    Neonatal growth of Steller sea lion (Eumetopias jubatus) pups in Alaska

    The growth rate of Steller sea lion (Eumetopias jubatus) pups was studied in southeast Alaska, the Gulf of Alaska, and the Aleutian Islands during the first six weeks after birth. The Steller sea lion population is currently stable in southeast Alaska but is declining in the Aleutian Islands and parts of the Gulf of Alaska. Male pups (22.6 kg [±2.21 SD]) were significantly heavier than female pups (19.6 kg [±1.80 SD]) at 1−5 days of age, but there were no significant differences among rookeries. Male and female pups grew (in mass, standard length, and axillary girth) at the same rate. Body mass and standard length increased at a faster rate for pups in the Aleutian Islands and the western Gulf of Alaska (0.45−0.48 kg/day and 0.47−0.53 cm/day, respectively) than in southeast Alaska (0.23 kg/day and 0.20 cm/day). Additionally, axillary girth increased at a faster rate for pups in the Aleutian Islands (0.59 cm/ day) than for pups in southeast Alaska v(0.25 cm/day). Our results indicate a greater maternal investment in male pups during gestation, but not during early lactation.     Multilocus sequence typing of Cronobacter sakazakii and Cronobacter malonaticus reveals stable clonal structures with clinical significance which do not correlate with biotypes

    Background: The Cronobacter genus (Enterobacter sakazakii) has come to prominence due to its association with infant infections, and the ingestion of contaminated reconstituted infant formula. C. sakazakii and C. malonaticus are closely related, and are defined according their biotype. Due to the ubiquitous nature of the organism, and the high severity of infection for the immunocompromised, a multilocus sequence typing (MLST) scheme has been developed for the fast and reliable identification and discrimination of C. sakazakii and C. malonaticus strains. It was applied to 60 strains of C. sakazakii and 16 strains of C. malonaticus, including the index strains used to define the biotypes. The strains were from clinical and non-clinical sources between 1951 and 2008 in USA, Canada, Europe, New Zealand and the Far East. Results: This scheme uses 7 loci; atpD, fusA, glnS, gltB, gyrB, infB, and pps. There were 12 sequence types (ST) identified in C. sakazakii, and 3 in C. malonaticus. A third (22/60) of C. sakazakii strains were in ST4, which had almost equal numbers of clinical and infant formula isolates from 1951 to 2008. ST8 may represent a particularly virulent grouping of C. sakazakii as 7/8 strains were clinical in origin which had been isolated between 1977 - 2006, from four countries. C. malonaticus divided into three STs. The previous Cronobacter biotyping scheme did not clearly correspond with STs nor with species. Conclusion: In conclusion, MLST is a more robust means of identifying and discriminating between C. sakazakii and C. malonaticus than biotyping.     3D geological models and their hydrogeological applications : supporting urban development : a case study in Glasgow-Clyde, UK

    Urban planners and developers in some parts of the United Kingdom can now access geodata in an easy-to-retrieve and understandable format. 3D attributed geological framework models and associated GIS outputs, developed by the British Geological Survey (BGS), provide a predictive tool for planning site investigations for some of the UK's largest regeneration projects in the Thames and Clyde River catchments. Using the 3D models, planners can get a 3D preview of properties of the subsurface using virtual cross-section and borehole tools in visualisation software, allowing critical decisions to be made before any expensive site investigation takes place, and potentially saving time and money. 3D models can integrate artificial and superficial deposits and bedrock geology, and can be used for recognition of major resources (such as water, thermal and sand and gravel), for example in buried valleys, groundwater modelling and assessing impacts of underground mining. A preliminary groundwater recharge and flow model for a pilot area in Glasgow has been developed using the 3D geological models as a framework. This paper focuses on the River Clyde and the Glasgow conurbation, and the BGS's Clyde Urban Super-Project (CUSP) in particular, which supports major regeneration projects in and around the City of Glasgow in the West of Scotland

    Quantifying tumour-infiltrating lymphocyte subsets : a practical immuno-histochemical method

    Background: Efficient histological quantification of tumour-infiltrating T and B lymphocyte (TIL) subsets in archival tissues would greatly facilitate investigations of the role of TIL in human cancer biology. We sought to develop such a method. Methods: Ten ×40 digital images of 4 μ sections of 16 ductal invasive breast carcinomas immunostained for CD3, CD4, CD8, and CD20 were acquired (a total of 640 images). The number of pixels in each image matching a partition of Lab colour space corresponding to immunostained cells were counted using the ‘Color range’ and ‘Histogram’ tools in Adobe Photoshop 7. These pixel counts were converted to cell counts per mm2 using a calibration factor derived from one, two, three or all 10 images of each case/antibody combination. Results: Variations in the number of labelled pixels per immunostained cell made individual calibration for each case/antibody combination necessary. Calibration based on two fields containing the most labelled pixels gave a cell count minimally higher (+ 5.3%) than the count based on 10-field calibration, with 95% confidence limits − 14.7 to + 25.3%. As TIL density could vary up to 100-fold between cases, this accuracy and precision are acceptable. Conclusion: The methodology described offers sufficient accuracy, precision and efficiency to quantify the density of TIL sub-populations in breast cancer using commonly available software, and could be adapted to batch processing of image files

    Objective. Determine which factors predict multiple pregnancy in gonadotropin-intrauterine insemination cycles so that cancellation criteria might be developed. Study Design. Retrospective chart review of all patients undergoing gonadotropin-intrauterine insemination over a continuous 36 month period. Results. No factors examined were able to predict the occurrence of multiple pregnancy. Conclusion. Multiple pregnancy is an unavoidable complication of gonadotropin-intrauterine insemination treatment

    Specific isoforms of the ubiquitin ligase gene <em>WWP2 </em>are targets of osteoarthritis genetic risk via a differentially methylated DNA sequence

    Background Transitioning from a genetic association signal to an effector gene and a targetable molecular mechanism requires the application of functional fine-mapping tools such as reporter assays and genome editing. In this report, we undertook such studies on the osteoarthritis (OA) risk that is marked by single nucleotide polymorphism (SNP) rs34195470 (A &gt; G). The OA risk-conferring G allele of this SNP associates with increased DNA methylation (DNAm) at two CpG dinucleotides within WWP2. This gene encodes a ubiquitin ligase and is the host gene of microRNA-140 (miR-140). WWP2 and miR-140 are both regulators of TGFβ signaling. Methods Nucleic acids were extracted from adult OA (arthroplasty) and foetal cartilage. Samples were genotyped and DNAm quantified by pyrosequencing at the two CpGs plus 14 flanking CpGs. CpGs were tested for transcriptional regulatory effects using a chondrocyte cell line and reporter gene assay. DNAm was altered using epigenetic editing, with the impact on gene expression determined using RT-qPCR. In silico analysis complemented laboratory experiments. Results rs34195470 genotype associates with differential methylation at 14 of the 16 CpGs in OA cartilage, forming a methylation quantitative trait locus (mQTL). The mQTL is less pronounced in foetal cartilage (5/16 CpGs). The reporter assay revealed that the CpGs reside within a transcriptional regulator. Epigenetic editing to increase their DNAm resulted in altered expression of the full-length and N-terminal transcript isoforms of WWP2. No changes in expression were observed for the C-terminal isoform of WWP2 or for miR-140. Conclusions As far as we are aware, this is the first experimental demonstration of an OA association signal targeting specific transcript isoforms of a gene. The WWP2 isoforms encode proteins with varying substrate specificities for the components of the TGFβ signaling pathway. Future analysis should focus on the substrates regulated by the two WWP2 isoforms that are the targets of this genetic risk
