    Passaging capability of human corneal endothelial cells derived from old donors with and without accelerating cell attachment

    In a recent report, we showed that it is possible to establish the culture of Human Corneal Endothelial Cells (HCEnCs) from older donor corneas (usually over 65 year olds) when left to attach in the presence of a viscoelastic solution, potentially increasing the donor pool for culturing HCEnCs. Therefore, we set out to evaluate the outcome of using a viscoelastic solution (Viscoat) to accelerate the attachment of passaged cultured human corneal endothelial cells (HCEnCs). The cells from 28 donor tissues were isolated using peel-and-digest method and evenly seeded into two wells of an 8-well chamber slide. The cells were left to attach after topical application of Viscoat. At confluence, one well was subjected to end-stage characterization, whereas the other well was passaged into another two wells. The cells at P1 were attached with and without the use of Viscoat. The growth rate was monitored; and at confluence, morphometric analysis, corneal endothelial specific (CD166-Tag1A3 & PRDX6-Tag2A12), mitochondrial and respiration assessment (Tom-20 and Seahorse); function-associated (Na+/K+ATPase & ZO-1); proliferative (Ki-67) marker analysis, and viability (Hoechst, Ethidium Homodimer and Calcein AM-HEC) studies were performed. Cells at P0 (with Viscoat) showed 100% confluence at day 9. Cells at P1 with and without Viscoat showed significant difference of confluence 67.0% v 18.8% respectively (p < 0.05). Confluence rate, cell density, hexagonality, Ki-67 positivity and mitochondrial intensity was significantly higher (p < 0.05), whereas cell-area and polymorphism was significantly lower (p < 0.05) in the cells attached with Viscoat compared with the cells attached without Viscoat. There was no significant difference in oxygen consumption rate between the groups. In conclusion, we observed that acceleration in the attachment of passaged HCEnCs with the assistance of Viscoat, could be beneficial for the propagation of HCEnCs isolated from older donors, to increase their propensity to proliferate, without loss of the expression of vital proteins and heterogeneity in cellular morphology

    Chondromyxoid fibroma of skull base: A tumour prone to local recurrence

    Chondromyxoid fibroma of the skull base is extremely uncommon. Sometimes involvement of the nasal cavity may occur and the patients may present with nasal symptoms. The biological behaviour of this tumour has not been well studied, primarily because of the limited number of reported cases and the short duration of follow-up. We report a histologically confirmed case of chondromyxoid fibroma of the skull base that recurred repeatedly over a 10-year period after the initial operation. Histologically it showed identical morphology to the original tumour with no evidence of histological progression or dedifferentiation. Ultrastructurally, the spindle tumour cells in the fibromyxoid area showed dual chondroblastic and fibroblastic differentiation, suggesting that these spindle fibroblastic cells and the better differentiated chondroid cells were of the same cell type with different histological morphology.published_or_final_versio

    Критическое значение высокотемпературных керамических покрытий заключается в их структурной целостности. На целостность, очевидно, влияет наличие дефектов, некоторые из которых могут быть нанесены как покрытия в результате технологических аберраций, тогда как другие развиваются в процессе эксплуатации. Покрытие на основе циркония является перспективным методом получения аварийно-стойкой топливной оболочки для легководных реакторов. Это исследование было мотивировано тем, чтобы оценить изготовленное покрытие с помощью легкодоступного, экономичного и быстрого подхода к неразрушающему контролю. В данном исследовании распыленный слой SiC был нанесен на подложку из сплава Zr-1Nb методом селективного лазерного спекания, а условия поверхности покрытия измерены с помощью метода иммерсионного ультразвукового контроля. Согласно полученным результатам, ультразвуковая иммерсионная методика может быть использована для контроля SiC-покрытия

    Topological Characterization of Hamming and Dragonfly Networks and its Implications on Routing

    Current HPC and datacenter networks rely on large-radix routers. Hamming graphs (Cartesian products of complete graphs) and dragonflies (two-level direct networks with nodes organized in groups) are some direct topologies proposed for such networks. The original definition of the dragonfly topology is very loose, with several degrees of freedom such as the inter- and intra-group topology, the specific global connectivity and the number of parallel links between groups (or trunking level). This work provides a comprehensive analysis of the topological properties of the dragonfly network, providing balancing conditions for network dimensioning, as well as introducing and classifying several alternatives for the global connectivity and trunking level. From a topological study of the network, it is noted that a Hamming graph can be seen as a canonical dragonfly topology with a large level of trunking. Based on this observation and by carefully selecting the global connectivity, the Dimension Order Routing (DOR) mechanism safely used in Hamming graphs is adapted to dragonfly networks with trunking. The resulting routing algorithms approximate the performance of minimal, non-minimal and adaptive routings typically used in dragonflies, but without requiring virtual channels to avoid packet deadlock, thus allowing for lower-cost router implementations. This is obtained by selecting properly the link to route between groups, based on a graph coloring of the network routers. Evaluations show that the proposed mechanisms are competitive to traditional solutions when using the same number of virtual channels, and enable for simpler implementations with lower cost. Finally, multilevel dragonflies are discussed, considering how the proposed mechanisms could be adapted to them

    Development of superlattice CrNNbN coatings for joint replacements deposited by High Power Impulse Magnetron Sputtering

    The demand for reliable coating on medical implants is ever growing. In this research, enhanced performance of medical implants was achieved by a CrN/NbN coating utilising nanoscale multilayer/superlattice structure. The advantages of the novel High Power Impulse Magnetron Sputtering technology, namely its unique highly ionised plasma were exploited to deposit dense and strongly adherent coatings on Co-Cr implants. TEM analyses revealed coating superlattice structure with bi-layer thickness of 3.5 nm. CrN/NbN deposited on Co-Cr samples showed exceptionally high adhesion, critical load values of LC2= 50 N in scratch adhesion tests. Nanoindentation tests showed high hardness of 34 GPa and Young's modulus of 447 GPa. Low coefficient of friction (µ) 0.49 and coating wear coefficient (KC) = 4.94 x 10-16 m3N-1m-1 were recorded in dry sliding tests. Metal ion release studies showed a reduction in Co, Cr and Mo release at physiological and elevated temperatures, (70 oC) to almost undetectable levels (<1 ppb). Rotating beam fatigue testing showed a significant increase in fatigue strength from 349±59 MPa (uncoated) to 539±59 MPa (coated). In vitro biological testing has been performed in order to assess the safety of the coating in biological environment, cytotoxicity, genotoxicity and sensitisation testing have been performed, all showing no adverse effects.     A comparative study of arbitration algorithms for the Alpha 21364 pipelined router

    Interconnection networks usually consist of a fabric of interconnected routers, which receive packets arriving at their input ports and forward them to appropriate output ports. Unfortunately, network packets moving through these routers are often delayed due to conflicting demand for resources, such as output ports or buffer space. Hence, routers typically employ arbiters that resolve conflicting resource demands to maximize the number of matches between packets waiting at input ports and free output ports. Efficient design and implementation of the algorithm running on these arbiters is critical to maximize network performance.This paper proposes a new arbitration algorithm called SPAA (Simple Pipelined Arbitration Algorithm), which is implemented in the Alpha 21364 processor's on-chip router pipeline. Simulation results show that SPAA significantly outperforms two earlier well-known arbitration algorithms: PIM (Parallel Iterative Matching) and WFA (Wave-Front Arbiter) implemented in the SGI Spider switch. SPAA outperforms PIM and WFA because SPAA exhibits matching capabilities similar to PIM and WFA under realistic conditions when many output ports are busy, incurs fewer clock cycles to perform the arbitration, and can be pipelined effectively.     Relationship Between Non-Hodgkin's Lymphoma and Blood Levels of Epstein-Barr Virus in Children in North-Western Tanzania: A Case Control Study.

    Non-Hodgkin's Lymphomas (NHL) are common in African children, with endemic Burkitt's lymphoma (BL) being the most common subtype. While the role of Epstein-Barr Virus (EBV) in endemic BL is known, no data are available about clinical presentations of NHL subtypes and their relationship to Human Immunodeficiency Virus (HIV) infection and Epstein Barr Virus (EBV) load in peripheral blood of children in north-western, Tanzania. A matched case control study of NHL subtypes was performed in children under 15 years of age and their respective controls admitted to Bugando Medical Centre, Sengerema and Shirati district designated hospitals in north-western, Tanzania, between September 2010 and April 2011. Peripheral blood samples were collected on Whatman 903 filter papers and EBV DNA levels were estimated by multiplex real-time PCR. Clinical and laboratory data were collected using a structured data collection tool and analysed using chi-square, Fisher and Wilcoxon rank sum tests where appropriate. The association between NHL and detection of EBV in peripheral blood was assessed using conditional logistic regression model and presented as odds ratios (OR) and 95% confidence intervals (CI). A total of 35 NHL cases and 70 controls matched for age and sex were enrolled. Of NHLs, 32 had BL with equal distribution between jaw and abdominal tumour, 2 had large B cell lymphoma (DLBCL) and 1 had NHL-not otherwise specified (NHL-NOS). Central nervous system (CNS) presentation occurred only in 1 BL patient; 19 NHLs had stage I and II of disease. Only 1 NHL was found to be HIV-seropositive. Twenty-one of 35 (60%) NHL and 21 of 70 (30%) controls had detectable EBV in peripheral blood (OR = 4.77, 95% CI 1.71 - 13.33, p = 0.003). In addition, levels of EBV in blood were significantly higher in NHL cases than in controls (p = 0.024). BL is the most common childhood NHL subtype in north-western Tanzania. NHLs are not associated with HIV infection, but are strongly associated with EBV load in peripheral blood. The findings suggest that high levels of EBV in blood might have diagnostic and prognostic relevance in African children

    A cyclin-binding motif in human papillomavirus type 18 (HPV18) E1^E4 is necessary for association with CDK–cyclin complexes and G2/M cell cycle arrest of keratinocytes, but is not required for differentiation-dependent viral genome amplification or L1 capsid protein expression

    Investigation into the effects the HPV E4 protein has in viral life cycleThe G2/M arrest function of human papillomavirus (HPV) E4 proteins is hypothesized to be necessary for viral genome amplification. Full-length HPV18 E1^E4 protein is essential for efficient viral genome amplification. Here we identify key determinants within a CDK-bipartite consensus recognition motif in HPV18 E1^E4 that are critical for association with active CDK–cyclin complexes and in vitro phosphorylation at the predicted CDK phosphorylation site (threonine 23). The optimal cyclin-binding sequence (43RRLL46) within this E4 motif is required for G2/M arrest of primary keratinocytes and correlates with cytoplasmic retention of cyclin B1, but not cyclin A. Disruption of this motif in the E4 ORF of HPV18 genomes, and the subsequent generation of stable cell lines in primary keratinocytes revealed that this motif was not essential for viral genome amplification or L1 capsid protein induction. We conclude that the HPV18 E4 G2/M arrest function does not play a role in early vegetative events
