    Cell orientation under stretch: Stability of a linear viscoelastic model

    The sensitivity of cells to alterations in the microenvironment and in particular to external mechanical stimuli is significant in many biological and physiological circumstances. In this regard, experimental assays demonstrated that, when a monolayer of cells cultured on an elastic substrate is subject to an external cyclic stretch with a sufficiently high frequency, a reorganization of actin stress fibres and focal adhesions happens in order to reach a stable equilibrium orientation, characterized by a precise angle between the cell major axis and the largest strain direction. To examine the frequency effect on the orientation dynamics, we propose a linear viscoelastic model that describes the coupled evolution of the cellular stress and the orientation angle. We find that cell orientation oscillates tending to an angle that is predicted by the minimization of a very general orthotropic elastic energy, as confirmed by a bifurcation analysis. Moreover, simulations show that the speed of convergence towards the predicted equilibrium orientation presents a changeover related to the viscous–elastic transition for viscoelastic materials. In particular, when the imposed oscillation period is lower than the characteristic turnover rate of the cytoskeleton and of adhesion molecules such as integrins, reorientation is significantly faster

    IDeS Method Applied to an Innovative Motorbike—Applying Topology Optimization and Augmented Reality

    This study is on the conception of the DS700 HYBRID project by the application of the Industrial Design Structure method (IDeS), which applies different tools sourced from engineering and style departments, including QFD and SDE, used to create the concept of a hybrid motorbike that could reach the market in the near future. SDE is an engineering approach for the design and development of industrial design projects, and it finds important applications in the automotive sector. In addition, analysis tools such as QFD, comprising benchmarking and top-flop analysis are carried out to maximize the creative process. The key characteristics of the bike and the degree of innovation are identified and outlined, the market segment is identified, and the stylistic trends that are most suitable for a naked motorbike of the future are analyzed. In the second part the styling of each superstructure and of all the components of the vehicle is carried out. Afterwards the aesthetics and engineering perspectives are accounted for to complete the project. This is achieved with modelling and computing tools such as 3D CAD, visual renderings, and FEM simulations, and virtual prototyping thanks to augmented reality (AR), and finally physical prototyping with the use of additive manufacturing (AM). The result is a product conception able to compete in the present challenging market, with a design that is technically feasible and also reaches new lightness targets for efficiency

    Post-hatching development of bovine embryos in vitro: the effects of tunnel preparation and gender.

    The objective of this study was to compare morphological characteristics, kinetics of development, and gene expression ofmale and female IVP embryos thatwere cultured until day (D)15 (fertilization = D0), using either phosphate-buffered saline (PBS) or Milli-Q water (MQW) to dilute the agarose gel used for tunnel construction. On D11, embryos (n = 286) were placed in agarose gel tunnels diluted in PBS and MQW. Embryos were evaluated for morphology, and embryo size was recorded on D11, D12.5, D14 and D15. Then, embryos were sexed and used for gene expression analyses (G6PD, GLUT1, GLUT3, PGK1, PLAC8, KRT8, HSF1 and IFNT). The percentage of elongated embryos at D15 was higher (p < 0.05) in the PBS (54%) than in theMQW(42%) gel. However, embryos produced inMQWwere bigger (p < 0.05) and had a lower expression of GLUT1 (p = 0.08) than those cultured in PBS. There was a higher proportion of male than female embryos at D15 in both treatments, MQW(65% vs. 35%; p < 0.05) and PBS (67% vs. 33%; p < 0.05); however, embryo size was not signi?cantly different between genders. Moreover, D15 female embryos had greater expression of G6PD (p = 0.05) and KRT8 (p = 0.03) than male embryos. In conclusion, the diluent used for tunnel construction affected embryo development in the post-hatching development (PHD) system, and the use ofMQWwas the most indicative measure for the evaluation of embryo quality. Male and female embryos cultured from D11 to D15, either in an MQW or PBS agarose gel, demonstrated similar development but different gene expression

    Supported tris-triazole ligands for batch and continuous-flow copper-catalyzed huisgen 1,3-dipolar cycloaddition reactions

    The lack of supported versions of the tris[(1-benzyl-1H-1,2,3-triazol-4-yl)methyl]amine (TBTA) ligand, suitable for flow-chemistry applications at scale, prompted us to develop a new route for the immobilization of such tris-triazole chelating units on highly cross-linked polystyrene resins. With this aim, the preparation of the known TBTA-type monomer 3 was optimized to develop a high-yield synthetic sequence, devoid of chromatographic purifications at any stage. Then, bead-type (P7) and monolithic (M7) functional resins were obtained by the easy and scalable suspension-or mold-copolymerization of 3 with divinylbenzene. Both types of materials were found to possess a highly porous morphology and specific surface area in the dry state and could be charged with substantial amounts of Cu(I) or Cu(II) salts. After treatment of the latter with a proper reducing agent, the corresponding supported Cu(I) complexes were tested in the copper-catalyzed alkyne-azide cycloaddition reaction (CuAAC). The immobilized catalysts proved active at room temperature and, in batch and with catalyst loadings as low as 0.6 mol%, afforded quantitative conversions within 20 h. Independent of the alkyne structure, extended use of the supported catalyst in flow was also possible. In the reaction of benzylazide and propargyl alcohol, this allowed a total turnover number larger than 400 to be reached

    Exercise training improves erectile dysfunction (ED) in patients with metabolic syndrome on phosphodiesterase-5 (PDE-5) inhibitors

    Introduction. Erectile dysfunction (ED) affects about 50% of males aged 40-70 years old. ED shares with atherosclerotic disease several common risk factors; therefore, it may be considered a surrogate marker of atherosclerosis. Sincephosphodiesterase-5 inhibitors are well known pharmacologic agents capable of significant improvement in ED, we designed this study to evaluate whether exercise training is of added value in patients with ED who are already on PDE-5 inhibitors. Methods. We recruited 20 male patients affected by ED with metabolic syndrome.At baseline, all patients underwent Cardio-Pulmonary Exercise Testing (CPET) and the International Index of Erectile Function (IIEF) test.After the initial evaluation, patients were subdivided into two groups: tadalafil group (group T, n=10), who were maintained only on tadalafil therapy, and a tadalafil/exercise training group (T/E group, n=10)who continued tadalafil but in addition underwent a2-month structured exercise training program. Results. Basal anthropometric characteristics of study population showed no significant differences. Although bothgroups showed at 2 months an improvement of the IIEF score, thiswas more evident in the T/E group (T group: 11.2 vs 14.2, P=0.02; T/E group: 10.8 vs 20.1, P&lt;0.001). There was an improvement of oxygen consumption at peak exercise (VO2peak) only in the T/E group patients (T group: 13.63±2.03 vs 14.24±2.98 mL/kg/min; P=0.521; T/E group: 13.41±2.97 vs 16.58±3.17 mL/kg/min; P=0.006). A significant correlation was found between the changes in VO2peak and the modifications in IIEF score (r=0.575; P=0.001). Conclusion. Exercise training in ED patients treated with PDE-5 inhibitors is of added valuesincefurther improves ED, as evaluated by IIEF score, and increases functional capacity

    Efeito da somatotropina bovina recombinante (RBST) sobre a curva de lactação de vacas da raça holandesas.

    Foi conduzido um estudo com o objetivo de avaliar os efeitos da aplicaç ão de rBST sobre a curva de lactação de vacas da raça Holandesa. O experimento foi desenvolvido no sistema de produção de leite do Centro de Pesquisa de Pecuária do Sudeste (Embrapa Pecuária Sudeste), em São Carlos, durante o período de abril a dezembro de 1999. Foram avaliadas 41 vacas, dos 60 aos 150 dias de lactação, distribuídas em um delineamento inteiramente casualizado, em três tratamentos: sem aplicação de rBST (controle) ? 15 vacas; aplicação de rBST 60 dias após o parto (rBST-60) ? 15 vacas; e aplicação de rBST 100 dias após o parto (rBST- 100) ? 11 vacas. Foram realizadas análises de regressão da produção de leite em função dos dias de lactação. A aplicação de rBST aos 60 dias de lactação apresentou maior resposta no aumento da produção de leite e menor queda na produção. Com relação ao tratamento rBST-100, foi verificada uma estabilização seguida da diminuição menos acentuada na curva de lactação quando comparado com o tratamento controle. Todos os tratamentos apresentaram diminuição na produção de leite em função dos dias de lactação. No tratamento com rBST-60 durante o período de 75 a 120 dias de lactação, a cada dia da lactação, a produção de leite diminuiu 25,7 g, enquanto que para o mesmo período no tratamento controle, a produção decresceu 80,9 g/dia. A suplementação com rBST aumentou a persistência de lactação e a produção de leite quando administrada aos 60 dias pós -parto
