Characterizing the stocks, flows, and carbon impact of dockless sharing bikes in China
The booming dockless sharing bikes (DSBs) in China, as a new sharing economy business model, have attracted increasing public and academic attention after 2015. The impact of DSBs development on the stocks and flows of bikes and the resource and climate consequences of short-lived DSBs, however, remain poorly understood. In this study, we characterized the stocks and flows of both DSBs and regular private bikes in China from 1950 to 2020 and evaluated the carbon cost and benefit of booming DSBs. We found China's bike consumption and stock decreased slightly after a fast development from the late 1970s and then a peak in the mid-1990s, resulting in a relatively low ownership of approximately 0.3 unit per person and 70% of production being exported in recent years. Despite a temporal boost, the unsustainable development of DSBs may affect the bike industry in the long term, because of its skyrocketing market share (from less than 1% to 80%) and short lifetime. Nevertheless, DSBs development still leads to an overall climate gain in China, due to its higher stock efficiency and potentials to substitute more carbon intensive trips. We suggest an urgent need for more empirical studies on the use (e.g., substitution ratio for other transportation models) of DSBs in China and a necessity for better management of DSB development with efforts of all relevant stakeholders
Stochastic assembly in a subtropical forest chronosequence: evidence from contrasting changes of species, phylogenetic and functional dissimilarity over succession

However, it has been widely held in previous studies that deterministic processes dominate forest succession. Furthermore, inference of mechanisms for community assembly may be misleading if based on a single axis of diversity alone. In this study, we evaluated the relative roles of deterministic and stochastic processes along a disturbance gradient by integrating species, functional, and phylogenetic beta diversity in a subtropical forest chronosequence in Southeastern China. We found a general pattern of increasing species turnover, but little-to-no change in phylogenetic and functional turnover over succession at two spatial scales. Meanwhile, the phylogenetic and functional beta diversity were not significantly different from random expectation. This result suggested a dominance of stochastic assembly, contrary to the general expectation that deterministic processes dominate forest succession. On the other hand, we found significant interactions of environment and disturbance and limited evidence for significant deviations of phylogenetic or functional turnover from random expectations for different size classes. This result provided weak evidence of deterministic processes over succession. Survivin as a therapeutic target in Sonic hedgehog-driven medulloblastoma.
Medulloblastoma (MB) is a highly malignant brain tumor that occurs primarily in children. Although surgery, radiation and high-dose chemotherapy have led to increased survival, many MB patients still die from their disease, and patients who survive suffer severe long-term side effects as a consequence of treatment. Thus, more effective and less toxic therapies for MB are critically important. Development of such therapies depends in part on identification of genes that are necessary for growth and survival of tumor cells. Survivin is an inhibitor of apoptosis protein that regulates cell cycle progression and resistance to apoptosis, is frequently expressed in human MB and when expressed at high levels predicts poor clinical outcome. Therefore, we hypothesized that Survivin may have a critical role in growth and survival of MB cells and that targeting it may enhance MB therapy. Here we show that Survivin is overexpressed in tumors from patched (Ptch) mutant mice, a model of Sonic hedgehog (SHH)-driven MB. Genetic deletion of survivin in Ptch mutant tumor cells significantly inhibits proliferation and causes cell cycle arrest. Treatment with small-molecule antagonists of Survivin impairs proliferation and survival of both murine and human MB cells. Finally, Survivin antagonists impede growth of MB cells in vivo. These studies highlight the importance of Survivin in SHH-driven MB, and suggest that it may represent a novel therapeutic target in patients with this disease
Scalar-mediated forward-backward asymmetry
A large forward-backward asymmetry in production, for large
invariant mass of the system, has been recently observed by the CDF
collaboration. Among the scalar mediated mechanisms that can explain such a
large asymmetry, only the t-channel exchange of a color-singlet weak-doublet
scalar is consistent with both differential and integrated cross
section measurements. Constraints from flavor changing processes dictate a very
specific structure for the Yukawa couplings of such a new scalar. No sizable
deviation in the differential or integrated production cross section

Hybrid Equation/Agent-Based Model of Ischemia-Induced Hyperemia and Pressure Ulcer Formation Predicts Greater Propensity to Ulcerate in Subjects with Spinal Cord Injury
Pressure ulcers are costly and life-threatening complications for people with spinal cord injury (SCI). People with SCI also exhibit differential blood flow properties in non-ulcerated skin. We hypothesized that a computer simulation of the pressure ulcer formation process, informed by data regarding skin blood flow and reactive hyperemia in response to pressure, could provide insights into the pathogenesis and effective treatment of post-SCI pressure ulcers. Agent-Based Models (ABM) are useful in settings such as pressure ulcers, in which spatial realism is important. Ordinary Differential Equation-based (ODE) models are useful when modeling physiological phenomena such as reactive hyperemia. Accordingly, we constructed a hybrid model that combines ODEs related to blood flow along with an ABM of skin injury, inflammation, and ulcer formation. The relationship between pressure and the course of ulcer formation, as well as several other important characteristic patterns of pressure ulcer formation, was demonstrated in this model. The ODE portion of this model was calibrated to data related to blood flow following experimental pressure responses in non-injured human subjects or to data from people with SCI. This model predicted a higher propensity to form ulcers in response to pressure in people with SCI vs. non-injured control subjects, and thus may serve as novel diagnostic platform for post-SCI ulcer formation. © 2013 Solovyev et al
Conjugation with L, L-diphenylalanine Self-Assemblies Enhances In Vitro Antitumor Activity of Phthalocyanine Photosensitizer
We present the synthesis and characterization of new peptide conjugates obtained by hierarchical co-assembly of L,L-diphenylalanine (FF) and zinc phthalocyanine complexes (ZnPc) in water. Self-assembly capabilities under defined conditions were investigated by scanning electron microscopy, and photophysical properties were evaluated using UV-Vis and fluorescence spectroscopy. AFM observations demonstrated that these ZnPcs form different highly ordered arrays on the crystalline faces of the FF microplates and that surface roughness significantly changes with the presence of differently substituted phthalocyanine units. XRD assays showed that the overall molecular packing of the conjugates is organized according to a hexagonal symmetry, with ZnPcs hosted in the interstices of the peptide phase. In vitro photodynamic studies were conducted on human breast cancer MCF-7 cells to investigate both cellular uptake and cytotoxicity. It was shown that FF self-assemblies are not toxicity and enhance accumulation of ZnPc in MCF-7 cells, improving apoptotic cell death upon irradiation. Our findings demonstrate enhancement of ZnPc antitumor efficiency by FF conjugates and a proof-of-concept for new photosensitizer carriers based on peptide conjugates

Musashi-2 controls cell fate, lineage bias, and TGF-β signaling in HSCs
Hematopoietic stem cells (HSCs) are maintained through the regulation of symmetric and asymmetric cell division. We report that conditional ablation of the RNA-binding protein Msi2 results in a failure of HSC maintenance and engraftment caused by a loss of quiescence and increased commitment divisions. Contrary to previous studies, we found that these phenotypes were independent of Numb. Global transcriptome profiling and RNA target analysis uncovered Msi2 interactions at multiple nodes within pathways that govern RNA translation, stem cell function, and TGF-β signaling. Msi2-null HSCs are insensitive to TGF-β–mediated expansion and have decreased signaling output, resulting in a loss of myeloid-restricted HSCs and myeloid reconstitution. Thus, Msi2 is an important regulator of the HSC translatome and balances HSC homeostasis and lineage bias
Deep generative modeling for single-cell transcriptomics.
Single-cell transcriptome measurements can reveal unexplored biological diversity, but they suffer from technical noise and bias that must be modeled to account for the resulting uncertainty in downstream analyses. Here we introduce single-cell variational inference (scVI), a ready-to-use scalable framework for the probabilistic representation and analysis of gene expression in single cells ( https://github.com/YosefLab/scVI ). scVI uses stochastic optimization and deep neural networks to aggregate information across similar cells and genes and to approximate the distributions that underlie observed expression values, while accounting for batch effects and limited sensitivity. We used scVI for a range of fundamental analysis tasks including batch correction, visualization, clustering, and differential expression, and achieved high accuracy for each task
Dynamics of multipartite quantum correlations under decoherence
Quantum discord is an optimal resource for the quantification of classical
and non-classical correlations as compared to other related measures. Geometric
measure of quantum discord is another measure of quantum correlations.
Recently, the geometric quantum discord for multipartite states has been
introduced by Jianwei Xu [arxiv:quant/ph.1205.0330]. Motivated from the recent
study [Ann. Phys. 327 (2012) 851] for the bipartite systems, I have
investigated global quantum discord (QD) and geometric quantum discord (GQD)
under the influence of external environments for different multipartite states.
Werner-GHZ type three-qubit and six-qubit states are considered in inertial and
non-inertial settings. The dynamics of QD and GQD is investigated under
amplitude damping, phase damping, depolarizing and flipping channels. It is
seen that the quantum discord vanishes for p>0.75 in case of three-qubit GHZ
states and for p>0.5 for six qubit GHZ states. This implies that multipartite
states are more fragile to decoherence for higher values of N. Surprisingly, a
rapid sudden death of discord occurs in case of phase flip channel. However,
for bit flip channel, no sudden death happens for the six-qubit states. On the
other hand, depolarizing channel heavily influences the QD and GQD as compared
to the amplitude damping channel. It means that the depolarizing channel has
the most destructive influence on the discords for multipartite states. From
the perspective of accelerated observers, it is seen that effect of environment
on QD and GQD is much stronger than that of the acceleration of non-inertial
frames. The degradation of QD and GQD happens due to Unruh effect. Furthermore,
QD exhibits more robustness than GQD when the multipartite systems are exposed

Color & Weak triplet scalars, the dimuon asymmetry in decay, the top forward-backward asymmetry, and the CDF dijet excess
The new physics required to explain the anomalies recently reported by the D0
and CDF collaborations, namely the top forward-backward asymmetry (FBA), the
like-sign dimuon charge asymmetry in semileptonic b decay, and the CDF dijet
excess, has to feature an amount of flavor symmetry in order to satisfy the
severe constrains arising from flavor violation. In this paper we show that,
once baryon number conservation is imposed, color & weak triplet scalars with
hypercharge can feature the required flavor structure as a consequence
of standard model gauge invariance. The color & weak triplet model can
simultaneously explain the top FBA and the dimuon charge asymmetry or the
dimuon charge asymmetry and the CDF dijet excess. However, the CDF dijet excess
appears to be incompatible with the top FBA in the minimal framework. Our model
for the dimuon asymmetry predicts the observed pattern in the
region of parameter space required to explain the top FBA, whereas our model
for the CDF dijet anomaly is characterized by the absence of beyond the SM
b-quark jets in the excess region. Compatibility of the color & weak triplet
with the electroweak constraints is also discussed. We show that a Higgs boson
mass exceeding the LEP bound is typically favored in this scenario, and that
both Higgs production and decay can be significantly altered by the triplet.
The most promising collider signature is found if the splitting among the
components of the triplet is of weak scale magnitude.Comment: references added, published versio
