens the possibility of tailoring the
Casimir force using lithographically defined components of non-conventional
shapes
Thalamic inputs to dorsomedial striatum are involved in inhibitory control: evidence from the five-choice serial reaction time task in rats
Rationale
Corticostriatal circuits are widely implicated in the top-down control of attention including inhibitory control and behavioural flexibility. However, recent neurophysiological evidence also suggests a role for thalamic inputs to striatum in behaviours related to salient, reward-paired cues.
Objectives
Here, we used designer receptors exclusively activated by designer drugs (DREADDs) to investigate the role of parafascicular (Pf) thalamic inputs to the dorsomedial striatum (DMS) using the five-choice serial reaction time task (5CSRTT) in rats.
Methods
The 5CSRTT requires sustained attention in order to detect spatially and temporally distributed visual cues and provides measures of inhibitory control related to impulsivity (premature responses) and compulsivity (perseverative responses). Rats underwent bilateral Pf injections of the DREADD vector, AAV2-CaMKIIa-HA-hM4D(Gi)-IRES-mCitrine. The DREADD agonist, clozapine N-oxide (CNO; 1 μl bilateral; 3 μM) or vehicle, was injected into DMS 1 h before behavioural testing. Task parameters were manipulated to increase attention load or reduce stimulus predictability respectively.
Results
We found that inhibition of the Pf-DMS projection significantly increased perseverative responses when stimulus predictability was reduced but had no effect on premature responses or response accuracy, even under increased attentional load. Control experiments showed no effects on locomotor activity in an open field.
Conclusions
These results complement previous lesion work in which the DMS and orbitofrontal cortex were similarly implicated in perseverative responses and suggest a specific role for thalamostriatal inputs in inhibitory control
Universal Vectorial and Ultrasensitive Nanomechanical Force Field Sensor
Miniaturization of force probes into nanomechanical oscillators enables
ultrasensitive investigations of forces on dimensions smaller than their
characteristic length scale. Meanwhile it also unravels the force field
vectorial character and how its topology impacts the measurement. Here we
expose an ultrasensitive method to image 2D vectorial force fields by
optomechanically following the bidimensional Brownian motion of a singly
clamped nanowire. This novel approach relies on angular and spectral tomography
of its quasi frequency-degenerated transverse mechanical polarizations:
immersing the nanoresonator in a vectorial force field does not only shift its
eigenfrequencies but also rotate eigenmodes orientation as a nano-compass. This
universal method is employed to map a tunable electrostatic force field whose
spatial gradients can even take precedence over the intrinsic nanowire
properties. Enabling vectorial force fields imaging with demonstrated
sensitivities of attonewton variations over the nanoprobe Brownian trajectory
will have strong impact on scientific exploration at the nanoscale
