Hubungan Leadership Competency Dan Leadership Wisdom Terhadap Trust Pada Tenaga Pendidik Dan Kependidikan P4tk Matematika YOGYAKARTA
The current research is intended to (1) to find out the correlation between leadership competency of the leader and the trust of the staffs, (2) to find out the correlation between wisdom of the leader and the trust of the staffs, (3) to find out the correlation among leadership competency, wisdom and trust of the teaching staffs and administration staffs at P4TK Matematika Yogyakarta Ditjen PMPTK Depdiknas. The design of the research is model testing. The population is 100 people who are selected by using purposive sampling. The data are collected using leadership competency, leadership wisdom, and trust scales. The data are analyzed by using structural equation model (SEM) using Amos version 16. The findings reveal that (1) there is a correlation between leadership competency of the leader and trust of the staffs (γ= 0,651, dan ρ= 0,000), (2) there is a correlation between wisdom of the leader and the trust of the staffs (γ= 0,430, dan ρ= 0,000); (3) there is no correlation between leadership competency and leadership wisdom, (4) the correlation between leadership and trust of the staffs is stronger than the correlation between leadership and wisdom, (5) in group, leadership competency is correlated to trust. However, in isolation, leadership has no correlation with trust
Aerodynamic Roughness Length Estimation with Lidar and Imaging Spectroscopy in a Shrub-Dominated Dryland
The aerodynamic roughness length (Z0m) serves an important role in the flux exchange between the land surface and atmosphere. In this study, airborne lidar (ALS), terrestrial lidar (TLS), and imaging spectroscopy data were integrated to develop and test two approaches to estimate Z0m over a shrub dominated dryland study area in south-central Idaho, USA. Sensitivity of the two parameterization methods to estimate Z0m was analyzed. The comparison of eddy covariance-derived Z0m and remote sensing-derived Z0m showed that the accuracy of the estimated Z0m heavily depends on the estimation model and the representation of shrub (e.g., Artemisia tridentata subsp. wyomingensis) height in the models. The geometrical method (RA1994) led to 9 percent (~0.5 cm) and 25% (~1.1 cm) errors at site 1 and site 2, respectively, which performed better than the height variability-based method (MR1994) with bias error of 20 percent and 48 percent at site 1 and site 2, respectively. The RA1994 model resulted in a larger range of Z0m than the MR1994 method. We also found that the mean, median and 75th percentiles of heights (H75) from ALS provides the best Z0 m estimates in the MR1994 model, while the mean, median, and MLD (Median Absolute Deviation from Median Height), as well as AAD (Mean Absolute Deviation from Mean Height) heights from ALS provides the best Z0m estimates in the RA1994 model. In addition, the fractional cover of shrub and grass, distinguished with ALS and imaging spectroscopy data, provided the opportunity to estimate the frontal area index at the pixel-level to assess the influence of grass and shrub on Z0m estimates in the RA1994 method. Results indicate that grass had little effect on Z0m in the RA1994 method. The Z0m estimations were tightly coupled with vegetation height and its local variance for the shrubs. Overall, the results demonstrate that the use of height and fractional cover from remote sensing data are promising for estimating Z0m, and thus refining land surface models at regional scales in semiarid shrublands
A regulatory role of polycystin-1 on cystic fibrosis transmembrane conductance regulator plasma membrane expression.
Autosomal dominant polycystic kidney disease (ADPKD) is caused by genetic mutations in either PKD1 or PKD2, the genes that encode polycystin-1 (PC-1) and polycystin-2 (PC-2), respectively. ADPKD is characterized by the formation of multiple, progressive, fluid-filled renal cysts. To elucidate the mechanism of fluid secretion by ADPKD cysts, we examined the effect of PC-1 on the plasma membrane expression of cystic fibrosis transmembrane conductance regulator (CFTR), a key Cl(-) secretory protein. Five stably transfected MDCK lines were used in this study: two transfected with empty vector (control cells) and three expressing human PC-1 (PC-1 cells). The cAMP-induced endogenous short circuit currents (I(sc)) were smaller in PC-1 cells than in control cells. Compared to control cells, PC-1 cells transiently expressing pEGFP-CFTR showed significant reduction of whole cell cAMP-activated Cl(-) currents. Cell surface biotinylation experiments also indicated a reduction in surface expression of CFTR in PC-1 cells compared to control. Furthermore, studies using CHO cells transiently expressing PC-1 and CFTR suggest the importance of the PC-1 COOH-terminus in the observed reduction of CFTR plasma membrane expression. No differences in either endogeneous K(+) currents or P2Y receptor responses were observed between PC-1 and control cells, indicating the specificity of PC-1's action. These results indicate that PC-1 selectively maintains low cell surface expression of CFTR. Moreover, these findings suggest that the malfunction of PC-1 enhances plasma membrane expression of CFTR, thus causing abnormal Cl(-)secretion into the cyst lumen
Engaging African American breast cancer survivors in an intervention trial: culture, responsiveness and community
Younger breast cancer survivors often lead extremely busy lives with multiple demands and responsibilities, making them difficult to recruit into clinical trials. African American women are even more difficult to recruit because of additional historical and cultural barriers. In a randomized clinical trial of an intervention, we successfully used culturally informed, population-specific recruitment and retention strategies to engage younger African-American breast cancer survivors
TRPP2 and TRPV4 form a polymodal sensory channel complex
The primary cilium has evolved as a multifunctional cellular compartment that decorates most vertebrate cells. Cilia sense mechanical stimuli in various organs, but the molecular mechanisms that convert the deflection of cilia into intracellular calcium transients have remained elusive. Polycystin-2 (TRPP2), an ion channel mutated in polycystic kidney disease, is required for cilia-mediated calcium transients but lacks mechanosensitive properties. We find here that TRPP2 utilizes TRPV4 to form a mechano- and thermosensitive molecular sensor in the cilium. Depletion of TRPV4 in renal epithelial cells abolishes flow-induced calcium transients, demonstrating that TRPV4, like TRPP2, is an essential component of the ciliary mechanosensor. Because TRPV4-deficient zebrafish and mice lack renal cysts, our findings challenge the concept that defective ciliary flow sensing constitutes the fundamental mechanism of cystogenesis
An Essential Role for DYF-11/MIP-T3 in Assembling Functional Intraflagellar Transport Complexes
MIP-T3 is a human protein found previously to associate with microtubules and the kinesin-interacting neuronal protein DISC1 (Disrupted-in-Schizophrenia 1), but whose cellular function(s) remains unknown. Here we demonstrate that the C. elegans MIP-T3 ortholog DYF-11 is an intraflagellar transport (IFT) protein that plays a critical role in assembling functional kinesin motor-IFT particle complexes. We have cloned a loss of function dyf-11 mutant in which several key components of the IFT machinery, including Kinesin-II, as well as IFT subcomplex A and B proteins, fail to enter ciliary axonemes and/or mislocalize, resulting in compromised ciliary structures and sensory functions, and abnormal lipid accumulation. Analyses in different mutant backgrounds further suggest that DYF-11 functions as a novel component of IFT subcomplex B. Consistent with an evolutionarily conserved cilia-associated role, mammalian MIP-T3 localizes to basal bodies and cilia, and zebrafish mipt3 functions synergistically with the Bardet-Biedl syndrome protein Bbs4 to ensure proper gastrulation, a key cilium- and basal body-dependent developmental process. Our findings therefore implicate MIP-T3 in a previously unknown but critical role in cilium biogenesis and further highlight the emerging role of this organelle in vertebrate development
MNS1 Is Essential for Spermiogenesis and Motile Ciliary Functions in Mice
During spermiogenesis, haploid round spermatids undergo dramatic cell differentiation and morphogenesis to give rise to mature spermatozoa for fertilization, including nuclear elongation, chromatin remodeling, acrosome formation, and development of flagella. The molecular mechanisms underlining these fundamental processes remain poorly understood. Here, we report that MNS1, a coiled-coil protein of unknown function, is essential for spermiogenesis. We find that MNS1 is expressed in the germ cells in the testes and localizes to sperm flagella in a detergent-resistant manner, indicating that it is an integral component of flagella. MNS1–deficient males are sterile, as they exhibit a sharp reduction in sperm production and the remnant sperm are immotile with abnormal short tails. In MNS1–deficient sperm flagella, the characteristic arrangement of “9+2” microtubules and outer dense fibers are completely disrupted. In addition, MNS1–deficient mice display situs inversus and hydrocephalus. MNS1–deficient tracheal motile cilia lack some outer dynein arms in the axoneme. Moreover, MNS1 monomers interact with each other and are able to form polymers in cultured somatic cells. These results demonstrate that MNS1 is essential for spermiogenesis, the assembly of sperm flagella, and motile ciliary functions

Episodic Occurrence of Favourable Weather Constrains Recovery of a Cold Desert Shrubland After Fire
Key to the long-term resilience of dryland ecosystems is the recovery of foundation plant species following disturbance. In ecosystems with high interannual weather variability, understanding the influence of short-term environmental conditions on establishment of foundation species is essential for identifying vulnerable landscapes and developing restoration strategies. We asked how annual environmental conditions affect post-fire establishment of Artemisia tridentata, a shrub species that dominates landscapes across much of the western United States, and evaluated the influence of episodic establishment on population recovery. We collected A. tridentata stem samples from 33 plots in 12 prescribed fire sites that burned 8–11 years before sampling. We determined individual establishment years using annual growth rings. We measured seasonal soil environmental conditions at the study sites and asked if these conditions predicted annual establishment density. We then evaluated whether establishment patterns could be predicted by site-level climate or dominant subspecies. Finally, we tested the effect of the magnitude and frequency of post-fire establishment episodes on long-term population recovery. Annual post-fire recruitment of A. tridentata was driven by the episodic availability of spring soil moisture. Annual establishment was highest with wetter spring soils (relative influence [RI] = 19.4%) and later seasonal dry-down (RI = 11.8%) in the year of establishment. Establishment density declined greatly 4 to 5 years after fire (RI = 17.1%). Post-fire establishment patterns were poorly predicted by site-level mean climate (marginal R2 ≤ 0.18) and dominant subspecies (marginal R2 ≤ 0.43). Population recovery reflected the magnitude, but not the frequency, of early post-fire establishment pulses. Post-fire A. tridentata density and cover (measured 8–11 years after fire) were more strongly related to the magnitude of the largest establishment pulse than to establishment frequency, suggesting that population recovery may occur with a single favourable establishment year. Synthesis and applications. This study demonstrates the importance of episodic periods of favourable weather for long-term plant population recovery following disturbance. Management strategies that increase opportunities for seed availability to coincide with favourable weather conditions, such as retaining unburned patches or repeated seeding treatments, can improve restoration outcomes in high-priority areas
